Oxaliplatin (Ox) is widely used for the treatment of various tumors. Since Ox prevents DNA replication and transcription, it may affect organs with rapid cell divisions such as the testes. Although its use during pregnancy has been reported, no information regarding its effects on the testes of the offspring is not available yet. Thirty-two mice were randomly divided into four groups. The control group (1) was administered intraperitoneally 0.2 ml of saline three times a week for the 21 days of pre-pregnancy, pregnancy and lactation. Experimental groups 2, 3 and 4 received 3 mg/kg of oxaliplatin three times a week for 21 days during pre-pregnancy, pregnancy and lactation, respectively. The left testis was removed from male offspring 30 and 60 days after birth. The volume of the testes, seminiferous tubules and interstitial tissue, the surface area and height of the seminiferous epithelium, as well as the length and diameter of seminiferous tubules, were analyzed by means of stereology. Results showed a decrease in the evaluated parameters in experimental groups, in comparison with the control group. Due to the ameliorating effect of Ox on offspring testes, cautiousness is needed during maternal administration in order to preserve the fertility of male offspring.

Infertility in cat becomes a serious problem in pet community since many compounds of infertility-caused can be found widely in the environment. One of the compounds causing infertility in mammals is Bisphenol-A (BPA), which is known as an estrogenic compound. The purpose of this research is to observe the changes in testicular histology of male domestic cat (Felis domestica) induced by BPA orally. Sample used in this study were 24 male domestic cats divided into 4 groups, namely the group 1 as control, group 2, 3 and 4 were treated with BPA about 5, 10 and 100 mg/kg bodyweight, respectively for 5 days. The results showed that the estrogenic effect of BPA affects the reproductive system of the cat. The effects of BPA in cat included decrease in the amounts of spermatozoa, vacuolization of seminiferous tubules cells, degeneration of Leydig cells, degeneration of tubular epithelial cells,  irregular forms of the germ, interstitial tissue hemorrhage, debris cell filled the lumen of seminiferous tubules. These results suggested that BPA can be harmful to the reproductive system of the cat

Introduction: The goal of the present study was to examine the effect of alcohol consumption on sperm count and motility and the morphological changes in the seminiferous tubules of parent mice and their offspring. Methods: Animals were divided into two groups, Group 1 (alcohol group) of twelve male and twelve female mice, were given a daily dose of (3 g/kg body weight as 25%, v/v) ethanol by gastric gavage for four and eight weeks. Group 2 (control group) also of twelve male and twelve female mice; received normal access of food and water. After four weeks of treatment, the males and females in each group were allowed to mate, and ethanol treatment continued for up to another four weeks. Twelve male offspring from group 1 and twelve male offspring from group 2 were selected randomly and allowed to become mature. Male parent mice were killed at the 4th and 8th weeks of treatment, and their male offsprings were killed when they reached maturity age. Results: Physiological examination of the sperm solution showed that there was a significant decrease in sperm count and motility after 4 and 8 weeks of ethanol treatment in parent male mice, but this decrease was not significant in their adult offspring. Furthermore, histological investigations indicated testicular lesions in the parent male mice and their adult male offspring. Conclusion: Alcohol abuse has deleterious effects on the testes structure and on the sperm count and motility of the epididymal spermatozoa of both parent mice and their offspring.

