Effect of leaf extracts of Carica papaya (Paw paw) and Pakia biglobosa (African locust bean) on some biochemical parameters of albino rats

In spite of the advances made in orthodox medicine, there has been an increasing interest in herbal medicine. The leaves of Carica papaya and Pakia biglobosa have been reported to contain lots of beneficial medicinal compounds, hence their use in the traditional prevention, management and treatment of ailments/diseases. In this study, the effect of varied concentrations of the ethanol leaf extract of the plants on some biochemical parameters of albino rats was assessed. The phytochemical compositions of the leaves were determined using established standard laboratory methods. Fifty four male Albino rats weighing between 150g-200g were randomly distributed into nine groups of six animals each. A daily single dose of 500mg/kg, 1000mg/kg, 1500mg/kg and 2000mg/kg body weight of either of the extracts was respectively, administered to the eight test groups for fourteen days. The control group was given only feed and water. Biochemical parameters such as the serum activities of Aspartate Aminotransferase, Alanine Aminotranferase and Alkaline phosphatase as well as the serum concentrations of Bilirubin, Albumin, HCO3-, Urea, Creatinine and Na+ were assessed. Result of the analyses showed that the administration of the extracts did not significantly raise the serum activity of ALT. The decreases in serum concentrations of Urea and increase in the concentration of HCO3- were directly proportional to the concentration of the extracts. It can be deduced from this study, that 500mg, 1000mg 1500mg and 2000mg/kg bw of either C. papaya or P. biglobosa did not elicit any marked hepatotoxicological or renotoxicological effect on the experimental animals

Plant-based products, believed to be less toxic have been employed as hepatoprotective agents in recent times. Polyherbal therapy is reported to have therapeutic benefits over single herb therapy. Allium sativum and Carica papaya are two known plants used traditionally for the treatment of liver related diseases. Thus, in this study, the hepatoprotective and antioxidant potential of hydroethanol extracts of Allium sativum cloves (Garlic) and Carica papaya (Pawpaw) leafs (singly and combined) against CCl4-induced toxicity in rats was evaluated. Thirty male albino rats used were classed into six (groups A- F) groups of five rats each and treated (orally) for 14 days. Group A- rats + distilled water (negative control); Group B- rats + CCl4 in olive oil (positive control); Group C- rats + 200 mg/ kg-day combined extract of A. sativum and C. papaya (1: 1 ratio); Group D- rats + 200 mg/ kg-day extract of A. sativum; Group E- rats + 200 mg/ kg-day extract of C. papaya; Group F- rats + 100 mg/kg bw of silymarin. CCl4 induce hepatotoxicity was characterized by an upsurge (P<0.05) in serum Alkaline Phosphatase (ALP), Aspartate Aminotransferase (AST) and Alanine Transaminase (ALT) activities, and total bilirubin. Also, CCl4 administration led to a substantial reduction (P<0.05) of reduced glutathione (GSH), Glutathione peroxidase (GPx), superoxide dismutase (SOD) and catalase (CAT) activities and an increase (P<0.05) in lipid peroxidation (MDA) in the liver tissue. However, treatment with hydroethanol extract of A. sativum and C. papaya prevented the CCl4 induced lipid peroxidation and returned the altered marker enzymes of serum and antioxidant enzymes level to near normal. The extracts of both plants demonstrated significant antioxidant and hepatoprotective activity as reflected by the results. Comparatively, the combined extract gave the most promising antioxidant and hepatoprotective potential, which may be due to positive synergistic effects of both plants over either plant used singly, thereby vindicating the use of polyherbal remedy over a single herb.

