The spleen is a separate organ of the teleost, playing an essential role in immune reactions. The morphology of the spleen is different from the fish species. Little knowledge about the spleen structure and the blood splenic barrier (BSB) in Nile tilapia has been reported. To address this issue, we studied the histology of the spleen and the BSB in healthy Nile tilapia. The morphology of the spleen was observed, then H&E staining, modified Jame’s staining, and ultrastructural techniques were performed to portion the spleen into three subregions and analyze the location of components and fibers. Thereafter, vital staining of Nile tilapia with Trypan blue was conducted to elucidate the composition and function of BSB. Histologically, the spleen could be divided into three subregions (inner, middle, and outer). The venules, clumps of lymphocytes, and vessels were separately characterized features of the outer, middle, and inner layers. Post injection, Trypan blue was intercepted in the endotheliocytes of ellipsoids in the middle layer (i.p.) or was deposited to the reticular fibers surrounding the ellipsoids (i.v.). Additionally, the amount of Trypan blue was shown to be positively correlated to that of the Acid phosphatase expressed. In conclusion, the spleen could be portioned into three subregions, and the BSB lay in the middle layer, composed of the cuboidal-shaped endotheliocytes and the surrounding reticular fibers of the ellipsoid capillaries. The present study enriched the research of immune tissues and system in tilapia and provided reference for the study of spleen in other fish species.

The emergence of a novel goose nephritic astrovirus (GNAstV) has caused economic losses to the Chinese goose industry. High viral load is found in the spleen of goslings infected with GNAstV, but pathological injuries to the spleen due to GNAstV are largely unknown. In this study, 50 two-day-old goslings were infected orally with GNAstV, and 50 goslings were treated with PBS as control. Spleens were collected at different times following infection to assess damage. GNAstV infection caused visceral gout and urate deposition in joints, and resulted in 16% mortality. GNAstV was found in the lymphocytes and macrophages within the spleen. Lymphocyte loss, especially around the white pulp, and destruction and decline in the number of reticular fibers was observed in GNAstV-infected goslings. Moreover, in GNAstV-infected goslings, ultrahistopathological examination found that splenic lymphocytes exhibited condensed chromatin and apoptotic bodies, and reticular cells displayed damage to plasma membrane integrity and swollen mitochondria. Furthermore, TUNEL staining confirmed apoptosis of lymphocytes, and the mRNA levels of Fas and FasL were significantly increased in the GNAstV-infected goslings. In addition, GNAstV infection reduced the number and protein expression of CD8. In conclusion, GNAstV infection causes lymphocyte depletion, reticular cell necrosis, reticular fiber destruction, lymphocyte apoptosis, and reduction in CD8 levels, which contribute to spleen injury.

Tenascin-C plays important roles in immunity. Toll-like receptor 4, integrin α9β1 and chemokines have already been identified as key players in executing the immune regulatory functions of tenascin-C. Tenascin-C is also found in reticular fibers in lymphoid tissues, which are major sites involved in the regulation of adaptive immunity. Did the “tool box” for reading and interpreting the immune-regulating instructions imposed by tenascins and tenascin-C co-evolve? Though the extracellular matrix is ancient, tenascins evolved relatively recently. Tenascin-like genes are first encountered in cephalochordates and urochordates, which are widely accepted as the early branching chordate lineages. Vertebrates lacking jaws like the lamprey have tenascins, but a tenascin gene that clusters in the tenascin-C clade first appears in cartilaginous fish. Adaptive immunity apparently evolved independently in jawless and jawed vertebrates, with the former using variable lymphocyte receptors for antigen recognition, and the latter using immunoglobulins. Thus, while tenascins predate the appearance of adaptive immunity, the first tenascin-C appears to have evolved in the first organisms with immunoglobulin-based adaptive immunity. While a C-X-C chemokine is present in the lamprey, C-C chemokines also appear in the first organisms with immunoglobulin-based adaptive immunity, as does the major histocompatibility complex, T-cell receptors, Toll-like receptor 4 and integrin α9β1. Given the importance of tenascin-C in inflammatory events, the co-evolution of tenascin-C and key elements of adaptive and innate immunity is suggestive of a fundamental role for this extracellular matrix glycoprotein in the immune response of jawed vertebrates.

Background: Bear specialist group recommended that the basic research on the Malayan sun bear is the highest priority need. Without such information, the establishment and implementation of scientifically-sound conservation plans is difficult. Therefore, present study was designed to provide information on gross morphological and light microscopic architecture of the spleen.Methods: The present study was conducted on the spleen of one Malayan sun bear. After doing the gross parameters the tissues were fixed in 10% neutral buffered formalin and were processed for light microscopic studies. Blocks were cut at 6μ thickness by Leica Semimotorized Rotary Microtome and stained by Harris’ haematoxylin and eosin for routine study. Result: The spleen of Malayan sun bear was located in the left hypogastric region and entirely intrathoracic as the stomach was almost empty. The parietal surface faces the diaphragm and left lateral abdominal wall, whereas the visceral surface was divided into gastric face and intestinal face by the ridge like hilus. The spleen of Malayan Sun Bear was surrounded by a thick connective tissue capsule invested by the peritoneum. The capsule, trabeculae and reticular fibers support the splenic parenchyma composed of a red pulp and a white pulp.

