The leaves of Lippia multiflora contain secondary metabolites including flavonoids which have an important antioxidant activity. This study aims to optimize the extraction conditions of total flavonoids and the antioxidant activity of these leaves. To achieve this, the Plackett-Burman design was used for the screening of the factors influencing the extraction, then the central composite design was implemented for the optimization itself. The effects of five factors, such as the plant-to-solvent ratio, the nature of the extraction solvent, the extraction time, the extraction method and the size of the L. multiflora leaves, on the extraction of total flavonoids and the antioxidant activity were studied. Results of Plackett-Burman design indicated that factors influencing both flavonoids extraction and antioxidant activity were the solid-liquid ratio and extraction time. The predicted optimal conditions for the highest flavonoids content from  L. multiflora leaf with better antioxidant activity were found with aqueous decoction for 30 min with 3.5 g of cut leaves in 100 mL of distilled water. Using the predicted conditions, experimental responses were 87.18 ± 1.03 mg/g QE and 372.34 ± 4.04 µmol/g TE for total flavonoids and antioxidant capacity, respectively. Under the above-mentioned conditions, the experimental results are very close to predicted one. Thus, L. multiflora leaf can be considered as a natural source of flavonoids content with good antioxidant activity.

The increase in durian production results in the accumulation of durian peel waste. The bioactive component of durian peel has the potential to be used as an antioxidant. Thus, there is a need to carry out an extraction process to obtain  bioactive compounds from durian peel. However, conventional extraction methods cause damage to phenolic compounds due to oxidation, hydrolysis and ionization reactions during the extraction process. Therefore, durian peel extraction was carried out using ultrasonic assisted extraction method (UAE) in this study. The purpose of this study was to investigate the effect of varying ratios of durian peel to ethanol solvents and extraction time on the yield, total phenolics content, total flavonoids content and antioxidant activity. The results of the research data were analyzed using a two-factor completely randomized design, which included variations of the ratio of durian peel to ethanol solvents and extraction time The Duncan’s New Multiple Range Test (DNMRT) was carried out as a follow up test to determine the differences in each treatment at a significance level of 0.05. The results showed that the best treatment for peel extraction using ultrasonic assisted extraction was a 1: 9 ratio of durian peel to ethanol at an extraction time of 20 minutes. The extraction of durian peel under this condition gave the highest yield of 12.77 ± 0.16%, antioxidant activity (IC50) of 38.33 ± 0.12 ppm, total phenolic content of 63.30 ± 0.08 mgGAE / g and total flavonoids content of 47.53 ± 0.48 mgQE / g. In addition, total phenolics content and total flavonoid content showed a strong correlation to the antioxidant activity of durian peel extract.

The effect of homogenization, ultrasound and microwave extraction methods and conditions on fucoxanthin content, total phenolic content and antioxidant activity of extracts obtained from Phaeodactylum tricornutum were investigated in this study. The solvent/biomass ratio was the most effective parameter on fucoxanthin content, total phenolic content and antioxidant activity. The maximum fucoxanthin content (5.60 ± 0.06 mg/g) and antioxidant activity (763.00 ± 15.88 EC50 μg/mL extract) were obtained with the homogenization extraction method whose optimum conditions were 1.93% biomass/solvent ratio, ~5200 rpm homogenization rate and 14.2 min extraction time. Although the ultrasonic extraction method has reached the approximately same level of fucoxanthin content (5.24 ± 0.07 mg/g)), TPC (67.68 ± 1.58 mg gallic acid/L) and antioxidant activity (619.90 ± 17.16 EC50 μg/mL extract) at an amplitude of 55.72%, a higher biomass/solvent ratio (2.72%) and a longer extraction time (17.37 min) have been required. The lowest fucoxanthin content, total phenolic content and antioxidant activity were determined for the microwave extraction method.

The adsorption efficiency of As (V) ions from an aqueous solution was investigated. The adsorption experiments were carried out in a batch reactor. The effects of operating parameters; i.e. the absorbent dose (g/L), the pH, the adsorption temperature (°C), the As (V) initial concentration (ppb), and type of adsorbents, on the adsorption efficiencies were studied. A PlackettBurman experimental design was used to screen for the important factors that influence As (V) adsorption. It has been found that the most important effect on the As (V) adsorption capacity was the type of adsorbent. The factor importance could be written in descending order as follows: the type of adsorbents, the adsorbent dose, the As (V) initial concentration, the adsorption temperature, and the pH.

