The present study was carried out to discover the protective and curative effects of alcoholic extracts of garlic (Allium sativum) and black seeds (Nigella sativa) in rabbits experimentally infected by Eimeria magna using in vivo, in vitro, and histopathological examination. Overall oocysts number per gram was significantly lower in the garlic treatment and pretreatment groups, compared to the control positive, sulfadimidine treatment, and black seed treatment groups. At the end of the experiment, the oocyst disappeared in garlic pretreatment, garlic treatment, and black seed pretreatment groups. While oocyst counts of control positive and sulfadimidine groups were increasing oocysts similarly at the end of the experiment with repeated cycles. In vitro sporulation inhibition of garlic extract showed significant efficacy on E. magna oocysts in comparison with black seed extract and high significant efficacy of sporulation inhibition, compared to sulfadimidine. While black seed extract showed high significant efficacy of sporulation inhibition, compared to sulfadimidine. Body weight gain increased in control negative, garlic pretreatment, and garlic treatment groups in comparison with other groups. The results showed that there were no significant differences in erythrocytes counts in all experimental groups while leukocyte counts showed a significant decrease in control positive and sulfadimidine groups, compared to the other groups. Similarly, the histopathological examinations on days 14 and 28 post-infection revealed pathological changes in intestinal villi of the control positive group that appeared thickened and deformed with hypertrophied enterocytes containing numerous developmental stages of E. magna. Both garlic and black seed extract had beneficial effects on improving the lesions grossly and microscopically. The results obtained in the present study proved that garlic pretreatment had a better effect on a prophylaxis and treatment for coccidiosis than garlic treatment and both had more beneficial effects, compared to black seed extract. Therefore, it is recommended to use garlic as a natural feed additive in rabbit feeding as a prophylaxis and treatment for coccidiosis to minimize the economic losses caused by this parasite.

Nigella sativa L. is a plant with high medicinal profile in the treatment of diseases. This study aimed to assess the chemical composition and in vitro antibacterial and cytotoxic effect of Nigella sativa L. seed extract. Seeds were purchased from supermarket and blended into powder using electric blender. The powder was macerated with 96% methanol. The extracts were then qualitatively screened to test the presence of secondary metabolites and agar well diffusion method was employed to study the antibacterial activity of extracts. In this evaluation, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was used to examine the cytotoxic effect of extract on peripheral blood mononuclear cells (PBMCs). MTT assay was done on different extract concentrations for 3 days and the results were recorded daily. The findings revealed the presence of tannins, terpenoids, steroids, saponins, alkaloids, phenolic compounds and flavonoids. The antibacterial activity was observed with the inhibition zones ranging from 11.3 ±1.1 to 16.3 ±1.5mm while the minimum inhibitory concentration (MIC) values of extract ranged between 0.5 and 1.25 μg/mL. The black seed extracts contain bioactive substances with germicidal activity and in vitro administration of black seed extract up to the dose of 100 μg/mL resulted no cytotoxic effect on PBMCs.

Osteoporosis is the most common reason for a broken bone among the elderly. Several experimental studies have been reported that Nigella sativa(NS) and/or its major component thymoquinone (TQ) have good effects on osteoporosis and bone healing. We conducted this systematic review toevaluate these relevant studies to prove whether NS and/or TQ has potential effect on osteoporosis and can stop pathogenesis of this disease or thismatter still just a fiction. A search on published studies was done using databases including Scopus, PubMed, Google Scholar, Web of Science, andCINAHIL. Terms searched included “Nigella sativa, black seed, TQ, osteoporosis, bone healing.” Initially, 213 articles were extracted. After reviewing their titles and abstracts, 124 articles (Medline, 43; Scopus, 67; EBSCO, 14) were retrieved for further evaluation. However, after excluding the clinical trial studies, human reviews, removal of abstracts and unrelated studies, seven studies were considered finally as eligible for our review. Finally, seven studies were considered eligible for our review. The total number of animals used was 220 (150 rats and 70 rabbits) from different experimental study. Based on the results of this systematic review, we conclude that NS or TQ extract therapy for osteoporosis cannot be recommended yet and these data will not suffice to exclude the beneficial effects of NS on bone turnover reliably. Therefore, more studies are required to explore the specific cellular and molecular targets of NS or TQ using animal osteoporosis models. Once the anti-osteoporotic effectiveness of NS or TQ will be established, human studies can be carried out.Keywords: Nigella sativa, Black seed, Thymoquinone, Osteoporosis, Bone healing.

Thymoquinone (TQ), the chief active constituent of Nigella sativa (NS), shows very valuable biomedical properties such as antioxidant, antimicrobial, anticancer, anti-inflammatory, antihypertensive, hypoglycemic, antiparasitic and anti-asthmatic effects. Several studies have examined the pharmacological actions of TQ in the treatment of oral diseases but its potential role in periodontal therapy and regeneration is not yet fully defined. The present investigation has been designed to review the scientific studies about the effects of TQ as an adjunct to periodontal treatment to promote healing and periodontal regeneration. Along with clinical experiments, in vitro studies exhibit the beneficial effects of TQ during periodontal therapy. Nevertheless, additional comprehensive clinical and preclinical studies at cellular and molecular levels are essential to examine the particular action mechanisms of Nigella sativa and its elements, particularly TQ, during periodontal treatment or regeneration.

