Antibiotic sensitivity of bacteria associated with selected waste dumpsites in Akure, Nigeria

A study was carried out to determine the antibiotic sensitivity pattern of bacteria isolates associated with selected waste dumpsites in Akure metropolis, Nigeria. Results revealed that the total plate count of bacteria ranged from 1.3×105 cfu/g to 3.0×106 cfu/g. Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella typhi, Streptococcus faecalis, Shigella dysenteriae, Escherichia coli, Micrococcus luteus, Serratia marcescens, Clostridium botulinum were the isolated bacterial species. The temperature and pH values ranged from 26˚C to 28˚C and 5.68 to 6.48 respectively. Pseudomonas aeruginosa and Serratia showed susceptibility to antibiotics while Micrococcus luteus exhibited resistance to the antibiotics. Some of these isolated bacteria are pathogenic and thus, poses potential threat to the local inhabitant of the area. Therefore, proper waste management practices must be put in place to avoid environmental health hazards. Keywords: bacteria, dumpsites, antibiotic, waste management, environmental health

The aim of the study was to investigate the microbial populations of water and biofilm samples from seven hand dug wells in Ilorin, Nigeria, which are for private and public use. Physicochemical parameters such as pH, turbidity, total dissolved solids, temperature, conductivity, dissolved oxygen and biological oxygen demand were determined. Biofilm growths were induced from water samples obtained from selected wells. Twenty bacteria isolated were identified using morphological and biochemical characteristics as well as molecular methods and includes Shigella sonnei, Shigella dysenteriae, Micrococcus luteus, Bacillus sphaericus, Salmonella enteritidis, Proteus mirabilis strain IK-MB4-518F, Bacillus licheniformis strain RH104, Bacillus subtilis, Erwinia sp., Proteus vulgaris, Yersina sp., Bacillus cereus, Staphylococcus aureus, Serratia marcescens, Pseudomonas aeruginosa strain GS1, Pseudomonas aeruginosa strain 218B, Staphylococcus epidermidis, Vibrio sp., Escherichia coli and Bacillus pasteurii. Total bacterial count ranged from 1.8×104-1.53×106 cfu/ml. Water samples from these wells are therefore not potable and some of the isolates are potential health hazards to human population.

This study investigated the prevalence and concentration of antibiotic residues in twenty brands of milk. Determination and quantification of antibiotic residues was done using High Performance Liquid Chromatography. Aerobic plate count was carried out and the associated bacteria isolated. Identification of bacteria was done using standard microbiological methods. Molecular characterization and identification of bacterial isolates was done using 16S rRNA gene sequencing method. Antibiotic sensitivity was carried out on bacterial isolates using disc diffusion method. Plasmid profile of drug resistant isolates was done using alkaline analysis method. Comparison of means was done using Analysis of Variance. Antibiotic residues were detected in 10 (50%) of the 20 milk brands analyzed. The residual levels of tetracycline, oxytetracycline and chlortetracycline ranged between 5ng/kg and 1569ng/kg while none of the samples had doxycycline residues. The prevalence of tetracycline residues in evaporated milk brands and powdered milk samples were 100 and 23 % respectively. The aerobic plate counts ranged from 2.5 × 102 to 6.5 × 102 CFU∕mL for evaporated milk and 2.5 × 101 to 6.0 × 101 CFU∕mL for powdered milk. A total of ten bacterial species were isolated and identified. Susceptibility result showed that 95% of the isolates were sensitive to erythromycin, cefuroxime and gentamycin; 12.5% were resistant to tetracycline and ampicillin, 10% were resistant to ofloxacin and 7.5% to doxycycline and penicillin. Five of the bacterial isolates were resistant to more than one class of antibiotics. Resistant isolates subjected to plasmid profiling had detectable plasmids with estimated sizes between 120bp and 1000bp. The obtained results provide evidence that the presence of antibiotic residues in evaporated and powdered milk is an indication that the public is exposed to the harmful effects of the residues.

Abstrak : Penyakit infeksi merupakan salah satu masalah kesehatan yang penting. Penggunaan antibiotik yang tidak rasional dan tepat guna pada pasien penyakit infeksi beresiko menyebabkan terjadinya resistensi. Tujuan dari penelitian ini yaitu untuk mengetahui pola kepekaan bakteri terhadap antibiotik pada pasien Ruang Rawat Intensif (ICU) RSPI Prof. Dr. Sulianti Saroso (RSPI-SS) Jakarta. Penelitian dilakukan deskriptif dan retrospektif terhadap data sekunder hasil uji kepekaan antibiotik dan jenis bakteri dari 107 pasien dalam kurun waktu 2011. Hasil menunjukkan 68 (65,4%) pasien mendapatkan hasil kultur positif dan uji kepekaan bakteri terhadap antibiotik. Jenis bakteri patogen yang dominan yaitu Acinetobacter baumannii (29,4%), disusul oleh Pseudomonas aeruginosa (27,9%), Klebsiella pneumoniae (13,2%) dan Escherichia coli (8,8%). Sebagian besar bakteri pada pasien ICU RSPISS telah berkurang kepekaannya (resisten) terhadap beberapa antibiotik. A. baumannii dan P. aeruginosa merupakan bakteri yang paling resisten terhadap antibiotik uji. Pola kepekaannya menunjukkan bahwa bakteri patogen mempunyai resistensi tertinggi terhadap erythromycin dan terendah terhadap amikasin.Infectious diseases is an important health problem. Irrational antibiotics usage is a leading cause in initiating drugs resistances. A preliminary study was conducted on the sensitivity pattern of microorganisms against antibiotics at the intensive care unit of Sulianti Infectious Diseases Hospital Jakarta. Retrospective. Secondary data were collected on the results of antibiotics sensitivity tests and species of microorganisms of 107 patients during the year 2011. Sixty eight (65,4%) patients were positive on microorganism culture test and tested on antibiotic sensitivity test. Predominance pathogenic species found were Acinetobacter baumannii (29,4%), followed by Pseudomonas aeruginosa (27,9%), Klebsiella pneumoniae (13,2%) and Escherichia coli (8,8%). Most species were less sensitive (resistant) to several antibiotics. The pattern of sensitivity showed that pathogenic microorganisms were the most resistant against erythromycin and the most sensitive antibiotics was amikacin.

