Background: Hemiscorpius lepturus is the most critical scorpion in Khuzestan, Iran, responsible for the majority of deaths caused by scorpion stings. Objectives: This prospective randomized experimental study was performed from June to August 2019 to investigate the effects of various venom fractions on the hemostatic system. Methods: Lyophilized venom of H. lepturus was separated into six fractions by Sephadex G-50 gel chromatography. A total of 96 Albino male rats were treated in eight equal groups; Control: 0.5 ml normal saline (IP); Whole venom group: H. lepturus Lyophilized venom, one mg/kg, IP; F1-F6 groups: Isolated venom fraction 1-6, 0.12 mg/kg, 0.43 mg/kg, 0.08 mg/kg, 0.18 mg/kg, 0.06 mg/kg and 0.13 mg/kg, IP, respectively. Blood sampling was performed at 1, 3, and 24 hours after injection. Results: Plasma fibrinogen was significantly elevated in the whole venom group, while it was significantly decreased in F3, F5, and F6 groups compared to the control group. Prothrombin time (PT) and activated partial thromboplastin time (APTT) assessment revealed that there was a significant prolongation of both tests in whole venom and fraction 5 and 6 receiving groups while fraction 2 only caused a significant PT extension, compared to the control group. Conversely, fraction 3 and 4 resulted in PT and APTT in comparison to the control group. The concentration of FDP and D-dimer was significantly increased in whole venom and F2 group compared to the control group at all sampling times. Conclusions: It seems that H. lepturus venom has both procoagulant and anticoagulant properties which are distinct characteristics of it. Further purification and sequencing of the amino acids in the fraction of peptides can lead to a more precise identification of the mechanisms of venom-induced coagulopathies.

Aim. To study the hemostatic system parameters and magnesium levels in patients with beta-thalassemia. Methods. The object of the study was the blood serum of 96 women with beta-thalassemia: 46 patients with intermediate beta-thalassemia and 50 patients with beta-thalassemia minor, without clinical manifestations of hypercoagulation. The blood serum of 30 healthy donors was used as the control group. It was studied hemostasis system parameters: platelet count activated partial thromboplastin time (aPTT), prothrombin time, plasma fibrinogen level, D-dimer level, euglobulin clot lysis time, antithrombin III activity. The serum magnesium level and risk of deficiency were determined using the MDQ questionnaire. Results. In patients with intermediate beta-thalassemia, an increase in the level of thrombinemia marker D-dimer (500 ng/ml) was revealed. Patients with intermediate beta-thalassemia were divided into two groups according to the revealed level of D-dimer: 14 (30.46.8%) patients with latent hypercoagulation in group 1 and 32 (69.66.8%) patients without latent hypercoagulation in group 2. It was found that in the group with a high levels D-dimer, fibrinogen level was increased (p 0.05), fibrinolysis time was prolonged (p 0.05), activated partial thromboplastin time was shortened (p 0.05), and antithrombin III activity was slightly reduced (p 0.05). The serum magnesium level in patients of the first group was lower (t=7.3; p 0.001), and the risk of deficiency in the questionnaire was higher than in patients of the second group (r=0.785, p 0.05). Hemostasis and magnesium levels in patients with beta-thalassemia minor did not differ from the control group (p 0.05). Conclusion. One-third of patients with intermediate beta-thalassemia have a pre-thrombotic state for hemostasis latent hypercoagulation and magnesium deficiency which can be predictors of clinical signs of thrombosis.

Objective: It is known that local or systemic injection of hemostatic drugs is used to reduce blood loss and number of blood transfusions in patients with hypocoagulation and hemorrhagic diathesis. However, the analysis of literature data shows that the drugs applied for the control of bleeding, traditionally used in medical practice, are not effective enough. This study deals with the systemic hemostatic activity of bis (2-aminoethan-1- sulfonate) calcium in the experiment.Methods: Experimental work in vitro is performed on the blood of healthy male donors, under conditions in vivo it is done on intraperitoneal injection in male rats. Thromboelastography was carried out with apparatus Thromboelastography (TEG) 5000 (Haemoscope Corporation, United States). The influence of first synthesized derivative and ethamsylate on functional activity of platelets was studied using a platelet aggregation analyzer “Biola 230LA” (Russia). Experimental evaluation of the system specific hemostatic activity in vivo was carried out using the model of parenchymatous bleeding in mature male rats. The interference came amid registration of bleeding stop time and extent of blood loss.Results: Bis (2-aminoethan-1-sulfonate) calcium shows procoagulation and proaggregant activity both in vitro and in vivo. Proaggregatory effect of bis (2-aminoethan-1-sulfonate) calcium is successfully implemented in the systemic hemostatic activity in terms of parenchymal bleeding, surpassing the control group and the group of etamsylate.Conclusion: The results of these studies reveal potentially high systemic hemostatic activity of bis (2-aminoethan-1-sulfonate) calcium, urging the need for further study on this compound and its analogs to create on their basis highly efficient, selective correctors of the hemostatic system.

