Effect of opioid peptides on liver function after patial hepatectomy

Цель исследования - сравнительный анализ влияния селективных агонистов отдельных классов опиоидных рецепторов на белковосинтетическую функцию печени, развитие цитолитического и холестатического синдромов у крыс, подвергшихся частичной гепатэктомии. Методика. Работа выполнена на 152 крысах-самцах Вистар массой 200-250 г. Частичную гепатэктомию выполняли по методу, описанному Higgins G.M. и Anderson R.M. с удалением 70% ткани печени. В плазме крови определяли концентрации общего белка, альбуминов, общего билирубина, активность аланинтрансаминазы (АЛТ), аспартаттрансаминазы (АСТ), лактатдегидрогеназы (ЛДГ) традиционными методами. Опиоиды: DAGO в дозе 6,3 мкг/кг, DSLET в дозе 10,0 мкг/кг, динорфин А (1-13) в дозе 20,1 мкг/кг, вводили внутрибрюшинно ежедневно 1 раз в сутки в течение 5 сут. эксперимента в объеме 0,2 мл. Контрольным животным аналогично вводили физраствор. Результаты. Удаление 70% ткани печени у крыс-самцов Вистар сопровождается развитием печеночной недостаточности, проявляющейся гипербилирубинемией, гипоальбуминемией, гипопротеинемией, повышением активности трансаминаз и лактатдегидрогеназы. Применение селективных агонистов опиоидных рецепторов у крыс, которым моделировали частичную гепатэктомию, оказывало гепатопротективное действие и снижало выраженность проявлений печеночной недостаточности, начиная с 3-х сут. после резекции. Активность трансаминаз, лактатдегидрогеназы и концентрация общего билирубина у животных, которым вводили опиоиды, были существенно ниже, чем в контрольной группе. Содержание общего белка и альбуминов было статистически значимо выше в группах, которые получали исследованные пептиды, по сравнению с контрольной группой на 7-е сут. после частичной гепатэктомии. Наиболее выраженное действие проявлял селективный агонист опиоидных мю-рецепторов DAGO. По нашему мнению, такое влияние пептидов объясняется присущими им антиоксидантным и антигипоксическим эффектами, что снижает повреждающее действие оперативного вмешательства на печень. Более выраженное влияние DAGO связано, по-видимому, с особенностями распределения опиоидных рецепторов или устойчивостью пептида к действию эндопептидаз благодаря модификациям в молекуле пептида. Заключение. Применение опиоидов стимулирует восстановление функциональной активности печени после частичной гепатэктомии. Наибольший эффект отмечается при введении мю-агониста DAGO. Aim. The aim of the study was to compare effects of selective agonists of opioid receptors from different classes on the protein-synthesizing function of liver and development of cytolytic and cholestatic syndromes in rats after partial hepatectomy. Methods. The study was conducted on 152 Wistar male rats weighing 200-250 g. The animals were subjected to partial hepatectomy according to the Higgins and Anderson method. Concentrations of total protein, albumin, total bilirubin, and activities of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase were measured in plasma using standard methods. The opioids, DAGO (6.3 mg/kg), DSLET (10.0 mg/kg), and dynorphin A (1-13) (20.1 mg/kg), were injected in 0.2 ml of saline daily for 5 days. Control animals were injected with 0.2 ml of saline for 5 days. Results. Resection of 70% of liver tissue resulted in development of liver failure as evidenced by hyperbilirubinemia, hypoalbuminemia, hypoproteinemia, and increased transaminase and lactate dehydrogenase activities. Selective agonists of opioid receptors administered to the rats after partial hepatectomy exerted a hepatoprotective effect and alleviated the signs of liver failure beginning from the 3 day after resection. Transaminase and lactate dehydrogenase activities were significantly lower in opioid-treated rats than in the control group. Levels of total protein and albumins were significantly higher in the groups injected with the study peptides compared to the control group on the 7 day after partial hepatectomy. The selective agonist of opioid m-receptors, DAGO, exerted the most pronounced effect. Apparently, the similar effects of peptides were due to their antioxidant and anti-hypoxic action, which alleviated the detrimental effect of liver surgery. The more pronounced effect of DAGO apparently resulted from peculiarities of opioid receptors distribution or peptide resistance to endopeptidase action due to modifications of the peptide molecule. Conclusion. Administration of opioids stimulated restoration of liver functional activity after partial hepatectomy. Injections of the m-agonist, DAGO, produced the most pronounced effect.

