Comparative Nephrotoxicity and Hepatotoxicity Effects of Kerosene, Gasoline, Liquefied Petroleum Gas and Biomass Fuel Exposure on Male Albino Rats

The effects of Kerosene, gasoline, and liquefied petroleum gas and biomass fuel exposure on biomarkers of kidney and liver were investigated in male wistar rats. Fifty adult male wistar rats were randomly assigned to five groups of ten animals each. Rats in group A served as control (exposed to fresh air). Group B, C, D and E were exposed to inhalation of kerosene, gasoline, liquefied petroleum gas and biomass fuel (wood smoke) respectively. All the exposures were done using whole body exposure chambers 70 cm x 60 cm x 60 cm measurement for six weeks, 6 days per week. Five millilitres of blood sample were collected and serum extracted at the end of six weeks. The serum concentration of urea, creatinine, uric acid and activities AST, ALT, γGT were determined using Cobas reagent kits manufactured by Roche Diagnostics GmbH, Sandhofer Strasse 116, D-68305 Mannheim, Germany. Values were analysed statistically using SPSS version 23.0.  The result shows significant increase in the serum  levels of urea, creatinine and uric acid of test groups relative to control (p<0.05), though the effect appear to be more pronounced with exposure to kerosene, gasoline and biomass fuel. The exposure also led to significant increase in activities of AST, ALT and γGT (p<0.05). These results suggest that repeated exposure to kerosene, gasoline and liquefied petroleum gas and biomass fumes may elicit hepatic and renal toxicity, thereby impairing the normal liver and kidney function.

Background: Trevo is a nutritional supplement with numerous bioactive natural products, with detoxifying and antioxidant properties. The purpose of this study was to investigate the ability of Trévo to protect against oxidative stress induced by lead in the kidneys of male Wistar rats. Methods: Thirty-five healthy male Wistar rats were divided into five groups of seven rats each, using a randomized design. I=control; II=15 mg/kg of lead acetate (PbA); III=2 ml/kg of trevo+PbA; IV=5 ml/kg of trevo+PbA; V=5 ml/kg of trevo. Animals were treated with trevo for five days before co-administration with lead intraperitoneally for 10 consecutive days. Animals were sacrificed 24hr after the last administration, blood samples were collected via cardiac puncture, and processed for assessment of urea, creatinine, and uric acid (UA), while the kidney samples were excised and processed for the following biochemical assays: Malondialdehyde (MDA), Glutathione-S-Transferase (GST), Catalase (CAT), Superoxide Dismutase (SOD), and Reduced Glutathione (GSH). Results: Injection of PbA caused a significant increase in the serum levels of urea, creatinine, and uric acid, and a significant increase (P<0.001) in the MDA concentration, and decreases in GSH concentration, CAT, SOD, and GST activities (P<0.05) as compared to the controls. Pretreatment with trevo prevented the oxidative stress induced by lead acetate in the kidney tissue samples and improve the renal function. The protective effect was evident at 5 ml/kg of trevo. Conclusion: The results showed that trevo was nephroprotective against lead toxicity and the activity might be linked to the presence of numerous antioxidant phytochemicals present in trevo.

In the present study, we examined gross changes in the mass of whole adrenal glands and that of the adrenal cortex and medulla in mature male Wistar rats subjected to three different stress types: acute, chronic, and combined, i.e., chronic followed by acute stress. These parameters were correlated with adrenal activity as judged from serum levels of corticosterone and catecholamine, respectively, as well as with serum levels of ACTH and glucose. Under all three conditions, we observed bilaterally asymmetric and stress-type-independent hypertrophy of whole adrenals, as well as adrenal cortices and medullas. Under acute and combined stress, adrenal hypertrophy was followed by increase of adrenal hormones in the blood serum. However, under chronic stress, both cortical and medullar activities as judged from low or unaltered levels of the respective hormones and glucose were compromised and disconnected from the input signal of ACTH. Since all of the studied adrenal activities could be restored by subsequent acute stress, it is concluded that chronic isolation can be viewed as partly maladaptive stress with characteristics resembling stress resistance rather than the stress exhaustion stage of the general adaptation syndrome.

SummaryIntroduction: The liver is involved in the metabolism of xenobiotics and their metabolites and it is vulnerable to oxidative damage. Hyperglycaemia is highly implicated in the progression of diabetes mellitus, and adversely affects the liver. Though, conventional hypoglycaemic drugs may be effective in reducing blood glucose, they do not appear to be effective in attenuating the progression of diabetes and its complications.Objective: This study evaluated the ameliorative effects of Anchomanes difformis on hyperglycaemia and hepatic injuries in type 2 diabetes.Methods: Type 2 diabetes was induced in male Wistar rats with a single intraperitoneal injection of streptozotocin (40 mg/kg BW) after two weeks of fructose (10%) administration. Aqueous extract of A. difformis (200 and 400 mg/kg BW) and glibenclamide (5 mg/kg BW) were administered orally for six weeks. Blood glucose concentrations were measured. Serum levels of liver dysfunction markers (ALT, AST, and ALP), total cholesterol, triglycerides, HDL- and LDL-cholesterol were investigated. Total protein, albumin, and globulin were also assessed. Antioxidant parameters: ORAC, GSH, GSSG, SOD, CAT and FRAP were evaluated in the liver while ORAC, FRAP and lipid peroxidation were determined in the serum. Histological examination of the liver tissue was carried out.Results: Treatment with aqueous extract of A. difformis significantly (p<0.05) reduced blood glucose and reversed steatosis in the diabetic-treated rats. The antioxidant status of diabetic-treated rats was significantly (p<0.05) improved. Serum levels of liver dysfunction markers were significantly (p<0.05) reduced in diabetic-treated rats.Conclusion: The findings in this study revealed that 400 mg/kgBW Anchomanes difformis was more effective than 200 mg/kg BW in ameliorating diabetes-induced hepatopathy, however, both doses of Anchomanes difformis demonstrated more antidiabetic ability than glibenclamide. Anchomanes difformis may be a novel and potential therapeutic agent in the management of diabetes and resulted hepatic injuries.

