Seed vitality and vigour are the two most common parameters related to seed quality. It is crucial to have reliable methods and tests for seed quality and seed vigour testing. The standard germination test can be used to predict field emergence, but laboratory seed testing conditions are often in conflict with field conditions. Validated tests for vigour evaluation in legumes are the conductivity test, the accelerated ageing test and the tetrazolium test. Also, other types of vigour tests have been used to solve different problems in the seed sector. The modern approach, the computerised image analysis of legume seeds and sprouts, based on interactive and traditional methods, is a promising alternative for vigour determination.

Studies were undertaken at the Department of Seed Science and Technology, Tamil Nadu Agricultural University, Coimbatore using different seed lots of blackgram cv. CO 6 with a view to elicit information on seed invigouration for maintenance of seed quality during storage period. Seeds soaking followed by drying for different durations ranging from 1h, 2h and 3h with different inorganic chemicals viz., Vitamin E (a- tocopherol), Para Amino Benzoic Acid (PABA) and Salicylic Acid at concentrations of 2.5 mM, 5.0 mM and 7.5 mM in 1:1 seed to solution ratio was attempted. The results shows that soaking of varying seed lots of freshly harvested, aged treated and treated aged seeds of blackgram (3 days of accelerated ageing) in low concentrations viz., 2.5 mM, 5.0 mM of salicylic acid followed by drying was effective in controlling seed deterioration and improving seed vigour and viability in all the seed lots.

AbstractSeed shelf-life or longevity is difficult to predict or to measure on a practical time scale. Predictive models suggest that water has the same effect on ageing rate for all seed lots within a species and that initial seed quality is the dominating factor explaining within-species variation. These assumptions are used in ‘accelerated ageing’ or ‘controlled deterioration’ (AA/CD) tests, which are commonly used to predict seed longevity in commercial and research applications. In this study, we describe within-species variation of longevity for seeds of Secale cereale and S. strictum (cultivated rye and its wild progenitor) under typical dry storage conditions of a genebank, and show that initial seed quality is an important, but not sole, factor explaining measured longevity. We also test the correlation of seed longevity, measured under humid and dry conditions, using 50 cultivars of rye, wheat (Triticum aestivum) and the intergeneric cross triticale, to assess how well AA/CD tests predict seed shelf-life during dry storage. Known differences in longevity between wheat and rye were confirmed at all water contents, and triticale seeds demonstrated intermediate behaviour. Longevity measured for humid and dry conditions were weakly correlated when comparisons included all grain types and were not correlated in within-grain-type comparisons. Response to moisture varied among cultivars. These findings do not support assumptions made in seed ageing models that use AA/CD tests. Our results suggest that more traits are involved in the expression of seed longevity than those typically measured in studies of initial seed vigour.

ABSTRACT The feasibility of extending the storability of fresh hot pepper seeds, by modifying the storage atmosphere, using the respiration inhibitors and seed desiccants was examined. Halogens such as chlorine, bromine and iodine and methanol, ethanol and potassium permanganate were used for a period of 22 months under accelerated ageing conditions of high seed moisture and temperature. The various seed quality parameters - viability, vigour, speed of germination, seedling dry weight, and moisture content were evaluated. In the untreated control, decreased seed viability was observed within 10 months of storage. However, iodine and chlorine were proved effective in retaining high seed viability up to 90.7 and 88.0%, respectively, even after 22 months of storage. Chlorine treatment was able to retain the seed vigour (904.0), iodine (766.4), KMnO4 (754.4) and methanol (566.7) whereas the value of vigour index in control was 72.0. Chlorine and iodine were on par in their ability to maintain the speed of seed germination (25.2 and 24.8, respectively), followed by KMnO4 (20.2). Seedling dry weight was proved as a parameter not sensitive in estimating the seed quality since even after 22 months storage of seeds at atmospheres of chlorine, iodine, KMnO4 and methanol did not show any differences. KMnO4 sharply reduced the seed moisture content from 9.83 to 7.89% providing better storability over the control.