Aims: Damage to liver tissue and its dysfunction is very important and if left untreated, it can cause serious problems and even death. In this study, we aimed to investigate the effects of the hydroalcoholic extracts of Linum usitatissimum and Rosa damascena on liver enzymes, total protein, bilirubin, albumin, and serum glucose levels. Materials and Methods: This is a non-randomized clinical trial conducted on 42 male rats divided into 6 groups; control group (group 1) received only sufficient water and food, groups 1 and 2 received 300 and 500 mg/ kgB.W Linum usitatissimum extract, groups 3 and 4 received 500 and 1000 mg/ kgB.W Rosa damascena, and group 6 received 100 mg/ kgB.W Linum usitatissimum plus 250 mg/ kgB.W Rosa damascena extracts intraperitoneally for 28 days. After the last injection, the rats were weighed and their blood samples were collected. The study parameters were measured using a colorimetric method by a spectrophotometer, and then were analyzed using ANOVA and Tukey’s test in SPSS V. 25 at a significance level of P<0.05. Findings: There was no significant difference between alanine aminotransferase, alkaline phosphatase, total and direct bilirubin levels in the control group in comparison with other groups (P>0.05). In the groups received Rosa damascena extract, there was a significant difference between total protein and albumin levels compared to the control group (P<0.05). Moreover, there was a significant difference between serum glucose and aspartate aminotransferase in the control group compared to other groups (P<0.05). Conclusion: Linum usitatissimum and Rosa damascena have no negative effect on the liver function. The probability of diarrhea occurrence and the possible effects on the total protein and serum albumin after using Rosa damascena, and the effects of different doses of Linum usitatissimum on the glucose levels should be taken into account.

          The experiment was conducted in two stages: first stage: determination of the most effective dose of Cyperus rotundus tubers aquatic extract in male rats for 5 days, which was concentrated as (200 mg/kg body weight). The second stage was designed to observe and test the protective effects of C. rotundus tubers aquatic extract in the liver and kidney functions of male rats exposed to cadmium chloride poisoning (5 mg/kg bw) for 30 days. The animals were divided into (4) groups within each group (5) animals weighted (200-220g). The results of oral dose of cadmium chloride showed a significant increase of (P˂0.05) in the activity of both enzymes Alanine aminotransferase (ALT), Aspartate aminotransferase (AST), Alkaline phosphatase (ALP), glucose, urea, uric acid, creatinine, malondialdehyde (MDA) and peroxynitrite (ONOO-). And a significant decrease (P˂0.05) in the values ​​of total protein, albumin, globulin and glutathione (GSH) compared with the control group. The oral dosage of the C. rotundus tubers aquatic extract resulted in a significant increase in GSH. The values ​​of each were not significantly different ALT, AST, ALP, Glucose, urea, uric acid, creatinine, total protein, albumin, globulin, MDA and peroxy nitrite Compare with control group. The results of the oral dosage with (C. rotundus tubers aquatic extract + cadmium chloride) showed a positive effect on these values ​​compared with the control group and cadmium chloride. It is concluded that the C. rotundus tubers aquatic extract has protective effects and reduces the effects that cadmium chloride can cause in rats liver and kidney functions through its antioxidant activity and removal of free radicals.

PURPOSE: To evaluate the protective action of alpha-tocopherol in ischemia/reperfusion injuries of pelvic member of rats. METHODS: Thirty adult male rats of the Wistar strain were randomized into three experimental groups of 10: Group I - control group with no ischemia or reperfusion. Groups II and III - four hours of ischemia and of hours of reperfusion by means of clamping of the infrarenal aorta. The animals of Group II were treated with saline and those of Group III were treated with i.v. alpha-tocopherol (50 mg/kg). Parameters studied were biopsies of the soleus muscle, dosing of creatine phosphokinase, lactate dehydrogenase, potassium, calcium and arterial blood gasometry. RESULTS: The results of biopsies of the soleus muscles studied by optical microscopy, were not significant in terms of presence of edema among the three groups studied. Variables inflammation and necrosis were not observed, therefore cannot be statistically analyzed. As to dosing of calcium and lactate dehydrogenase, the pH, pO2 and pCO2 values were not significant for all groups studied. We observed that the levels of potassium (Group II > Group I, Fcalculated = 5.84; Fcritical = 3.33), creatine phosphokinase (Group II > Groups I and III, Hcalculated = 13.92; Hcritical = 5.99) and bicarbonate (Groups I and III > Group II, Hcalculated = 11.98; Hcritical = 5.99) presented significant results among groups. CONCLUSION: From the serum biochemical perspective, the treatment with alpha-tocopherol has attenuated the metabolic injuries in the ischemia/reperfusion syndrome in this experimental model.

