The Phytochemical Study of the Populus nigra L. Leaves

From the leaves of the black poplar (Populus nigra L., Salicaceae family) there were for the first time isolated rutin (quercetin-3-O-rutinoside), calendoflavobioside (quercetin-3-O-neohesperidoside) (flavonoids) and ferulic acid (phenylpropanoid) by using of liquid column chromatography. The chemical structures of the isolated compounds there were elucidated by means of 1H-NMR-, 13C-NMR-, UV-spectroscopy, mass spectrometry and results of chemical transformations. The isolated flavonoids are the dominant phenolic components of the raw material of the investigated plant. It was determined, that the isolated flavonoids cause the spectral characteristics of the UV spectra of the water-ethanolic extract from the Populus nigra leaves. It was substantiated the expediency of the estimation of the total flavonoids in the leaves of Populus nigra calculated on rutin. As results there was development the method of the quantitative determination of the total flavonoids in the leaves of Populus nigra by the using of the differential spectrophotometry at an analytical wavelength of 414 nm. It was determined that the content of the total flavonoids in the raw materials of the studied plant varies from 2.04±0.09% to 2.99±0.11%. The relative degree of the determination of the total flavonoids in the leaves of black poplar in developed method with confidence probability 0.95 is no more than ± 6.26%.

In the present paper are discussed the results of the study on the development of the new approaches to the standardization of the Monarda fistulosa L. herbs. From the herb of the Monarda fistulosa L. there were isolated for the first time isorhoifolin (apigenin-7-O-rutinoside) and linarin (acacetin-7-O-rutinoside) chemical structures of which there were elucidated by means of 1H-NMR-, 13C-NMR-, UV-spectroscopy, mass spectrometry and results of chemical transformations. It was determined, that the isolated flavonoids cause the spectral characteristics in the UV spectra of the water-ethanolic extracts from the Monarda fistulosa L. herbs. It was substantiated the expediency of the estimation of the total flavonoids in the herbs of Monarda fistulosa L. calculated on isorhoifolin. As results there was development the method of the quantitative determination of the total flavonoids in the herbs of Monarda fistulosa L. by the using of the differential spectrophotometry at an analytical wavelength of 394 nm. It was determined that the content of the total of flavonoids in the raw materials of the studied plant varies from 4.23±0.08% to 7.68±0.12%.

This paper presents the findings of the study of the composition of phenolic compounds in black poplar leaves (Populus nigra L.). For the first time flavonoid compounds such as сalendoflavobioside (quercetin-3-O-neohesperidoside), rutin (quercetin-3-O-rutinoside), and ferulic acid (phenylpropanoid) have been extracted from the leaves of the black poplar by the methods of extraction and the following adsorption column chromatography on silica gel and on polyamide sorbent. The obtained substances were characterized with the use of UV-, 1H-NMR-, 13C-NMR-spectroscopy and mass spectrometry and also by means of the results of acid hydrolysis. The flavonoids are the dominant phenolic components of the raw material of this plant.

Relevance. Standardization of plant raw material derived from stevia is a topical issue, since this plant (Stevia rebaudiana Bertoni) contains sweet diterpene glycosides and is used as a natural sugar substitute. In addition, stevia contains flavonoids which are biologically active compounds that exert antioxidant, anti-inflammatory and capillary-strengthening properties. Currently, spectrophotometric methods are used to identify and quantify flavonoids in medicinal plants. They are fast, convenient and do not require sophisticated equipment. The aim of our paper is comparative phytochemical study of the content of the total flavonoids in stevia raw material of different origin. Materials and methods. The study material was dried stevia leaves (Stevia rebaudiana Bertoni), grown in the Penza Region, the Republic of Crimea, the Krasnodar territory, and raw materials imported from Paraguay and India. The content of the total flavonoids was estimated by differential spectrophotometry calculated with reference to cynaroside. Results. The study of the quantity of flavonoids in the raw material of stevia of different origin was carried out. In the presence of aluminum chloride stevia flavonoids form a complex compound at the absorption maximum of 408 2 nm. The content of flavonoids in different species of stevia are found to range from 1.13% to 1.74%. Conclusion. The obtained results allow to recommend stevia leaves as a source of flavonoids along with other available medicinal plants. It is advisable to classify the complex of flavonoids as the second group of biologically active compounds of stevia leaves.