Plant bioactive compounds are currently viewed as possible feed additives in terms of methane mitigation and improvement of ruminal fermentation. A range of analyses, including the botanical characterisation, chemical composition and in vitro efficiency, have to be conducted before testing the compounds in vivo. Therefore, the aims of this study were (1) to identify the main bioactive components of black seed (Nigella sativa) oil (BO) and of the root powder of Ferula elaeochytris (FE), and (2) to investigate their effects on ruminal fermentation in vitro, when supplemented in different dosages to a diet (1 : 1, forage : concentrate), using the rumen simulation technique (Rusitec). Main compounds of BO were thymoquinone and p-cymene and α-pinene in FE. Supplementation of the diet with BO and FE did not affect concentration of volatile fatty acids but ammonia concentrations decreased with both supplements (P < 0.001). No effects of supplements on protozoal counts were detected but in vitro disappearance of DM and organic matter tended to increase with 50 mg/L FE (P < 0.1), compared with the control.

Neurotrophin-4 (NT-4) is a neurotrophic factor that plays an important role in follicular development and oocyte maturation. However, it is not yet known whether NT-4 is related to oocyte maturation and follicular development in pigs. This study aims to investigate the effects of NT-4 supplementation during in vitro maturation (IVM) of porcine oocytes and subsequent embryonic development after parthenogenetic activation (PA). First, NT-4 and its receptors (TrkB and p75NTR) were identified through fluorescent immunohistochemistry in porcine ovaries. NT-4 was mainly expressed in theca and granulosa cells; phospho-TrkB and total TrkB were expressed in theca cells, granulosa cells, and oocytes; p75NTR was expressed in all follicular cells. During IVM, the defined maturation medium was supplemented with various concentrations of NT-4 (0, 1, 10, and 100 ng/mL). After IVM, the nuclear maturation rate was significantly higher in the 10 and 100 ng/mL NT-4 treated groups than in the control. There was no significant difference in the intracellular reactive oxygen species levels in any group after IVM, but the 1 and 10 ng/mL NT-4 treatment groups showed a significant increase in the intracellular glutathione levels compared to the control. In matured cumulus cells, the 10 ng/mL NT-4 treatment group showed significantly increased cumulus expansion-related genes and epidermal growth factor (EGF) signaling pathway-related genes. In matured oocytes, the 10 ng/mL treatment group showed significantly increased expression of cell proliferation-related genes, antioxidant-related genes, and EGF signaling pathway-related genes. We also investigated the subsequent embryonic developmental competence of PA embryos. After PA, the cleavage rates significantly increased in the 10 and 100 ng/mL NT-4 treatment groups. Although there was no significant difference in the total cell number of blastocysts, only the 10 ng/mL NT-4 treatment group showed a higher blastocyst formation rate than the control group. Our findings suggest that supplementation with the 10 ng/mL NT-4 can enhance porcine oocyte maturation by interacting with the EGF receptor signaling pathway. In addition, we demonstrated for the first time that NT-4 is not only required for porcine follicular development, but also has beneficial effects on oocyte maturation and developmental competence of PA embryos.

Objective:  The aim of this study is to investigate the protective effects of methanolic extract of Nigella sativa (MENS) (Black seed) against cardiotoxicity of cadmium in albino rats.  Methods: Twenty five (25) male albino rats, weighing (150-170g), were randomly grouped into five groups: A-E. Group B (Negative Control) received intraperitoneal administration of cadmium chloride (CdCl2, 5mg/kg) only, group C received CdCl2 and low dose MENS (300mg/kg, oral), group D received CdCl2 and high dose MENS (600mg/kg, oral), group E (Positive control) received CdCl2 and Vitamin C (200mg/kg, oral), for 7 days. No treatment was administered to group A (Normal control). Cardiac injury was assessed by measuring serum levels of Aspartate aminotransferase (AST), Lactate dehydrogenase (LDH) and Creatine kinase (CK-MB) using standard methods. The heart was harvested for histopathological examination. Results: CdCl2 induced significant cardiotoxicity with marked elevation in the levels of biochemical markers of cardiac functions (p<0.05 or p<0.01); these were however attenuated by MENS. Histopathological examination of the heart sections supported the biochemical findings. Conclusion: Nigella sativa seed extract is potentially cardioprotective against harmful chemical toxins such as cadmium. Keywords: Cadmium, CdCl2, Nigella sativa, Medicinal food, cardiotoxicity

Introduction: Nigella Sativa or Black Cumin is an annual flowering plant that can be used for wound treatment. In the treatment for wound healing, this plant contains many beneficial components that can help with the treatment but this plant utilisation in the medical field is still limited. This review will provide an overview of the advantages when this plant is used in the wound healing treatment. Method: The review was based on the ROSES protocol, and the databases used were ScienceDirect, PubMed, and Google Scholar. After the search, only 11 papers had been chosen to be included in the results and they were divided into four main themes that were used for the analysis of the results. Results: The themes were gross analysis, microscopic analysis, biochemical analysis and antimicrobial analysis. Conclusion: In conclusion, the black seed extract contains several valuable properties, such as antimicrobial and antioxidant properties, which help improve the wound healing process. Further study needs to be done to discover more potential of Nigella Sativa in treating the wound.