The objective of this work was to investigate certain biochemical parameters in the blood of sheep in conditions of different intensity of parasitic infection with Strongyloides papillosus, and also after the therapy with albendazole. The research was carried out on the sheep of Wurtemberg breed (n = 30) in which there was determined low, medium and high intensity of parasitic infection with S. papillosus. The control group (n = 10) consisted of the sheep that were negative to the presence of the parasitic infection. The degree and type of changes were monitored by determining the concentration of glucose, total protein, albumin, Albumin/Globulin ratio, AST, urea, total bilirubin, calcium, phosphorus, lactate dehydrogenase (LDH) total activity and isoenzyme distribution LDH1-5. On the basis of the obtained results it was concluded, through isoenzyme distribution of lactate dehydrogenase, that parasitic infection with S. papillosus can cause damage to the liver, heart muscle and lungs, while after the therapy with albendazole, the most affected organ is the liver. The concentration of glucose, total protein and albumin linearly decreased with the increase of intensity of the parasitic infection (p < 0,05), and after the therapy with albendazole, a drop in concentration of these parameters was at a statistically significantly higher level (p< 0,01). AST activity as well as the concentration of urea and total bilirubin grew linearly along with the intensity of the parasitic infection (p < 0,05). After the therapy with ABZ, AST activity and concentration of total bilirubin were at a statistically significantly higher level (p < 0,001), while urea concentration kept the same level as in the case of the parasitic infection. The values of calcium concentration (p > 0,05) as well as phosphorus (p < 0,05) linearly decreased with the parasitic infection intensity growth. Trend towards the decline of the concentration of these macroelements continues even after the therapy with albendazole (p < 0,001). Considering our previous research in the field of oxidative stress, a phenomenon that is fundamentally related to the changes determined in this work, we strongly recommend that besides antihelmintics, preparations with antioxidative properties should be used in antiparasitic protocols as well. <br><font color="red"><b> This article has been retracted. The purpose of the experiment is an investigate the association of walnut oil with lipid profiles, glucose as well as total proteins and assessment its side effect on some sensitive organs such as the liver and spleen tissues. The experiment divided into two main groups are treated group with walnut oil and the control group, where the former divided into two subgroups 0.25 and 0.5 ml of oil per each. We found a statistically non-significant difference between treated and control groups (for total protein, lipid profile, and glucose). There was no change in total protein, but cholesterol decreased by 0.25 ml but a little increased by 0.5 ml. HDL was increased for the treated group. While on 0.25 ml LDL decreased in treated animals, on another hand no change for 0.5 ml. Also, no change for VLDL between treated and control. The only triglyceride was increased but non-significant for the treated group compared with the control. Both doses decreased in treated animals for glucose. We also found an increase in whole-body weight and on sensitive organs such as the liver and spleen. Even no change in a histological study for the mentioned organs. The conclusion: By walnut oil, all parameters have changed despite the treated group was normal without any induced diseases. So, recommended the researchers induce the disorders in the liver and assessment the extracted oil on the lipid profile.

The present study was carried out in the animal house and laboratories of the College of Veterinary Medicine/ University of Basrah to investigate the role of the administration of vitamin D3 on liver and kidney function of alloxan induced toxic in mature male rats. For this purpose, 48 male rats were divided into 4 groups with 12 animals in each as the following; group (1), animals of were administered with D.W(0.2ml)/day by oral gavages for 6 weeks as control group, group(2) were injected intraperitoneally with 150 mg/kg b.w of alloxan as diabetic group, group (3) were injected with alloxan then administered with 500 UI/kg/day of alpha 25-hydroxy vitamin D3 by gavages for 6 weeks orally, group (4) were injected with alloxan then administer 1000 UI/kg/day of alpha 25-hydroxy vitamin D3 by gavages for 6 weeks orally. At the end of the experiment, animals were euthanized and blood samples were obtained for some biochemical assessment of liver and kidneys.In compares with control, alloxan treated rats recorded a significant increase (p≤0.05)in serum concentration of ALT, AST,ALP ,urea and creatinine, while there was a significant decrease (p≤0.05) in serum total protein. On other hand, administration of vitamin D3 for diabetic rats induced a significant improvement (P≤0.05) of serum ALT, AST, ALP, total protein urea and creatinine.

