ABSTRACTOrganelle heterogeneity and inter-organelle associations within a single cell contribute to the limited sensitivity of current organelle separation techniques, thus hindering organelle subpopulation characterization. Here we use direct current insulator-based dielectrophoresis (DC-iDEP) as an unbiased separation method and demonstrate its capability by identifying distinct distribution patterns of insulin vesicles from pancreatic β-cells. A multiple voltage DC-iDEP strategy with increased range and sensitivity has been applied, and a differentiation factor (ratio of electrokinetic to dielectrophoretic mobility) has been used to characterize features of insulin vesicle distribution patterns. We observed a significant difference in the distribution pattern of insulin vesicles isolated from glucose-stimulated cells relative to unstimulated cells, in accordance with functional maturation of vesicles upon glucose stimulation, and interpret this to be indicative of high-resolution separation of vesicle subpopulation. DC-iDEP provides a path for future characterization of subtle biochemical differences of organelle subpopulations within any biological system.

One of the most common soft soil enhancement techniques used to expedite the consolidation time significantly is Prefabricated Vertical Drains (PVD). This technique needs a sufficient discharge capacity value because it primarily functions as a drainage channel. The deformation of PVD is considered as one of the primary factors which affect discharge capacity. Therefore, this research determined the influence of upper-side deformation on PVD's discharge capacity (qw) using a specific design apparatus known as ASTM D4716, which manages the determination of transmissivity and flow rate at the longitudinal direction of geosynthetics. Furthermore, two PVD samples with dimensions of 3 and 4 mm thickness, 100 mm width, and 1000 mm length were examined under straight and buckled conditions. Stepwise confining pressures from 50 to 200 kPa were subjected to the samples under hydraulic gradients with values of 0.2, 0.5, and 1.0. The results showed that samples with greater thickness had higher discharge capacity, which significantly reduced in the lower hydraulic gradient. The deformation on the upper side of PVD induced a decrease of discharge capacity by approximately 13-16%, which led to a delay in the consolidation time. The discharge capacity values obtained from the experiments were employed as parameters in a time factor ratio of Th,w/Th. The analysis results show that the buckled PVD has a more considerable consolidation time due to the increase in the Th,w/Th ratio, with a discharge capacity value below 10-4 m3/s. It can be concluded that the deformation in the form of buckled conditions on the upper side of PVD had a considerable impact on PVD effectiveness.

AbstractSuper-hydrophobic textured surfaces reduce hydrodynamic drag in pressure-driven laminar flows in micro-channels. However, despite the wide usage of non-Newtonian liquids in microfluidic devices, the flow behaviour of such liquids was rarely examined so far in the context of friction reduction in textured super-hydrophobic micro-channels. Thus, we have investigated the influence of topologically different rough surfaces on friction reduction of shear-thinning liquids in micro-channels. First, the friction factor ratio (a ratio of friction factor on a textured surface to a plain surface) on generic surface textures, such as posts, holes, longitudinal and transverse ribs, was estimated numerically over a range of Carreau number as a function of microchannel constriction ratio, gas fraction and power-law exponent. Resembling the flow behaviour of Newtonian liquids, the longitudinal ribs and posts have exhibited significantly less flow friction than the transverse ribs and holes while the friction factor ratios of all textures has exhibited non-monotonic variation with the Carreau number. While the minima of the friction factor ratio were noticed at a constant Carreau number irrespective of the microchannel constriction ratio, the minima have shifted to a higher Carreau number with an increase in the power-law index and gas fraction. Experiments were also conducted with aqueous Xanthan Gum liquids in micro-channels. The flow enhancement (the flow rate with super-hydrophobic textures with respect to a smooth surface) exhibited a non-monotonic behaviour and attenuated with an increase in power-law index tantamount to simulations. The results will serve as a guide to design frictionless micro-channels when employing non-Newtonian liquids.

Abstract Matrix cooling has opened new possibilities for enhancing the convective heat transfer coefficients without compromising upon the structural rigidity and the life of the gas turbine blade at elevated temperatures. However, the dense structure of the matrix significantly increases the flow resistance, and imposes the limitation to its usage. Recently, a matrix with a gap on the sidewalls called open matrix has been proposed by few researchers to reduce the associated pressure penalties. This detailed experimental investigation aims to study the open matrix channel flow, and presents the effects of varying sidewall gaps on heat transfer characteristics and friction factor in the open matrixes having rib angle 45o for three different sub-channel aspect ratios 1.2, 0.8, and 0.4. Liquid crystal thermography has been utilized to discern the detailed heat transfer characteristics. Results have been evaluated in terms of augmentation Nusselt number, friction factor ratio, and overall thermal performance factor over the Reynolds numbers 5800 -14000. The closed matrixes provided the highest augmentation in Nusselt number, and the gaps on the sidewall have shown an overall reduction in augmentation Nusselt number in most cases. However, the suitable sidewall gap showed the effective reduction in pressure penalties for the smaller sub-channel aspect ratios. The highest augmentation Nusselt number amongst the open matrixes has been found as 3.83 with a reduced friction factor ratio for the matrix with a 4 mm gap in sub-channel aspect ratio = 0.8 (i.e. 4 sub-channels) at Re = 8100.