1. The effects of dietary polychlorinated biphenyls (PCB) and excess tyrosine on serum and liver lipids, urinary ascorbic acid and catecholamines were compared in male Wistar rats.2. Serum levels of cholesterol, urinary ascorbic acid, norepinephrine, epinephrine, dopamine and histamine were significantly increased in rats given either PCB or excess tyrosine.3. The hypercholesterolaemia induced by PCB or excess tyrosine was blocked by the adrenergic α-blocker, phenoxybenzamine.4. The present results suggest causal interrelations between the hypercholesterolaemia induced by dietary PCB or excess tyrosine and the secretion of catecholamines.

The present study investigated the anti-atherosclerotic and antioxidative effect of the methanolic extracts (MExt) of Cochorous olitorous (CO) and Adansonia digitata (AD) leaves on irradiation-induced atherosclerosis in male Wistar rats. Atherosclerosis was induced in male rats by a single dose of 6 gray whole body gamma radiation. MExt of C. olitorous and A. digitata leaves at 500 and 1,000 mg/kg bwt were administered as treatment for 7 days. Blood serum was analysed for lipid profile, MDA (malondialdehyde) and liver tissue for antioxidants enzymes, whereas the therapeutic potential was compared to the lipids-lowering drug lovastatin at 10 mg/kg/bwt. The phytochemical studies showed the presence of polyphenols, flavonoids, tannins and saponins. Treatment with MExt of CO and AD normalized the elevated MDA level, whereas the activities of superoxide dismutase, catalase and glutathione peroxidase in the treated rats increased. Pronounced changes were observed at 1,000 mg/kg bwt mixture of MExt of CO and AD for 1 weeks and it was more potent than the standard drug. The current study provided strong evidence that MExt of CO and AD might be important in the treatment of atherosclerosis and ROS without any side effects at the studied dosage and duration.

ABSTRACT: Flaxseed is considered a functional food with several health benefits. However, because of its high phytoestrogen content, flaxseed influences hormone metabolism and affects the gonadal biomorphology. In this study, computerized histomorphometry was used to evaluate seminiferous and epididymal tubules, considering the different regions of the epididymis (head, body and tail) of rats subjected to a prolonged diet of flaxseed. Young adult male Wistar rats (n=20) were divided into 2 groups during their lactation period: Control Group (CG), fed casein-based meals and Flaxseed Group (FG), fed a 25% flaxseed meal. After 250 days of continuous ingestion, the animals were euthanized and a blood sample was collected. The testicles and epididymis were removed and fixed in buffered formalin solution. The samples were subjected to routine histological paraffin techniques and stained with hematoxilin and eosin. Immunostaining was performed using an antivimentin antibody for Sertoli cell identification. For morphometry, images of the slides were scanned and analyzed using Image J to determine the epithelial height, tubular and luminal diameter and tubular and luminal area. In the hormonal evaluation, FG had a higher serum concentration of estrogen (P=0.001), but no change was observed in the concentration of testosterone. The morphometric assay of seminiferous tubules and epididymal regions revealed no significant differences between the analyzed groups. Similarly, Sertoli cell quantification showed no significant differences in the FG (P=0.98). These results revealed that the continuous and prolonged intake of 25% flaxseed meals from gestation to 250 days of age, even with a significant increase in serum levels of estradiol, does not exert adverse effects on the testicular and epididymal structure or on the cells participating in the spermatogenesis of rats.

Safety evaluation of Waltheria indica Linn. root was carried out in Wistar rats. Twenty adult male Wistar rats were randomly divided into four groups (n=5); A–D. Group A served as control group while groups B, C and D were administered with 200, 400 and 800 mg/Kg of crude ethanol extract of Waltheria indica Linn. root. After 28 days of administration, the rats were sacrificed and blood samples were collected and serum prepared. Haematological and biochemical parameters, as well as serum electrolyte were determined according to standard procedures. The 200 mg/Kg of Waltheria indica Linn. root caused the highest percentage increase in body weight. The kidney was also significantly increased at 400 mg/kg. The total WBC and lymphocytes counts were significantly increased at 400 mg/kg. The serum levels of ALT and chloride were significantly increased at 800 mg/kg. Waltheria indica Linn. root also caused hepatic necrosis and renal tubular degeneration in Wistar rats. Therefore, Waltheria indica Linn. root should be used with caution especially at higher dose despite its therapeutic potentials.