The flowers and fruits of various species of the hawthorn genus (Crataegus L.) are widely used in domestic and foreign medical practice as cardiotonic preparations. In our opinion, Crataegus submolls Sarg. is a perspective species for collection of the raw materials. Crataegus submolls Sarg. is successfully cultivated in the Russian Federation and is distinguished by its rapid growth and high yield compared with wild-growing species. Moreover, the fresh fruits of Crataegus submolls Sarg. are used in nutrition. The flowers of Crataegus submollis Sarg. contain flavonoids, among of which, like in the case of Hawthorn blood-red the hyperoside is present. It was determined that the maximum of the absorption curve of the solution of Crataegus submollis Sarg. flowers with a solution of aluminum chloride is 412 nm. The method of quantitative determination by the differential spectrophotometry of the total flavonoids calculated on hyperoside in the Crataegus submollis Sarg. flowers has been developed. It was established that the appropriate extragent for this raw material is 70% ethanol. The level of total flavonoid in Crataegus submollis Sarg. flowers has been studied. It was established that separate parts of Crataegus submollis Sarg. flowers differ markedly in the content of total flavonoids and their specific weight in raw materials. It was established that the content of total flavonoids calculated on hyperoside is varied from 2.05±0.08% tо 3.00±0.12%. It was determined that the content of total flavonoids calculated on hyperoside in different parts of hawthorn flowers ranges from 0.98% (receptacle and calyx) to 4.53% (petals). In our opinion, Crataegus submollis Sarg. flowers are the promising type of medicinal plant raw materials.

The article proposes a method of increasing the amount of OH-phenolic functional groups in the composition of a macromolecule lignosulphonate, a sulfo-derivative natural lignin polymer, tested under laboratory conditions by step-by-step chemical treatment of the lignosulphonate matrix with a solution of a bromine derivative, then with a solution of neutral sodium sulfite. This makes it possible to solve the problem characteristic of modern lignosulphonates of neutral sulfite production method, which have low values of tannidicity index: content of OH-phenolic groups, quantitative content of which forms inhibitory (tanning) capacity of lignosulphonates. The proposed procedure involves carrying out said demethylation reaction with cleavage of methyl group contained in phenylpropane unit of lignosulfonate. Pyrocatechin groups are formed and methanesulfonic acid molecule is cleaved off. The obtained product was analyzed by UV spectroscopy and the increase in optical density in the area of absorption of 280 nm characteristic of OH-phenolic functional groups was shown, which is the basis for the appearance of pyrocatechin structures in the phenylpropane unit of lignosulfonate. Proposed method allows increasing quantitative content of OH-phenolic groups in macromolecule composition from 1.7% of initial to 6.5%. The obtained results are correlated with the data on determination of tannidicity index - one of objective characteristics of inhibitory (tanning) capacity of lignosulfnates, which is 32%, compared to similar characteristic of undemethylated neutral lignosulfonate, where the value did not exceed 21%. The data show that demethylation of the lignosulphonate matrix is possible by converting modern inactive neutral lignosulphonates from the low-demand waste category of the pulp and paper industry to a promising raw material component.