Changes in the priorities of the treatment of animals in the conditions of intense technology of production of livestock products are based on the necessity of wellbeing of animals. Therefore, there is a need of search for and broad introduction of generally accepted alternatives to surgery, which would use modern means of castration, particularly immunocastration. The study presents morpho-functional substantiation of practicability of using immunological castration in the conditions of industrial production of pork. At the same time, we studied immunological castration using Improvak on the morphological and physiological condition of the testicles of male pigs. The testosterone level was determined using radioimmunologic method after 2, 4, 6, 8 weeks of immunological and surgical castrations, and also in intact boars of the control group. During the slaughter, we selected biopates of the testicles in immunological castrates and pigs of the control group for histological examination. The testosterone level 2 weeks after castration was the lowest in the animals castrated using Improvak. This indicator gradually increased, and after 8 weeks was higher than in the surgically castrated pigs. In the latter, the level of testosterone gradually decreased for 8 weeks, and did not significantly change in non-castrated pigs. The last stages of spermatogenesis in immunocastrates were inhibited after the second vaccination. As a result of immunological castration, the interstitial tissue of the testicle underwent changes. Between the tubules, a spreading of the loose connective tissue was observed. Leydig cells lost hyper chromaticity of the cytoplasm and typical polygonal profile, and their functional potential decreased. This fact was confirmed by the changes in the Hertwig’s ratio. In particular, we observed decrease in the value of the nuclear-cytoplasmic ratio. There were also a time shift of mitotic cycle, low degree of differentiation of spermatogonia and rupture of the course of the subsequent stages of spermatogenesis. However, there occurred multiplication of primary spermatogonia, single cellular divisions, and in the ductus deferentes, there could be found single spermatids. Some of them formed specific cellular groups of rounded and elliptic shapes in the seminiferous tubules. These structures were absent in the testicles of the control animals. Microstructural changes in the swine after injecting Improvak were characterized by deficiency of Leydig cells, indicating absence of the normal hormonal background, as confirmed by the results of the study of testosterone level. The epithelium-spermatogenic layer was underdeveloped, and the lumens of the tubules were in some places filled with generations of spermatocytes. In some places, meiosis was observed, which also indicates insignificant functioning of the testicles. Vaccination with Improvak caused atrophy of the testicles in swine and decrease in their functional condition, allowing it to be recommended it for broader application as an alternative to surgical castration.

Anticancer drugs like doxorubicin have been found to affect male gonads thereby leading to infertility. This study was conducted to evaluate the effects of doxorubicin over short, mid and long term on testes of male albino rats. Sixty male albino rats aged 6-8 weeks were taken for study. The rats were randomly divided into 3 groups of experimental (each group containing 10 rats) and 3 groups of control (each group containing 10 rats). The experimental groups were given a single dose of doxorubicin i.e. 10 mg/kg body weight intra-peritoneally and sacrificed after 3 different duration for each group (second week, eighth week and sixteenth week). All rats under 3 control groups were given a single intra-peritoneal dose of 2.5 ml/kg body weight normal saline and sacrificed with their respective experimental groups. Significant difference in diameters (p=0.029) and cross-sectional area (p=0.028) of seminiferous tubules was observed between short term experimental and short term control rats. For both between midterm experimental and midterm control group, and between long term experimental and long term control group, a significant difference in right testis weight (p<0.001 for both), left testis weight (p<0.001 for both), volume of testis (p<0.001 and p=0.038), diameter (p<0.001 for both) and area (p<0.001 for both) of seminiferous tubules was observed. As compared to short term experimental group, midterm experimental group and long term experimental group had significantly lower right testis weight (p<0.001 for both), left testis weight (p<0.001 for both), diameter of seminiferous tubule (p<0.001 for both) and cross-sectional area of seminiferous tubule (p<0.001 both). Cross-sections of the seminiferous tubules of all the control groups had normal architecture. However, there was progressive destruction of seminiferous tubules structure across the experimental groups. The effect of obesity on testicular activity in prepubertal and pubertal rats was investigated in the present study. Obesity was induced in adult females by feeding a high-calorie diet (HCD). These females were mated with normal males and were fed an HCD during pregnancy and lactation. The male offspring born to obese mothers and fed an HCD after weaning were found to be obese. Seminiferous tubules of offspring from control mothers (OCM) and offspring from HCD-fed mothers (OHCDM) had the same set of germ cells at different age intervals, namely spermatogonia, leptotene spermatocytes, zygotene spermatocytes, pachytene spermatocytes and round and elongated spermatids on postnatal days (PND) 7, 13, 17, 24 and 36, and on the day of preputial separation, respectively. However, there was a significant decrease in round and elongated spermatids and the epididymal sperm count, coupled with a significant decrease in testosterone and an increase in leptin serum concentrations in OHCDM compared with OCM. These results show that obesity in prepubertal rats does not affect the age-dependent appearance of germ cells according to developmental hierarchy, but it does interfere with spermatid formation, resulting in a reduced sperm count, which may be due to a deficiency of testosterone mediated by hyperleptinaemia.

