The present study investigated the toxicological potential of the oral administration of the stem bark aqueous extract of R. vomitoria on the liver and kidney in rats. Acute oral toxicity study of the extract to a single dose of 2000 mg/kg was studied in 10 rats of both sexes. Sub –acute oral toxicity of aqueous extract of was carried out on 60 rats. We constituted 4 groups of 10 rats each (5 males and 5 females) which were orally administered 300, 600, and 900 mg/kg of aqueous extract and control group received water. 2 group satellites (SAT) of 10 rats each (5 males and 5 females) in which one group (SAT 900 mg/kg) was received orally 900 mg/kg of aqueous extract and another (SAT control) water. Serum blood was collected for biochemical and haematological parameters. The liver and kidney served for histological examination. No deaths of acute oral toxicity were recorded. In female rats, Aspartate Aminotransferase (ASAT) activity increased by 31.20 % and Alamine Aminotransferase (ALAT) increased by 37.20 %. In male rats, only ALAT activity increased significantly by 35.37 % compared to control. Haematological analysis revealed in male rats treated at the dose of 900 mg/kg an increase significant (p<0.001) level of white blood cells with 52.20 %, compared to control group. Histological examination of liver and kidney showed normal architecture. Aqueous extract has untoward effect on liver and kidney, could be considered non-toxic.

Aristolochia longa L. (Aristolochiaceae) is widely used to treat breast cancer in Algerian traditional medicine. The present study was conducted to investigate the effects of ingestion of aqueous extracts of different parts of the plant on liver and kidney functions in wistar albinos mice. Acute oral toxicity was performed to determine DL50, this toxicity was carried out by the oral administration in single doses of 0–12 g/kg for tubers aqueous extracts and 2 g/kg and 5 g/kg for aerial and fruit aqueous extract respectively.  General behavior, adverse effects and mortality were determined for up to 14 days. The animals were sacrificed and biochemical study was done. The acute oral toxicity result revealed that LD50 of the tubers and fruit aqueous extracts was respectively more than 12 g/kg and 5 g/kg, but DL50 of the aerial aqueous extract was at 5 g/kg. The result revealed also that liver and kidney function of different groups receiving high doses was affected as ASAT, ALAT, Urea, creatinine was significantly increased as compared to control group. Histological examination showed alterations of the renal parenchyma and the liver which is greater in animals treated with high dose. Thus, caution should be exercised with its usage. Abstract Objectives Several studies have established the ethnobotanical benefits of Pupalia lappacea (PL) in laboratory animals without extensive toxicological evaluation of its safety profiles. Thus, an extensive toxicological investigation of sub-chronic oral administration of the hydroethanol leaf extract of P. lappacea in rodents was carried out in this study. Methods Different groups of rats were treated orally with the extract (10, 50 and 250 mg/kg) daily for 90 consecutive days. The control group received distilled water (10 mL/kg). After 90 days, some rats were left for additional 30 days without treatment for reversibility study. Blood and organs samples were collected for different evaluations at the end of study periods. Results The extract decreased the bodyweights, feeding and water intakes in female rats. PL increased the weights of the liver and kidney in male rats. PL increased the red blood cell (RBC), packed cell volume (PCV), hemoglobin (Hb), triglycerides (TRIG), cholesterol and high density lipoprotein (HDL) contents in rats. PL (250 mg/kg) significantly reduced the sperm motility and serum testosterone level. Cyto-architectural distortions of the testes, liver and spleen were visible. Conclusions The findings showed that P. lappacea is relatively safe at lower doses but cautions should be taken at higher dose.

