Cerebral organoids are three-dimensional cell-culture systems that represent a unique experimental model reconstructing early events of human neurogenesis in vitro in health and various pathologies. The most commonly used approach to studying the morphological parameters of organoids is immunohistochemical analysis; therefore, the three-dimensional cytoarchitecture of organoids, such as neural networks or asymmetric internal organization, is difficult to reconstruct using routine approaches. Immunohistochemical analysis of biological objects is a universal method in biological research. One of the key stages of this method is the production of cryo- or paraffin serial sections of samples, which is a very laborious and time-consuming process. In addition, slices represent only a tiny part of the object under study; three-dimensional reconstruction from the obtained serial images is an extremely complex process and often requires expensive special programs for image processing. Unfortunately, staining and microscopic examination of samples are difficult due to their low permeability and a high level of autofluorescence. Tissue cleaning technologies combined with Light-Sheet microscopy allows these challenges to be overcome. CLARITY is one of the tissue preparation techniques that makes it possible to obtain opaque biological objects transparent while maintaining the integrity of their internal structures. This method is based on a special sample preparation, during which lipids are removed from cells and replaced with hydrogel compounds such as acrylamide, while proteins and nucleic acids remain intact. CLARITY provides researchers with a unique opportunity to study three-dimensional biological structures while preserving their internal organization, including whole animals or embryos, individual organs and artificially grown organoids, in particular cerebral organoids. This protocol summarizes an optimization of CLARITY conditions for human brain organoids and the preparation of Light-Sheet microscopy samples.

This article deals with the industrial heritage of the 20th century in Catanduva-SP, having as object of analysis the Railway Villages of the old company Estrada de Ferro Araraquara (EFA), located in the São Francisco and Higienópolis neighborhoods. The justification of the theme is supported by the lack of research on the railway village of Catanduva, the lack of knowledge of the complex and the lack of recognition of its value as cultural heritage. In this context, the work aims to document and inventory railroad villages in the São Francisco and Higienópolis de Catanduva neighborhoods, and to propose guidelines for their safeguarding. The working methodologies used were Historical Research and Case Study, based on a documentary survey on the object of analysis and on-site visit. As a result, 13 bungalow-style residences were identified, most of them in good condition. As safeguard guidelines, the conservation, preservation, restoration and rehabilitation of these residences were proposed so that they maintain their physical structures and the internal organization of the rooms, meet the needs and expectations of users and preserve their cultural significance.

This paper is concerned with the study of the modifications of consonants caused by their mutual influence, taking into account the varying degrees of connectivity of the elements of the speech chain. As such, this study of vocal connections of various types is carried out from the point of view of the nature of the junction of the elements – more or less close. Thus, this paper is devoted to problems concerning the phenomenon of vocal junction as it is established in linguistics in general and in phonetics in particular. The relevance of these problems stems from the need for disclosure and knowledge of the mechanism of internal organization of speech flow. This paper is aimed at analyzing the features of the vocal stitch and its components, which allow you to delve deeper into the mechanism of production of related speech. Thereby, this study attempts to identify modifications of elements of interdisciplinary vocal combinations, which are important for the implementation of their delimitative function, as these are manifested in the distinction of different units of speech.

Along with the development of information and communications technology being popular, types of data become rich and multiple, the analysis scope changes from structured data to non-structured data without sorting context, and the data volume becomes huge and is continuously growing. When data application cases and value benefit are gradually noticed in past years, government agencies realize that data could develop the value through cross-boundary collaboration, rather than simple relying on internal processing and analyses, and the collaboration process allows the government cultivating to apply data with added-value and establish evidence-based governance. Aiming at employees in public sectors in the central government of Taiwan, total 320 copies of questionnaire are distributed and 247 valid copies are retrieved, with the retrieval rate 77% . The research results are summarized as below: (1) Regarding the curiosity and expectation of data analysis of the government, either directors of agencies or key case officers, with the expectation of applying the possessed data with added value, do not simply regard data as dead records, but attempt to apply data to solve specific public issues; (2) In addition to inducing the curiosity about data application in the internal organization, success cases of other agencies could facilitate the action of an organization participating in the project for expanding to central and local levels or cross-units inducing the agencies with similar businesses engaging in the project as well as accelerate project influence through experience sharing and reinforce the confidence of other units in information and data governance; (3) Information and data governance could benefit the government shaping positive image to interpret outcomes through data for the reference of future policies, strengthen the industrial and academic research energy of the business, as well as enhance public trust and agency transparency through cooperation with experts. According to the results to propose suggestions, it is expected to provide related policy suggestions for the government examining the internal organization from management to execution about the preparation for data governance and the reference for top decision-makers planning data application strategies, applying data to public-private collaboration, and improving existing data governance structure.

Abstract This study concentrates on the alternation between two dorsal fricatives: [x] and [ç] in Modern Standard German. The primary source of data for the analysis include both native German words and loanwords. Moreover, the discussion encompasses some strictly related processes such as g-spirantization [g] > [x]/[ç], e.g. Ber[ç] ‘mountain’ and Ta[x] ‘day’, and coronalization [ç] > [ʃ], e.g. mi[ʃ] ‘me’ found in various German dialects and colloquial variants. Finally, since it is responsible for the appearance of both alternants in an identical context, e.g. Frau[ç]en ‘woman, dim’ and rau[x]en ‘to smoke’, a brief look is taken at the historical development of the diminutive suffix chen. It is argued here that in contemporary German there are two palatovelar fricatives [ç] which differ in their internal organization of the melodic content. In loanwords and in the diminutive suffix -chen, [ç] is lexically specified for palatality, while after front vowels and coronal sonorants the same fricative shares the palatality element with the preceding segment. The analysis is couched in a recent version of Element Theory and it proceeds on two assumptions: a) front vowels and coronal sonorants [l ʁ n] in German are defined by the resonance element |I| and b) [ç] is a complex segment containing two resonants |I| and |U|.

Aesthetic emotions are elicited by different sensory impressions generated by music, visual arts, literature, theater, film, or nature scenes. Recently, the AESTHEMOS scale has been developed to facilitate the empirical assessment of such emotions. In this article we report a semantic profile analysis of aesthetic emotion terms that had been used for the development of this scale, using the GRID approach. This method consists of obtaining ratings of emotion terms on a set of meaning facets (features) which represent five components of the emotion process (appraisal, bodily reactions, action tendencies, expression, and feelings). The aims here were (a) to determine the dimensionality of the GRID features when applied to aesthetic emotions and compare it to published results for emotion terms in general, and (b) to examine the internal organization of the domain of aesthetic emotion terms in order to identify salient clusters of these items based on the similarity of their feature profiles on the GRID. Exploratory Principal Component Analyses suggest a four-dimensional structure of the semantic space consisting of valence, power, arousal, and novelty, converging with earlier GRID studies on large sets of standard emotion terms. Using cluster analyses, 15 clusters of aesthetic emotion terms with similar GRID feature profiles were identified, revealing the internal organization of the aesthetic emotion terms domain and meaningful subgroups of aesthetic emotions. While replication for further languages is required, these findings provide a solid basis for further research and methodological development in the realm of aesthetic emotions.