Objective: To evaluate the potential hazards of cadmium and/or chromium on the reproductive system of adult male albino rat. Design: Randomized controlled study. Animals: Forty mature male albino rats weighing 260 ± 10 g. Procedures: Rats were allocated into four groups (ten animals each). Control group (group 1), group 2 received 4.4 mg kg-1 cadmium chloride, group 3 was given 2.5 mg kg-1 sodium dichromate and group 4 received combination of Cd (2.2 mg kg-1) and Cr (1.25mg kg-1) orally, once daily for 65 consecutive days. Results: Exposure to Cd or Cr, in particular their combination, caused a reduction in the index weights of testes, epididymis, seminal vesicle and prostate glands. They induced a reduction of sperm count and viability with an increase of abnormal sperm morphology. Interestingly, in the combination group (Cd and Cr together), the deleterious effects were more noticeable. Pathologically, both Cd and Cr produced degenerative changes in seminiferous tubules, necrosis of spermatogenic epithelium within the testis. Moreover, the interstitial tissue of epididymis showed marked edema and prostate showed necrosis and serous exudate of lining epithelium. In the interaction group, testis showed complete degenerative changes and necrosis of spermatogenic epithelium, with marked interstitial edema and hyperplastic epithelial lining of epididymal tubules. Conclusion and clinical relevance: The present results support the hypothesis that the testis is one of the most sensitive organs to Cd and/or Cr and that the exposure to any of them or to their combination lead to testicular damage and thereby male infertility.

     The present study was carried out to investigate the protective effect of Eruca sativa seeds against changes of pituitary and testes in cadmium treated rats. Forty adult male albino rats were used and divided into 4 equal groups and treated for 8 weeks as follow: The first group was given distilled water and used as a control group. The second group (G1) was administered tap water containing 30 ppm/ L of cadmium chloride. Animals in third group (G2) received tap water containing Cadmium chloride as in G1 plus 250 mg/Kg B.W ethanolic extract of Eruca sativa seeds, while rats in fourth group (G3) administered 250 mg/Kg B.W of ethanolic extract of Eruca sativa seeds only. After 8 weeks of experiment the animals were anesthetized, the pituitary and testes were excised for histological studies. Results showed that cadmium chloride intake caused severe alterations in the pituitary gland manifested by a marked  apoptotic  cells  of  pars distalis, accompanied with marked necrosis leading to left large multiple spaces in their parenchyma. Whereas testes revealed shrinkage, irregular arrangement of seminiferous tubules and increase intertubular spaces. A considerable changes was also found in the seminiferous tubules with loss most of their epithelial layer and filled with cellular debris, incomplete spermatogenesis, congested blood vessels and few Leydig's cells. Administration of Eruca sativa extract reduce the deleterious effects of cadmium chloride on pituitary gland as well as more or less complete spermatogenesis process with thickness of interstitial tissue in most of seminiferous tubules. It could be suggested that the antioxidant properties of Eruca sativa extract protects the deleterious histological damage induced by cadmium chloride in adult rats.