The ability of a single early count of radicle emergence (RE) to predict differences in the production of normal seedlings (NG) and seed vigour was examined in four seed lots from each of three varieties of cauliflower (Brassica oleracea var botrytis). Initial seed quality was assessed in germination and controlled deterioration (CD) vigour tests. RE (first appearance of the radicle) was counted manually after 48 hours in the germination test (RE 48h 20/30°C) and every two hours for 144 hours at 20°C using automated image analysis. Seed vigour was assessed in field and glasshouse trials. A large proportion of the variance in NG was explained by the RE count. The same RE counts at 20/30°C and 20°C predicted seed vigour in the field and glasshouse and revealed the same vigour differences as the validated CD test. Differences in vigour (field and glasshouse emergence) observed between varieties were related to the extent of previously sustained seed ageing revealed by the CD test, although genotypic differences in vigour could not be excluded. We propose that single counts of RE taken manually or by image analysis can be used to reveal differences in the NG and vigour of seed lots of cauliflower.

Investigations were made on the seed viability (standard germination test and vigour after accelerated ageing) and seed quality (starch, protein, lipid, sucrose, glucose and fructose) of seeds of maize (cv Melkassa-2) and sorghum (cv Macia) harvested from plants after exposure to and recovery from pre- and post-flowering dehydration. The objectives of the study were to achieve a better understanding of 1) the effects of water deficit during the pre- and post-flowering stages on the seed viability and food (storage reserves) quality, and 2) the effects of dehydration and rehydration cycles at critical growth stages on subsequent seed performance and production, which could lead to the development of cultivars more able to efficiently partition assimilates to the reproductive organs in the field. The experiment was conducted in a controlled environment growth chamber under constant environmental conditions (12/12 h day/night, 28–32/17°C day/night temperature, 60–80% RH and 1200–1400 μmol m −2 s −1 PPDF). The seed viability and vigour tests were done on air-dried seeds and the seed quality analysis on freeze-dried seeds of both species.The results of the standard germination test indicated that sorghum seeds harvested after both pre- and post-flowering dehydration were not affected by the treatment, while maize seeds had reduced % germination. Sorghum seeds harvested after post-flowering dehydration had significantly decreased vigour after accelerated ageing. Dehydration during both the pre- and post-flowering stages resulted in reduced contents of protein, lipid and soluble carbohydrates (sucrose, glucose and fructose) in both species as compared to the control seeds. The species differed in the extent to which these reserves were reduced.

The study was aimed at determining potential differences in seed quality of spring and winter barley in relation to varieties (four winter and two spring barley varieties) and growing conditions (six locations, two years). Three tests were performed on each seed sample, i.e. (1) the standard germination test; (2) the low temperature and drought stress germination test and (3) the germination test in a salt solution. The effects of the factors of variety, provenance and year on seed vigour were estimated by the analysis of variance and Tukey’s post hoc test. The germination of winter varieties was influenced by provenance, both measures of vigour (2 and 3) by year, and significant differences between varieties were found in all three traits. Spring varieties differed only in germination and vigour (3) in one year when the influence of provenance prevailed. The results show different effects of variety and provenance on seed vigour. This could be promising for the breeding of high vigour barley varieties. Some preliminary results were obtained by using controlled deterioration (CD) as a vigour testing method for barley. The suitable conditions seem to be the combination of 45°C and 24% moisture content for the period of 24 hours.