Background: Scorpion stings are responsible for many deaths in humans; however, the toxicity mechanisms of the venoms from many species are not well studied. We investigated the cardiotoxicity of the crude venom from H. lepturus scorpion and its isolated fractions, F-I to F-VI. Methods: The scorpion’s venom was extracted into six fractions by chromotagraphy. Healthy male Wistar rats (N=72) were equally divided into eight groups of nine: G1: Controls (0.5ml. normal saline), G2: Crude venom (1000µg/kg), G3: F-I (120µg/kg), G4: F-II (430µg/kg), G5: F-III (80 µg/kg), G6: F-IV (180µg/kg), G7: F-V (60µg/kg), and G8: F-VI (130µg/kg). Blood samples were obtained by cardiac puncture at 1, 3 and 24 hours after the venom injection. The serum levels of AST, LDH, CPK, CK-MB and troponin-I were determined. Upon euthanasia, the hearts were removed from the rats and examined microscopically for histopathology. Results: In groups receiving crude venom and F-VI, we observed multifocal fragmentation of myocardial fibers, hemorrhage, degeneration and disappearance of striations in cardiac muscles as compared to the controls. The findings showed that AST and LDH activity in groups 2, 4 and 8, CPK activity in groups 2, 4, 6 and 8 and CK-MB activity and troponin-I levels in groups 2 and 8 increased significantly compared to those in the control group. Conclusion: There was evidence of significant cardiotoxicity in the group receiving crude venom and F-VI. Although alterations in the enzymatic and troponin-I levels were observed in other groups, the greatest cardiotoxicity of H. lepturus venom was caused by fraction VI.

The objective: to evaluate the performance of screening coagulation tests in normal pregnancy. Materials and methods. A study of screening coagulation tests was conducted with the participation of 124 patients with normal pregnancy in terms of 12–14, 20–22, 28–30 and 37–40 weeks of gestation. The control group consisted of 82 patients who were at the pregravid stage of pregnancy planning. Coagulation tests were determined on a HELENA 2000 coagulometer. Namely, the activated partial thromboplastin time, the percentage of prothrombin by Quick and fibrinogen were examined. Results. During normal pregnancy, a dynamic, statistically significant increase in plasma fibrinogen level was determined. This indicator during full-term pregnancy differed from that of non-pregnant women by 1.8 times. During all normal pregnancies, there was no statistically significant dynamics of the activated partial thromboplastin time and the percentage of prothrombin by Quick, but there was a tendency towards these indicators increase in coagulation potential with an increase in the gestation term. Conclusion. When assessing a screening coagulation tests, pregnancy term should be taken into account first of all, rather than comparing the obtained values with reference indices for non-pregnant women. The CoViD-19 pandemic have been lasting since 2019 year. It is a serious threat for health care system around the world because a huge number of patients requires of intensive assistance. Firstly, SARS-CoV-2 causes damage of the respiratory system. However, researches demonstrate that CoViD-19 is the hidden enemy for our body. It causes suffering of other organs. Coagulopathy often develops in SARS-CoV-2 patients. It includes acute inflammation, hypercoagulation and violation of the integrity and functions of the endothelium. A lot of scientific information has accumulated about changed in the hemostatic system during deveploment of CoViD-19. However, clinical researches are ambiguous, but abnormal blood clotting is demonstrated in most hospitalized patients with acute CoViD-19. Our research is aimed to analyse the main parameters of the hemostatic system in the pregnant women with infection of SARS-CoV-2 and the comparison with conventionally healthy pregnant patients. We have revealed the concentration of fibrinogen is increasing by 2,1 g/L, levels of D-dimer are increased by 500 ng/mL, the prothrombic time is rising by 1,2 seconds in plasma of the CoViD-19 pregnant patients unlike by conventionally healthy pregnant patients. Changes in the duration of activated partial thromboplastin time weren`t detected in plasma of the CoViD-19 pregnant patients unlike by conventionally healthy pregnant patients. Our research could be useful for diagnostic complications of the hemostatic system in CoViD-19 patients, for detection of blood clotting disorders during early stages of development, suppression of the spread of pathological processes of the fetus in pregnant women and for creation of effective treatment