<p><strong>Methodology:</strong> Wistar rats were randomly assigned into eight groups of five animals each: four male groups and four female groups. Each sex group had a control group receiving distilled water and three test groups receiving 200, 500 and 1000mg/kg respectively. Animal’s body weights were recorded on the first day and once a week for the four experiment weeks. The hematological analysis included total WBC count, total RBC count, Hb, %HCT, MCV, MCH and MCHC. Biochemical/serum profile studies include TG, TC, ALT, AST, urea and TP. Tissue specimens of the liver, kidney and lung were subjected to histological examination using standard hematoxylin-eosin staining.</p><p><strong>Results:</strong> In male rats, aqueous extract showed significant decreases in relative weight of liver with extreme significance P&lt;0.001 at a dose of 200mg/kg (vs. control group), P&lt;0.001 of lung at all the doses, P&lt;0.05 (200 and 500mg/kg) and P&lt;0.01 (1000mg/kg) in heart weight. In relative kidney weight, only the dose of 1000mg/kg showed a significant increase vs. normal control male rats. Unlike male rats, only relative kidney weight in female rats was significantly different from the control group in a dose-dependent manner. The aqueous extract treated male groups showed significant increases P&lt;0.001 (1000mg/kg) of total WBC count and MCHC, significant decreases of %HTC (dose response manner), P&lt;0.05 total RBC count (at doses of 500 and 1000mg/kg) and Hb P&lt;0.01 (500mg/kg) vs. normal male rats. In female rats, the haematological study showed significant increase P&lt;0.01 of total WBC count (at the doses of 500 and 1000mg/kg), significant decreases P&lt;0.05 and P&lt;0.01 of total RBC respectively at the doses of 200 and 1000mg/kg, significant decrease of Hb with extreme significance P&lt;0.001 at the dose 1000mg/kg, %HTC also decrease dose response manner vs. control female rats. Biochemical study showed in male rats significant decreases in level of TG P&lt;0.001 (at the doses of 200 and 500mg/kg) and urea, although it showed any dose-dependent effect vs. control male rats. AST also decreases (P&lt;0.05) in male rats at the dose of 200mg/kg but significantly increase P&lt;0.001 at the dose of 500mg/kg. In the female rats, biochemical study revealed significant increases in level of TG P&lt;0.001 and urea P&lt;0.01 at the dose of 200mg/kg and significant decreases in level of TG P&lt;0.01, AST P&lt;0.05 and urea P&lt;0.05 at the dose of 500mg/kg (vs. control female rats). Microscopically, there were mild hepatic and renal tissue injuries supporting the hematological analysis.</p><p><strong>Conclusion:</strong> The results indicated that aqueous extract of <em>Alstonia boonei</em> De Wild is toxic in high doses.</p>

Abstract Background Several local communities in Central, Western, Eastern, and Northern regions of Uganda have been using the whole leaf extracts of Aloe vera (L.) Burm. f. (Asphodelaceae) in the treatment of various ailments. Also, several commercial companies sell A. vera as soft drinks in Uganda. However, there are inadequate reports on the toxicities of such preparations. This paper reports the acute and sub-acute oral toxicity of aqueous extracts of whole leaf and green rind of A. vera in Wistar rats. Methods Acute oral toxicity test was carried out in female Wistar rats at doses of 175, 550, 1750, and 5000 mg/kg, p.o. The animals were observed for signs of toxicity for 14 days. Similarly, a sub-acute oral toxicity test was performed in both sexes of rats at doses of 200, 400, and 800 mg/kg, p.o. daily for 28 days. All the groups of animals were monitored for behavioral, morphological, biochemical, and physiological changes, including mortality and compared with respective controls. Body weights were measured weekly while the animals’ relative organ weights, hematological, biochemical, gross, and microscopic pathology were examined on day 29. Results There was no mortality or apparent behavioral changes at the doses tested in acute and sub-acute oral toxicity tests. Thus, the Median Lethal Dose (LD50) of green rind and whole leaf aqueous extracts was above 5000 mg/kg. Gross anatomy revealed that the rats’ relative spleen weight in green rind extract at 200 mg/kg significantly decreased compared to the control group. The creatinine levels in female rats that received green rind extract and the chloride ion levels in male rats administered whole leaf extract were significantly elevated. Conversely, Mean Corpuscular Hemoglobin Concentration (MCHC) levels significantly decreased at lower doses of the green rind extract compared to the control. Histopathology of the kidney revealed the renal interstitium’s inflammation at doses of 200 and 800 mg/kg of the whole leaf extract. Conclusion The findings demonstrated that A. vera green rind and whole leaf extracts are non-toxic at relatively high doses when used for a short duration. Prolonged use of the aqueous whole leaf extract might be associated with kidney toxicity.