The current study focused on testing the hypolipidemic activity of two doses of barley bran on hypercholesterolemic male rats. Twenty-four male albino rats weighing 180–200 gm were divided into four groups. The first group (G1) was the negative control, the second group (G2) was the positive control group fed 2% cholesterol in the diet, and rats of the third and the fourth groups were fed 2% cholesterol and were cosupplemented with 5% and 10% barley bran, respectively, for 8 weeks. The hypercholesterolemic rats of (G2) showed an increase in lipid profile, liver enzymes, lactate dehydrogenase, creatine kinase-MB, and lipid peroxide and a decrease in antioxidant enzymes, whereas kidney function, fasting blood sugar, glycated hemoglobin total protein, and total bilirubin were not significantly affected compared with the negative control group in G1. Moreover, histology of heart, liver, and kidney of G2 rats showed histopathological changes compared with the negative control. Administration of the two doses of barley bran in G3 and G4 to the hypercholesterolemic rats ameliorated the level of lipids, liver enzymes, lactate dehydrogenase, and creatine kinase-MB. In addition, the histology of heart, liver, and kidney tissues nearly restored the normal state as in G1.

Abstract. Alterations of essential elements in the brain are associated with the pathophysiology of many neuropsychiatric disorders. It is known that chronic/overwhelming stress may cause some anxiety and/or depression. We aimed to investigate the effects of two different chronic immobilization stress protocols on anxiety-related behaviors and brain minerals. Adult male Wistar rats were divided into 3 groups as follows ( n = 10/group): control, immobilization stress-1 (45 minutes daily for 7-day) and immobilization stress-2 (45 minutes twice a day for 7-day). Stress-related behaviors were evaluated by open field test and forced swimming test. In the immobilization stress-1 and immobilization stress-2 groups, percentage of time spent in the central area (6.38 ± 0.41% and 6.28 ± 1.03% respectively, p < 0.05) and rearing frequency (2.75 ± 0.41 and 3.85 ± 0.46, p < 0.01 and p < 0.05, respectively) were lower, latency to center area (49.11 ± 5.87 s and 44.92 ± 8.04 s, p < 0.01 and p < 0.01, respectively), were higher than the control group (8.65 ± 0.49%, 5.37 ± 0.44 and 15.3 ± 3.32 s, respectively). In the immobilization stress-1 group, zinc (12.65 ± 0.1 ppm, p < 0.001), magnesium (170.4 ± 1.7 ppm, p < 0.005) and phosphate (2.76 ± 0.1 ppm, p < 0.05) levels were lower than the control group (13.87 ± 0.16 ppm, 179.31 ± 1.87 ppm and 3.11 ± 0.06 ppm, respectively). In the immobilization stress-2 group, magnesium (171.56 ± 1.87 ppm, p < 0.05), phosphate (2.44 ± 0.07 ppm, p < 0.001) levels were lower, and manganese (373.68 ± 5.76 ppb, p < 0.001) and copper (2.79 ± 0.15 ppm, p < 0.05) levels were higher than the control group (179.31 ± 1.87 ppm, 3.11 ± 0.06 ppm, 327.25 ± 8.35 ppb and 2.45 ± 0.05 ppm, respectively). Our results indicated that 7-day chronic immobilization stress increased anxiety-related behaviors in both stress groups. Zinc, magnesium, phosphate, copper and manganese levels were affected in the brain.

Abstract. It has been suggested that treatment of intact male rats with oestradiol benzoate (OeB) causes an interference with testosterone (T) production by the testes by a direct inhibitory effect on steroidogenesis. To test this hypothesis, different doses (5, 10 or 25 IU) of hCG were administered concomitantly with 50 μg of OeB to adult intact or hypophysectomized male rats. The testicular and plasma testosterone, and serum hCG levels were determined. The sex accessory weights were recorded. In the intact OeB-treated group of animals, hCG stimulated both the secondary sex organs and plasma testosterone levels above the intact control group. However, in hypophysectomized animals, although plasma testosterone levels increased above that of intact controls, their secondary sex organ weights did not. Moreover, inspite of high circulating hCG levels, the testicular testosterone content and concentration remained suppressed in OeB-treated animals. The reason for such dichotomy of hCG action on OeB-treated animals is not clear at present.