An experiment was carried out at Germplasm Centre and Plant Biotechnology Lab, Department of Horticulture, PSTU to evaluate the growth, seed yield and quality of hybrid F1 chilli (cv. Sonic) crossing male line (LTSL-004-M) and female line (LTSL-004-F) along the viability of productive seeds influenced by NPK fertilizers during the period from November 2015 to October 2016. Ten different treatments of NPK fertilizers were used. Result showed that all the characteristics except primary branches and1000 seed weight were influenced significantly due to the application of NPK fertilizers. The tallest plant (69.88 cm), more leaves plant–1 (2491.33), highest number of secondary branches plant-1, tertiary branches plant-1 (5.00 and 88.00, respectively), number of flowers and hybrid fruits plant–1 (1533.00 and 12.33, respectively) and weight of seeds (4.03 g plant–1) along with required more time (54.67 days) for flowering were obtained from the application of 145 kg N ha–1, 175 kg P ha–1 and 96 kg K ha–1 (T4) which produced the highest final seed yield of chilli (4.03 kg ha–1). Application of T5 and T9 showed the highest number of primary branches (2.33). In seed viability characteristics T9 treated seeds showed the highest germination (100, 97.33 and 94.97%) at 1st, (1 MAH - Month After Harvest), 2nd (2 MAH) and 3rd (4 MAH) observations, respectively. The T3 treated seeds performed well in respect of Seed Vigour Index (SVI) at 1st and 2nd observations (8.00 and 7.48, respectively) but T9 treated seeds showed the highest SVI (6.32) at 3rd observation. Above indicating all characteristics were lowest under only recommended doses of NPK T1 as control treatment except days to first flowering. The observations suggested that production of hybrid chilli seeds could be enhanced by applying T4. While seeds of chilli treated by T9 in field level showed long time viable. Application of 145 kg N ha–1, 175 kg P ha–1 and 96 kg K ha–1 may be suggested for seed production of chilli and 110 kg N ha–1, + 175 kg P ha–1 + 115 kg K ha–1 for keeping the long time seed viability. The use of actinomycetes for improving soil fertility and plant production is an attractive strategy for developing sustainable agricultural systems due to their effectiveness, eco-friendliness, and low production cost. Out of 17 species isolated from the soil rhizosphere of legume crops, 4 bioactive isolates were selected and their impact on 5 legumes: soybean, kidney bean, chickpea, lentil, and pea were evaluated. According to the morphological and molecular identification, these isolates belong to the genus Streptomyces. Here, we showed that these isolates increased soil nutrients and organic matter content and improved soil microbial populations. At the plant level, soil enrichment with actinomycetes increased photosynthetic reactions and eventually increased legume yield. Actinomycetes also increased nitrogen availability in soil and legume tissue and seeds, which induced the activity of key nitrogen metabolizing enzymes, e.g., glutamine synthetase, glutamate synthase, and nitrate reductase. In addition to increased nitrogen-containing amino acids levels, we also report high sugar, organic acids, and fatty acids as well as antioxidant phenolics, mineral, and vitamins levels in actinomycete treated legume seeds, which in turn improved their seed quality. Overall, this study shed the light on the impact of actinomycetes on enhancing the quality and productivity of legume crops by boosting the bioactive primary and secondary metabolites. Moreover, our findings emphasize the positive role of actinomycetes in improving the soil by enriching its microbial population. Therefore, our data reinforce the usage of actinomycetes as biofertilizers to provide sustainable food production and achieve biosafety.

Selecting genotypes that combine high productivity with high seed quality is a challenge. The exploration of intracultivar genetic variation is an alternative to significantly increase the annual genetic gain and maximize the selection efficiency for both characteristics. The present study aimed to identify intracultivar variation to improve the seed quality of soybean genotypes derived from the commercial cultivar BRS 284, selected using the HSD (Honeycomb Selection Designs) model. Soybean genotypes selected for two years from two environments with contrasting edaphoclimatic characteristics, according to the principles of the HSD selection model, were cultivated under competition by using the experimental model in randomized blocks with four replicates and evaluated regarding the productivity and physiological quality of seeds. The results showed that genotype 284-3 presented a greater mass of 100 seeds, germination, vigour after accelerated ageing test, seedling emergence and emergence speed index than the other genotypes in both environments, with no significant difference in the standard cultivar regarding seed yield. The HSD method was an efficient selection method to identify intracultivar variation to improve cultivar performance.

Plant organs, such as seeds, are primary sources of food for both humans and animals. Seed size is one of the major agronomic traits that have been selected in crop plants during their domestication. Legume seeds are a major source of dietary proteins and oils. Here, we report a conserved role for the BIG SEEDS1 (BS1) gene in the control of seed size and weight in the model legume Medicago truncatula and the grain legume soybean (Glycine max). BS1 encodes a plant-specific transcription regulator and plays a key role in the control of the size of plant organs, including seeds, seed pods, and leaves, through a regulatory module that targets primary cell proliferation. Importantly, down-regulation of BS1 orthologs in soybean by an artificial microRNA significantly increased soybean seed size, weight, and amino acid content. Our results provide a strategy for the increase in yield and seed quality in legumes.