Ficus vogelii is a medicinal plant mainly found in tropical Africa and reported to treat inflammatory complaints. This study aims to evaluate the acute and sub-chronic toxicity of the aqueous extract of Ficus vogelii stem bark in wistar rats. For acute study, aqueous extract at a single dose of 5000 mg/kg body weight was administered to female rats and observed for 14 days. In the sub-chronic study, the extract was administered daily to both sex rats at the doses of 100, 200, 400, and 600 mg/kg body weight for 28 consecutive days. Body weight was measured weekly, while hematological, biochemical, and histopathological parameters were analyzed after euthanize. Aqueous extract of Ficus vogelii at all tested doses didn’t produced any mortality or significant change on the body weight and relative weight of rats on acute and sub-chronic studies. The lethal dose 50 was estimated greater than 5000 mg/kg (DL50˃5000 mg/kg). Hematological parameters were recorded non-significant in all treated rats. Aqueous extract at 600 mg/kg significantly changed transaminases and alkaline phosphatase activities, these changes were reversible in satellites. The concentrations of bilirubin was increased at 200 and 600 mg/kg in male rats, at 100, 400 mg/kg in female rats. The levels of lipids markers didn’t changed, except the significant decrease of LDL-cholesterol. Histological examination didn’t showed any change in the architecture of the liver and kidney of rats treated compared to control. Thus aqueous extract of Ficus vogelii stem bark didn’t produced adverse effects in rats after oral acute and sub-chronic treatment.

Abstract. The aims of this study were to determine the potential for acute oral toxicity of ethanolic extract of A.squamosa leaves with LD50 and the histopathological changes in liver and kidney of mice.This research used experimental method as per fixed dose method. The number of animals used in this research were 20 female mice. The study was divided into 2 steps, there were sighting and main studies. The control group was given Na-CMC 0.5%, the treatment groups were given ethanolic extract of A.squamosa leaves with doses of 5, 50, 300,  2,000 and 5,000 mg/kg bw. The results showed that the ethanolic extract of A.squamosa leaves with doses of 2,000 and 5,000 mg/kg bw did not show any toxicity signs. At a dose of 5,000 mg/kg bw caused hydropic degeneration, necrosis hepatocyte, glomerular atrophy, and tubular dilatation.  There was no mortality was observed.It was estimated that LD50 of ethanolic extract of A.squamosa leaves was higher than 5,000 mg/kg bw and the extract were practically non-toxic. Halocarbon 27-S (HC 27-S), a polymer of chlorotrifluoroethylene (CTFE), is used as a lubricating oil for pumps in hyperbaric chambers. Although monomeric CTFE has been shown to produce renal lesions in rats, the toxicity of CTFE polymers have not been investigated. To assess the toxicity of repeated exposure to HC 27–S, three groups (N = 6/group) of male and female Fischer-344 rats were dosed with 2.5 g HC 27-S/kg for 7 or 21 consecutive days. Groups were sacrificed at 7, 21, and 35 days (14 days after the 21-day dosing). Corresponding control groups (N = 6) were dosed with deionized water. Decreased water consumption and urine output were apparent in all test groups. Statistically significant increases in fluoride excretion were noted in 24-hr urine samples assessed periodically during the study. Neurotoxic signs were observed in female rats but not in male rats. Significant increases in liver and kidney weights were seen in all rats, regardless of number of dosing days. The increased fluoride burden in treated animals appeared sufficient to alter bone calcium/phosphate ratios in male rats but not female rats. Gross liver enlargement and hepatocellular cytomegaly indicated that the liver was probably the primary target organ following repeated administration of HC 27-S. Probiotics Bamboo shoot (Dendrocalamus hamiltonii), the young tender and immature culm, is being considered as a complete health food due to its high nutritional and bioactive content. Although, freshly harvested bamboo shoots are nutritionally richer; anti-nutritional factors and toxicity in fresh shoots used for consumption are a matter of concern. This study was conducted to evaluate the sub-acute oral toxicity of aqueous extract in shoots of Dendrocalamus hamiltonii. The bamboo shoot extract was administered orally to Balb/c mice at doses of 400, 800 and 1600 mg/kg, body weight for 28 days. Results revealed a significant enhancement in the level of reduced glutathione and superoxide dismutase accompanied by a significant reduction in the level of lipid peroxidation. A dose-dependent increase was observed in the activity of glutathione peroxidase and glutathione reductase while, a marked reduction was seen in the activity of enzyme catalase. None of any mortality and behavioral changes was observed. At higher dose (1600 mg/kg, body weight) the level of serum creatinine, serum glutamic pyruvic transaminase and lactate dehydrogenase increased significantly however, the histo-architecture of liver and kidney was normal. Therefore, it is concluded that the medium-term oral administration of the bamboo shoot extract for 28 days does not cause toxicity.