Abstract. Alterations of essential elements in the brain are associated with the pathophysiology of many neuropsychiatric disorders. It is known that chronic/overwhelming stress may cause some anxiety and/or depression. We aimed to investigate the effects of two different chronic immobilization stress protocols on anxiety-related behaviors and brain minerals. Adult male Wistar rats were divided into 3 groups as follows ( n = 10/group): control, immobilization stress-1 (45 minutes daily for 7-day) and immobilization stress-2 (45 minutes twice a day for 7-day). Stress-related behaviors were evaluated by open field test and forced swimming test. In the immobilization stress-1 and immobilization stress-2 groups, percentage of time spent in the central area (6.38 ± 0.41% and 6.28 ± 1.03% respectively, p < 0.05) and rearing frequency (2.75 ± 0.41 and 3.85 ± 0.46, p < 0.01 and p < 0.05, respectively) were lower, latency to center area (49.11 ± 5.87 s and 44.92 ± 8.04 s, p < 0.01 and p < 0.01, respectively), were higher than the control group (8.65 ± 0.49%, 5.37 ± 0.44 and 15.3 ± 3.32 s, respectively). In the immobilization stress-1 group, zinc (12.65 ± 0.1 ppm, p < 0.001), magnesium (170.4 ± 1.7 ppm, p < 0.005) and phosphate (2.76 ± 0.1 ppm, p < 0.05) levels were lower than the control group (13.87 ± 0.16 ppm, 179.31 ± 1.87 ppm and 3.11 ± 0.06 ppm, respectively). In the immobilization stress-2 group, magnesium (171.56 ± 1.87 ppm, p < 0.05), phosphate (2.44 ± 0.07 ppm, p < 0.001) levels were lower, and manganese (373.68 ± 5.76 ppb, p < 0.001) and copper (2.79 ± 0.15 ppm, p < 0.05) levels were higher than the control group (179.31 ± 1.87 ppm, 3.11 ± 0.06 ppm, 327.25 ± 8.35 ppb and 2.45 ± 0.05 ppm, respectively). Our results indicated that 7-day chronic immobilization stress increased anxiety-related behaviors in both stress groups. Zinc, magnesium, phosphate, copper and manganese levels were affected in the brain.

SummaryIncreased thrombin generation occurs in many individuals with inherited defects in the antithrombin or protein C anticoagulant pathways and is also seen in patients with thrombosis without a defined clotting abnormality. Hyperhomocysteinemia (H-HC) is an important risk factor of venous thromboembolism (VTE). We prospectively followed 48 patients with H-HC (median age 62 years, range 26-83; 18 males) and 183 patients (median age 50 years, range 18-85; 83 males) without H-HC for a period of up to one year. Prothrombin fragment Fl+2 (Fl+2) was determined in the patient’s plasma as a measure of thrombin generation during and at several time points after discontinuation of secondary thromboprophylaxis with oral anticoagulants. While on anticoagulants, patients with H-HC had significantly higher Fl+2 levels than patients without H-HC (mean 0.52 ± 0.49 nmol/1, median 0.4, range 0.2-2.8, versus 0.36 ± 0.2 nmol/1, median 0.3, range 0.1-2.1; p = 0.02). Three weeks and 3,6,9 and 12 months after discontinuation of oral anticoagulants, up to 20% of the patients with H-HC and 5 to 6% without H-HC had higher Fl+2 levels than a corresponding age- and sex-matched control group. 16% of the patients with H-HC and 4% of the patients without H-HC had either Fl+2 levels above the upper limit of normal controls at least at 2 occasions or (an) elevated Fl+2 level(s) followed by recurrent VTE. No statistical significant difference in the Fl+2 levels was seen between patients with and without H-HC. We conclude that a permanent hemostatic system activation is detectable in a proportion of patients with H-HC after discontinuation of oral anticoagulant therapy following VTE. Furthermore, secondary thromboprophylaxis with conventional doses of oral anticoagulants may not be sufficient to suppress hemostatic system activation in patients with H-HC.

Abstract. It has been suggested that treatment of intact male rats with oestradiol benzoate (OeB) causes an interference with testosterone (T) production by the testes by a direct inhibitory effect on steroidogenesis. To test this hypothesis, different doses (5, 10 or 25 IU) of hCG were administered concomitantly with 50 μg of OeB to adult intact or hypophysectomized male rats. The testicular and plasma testosterone, and serum hCG levels were determined. The sex accessory weights were recorded. In the intact OeB-treated group of animals, hCG stimulated both the secondary sex organs and plasma testosterone levels above the intact control group. However, in hypophysectomized animals, although plasma testosterone levels increased above that of intact controls, their secondary sex organ weights did not. Moreover, inspite of high circulating hCG levels, the testicular testosterone content and concentration remained suppressed in OeB-treated animals. The reason for such dichotomy of hCG action on OeB-treated animals is not clear at present.