The present study investigated the unsafe impacts of sodium nitrite, sodium benzoate and their mixture which is utilized in fabricating of the food on some biochemical parameters in male albino rats. Rats (40) were divided into four groups as follows: group 1 used as the control, group II and III were treated orally with sodium nitrite nano2 (80 mg/kg BW) and sodium benzoate (SB) (200 mg/kg BW), respectively. Group IV was treated orally with a mixture of sodium nitrite and sodium benzoate. Rats took their respective doses every day for 8 weeks. The obtained results showed that sodium nitrite, sodium benzoate and their mixture (nano2 and SB)  initiated a diminished within in the activity of antioxidant enzymes (SOD, CAT, and GSH) within the kidney, while, MDA recorded a highly significant activity level within experimental groups. Urea and creatinine mean levels were were expanded within plasma of the experimental rats. In the histology of the kidney, sections appeared edema with few mononuclear leukocyte cellular infiltrations, shrinkage of glomeruli. The severity of these changes increased in the experimental group which treated by the mixture of sodium nitrite and sodium benzoate. Overexpression in p53 occurred in experimental groups that were treated by nano2, SB and their mixture. The present study concluded that the mixture of food additives can induce toxicity in the kidney of rats. It is obvious that food additives induced nephrotoxicity  within the kidney. It decreased the antioxidant enzymes (GSH, CAT, and SOD) and increased MDA. Increase tumor suppressor gene p53 in kidney tissue. Food added substances caused changes in biochemical parameters as in creatinine and urea. The utilization of food additives must be decreased. The presence of more than one type of food additives on our food and the usage of the mixture of sodium nitrite and sodium benzoate initiated changes in biochemical parameters and immune-histopathological changes.

Aim: Diabetes mellitus (DM) is a prevalent metabolic disorder that leads to other microvascular and macrovascular complications. Diabetes affects fertility and blood clotting, and also cause impaired lipid profile thus leading to increased atherogenic risks and coronary diseases. This research investigates the effects of Carica papaya leaf methanol extract on fertility indices and lipid profile of male diabetic rats. Methodology: Male Wistar albino rats were randomly divided into five groups of six rats each. Diabetes was induced in the rats by a single intraperitoneal injection of streptozotocin (55 mg/kg). Diabetic rats were treated orally with 100 and 200 mg/kg C. papaya methanol extract for 14 days. At the end of administration, the plasma glucose concentration and lipid profile were assayed by spectrophotometric methods; seminal analysis was carried out for evaluation of morphology, motility and sperm count under the microscope. The bleeding and clotting times of the rats were also determined. Results: C. papaya leaf methanol extract caused significant (p = 0.05) reduction in plasma glucose, total cholesterol, triglycerides, VLDL-C, LDL-C, bleeding and clotting times of diabetic treated rats, while the HDL-C of treated groups were significantly (p = 0.05) elevated compared to the diabetic control. Percentage normal cells were lower in diabetic control rats (41.4±4.4%) and C. papaya treated groups (50.0±8.5% for 100 mg/kg; 47.5±9.1% for 200 mg/kg) compared with the normal control group (69.5±5.6%). Similar results were recorded for sperm count. The qualitative phytochemical screening showed the presence of steroids, anthraquinone, tannin, and other bioactive compounds. Conclusion: findings from this study indicated that C. papaya leaf methanol extract could possess hypoglycemic and hypolipidemic activities. Thus, could be considered as a potential source of bio pharmacological agent for management and control of DM and its complications. Prolonged administration of C. papaya leaves may negatively affect male fertility.

