The spleen is a separate organ of the teleost, playing an essential role in immune reactions. The morphology of the spleen is different from the fish species. Little knowledge about the spleen structure and the blood splenic barrier (BSB) in Nile tilapia has been reported. To address this issue, we studied the histology of the spleen and the BSB in healthy Nile tilapia. The morphology of the spleen was observed, then H&E staining, modified Jame’s staining, and ultrastructural techniques were performed to portion the spleen into three subregions and analyze the location of components and fibers. Thereafter, vital staining of Nile tilapia with Trypan blue was conducted to elucidate the composition and function of BSB. Histologically, the spleen could be divided into three subregions (inner, middle, and outer). The venules, clumps of lymphocytes, and vessels were separately characterized features of the outer, middle, and inner layers. Post injection, Trypan blue was intercepted in the endotheliocytes of ellipsoids in the middle layer (i.p.) or was deposited to the reticular fibers surrounding the ellipsoids (i.v.). Additionally, the amount of Trypan blue was shown to be positively correlated to that of the Acid phosphatase expressed. In conclusion, the spleen could be portioned into three subregions, and the BSB lay in the middle layer, composed of the cuboidal-shaped endotheliocytes and the surrounding reticular fibers of the ellipsoid capillaries. The present study enriched the research of immune tissues and system in tilapia and provided reference for the study of spleen in other fish species.

Introduction of nile tilapia (Oreochromis niloticus) in Lake Paniai known as “serapia” is aimed for improving the diversity of catches and reducing the exploitation pressure on the endemic fish species. The rapid growth, reproduction and production of nile tilapia without its management efforts is a new threat to the sustainability of the existence and utilization of endemic fish species in the lake. Management efforts of fish resources require population dynamics information. Research on the dynamics and management of nile tilapia populations was conducted from February to October 2016. Fish samples were collected from fishermen catches using nets with various mesh sizes and from four enumerators at seven observation stations. The results showed that the population of nile tilapia in Lake Paniai was dominated by individual length between 15-25 cm with frequency of 67,24%. The growth pattern of male and female fish were isometric, the average maximum length (L) was 37.28 cm and the growth coefficient (K) was 0.50 per year. Natural mortality (M) and fishing mortality (F) were 0.99 and 0.54 per year respectively. Exploitation rate (E) of 0.35 was smaller than the optimum value (E=0.5). The average size of nile tilapia captured (Lc) was 20.55 cm larger than the first size of gonad maturity (Lm) of 14.73 cm. The Lc value was higher than that the Lm value(Lc>Lm) indicating that most of nile tilapia population in Lake Paniai has spawned so that increasing the more or equal to the optimum value was expected to improve the of nile tilapia fish in Lake Paniai.

Nile tilapia (Oreochromis niloticus) has always been considered more resistant to diseases compared to other fish species. However, in recent decades, this fish species has also been susceptible to many bacterial, fungal, parasitic, and nutritional diseases. In Brazil, streptococcosis is one of the main diseases that affect tilapia and has caused many economic losses. A resistance test was conducted with 93 tilapia of four varieties (UFLA strain and three commercial varieties: SC1, SC2, and SC3) to evaluate resistance to experimental inoculation with Streptococcus agalactiae. Each variety was distributed in five aquariums, with nine specimens of the same variety. Among the five aquariums, four housed fish were intraperitoneally inoculated with 107 CFU/fish of S. agalactiae and one aquarium harbored fish from the control group, which received 0.1 mL of sterile BHI (Brain Heart Infusion). Significant differences were observed in the survival rates accumulated at the end of the experiment (day 15), with values of 22, 36, 36, and 15% for SC1, SC2, SC3, and UFLA, respectively. The UFLA strain presented lower survival than other strains, which did not differ among the 15 days of challenge. These results demonstrate that there are animals with a better response to the resistance test than others.

