Hepatho-nephroprotective and antioxidant effect of stem bark of <i>Allanblackia gabonensis</i> aqueous extract against acetaminophen-induced liver and kidney disorders in rats

The present study investigated the toxicological potential of the oral administration of the stem bark aqueous extract of R. vomitoria on the liver and kidney in rats. Acute oral toxicity study of the extract to a single dose of 2000 mg/kg was studied in 10 rats of both sexes. Sub –acute oral toxicity of aqueous extract of was carried out on 60 rats. We constituted 4 groups of 10 rats each (5 males and 5 females) which were orally administered 300, 600, and 900 mg/kg of aqueous extract and control group received water. 2 group satellites (SAT) of 10 rats each (5 males and 5 females) in which one group (SAT 900 mg/kg) was received orally 900 mg/kg of aqueous extract and another (SAT control) water. Serum blood was collected for biochemical and haematological parameters. The liver and kidney served for histological examination. No deaths of acute oral toxicity were recorded. In female rats, Aspartate Aminotransferase (ASAT) activity increased by 31.20 % and Alamine Aminotransferase (ALAT) increased by 37.20 %. In male rats, only ALAT activity increased significantly by 35.37 % compared to control. Haematological analysis revealed in male rats treated at the dose of 900 mg/kg an increase significant (p<0.001) level of white blood cells with 52.20 %, compared to control group. Histological examination of liver and kidney showed normal architecture. Aqueous extract has untoward effect on liver and kidney, could be considered non-toxic.

Ricinodendron rautanenii is a plant, used in traditional medecine to treat fever, eczema, back pain, cancer and stomacal disorders. This work was carried out to evaluate the safety of the aqueous extract from the stem bark of Ricinodendron rautanenii by determining its potential toxicity after acute and subacute administration in Wistar rats. In Acute toxicity assay, the animals received the extract at the single dose of 5000 mg/Kg and were observed during 48h for mortality and any toxicity manifestations. General behavior, adverse effects and mortality were determined for up to 14 days post treatment. In subacute toxicity assay, extract was given orally to rats at doses of 6, 20, 40 and 80 mg/kg/day for 28 days respectively. Animal body weight, water and food intake, biochemical and hematological parameters were determined. Liver and kidney were examined histologically for any signs of organ damage. No behavioral changes or mortality were recorded in the treated groups. No significant hematological changes were observed in the both sex. The biochemical analysis indicated a significant increase of high density lipoprotein levels in both male and female rats. Histopathological analysis of the liver and kidney did not show any observable cellular damages. The overall finding of this study suggest that, extract of Ricinodendron rautanenii did not cause any death up to a dose of 5000 mg/kg and can be considered non-toxic. Biochemical and histological studies of the extract did not revealed major signs in subacute toxicity.

Background: The use of herbal medicines for treating ailments is rampant in recent years, and the toxicity implications of various plant preparations are sparingly reported. We investigated the potential effect of daily administration of aqueous extract of stem-bark of cashew tree on the liver and kidney status of rats. Methods: Rats were divided into 4 groups as follows: control rats received 1 mL of distilled water, G1 received 100 mg/kg, G2 received 200 mg/kg, while G3 received 400 mg/kg body weight of the extract for 28 consecutive days. The tissue homogenate supernatants were analysed for liver enzymes-alanine aminotransferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST) and gamma glutamyl transferase (GGT) and kidney function indices- urea and creatinine. Results: In comparison to control, total protein increased significantly (P< 0.05) at 400 mg/kg extract, whereas albumin level significantly decreased (P<0.05) in rats treated with extract. .Activities of AST, ALP and GGT increased markedly (P< 0.05) at 400 mg/kg, whereas a significant decrease was observed in bilirubin level when compared with the control. Levels of urea and creatinine in kidney tissue were significantly higher in extract-treated rats compared to control. Conclusion: The findings suggest that the extract dose at 400 mg/kg may cause alterations with toxic implications in the liver and kidney of rats.

Abstract Objective This study was aimed at evaluating the toxicity and the antioxidant property of Cassia fistula stem bark extracts in zebrafish. Materials and Methods Crude aqueous and ethanol extracts of C. fistula stem bark were obtained following a standard solvent-based extraction method. The toxicity of these extracts on zebrafish embryonic development was determined and the LC50 values were calculated. Finally, the antioxidant property of C. fistula stem bark extracts was determined in arsenic-induced oxidative stress models of zebrafish. Results The aqueous extract of C. fistula stem bark showed a slightly larger LC50 value (213.6 ppm) compared with the ethanol extract (LC50 = 63.5 ppm), suggesting a lower toxicity of the aqueous extract. A significant reduction of reactive oxygen species (ROS) signal was observed in arsenic-exposed embryos treated with the aqueous extract, but not the ethanol extract, indicating that the antioxidant activity is present only in the aqueous extract of C. fistula stem bark. Conclusion Identification of antioxidants from natural sources is desirable because of increasing safety concerns associated with synthetic antioxidants. This study demonstrated that aqueous extract from C. fistula stem bark possesses antioxidant properties, which can be further characterized for mechanism of action and potential applications.

The decoction of the stem barks from Bowdichia virgilioides KUNTH is a folk remedy used to treat inflammatory disorders in Latin American and Brazil. In the present study, the wound healing activity of aqueous extract of the stem bark from B. virgilioides, called AEBv, was evaluated by the rate of healing by wound contraction and period of epithelization at different days post-wound using the wound excisional model. On day 9, the AEBv-treated animals exhibited significative reduction in the wound area when compared with controls. In wound infected with S. aureus, the AEBv significantly improved the wound contraction when compared to the saline-treated mice. The histological analysis showed that AEBv induced a collagen deposition, increase in the fibroblast count and few inflammatory cells than compared to saline-treated group. The expression of collagen type I was increased in the group treated with AEBv as indicated by immunohistochemical staining. In vitro, the AEBv was effective only against S. aureus but not against P. aeruginosa. Together, the results of this study demonstrate, for the first time, the healing and antimicrobiological effects of aqueous extract of the stem bark from B. virgilioides in the therapy of skin wounds.