Background: Dengue is an infectious disease associated with high mortality and morbidity. Being a viral disease, there is no specific drug available for treatment. There are some reports that Carica papaya leaf extract may improve the clinical condition of dengue patients. However, to support this, at present, there is no systematically searched and synthesized evidence available. Hence this study was undertaken to compare the efficacy of commercial preparation of Carica papaya leaves with freshly prepared Carica papaya leaf extracted.Methods: 48 albino rats were randomly divided into eight groups of six each. Thrombocytopenia was induced by giving hydroxyurea (15mg/kg) orally. Group I and II served as saline and toxic control group respectively. Other six groups were given two different doses of either commercial extract or fresh extract orally for five days. 1ml of blood was withdrawn at baseline,3rd and 6th day of the study. Platelet, WBC, RBC count, clotting and bleeding time were determined.Results: Mean platelet count increased significantly on day 6 in both low dose (2.06 to 4.93lakh/mm3) and human equivalent dose (2.73 to 7.66lakh/mm3) of commercial extract groups compared to the toxic control group (p<0.05). Similarly, the mean platelet count increased significantly for human equivalent dose in fresh leaf extract group (3.17 to 4.69lakh/mm3) but the increase in low dose fresh extract (3.28 to 3.76lakh/mm3) was not significant. There was no significant rise in mean platelets count, mean RBC count, WBC count, decrease in mean bleeding and clotting time between commercial extract and fresh leaf extract group for both low dose and human equivalent dose.Conclusions: Efficacy of fresh leaf extract of Carica papaya was not inferior to commercial available preparation. Fresh Carica papaya leaf extract no doubt offers a potential therapeutic efficacy which is cost effective, more affordable and accessible treatment in patients with thrombocytopenia.

Background: With the increased trend of urbanization of our society, the incidence of various stress related diseases are increasing day by day. People under chronic stress often tends to seek relief through drugs, which may have side effects. But some natural supplements called adaptogen, can be more beneficial in reducing symptoms of stress. Objective: The present animal model experimental study was designed to observe the effects of restraint stress on body weight, serum ALT, AST, Glucose, Cholesterol and their modifications by Ocimum sanctum Linn (tulsi) pretreatment which is an Ayurvedic adaptogen and has a long history of therapeutic use. Study design: For this purpose, 30 albino rats aged 90 to 120 days were included in this study. Twenty rats of experimental group were further subdivided into two groups. One group consisted of 10 rats exposed to one hour restraint stress daily for 7 days and 10 rats of the second group were pretreated with tulsi for 7 days before exposure to stress in the same way. 10 non stressed , non pretreated rats were taken into control group.This study was undertaken in the laboratory of the Department of Physiology,BSMMU during the period from July 2003 to June 2004. Methods: The body weight of all control rats were recorded daily in the morning. Body weight of the experimental rats were recorded just before exposure to stress. After completing the experiment , the animals were sacrificed and blood was collected . Serum glucose, cholesterol and ALT,ASTwere determined by standard laboratory technique. Data were compared among the groups and the results were statistically analyzed using unpaired student t' test. Results : The body weight in untreated stressed group was significantly lower (p < 0.001) than those of the control group and tulsi pretreated group. Serum levels of glucose, cholesterol, aminotrasferases (ALT and AST) were significantly higher (p < 0.001) in stressed group than those of control . Again in Tulsi treated group all these biochemical parameters were significantly lower (p < 0.001) than those of stressed group. This restraint stress-induced changes in body weight and biochemical parameters may be due to hypophagia, altered secretion of various metabolic hormones and neurotransmitters, changes in membrane permeability & hypovolaemia resulting from stress induced secretion of corticosterone and epinephrine through hypothalamo-pituitary adrenal axis activation. Conclusion: Prevention of stress induced changes in biochemical parameter by tulsi pre treatment indicates its anti stressor effect. The present study was conducted to evaluate the impact of feeding fermented karonda whey beverage(FKWB) on biochemical parameters of albino rats. A total of 24 albino rats (male) of age 28±3 days and weighing 80 to 95 gm were randomly divided into four groups (n=6) and assigned to experimental dietary treatment. Experimental groups consisted of control group fed with basal ration only and groups fed with basal ration containing 10%, 20% and 30% FKWB. Results showed that levels of BUN, creatinine, SGOT, SGPT were significantly (p>0.05) lower in FKWB fed groups, signifying positive impact of FKWB on functioning of kidney and liver. FKWB consumption had no significant (p>0.05) effect on cholesterol and protein levels, while its consumption reduced triglyceride level.