Background: India, has an estimated burn incidence of 6-7 million annually. Nearly 10% of these are life threatening and require hospitalization, and main cause of mortality and morbidity of these burn patients were wound infection and sepsis after 1st 24 hours. The present study was tried to determine specific pattern of burn wound infections, and antibiotic susceptibility of those isolates.Methods: After matching inclusion and exclusion criteria, total 55 patients were taken for this institution based, prospective observational study. Wound swabs were collected on day 7 and cultured aerobically in MacConkey agar and 5% blood agar and antibiotic susceptibility testing was done on Muller Hinton agar using Kirby-Bauer disc diffusion method.Results: Among study population 69.1% patients were female and majority (29.1%) of the patients belongs to age group from 21 to 30. It was found that 48 wound swabs were positive for microorganisms, of which Pseudomonas aeruginosa was most common isolated organism (23.6%), followed by Klebsiella pneumoniae (16.4%) and Staphylococcus aureus (14.5%). The most effective antibiotic found in this study was piperacillin/tazobactam, followed by imipenem/cilastatin.Conclusions: It was seen that gram-negative organisms were more prevalent. Pseudomonas aeruginosa was the most common microorganism and piperacillin/tazobactam was most effective antibiotic.

Solid waste management system is the most difficult task that many countries face. The aim of this study was to evaluate the effectiveness of solid waste management practices of households and analyze dumpsites' environmental suitability in Debre Markos town. Primary data were collected from randomly selected households using structured questionnaire and from secondary data from different bureaus, from research and other reports in 2017. Qualitative and quantitative techniques were used to evaluate solid waste management practices. Data were analyzed using statistical package for social Science and Geographical Information System computer program. The results revealed that households dominantly produced biodegradable solid wastes (87.6%) with the generation rate of 0.3 kg/person/day. This made the daily total solid waste generation of households up to 6210 kg (17 m3). The suitability map of the area for landfill showed that 1.4% of the study area was highly suitable, 36% was moderately suitable and 58% was less suitable. Moreover, 4.7% of the area was found unsuitable to be used for waste disposal. In conclusion, sustainable solid waste management systems must be executed, the structure and capacity of the municipality improved and integrated solid waste management practice must be applied for a healthy environment. Moreover, the municipality should make dumpsites environmentally sound and socially acceptable to decrease impacts of solid wastes on the communal land, on human, on animal and on environmental health. A retrospective study was conducted among out-patient and in-patient in Kathmandu medical collegeTeaching hospital of one month from the duration 2060-11-15 to 2060-12 -15. Mid-stream urine [MSU] of300 patient were cultured. Out of these, 75 patients’ urine were found to have significant bacterial growth.Total of five species of bacterial species isolated were viz.E.coli 37(33.3%), Proteus species 25(27.7%),Klebsiella species 15(16.6%), Staphylococcus aureus 8(8.8%) and Pseudomonas aureginosa 1(1.1%).Regarding their antibiotic sensitivity pattern, E. coli were sensitive to Nitrofurantoin 31 (83.8% ), Norfloxacin25 (67.5%), Ofloxacin 30(81.0%), Amoxycillin 16(43.2%), Nalidixic acid 15 (40.5%).proteus species weresensitive to Nitrofurantoin 15 (60.0%), Norfloxacin 15 (60.0%), Ofloxacin 12(48.0%), Amoxycillin 13(52.0%),Nalidixic acid 10 (40.0%), Ciprofloxacillin 12(48.0%). Klebsiella species were sensitive to Nitrofurantoin 9(60.0%), Norfloxacin 5 (33.3%), Oflaxin 6(40.0%), Amoxycillin 5(33.3%), Nalidixic acid 4 (26.6%),ciprofloxacillin 6(40.0%).Staphylloccus aureus were sensitive to Nitrofurantoin 8(100.0%), Norfloxacin 6(75.0%), Oflaxin 8(100.0%),Amoxycillin 7(87.5%), Nalidixic acid 6(75.0%), Ciprofloxacillin 8(100 %). And Pseudomonas aureginosawere resistant to all antibiotics which we are using in this research work.Key Words: Urine sample, Identification, Sensitivity pattern.