Myrtle (Myrtus communis L.) is an evergreen plant, a small tree or shrub endemic to the Mediterranean region. In the area of growth since ancient times, various parts (berries, fruits and leaves) of myrtle are used in folk medicine. The therapeutic properties of varies parts of the myrtle can be caused with different biologically active compounds, such as terpenoids (α-pinene, 1,8-cineole, geranyl acetate, linalool, limonene, trans-myrtanol acetate), polyphenolic compounds named flavonoids (quercetin, myricetin, myrtocommulones etc. At present time the myrtle is successfully cultivated in the coastal zone of the southern coast of Crimea. We studied samples of leaves of myrtle, harvested in the Nikitsky Botanical Garden – National Science Center of the Russian Academy of Sciences. The optimal conditions for the extraction of flavonoids from the leaves of were determined. It was established that the appropriate extragent for this raw material is 80% ethanol. The extraction time is 90 min, the ratio of "raw material-extragent" is 1 : 50. The method has been developed for the quantitative determination of the total flavonoids (calculated on rutin) in the leaves of myrtle by the method of differential spectrophotometry, at an analytical wavelength of 414 nm. Using the developed methodic, a number of samples of the myrtle leaves were analyzed, and it was established that the content of the total flavonoids in the leaves of myrtle varies from 2.48±0.08% to 3.64±0.12% (calculated on rutin) .

Bracken (Pteridium aquilinum (L. Kuhn)), use widely in the eastern and western medicine and food, has attracted researchers interest. It is related with norsesquiterpene glucoside ptaquiloside, which is carcinogen, contains in the bracken. Therefore, our aim of the study was quantitatively and qualitatively to determine ptaquiloside in the bracken and relationship between the ptaquiloside and it’s degrade or inactive way, which is “Nomkhotgol”. Nomkhotgol is a way of decreasing or inactivating toxic effect and increasing the treatment activity in raw material of drug. Ptaquiloside was present in all samples, with a minimum of 0,41ug/g in sample 1-1 (used the Nomkhotgol), with a maximum of 49.68 ug/g in spore and 10.06-32.01 ug/g in other samples. Total proteins of sample (1-1 ) were 0,01% of low compared to sample 1-2 and 0.01% of high compared to sample 5 (not spore), sample 6 (with spore). Also, total flavonoids were 1,39-5,3 times and total saponins were 1,26-53,8 times low compared to other samples. Balsam poplar and black poplar (Populus balsamifera L. and Populus nigra L.) buds that grow in Lithuania are the primary source of propolis, therefore it is proper to evaluate and compare the composition of these raw plant materials and propolis quantitatively and qualitatively. Propolis and balsamic poplar bud extract are dominated by p-coumaric acid and black poplar-caffeic acid. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), FRAP (ferric-reducing antioxidant power) and CUPRAC (cupric reducing antioxidant capacity) methods and all extracts showed antioxidant activity, and obtained results correlated with the obtained amounts of phenolic compounds and flavonoids in the extracts. Studies of antimicrobial activity have shown that all extracts have a growth inhibitory effect against Staphylococcus aureus and Candida albicans, but the extract of balsam poplar buds showed the most significant effect of such kind. Considering the results of the research, it can be stated that balsam poplar buds cultured in Lithuania are the primary raw material of propolis, which is rich in phenolic compounds with antioxidant properties and is a promising raw material for pharmaceutical purposes.

The total flavonoid content of various species of Crataegus novel raw material– «flowers with leaves» was conducted to a comparative study in order to identify the possibility of co-harvesting material from different species. 27 samples of 6 pharmacopoeial and 7 nonpharmacopoeial Crataegus species obtained from the Biological Collections of VILAR, Tsitsin Main Botanical Garden RAS, Kuzbass, Penza and Stavropol Botanical Gardens, Altai Territory, Voronezh region, Kemerovo and Moscow harvested in Russian Federation were analyzed. Spectrophotometric assay was based on aluminium complex formation was used for determination of total flavonoid expressed as hyperoside. The fractional composition of the raw material (percentage of leaves and stems) and its influence on the content of flavonoids was studied. The total flavonoids expressed as hyperoside of the raw material "flowers with leaves" of pharmacopoeial Crataegus species ranged from 1.40±0.05% (C. chlorocarpa) to 2.20±0.11% (C. monogyna), nonpharmacopeal species – from 1.21±0.11% (C. maximowiczii) to 2.21±0.09% (C. ambigua). The average quantity was 1.65%. The total flavonoid values of non-pharmacopoeial species raw material were comparable with pharmacopoeial species thus it allows to consider them promising for further study and use of their parts for medical purpose.