Virgin olive oil has demonstrated its effective activity against oxidative stress. However, data on the bioactive effect of olive leaves or their major constituents on the liver are scarce. The present research work was conducted to evaluate the hepatoprotective effects of supercritical carbon dioxide (SC-CO2) extracts from fresh and dried olive leaves on hepatotoxicity caused by carbon tetrachloride (CCl4) in rat models. For this purpose, healthy albino rats of 180–250 g weight were used. The assessment of biochemical markers was carried out on blood and liver tissue. Then, a histopathological study was carried out on liver tissue. The obtained results showed that fresh and dried olive leaf extracts ameliorate the perturbed biochemical parameters caused by CCl4 treatment. Furthermore, the results registered for the histopathological study are in accordance with the biochemical parameters and the protective capacity of SC-CO2 extracts against DNA damage, indicating that olive leaf extracts helped to improve liver fibrosis caused by CCl4 treatment.

AbstractHyperhomocysteinemia (HHC), both basal and after methionine load, may occur due to genetic disorders or deficiencies of nutrients that affect the remethylation or trans-sulphuration pathways during methionine metabolism. HHC is involved in the pathogenesis of many illnesses as a result of its prooxidative effect and its impairment of antioxidative protection. The aim was to examine the effects of subchronic methionine overload on the body weight and standard biochemical parameters in rat serum and to examine whether simultaneous subchronic intraperotoneal administration of methionine alone or together with L-cysteine or N-acetyl-cysteine resulted in a change in the body weight and biochemical parameters in the rat serum. The research was conducted during a three-week period (male Wistar albino rats, n=36, body weight of approximately 160 g, age of 15-20 days), and the animals were divided into a control group and three experimental groups of 8-10 animals each: a) control group (0.9% sodium chloride 0.1-0.2 ml/day); b) methionine (0.8 mmol/kg/bw/day) (MET group); c) methionine (0.8 mmol/kg/bw/day) + L-cysteine (7 mg/kg/bw/day) (L-cys+MET group); and d) methionine (0.8 mmol/kg/bw/day) + N-acetyl-L-cysteine (50 mg/kg/bw/day) (NAC+MET group). In addition to the body weight monitoring, the levels of total homocysteine and the standard biochemical parameters in blood samples (plasma or serum) were determined. The results indicated that monitoring the homocysteine levels and standard biochemical parameters in blood could be used for analysis and could provide an excellent guideline for distinguishing between toxic and non-toxic doses of methionine intake, which may be meaningful for clinical applications.

Background: Literature has reported thyroid functional abnormalities in diabetes mellitus. The objectives of this study were to determine and compare the serum concentrations of T3, T4 and TSH in alloxan-induced type 1 diabetic and control Wistar albino rats. Materials & Methods: It was an experimental animal study on 20 Wistar albino rats, extending over a period of eight weeks. Alloxan, a diabetogenic agent, was used to produce animal models of type 1 diabetes. Animals were divided equally into two groups: control and diabetic. The animals in the diabetic group were injected intraperitoneally with 150 mg/kg body weight of 10% alloxan to induce diabetes. After 72 hours, diabetes was confirmed with glucometer (glucose >350mg/dl). During the course of experiment, one rat in control group and 2 rats in diabetic group died. Blood was collected for estimation of serum concentrations of thyroid hormones, thyroid stimulating hormone at the end of experimental period. Serum T3, T4, and TSH were measured using ELISA kits. Results: At the end of eight weeks, the mean concentration of serum T3 was 0.69 ±0.29 ng/ml and 0.44±0.02 ng/ml in control and diabetic groups, respectively. The mean concentration of T4 was 3.78±1.16 μg/dl and 2.24±0.86 μg/dl in control and diabetic groups respectively. The mean concentration of TSH was 0.77±0.20 μU/ml and 1.41±0.23 μU/ml in control and diabetic groups respectively. The mean serum concentrations of T3 (p=.0025) and T4 (p=<.00001) were significantly lower in diabetic and that of TSH (p=<.00001) were significantly higher in diabetic than control group. Conclusion: This study concludes that the serum concentrations of both T3 and T4 are significantly lower and that of TSH is significantly higher in alloxan-induced type 1 diabetic as compared to control group in Wistar albino rats.