Objective: To enhance the antioxidant potential of Xylaria feejeensis by statistical optimization. The components of potato dextrose yeast extract (PDYEB) medium: potato infusion, dextrose and yeast extract were investigated to optimize antioxidant activity by Plackett-Burman design (P-B design). The interaction between the critical components of the medium and incubation time was further investigated by Response Surface Method (RSM) and the culture conditions were optimized for enhancing antioxidant activity. Central composite design (CCD) was employed and quadratic response surface model was used to analyze the data using analysis of variance (ANOVA).Methods: The antioxidant potential was measured by 1-1diphenyl-2-picryl-hydrazil (DPPH) radical scavenging assay, nitric oxide (NO) scavenging assay. The antioxidant activity was compared with total phenolic content measured by Folin-Ciocalteau (FC) reagent based assay.Results: Plackett-Burman design revealed dextrose and yeast extract to be the most significant components of PDYEB medium (p<.0001). ANOVA analysis showed that the model was highly significant (p<0.0001) for antioxidant activity measured by DPPH, NO scavenging and TPC (total phenolic content). The model was justified by applying the optimized conditions and values of 87.38%, 56.43%, 27.28 mg/g for DPPH, NO scavenging and TPC respectively were obtained. The estimated optimum conditions of the variables for the antioxidant activity and phenolic content are 12.72g of dextrose, 1.67g yeast extract, and incubation period of 23.43 d.Conclusion: The study highlights the importance of various components of the medium and the physiochemical limitations in antioxidant potential and phenol production of the fungal strain.

Ultrasonic-assisted extraction (UAE) was applied to extract total carotenoids from dried peach palm peel using soy oil as extraction solvent. Optimization of UAE parameters of total carotenoid extraction from dried peel was done using Response Surface Methodology (RSM) and Central Composite Design (CCD). The extraction parameters were temperature, extraction time and solid to solvent ratio. The optimum UAE conditions were 48°C, extraction time of 28 min, and solid to solvent ratio of 0.0037 g/mL, with a predicted carotenoids content of 151.50 mg/100 g dried peel. To validate the optimized model, the experimental values were compared with the predicted values to check the adequacy of the model. Ultrasound extraction was 33.60% higher than maceration technique. This study indicated that UAE should contribute to a green process for valorization of peach palm by-products.

The extraction processing of lycopene from watermelon was investigated in this paper. Based on single factor experiments, response surface methodology (RSM) was adopted to study the key parameters such as number of extraction, extraction temperature, extraction time in order to increase extraction rate of lycopene. A second order quadratic equation was established and the applicability of model and interaction involved factors on predicting the lycopene extracting content was verified. The results indicated that the optimum extraction conditions were hexane (containing 2% dichloromethane) as extraction solvent, ratio of solvent to raw material of 3:1 (mL/g), number of extraction of 2, extraction time of 1.9 h and extraction temperature of 29.8°C. Under these conditions the extracting content of lycopene was 14.71±0.22 mg/kg, which is well in agreement with value predicted by the model.

In the last decade by-products of food and beverage processing have attracted much attention due to their functionality and potential as food ingredients. Brewers’ spent grain is the major by-product of the brewing industry representing a valuable source of bioactive ingredients. The aim of this study was to assess the effect of extraction time and temperature on the efficiency of water as solvent for the extraction of bioactive compounds from brewers’ spent grain (BSG). In terms of extraction efficiency, the results from polyphenols, flavonoids and antioxidant activity, showed that the best extraction parameters for aqueous extracts are 90⁰C and 60 minutes. In comparison with the control, the best extraction method generated 87% of the phenolics and 43.46% of the flavonoids obtained by a methanol extraction. The preliminary results for the aqueous extracts showed that water can be used as extraction solvent, but a higher extraction time and temperature are needed in order to have a content in bioactive compounds similar to that of methanolic extracts. The obtained values for polyphenols, flavonoids and antioxidant activity, emphasize the importance and the opportunities of the reuse of this agro-industrial waste.

This research was conducted to determine temperature and time extraction with ultrasonic waves on the antioxidant activity of guajava leaf extract and obtain the temperature and extraction time with ultrasonic waves that are appropriate to produce the highest antioxidant activity. The experimental design used in this research was a Factorial Completely Randomized Design with time treatment consisting of 3 levels, namely 10 minutes, 20 minutes and 30 minutes and temperatures consisting of 3 levels, namely 40°C, 45°C and 50°C. The treatment was repeated two times, so that 18 experimental units were obtained. Data were conducted using analyzed of variance and if treatment had a significant effect, followed by Duncan's test. The results showed that the interaction between temperature and time of extraction of guajava leaves with ultrasonic waves had a very significant effect (P<0.01) on yield, total phenol, total flavonoids, total tannins and antioxidant activity of guajava leaves. The treatment of extraction temperature and extraction time of guajava leaves with ultrasonic waves had very significant effect (P<0.01) on yield, total phenol, total flavonoids, total tannins and antioxidant activity of guajava leaves. The best results showed that temperature of 45oC with time 20 minutes result antioxidant activity based on 89.03 %, free radical inhibition percentage with IC50 values 3.55 mg/L, yield 16.26 %, total phenol was 331.77 mgGAE/g extract, total flavonoids was 637.33 mgQE/g extract and total tannins were 583.75 mgTAE/g.