The present study aimed to investigate the effect of turmeric and ginger on lipid profile of male rats exposed to oxidative stress induced by hydrogen peroxide H2O2 at a concentration of 1% given with consumed drinking water to male rats. Methods: 200 mg/kg from turmeric and ginger were used, and the animals were treatment for 30 days. Results: the results showed a significant increase in cholesterol, triglycerides, low density lipoprotein (LDL), very low density lipoprotein (VLDL), whereas it explained a significant decrease in high density lipoprotein (HDL) of male rats exposed to oxidative stress when compared with control group. the results showed a significant decrease in cholesterol, triglycerides, (LDL), (VLDL), whereas it explained a significant increase in (HDL) of rats treated with turmeric and ginger at dose 200 mg/kg when compared with male rats exposed to oxidative stress.

Цель исследования - сравнительный анализ влияния селективных агонистов отдельных классов опиоидных рецепторов на белковосинтетическую функцию печени, развитие цитолитического и холестатического синдромов у крыс, подвергшихся частичной гепатэктомии. Методика. Работа выполнена на 152 крысах-самцах Вистар массой 200-250 г. Частичную гепатэктомию выполняли по методу, описанному Higgins G.M. и Anderson R.M. с удалением 70% ткани печени. В плазме крови определяли концентрации общего белка, альбуминов, общего билирубина, активность аланинтрансаминазы (АЛТ), аспартаттрансаминазы (АСТ), лактатдегидрогеназы (ЛДГ) традиционными методами. Опиоиды: DAGO в дозе 6,3 мкг/кг, DSLET в дозе 10,0 мкг/кг, динорфин А (1-13) в дозе 20,1 мкг/кг, вводили внутрибрюшинно ежедневно 1 раз в сутки в течение 5 сут. эксперимента в объеме 0,2 мл. Контрольным животным аналогично вводили физраствор. Результаты. Удаление 70% ткани печени у крыс-самцов Вистар сопровождается развитием печеночной недостаточности, проявляющейся гипербилирубинемией, гипоальбуминемией, гипопротеинемией, повышением активности трансаминаз и лактатдегидрогеназы. Применение селективных агонистов опиоидных рецепторов у крыс, которым моделировали частичную гепатэктомию, оказывало гепатопротективное действие и снижало выраженность проявлений печеночной недостаточности, начиная с 3-х сут. после резекции. Активность трансаминаз, лактатдегидрогеназы и концентрация общего билирубина у животных, которым вводили опиоиды, были существенно ниже, чем в контрольной группе. Содержание общего белка и альбуминов было статистически значимо выше в группах, которые получали исследованные пептиды, по сравнению с контрольной группой на 7-е сут. после частичной гепатэктомии. Наиболее выраженное действие проявлял селективный агонист опиоидных мю-рецепторов DAGO. По нашему мнению, такое влияние пептидов объясняется присущими им антиоксидантным и антигипоксическим эффектами, что снижает повреждающее действие оперативного вмешательства на печень. Более выраженное влияние DAGO связано, по-видимому, с особенностями распределения опиоидных рецепторов или устойчивостью пептида к действию эндопептидаз благодаря модификациям в молекуле пептида. Заключение. Применение опиоидов стимулирует восстановление функциональной активности печени после частичной гепатэктомии. Наибольший эффект отмечается при введении мю-агониста DAGO. Aim. The aim of the study was to compare effects of selective agonists of opioid receptors from different classes on the protein-synthesizing function of liver and development of cytolytic and cholestatic syndromes in rats after partial hepatectomy. Methods. The study was conducted on 152 Wistar male rats weighing 200-250 g. The animals were subjected to partial hepatectomy according to the Higgins and Anderson method. Concentrations of total protein, albumin, total bilirubin, and activities of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase were measured in plasma using standard methods. The opioids, DAGO (6.3 mg/kg), DSLET (10.0 mg/kg), and dynorphin A (1-13) (20.1 mg/kg), were injected in 0.2 ml of saline daily for 5 days. Control animals were injected with 0.2 ml of saline for 5 days. Results. Resection of 70% of liver tissue resulted in development of liver failure as evidenced by hyperbilirubinemia, hypoalbuminemia, hypoproteinemia, and increased transaminase and lactate dehydrogenase activities. Selective agonists of opioid receptors administered to the rats after partial hepatectomy exerted a hepatoprotective effect and alleviated the signs of liver failure beginning from the 3 day after resection. Transaminase and lactate dehydrogenase activities were significantly lower in opioid-treated rats than in the control group. Levels of total protein and albumins were significantly higher in the groups injected with the study peptides compared to the control group on the 7 day after partial hepatectomy. The selective agonist of opioid m-receptors, DAGO, exerted the most pronounced effect. Apparently, the similar effects of peptides were due to their antioxidant and anti-hypoxic action, which alleviated the detrimental effect of liver surgery. The more pronounced effect of DAGO apparently resulted from peculiarities of opioid receptors distribution or peptide resistance to endopeptidase action due to modifications of the peptide molecule. Conclusion. Administration of opioids stimulated restoration of liver functional activity after partial hepatectomy. Injections of the m-agonist, DAGO, produced the most pronounced effect.