Background: Chabro is a strain of poultry birds especially designed for backyard farming and is more adoptive to climatic variations in the tropics. The thyroid gland plays an important role in controlling basal metabolic rate. The histoarchitectural changes of the gland in association with seasonal changes has not been studied so far in Chabro. The present study describes the season related variations in the histometry of thyroid gland.Methods: Micrometrical studies were conducted on thyroid gland of eight to ten weeks old 24 apparently healthy Chabro chickens procured from Poultry Farm, DUVASU, Mathura after approval of CPCSEA. For this study the chickens were divided into two groups consist of 12 chickens in each group reared in summer and winter seasons.Result: Histologically, the thyroid gland was composed of stroma and parenchyma. The capsule had outer thick adipose and inner thin fibrous layers. The follicles were filled with colloid produced by the follicular cells. The percentage of small follicles was more followed by medium and large follicles. The follicles were lined by simple squamous epithelium in summer and cuboidal epithelial cells in winter. All micrometrical parameters, amount of reticular fibers, percentage of large and active follicles were higher in winter.

Trehalose is a disaccharide molecule consisting of two molecules of glucose. Industrially, trehalose is derived from corn starch and utilized as a drug. This study aims to examine whether the integration of nanoparticle-encapsulated trehalose to the Ice-Free Cryopreservation (IFC) method for preserving heart valves has better cell viability, benefits to protect the extracellular matrix (ECM), and reduce immune response after storage. For the experiment to be carried out, we obtained materials, and the procedures were carried out in the following manner. The initial step was the preparation of hydroxyapatite nanoparticles, followed by precipitation to acquire Apatite colloidal suspensions. Animals were obtained, and their tissue isolation and grouping were done ethically. All samples were then divided into four groups, Control group, Conventional Frozen Cryopreservation (CFC) group, IFC group, and IFC + T (IFC with the addition of 0.2 M nanoparticle-encapsulated Trehalose) group. Histological analysis was carried out via H&E staining, ECM components were stained with Modified Weigert staining, and the Gomori Ammonia method was used to stain reticular fibers. Alamar Blue assay was utilized to assess cell viability. Hemocompatibility was evaluated, and samples were processed for immunohistochemistry (TNFα and IL-10). Hemocompatibility was quantified using Terminal Complement Complex (TCC) and Neutrophil elastase (NE) as an indicator. The results of the H&E staining revealed less formation of extracellular ice crystals and intracellular vacuoles in the IFC + T group compared with all other groups. The CFC group’s cell viability showed better viability than the IFC group, but the highest viability was exhibited in the IFC + T group (70.96 ± 2.53, P < 0.0001, n = 6). In immunohistochemistry, TNFα levels were lowest in both IFC and IFC + T group, and IL-10 expression had significantly reduced in IFC and IFC + T group. The results suggested that the nanoparticle encapsulated trehalose did not show significant hemocompatibility issues on the cryopreserved heart valves.

Abstract Introduction The aim of the present study was to determine the morphological features of the pineal gland in three closely related Canidae species (raccoon dog, Nyctereutes procyonoides Gray, 1834; silver fox, Vulpes vulpes L., 1758; and blue fox, Vulpes lagopus L., 1758) of different ages during the breeding (spring) and nonbreeding (winter) periods. Materials and Methods Histological analysis of the pineal glands of canids was performed. Results The morphological changes in the pineal gland detected in the current study are either age-associated, including increase in the reticular fibers and vascularization in the studied species, as well as increase in the amount of the protruding septae in the blue fox, or seasonally related, including an increase in the number and size of blood vessels. The present work reported two types of pigments: lipofuscin (primarily in the silver fox) and melanin (primarily in the raccoon dog and in the blue fox). The pineal gland in the blue fox is characterized by the ability to form corpora arenacea. Conclusions The present study provides the first insight into the morphological changes of the pineal gland in three closely related Canidae species of different ages during the breeding (spring) and nonbreeding (winter) periods, and showed some species-specific features of gland morphology. The aspects concerning the biogenesis of the calcium concretions and the factors influencing the accumulation of pigments need further investigation.

ABSTRACT We describe and compare the histology of liver and spleen ofGeophagus brasiliensis (Perciformes), Hypostomus francisci (Siluriformes) and Hoplias aff. malabaricus (Characiformes), tropical freshwater fishes. InG. brasiliensisandH. aff. malabaricusthe hepatocytes were arranged in tubular form whereas in H. franciscithey cord-like. In all species, hepatocytes presented glycogen, but in G. brasiliensis and H. aff. malabaricus they showed strong stained for hemossiderin in the cytoplasm. InG. brasiliensis and H. aff. malabaricus, melanomacrophage centres (MMCs) were associated to hepatic structures and only in G. brasiliensis was observed intrahepatic exocrine pancreas. The spleen, in all species, was characterized by red and white pulp without boundary between the two regions, but only in H. francisci was recorded nodular organization in splenic parenchyma. The G. brasiliensisandH. aff. malabaricuspresented in the white pulp MMCs linked mainly to ellipsoids. Besides, we observed large MMCs in the spleen in relation to liver of G. brasiliensis and H. aff. malabaricus. In liver, highest values of reticular fibers and collagen were observed inG. brasiliensis. In spleen, highest values of reticular fibers and collagen were recorded inH. aff. malabaricusandH. francisci, respectively. Histological differences confirm the hypothesis that the phylogenetic distance is reflected in liver and spleen.