Ricinodendron rautanenii is a plant, used in traditional medecine to treat fever, eczema, back pain, cancer and stomacal disorders. This work was carried out to evaluate the safety of the aqueous extract from the stem bark of Ricinodendron rautanenii by determining its potential toxicity after acute and subacute administration in Wistar rats. In Acute toxicity assay, the animals received the extract at the single dose of 5000 mg/Kg and were observed during 48h for mortality and any toxicity manifestations. General behavior, adverse effects and mortality were determined for up to 14 days post treatment. In subacute toxicity assay, extract was given orally to rats at doses of 6, 20, 40 and 80 mg/kg/day for 28 days respectively. Animal body weight, water and food intake, biochemical and hematological parameters were determined. Liver and kidney were examined histologically for any signs of organ damage. No behavioral changes or mortality were recorded in the treated groups. No significant hematological changes were observed in the both sex. The biochemical analysis indicated a significant increase of high density lipoprotein levels in both male and female rats. Histopathological analysis of the liver and kidney did not show any observable cellular damages. The overall finding of this study suggest that, extract of Ricinodendron rautanenii did not cause any death up to a dose of 5000 mg/kg and can be considered non-toxic. Biochemical and histological studies of the extract did not revealed major signs in subacute toxicity.

BACKGROUND: Moringa stenopetala is used as nourishments, and treatment of various diseases. However, there is no much information on its safety. Hence, this study aimed to investigate the chronic administration of aqueous leaves extract of the plant.MATERIALS AND METHODS: Twenty-four rats were divided into: a control group administered with distilled water and three experimental groups, respectively, administered with the extract at doses of 500, 1000, and 2000 mg/kg orally for six months were investigated. Various hematological and biochemical parameters followed by histopathological analysis were evaluated.RESULTS: Treatment with the extract did not significantly affect most of the hematological parameters. However, there were a significant decrease of MCH at doses of 1000 mg/kg and 2000 mg/kg in male rats and increase of MCV at all doses in female rats. Levels of ALP at 2000 mg/kg and those of AST and ALT at 1000 and 2000 mg/kg were significantly increased in male rats. Furthermore, significant decrease in urea and increase in creatinine levels at the dose of 2000 mg/kg occurred in female rats. Mild histopathological changes were also observed in the liver of male rats and kidney of female rats treated with the extract, respectively at doses of 1000 and 2000 mg/kg, and 2000 mg/kg.CONCLUSION: Findings from the present study suggest that prolonged administration of extract of Moringa stenopetala at therapeutic doses is safe, but shows sign of mild toxicity as dose increases, with differential effect on male verses female rats.