Chloroquine has gained great emphasis in the treatment of malaria. This study sought experimentally to determine the histomorphological and weight changes in the testes of male mice infected with Plasmodium berghei and treated with chloroquine. The study used 30 Swiss mice divided into five groups. Group 1 is Control that was not infected with Plasmodium berghei and not treated with chloroquine as control, Group 2 is Plasmodium (Plasmodiul berghei) Infected animals but not treated, Group 3 is Plasmodium Infected animals + Chloroquine (5mg/kg), Group 4 is Plasmodium Infected animals + Chloroquine (10mg/kg) and Group 5 is Plasmodium Infected animals + Chloroquine (15mg/kg). The mice were treated for 7 days after parasitaemia was confirmed and the Group 2-5 testes studied with reference to Group 1. The results showed that the Group 2 showed a little distortion, difference in spermatogenic activities and increased cellular activities; Group 3 showed large, convoluted tubules, moderate number of spermatids and large interstitial spaces, Group 4 showed Large seminiferous tubules, large spermatids, increased distortion and group 5 showed shrinking of seminiferous tubules, degeneration of interstitial cells of Leydig cells and Sertolic cells with spermatids. Groups 1-5 showed no significant effect in in body weights and testes weights of Swiss mice. Plasmodium berghei (malaria) and chloroquine have effects on histomorphological structures of Swiss mice testes but not on their teste’s weights. The testicular section from Swiss mice infected with malaria and treated at various doses when compared with the Control (Group 1) showed some moderate distortion in some structures like seminiferous tubules, connective tissues between the tubules, lumen and interstitial spaces. It can be deducted that Plasmodium berghei which caused parasitaemia in mice could cause a little tissue effect on mice if not treated.

AbstractThis study investigated the effect of a high-fat (HF) diet on protein expression of leptin and its receptor in the gonads of dams and their offspring. Female Wistar rats were fed a HF diet (30% fat) or a standard breeding (BD) diet (5% fat) during pregnancy and lactation. At 21 days of lactation, mothers and both sexes of prepubertal offspring were killed by decapitation. The protein expression of leptin and its receptor was assayed by Western blot and immunohistochemistry in gonadal, periovarian, and epididymal white adipose tissue (WAT). We demonstrated that leptin protein expression in ovary, and both leptin and ObRb expression in periovarian WAT was decreased in HF dams compared with BD animals. Immunohistochemistry showed lower leptin expression in growing antral follicles and corpora lutea of HF dams. Conversely, in both gonads and epididymal WAT of HF offspring, leptin and its receptor were significantly higher expressed compared with BD. Immunolocalization of leptin system in HF offspring gonads showed higher expression in growing and antral follicles of the ovary, seminiferous tubules, and interstitial tissue of testes. In conclusion, high gonadal and gonadal-WAT expression of leptin system was observed in the offspring of dams fed a HF diet during pregnancy and lactation.

Thymoquinone is the major active component derived from Nigella sativa. Methotrexate is a folic acid antagonist widely used in clinic. Aim of this study was to investigate the possible protective role of thymoquinone on testicular toxicity of methotrexate. Experiments were performed on male C57BL/6 mice (6 weeks old, 20 ± 2 g). The animals were divided into four groups with six mice in each group. Equivalent volumes of saline were injected intraperitoneally (i.p.) in the control group. In the thymoquinone group, mice received thymoquinone i.p. with a dose of 10 mg/kg/day for 4 days. Mice in the methotrexate group received single dose of methotrexate i.p., with a dose of 20 mg/kg. Finally, in the methotrexate plus thymoquinone group, in the first and the following 3 days after methotrexate administration, thymoquinone was injected with a dose of 10 mg/kg/day, i.p. At the end of the experiment, the left testis was quickly removed and divided into two parts for histological examination and biochemical analysis. Methotrexate alone increased total antioxidant capacity and myeloperoxidase activity compared to the controls. Thymoquinone treatment decreased total antioxidant capacity and prevented the increase in the myeloperoxidase activity. Light microscopy showed in mice that receiving methotrexate resulted in interstitial space dilatation, edema, severe disruption of the seminiferous epithelium and reduced diameter of the seminiferous tubules. Administration of thymoquinone reversed histological changes of methotrexate significantly. We suggest that thymoquinone use may decrease the destructive effects of methotrexate on testicular tissue of patients using this agent.