Aim: The aim of the study is to identify helminth parasites in the gastrointestinal tract of Oreochromis niloticus in Dadin Kowa Dam, Yamaltu Deba Local Government Area, Gombe State. Study design: Oreochromis niloticus fish species were randomly obtained from the local fishermen in the landing sites; between 7:00am-10:00am weekly. Place and duration of study: The study was carried out at Dadin Kowa Dam in Yamaltu Deba Local Government Area of Gombe State between the months of March to June, 2019. Methodology: 60 Oreochromis niloticus (Nile Tilapia) were obtained and examined for gastro intestinal parasites. Sex of the fish was determined by the presence and absent of an intromittent organ. The total length, standard length and body weight was measured using standard method. Dissection was done using standard procedure and the Parasites recovered were identified using the helminth parasites of fish identification guide. Results: Out of the 60 fish species examined. (23 female and 37 males), 21 (35.00%) were infected and were observed to habour six (6) endoparasites; viz nematodes, Cestodes and trematodes. The results show that there was a significant difference between infection rate, total length and weight of fish with female species having relatively higher percentage prevalence 11(43.50%) than the males 10 (29.70%). It was observed that the prevalence of parasitic infection increase with length and weight of the fish. Conclusion: Presence of these fish helminth parasites may lead to devaluation in protein constant in the body of the fish. The work therefore recommends water quality assessment of Dams as it is one of the factors that bring about the increase in parasitic infection.

Francisellosis in fish is caused by the facultative intracellular Gram-negative bacterial pathogens Francisella noatunensis ssp. noatunensis and Francisella orientalis. The disease is affecting both farmed and wild fish worldwide and no commercial vaccines are currently available. In this study, we tested isolated membrane vesicles (MVs) as possible vaccine candidates based on previous trials in zebrafish (Danio rerio) indicating promising vaccine efficacy. Here, the MV vaccine-candidates were tested in their natural hosts, Atlantic cod (Gadus morhua L.) and Nile tilapia (Oreochromis niloticus). Injection of MVs did not display any toxicity or other negative influence on the fish and gene expression analysis indicated an influence on the host immune response. However, unlike in other tested fish species, a protective immunity following vaccine application and immunization period could not be detected in the Atlantic cod or tilapia. Further in vivo studies are required to achieve a better understanding of the development of immunological memory in different fish species.

A control diet containing 29.03% crude protein was used in this experiment. Experimental diet was made by mixing 30% biofloc waste meal with the control diet and repelleted after the addition of 2% of binder. To determine the experimental feed digestibility, 0.5% of Cr2O3 was added as a marker for digestibility. The feed was offered to satiation at a frequency of 3 times a day for 28 days of experimentation. Nile tilapia and common carp juveniles with an initial average body weight of 11.72±0.04 g and 8.81±0.04 g, respectively, were used as the experimental animals. Each fish species were randomly stocked with a density of 10 fish/aquarium (30´45´30 cm3). The results showed that dry matter digestibility of diets with 30 % biofloc waste meal in both fish species were significantly lower than those of the controls (P<0.05). However, protein, lipid and phosphorus digestibilities of diets containing biofloc waste meal were significantly higher than those of the controls (P<0.05). Feeding with biofloc waste meal mixed feed to tilapia resulted in lower growth rate compared to that to fed control feed. ABSTRACT The Nile tilapia (Oreochromis niloticus) and African catfish (Clarias gariepinus) from the Golinga reservoir in Northern Ghana are consumed as an alternative source of protein. In the current study, levels of selected metals (As, Cd, Fe, Co, Cr, Cu, Mn, Mo, Ni and Pb) in the two fish species and the fish condition were determined, and potential human health effects assessed. The metal concentrations in the edible fish muscles ranged from 0.16 mg kg-1 (Mn) to 101 mg kg-1 (Pb). The weight-length relationships showed that both fish were growing isometrically, and bioaccumulation of metals from the reservoir was considered insignificant with bioconcentration factors below 100 except for Mn, whose value was 161 in the gills of Oreochromis niloticus. The health risk assessment showed that Pb, Cd and As had Target Hazard Quotient values above 1 for both fish species, with that of As being as high as 19.6 for Oreochromis niloticus. The target cancer risk values for As, Cd, Cr and Pb were above the non-risk threshold of <10-6, indicating the risk of cancer developing over the years due to fish consumption. The metal concentration in the fish in the reservoir should therefore be constantly monitored and potential health risks associated with frequent consumption of the fish assessed. Abstract Steroidogenic factor 1 (sf1) is an important regulator of gonad development and function in mammals. However, study of sf1 in fish is limited to cloning and expression and in vitro experiments. Using antisense RNA we knockout transcription of the sf1 gene in Nile tilapia Oreochromis niloticus, and obtain experimental fish in vivo. We demonstrate that antisense RNA can silence sf1 transcription and protein expression, and report suppression of sf1 transcription to affect gonad development and external genitalia formation in Nile tilapia. We also report disfunction of retinal metabolism and fatty acid metabolism to be important causes of weight gain and gonad abnormality with sf1 suppression. The feasibility of using antisense RNA for gene editing in fish is verified, and a new way of studying gene function and performing biological breeding is presented.
