Background. To obtain reliable results of laboratory studies on the identification of Listeria, the presence of certified diagnostic agglutinating Listeria sera is required. An important step in the manufacturing process of such medical devices for in vitro diagnostics requires effective nutrient media for the accumulation of listeriosis microbe. Aim of the research. To develop an effective nutrient medium for the accumulation of bacterial mass of Listeria. Materials and methods. The object of the study was an experimental culture medium for Listeria cultivation. As a control, we used nutrient agar for the cultivation of microorganisms (fish meal hydrolysate, FMH-agar) and meat-peptone agar with 1 % glucose (MPA with 1 % glucose). The specific activity of nutrient media during cultivation of the test strain Listeria monocytogenes 766 was evaluated using a complex of microbiological methods. Results. The optimal base of the nutrient medium for Listeria cultivation has been selected: pancreatic hydrolysate of river magpie fish (Rutilus rutilus lacustris) and hydrolysate of meat water production waste. The qualitative and quantitative composition of the nutrient medium has been developed, its physical, chemical and biological properties have been studied. It was found that after 24 hours of incubation at 37 °C, the nutrient medium provided the growth of typical Listeria colonies. The germination rate was 85 %, which is higher compared to the growth of the culture on MPA with 1 % glucose and GRM agar by an average of 21 % (p < 0,05). Conclusion. The experimental culture medium for Listeria cultivation provided growth of colonies of the test strain L. monocytogenes 766 with the preservation of characteristic cultural, morphological and biochemical properties, and, in terms of germination and growth rate, exceeded the control media. The developed nutrient medium provides effective growth of Listeria and can be used as a medium for the accumulation of microbial mass.

The results over the test of new domestic vaccine against to the blackleg – “ЕМКАРВАК” was  brought. The local productive strain C. chauvoei of Л– 7 has high immunogenicity, protective  capacity and  create  immunity duration over 9 months. The corresponding changes in regulation technological process was enable to use  of the newest data in relation to immunogenicity capacity of C. chauvoei. In particular, duration of accumulation of microbial mass of C. chauvoei in a reactor diminished from 36–42 hours to 16–18 hour, as an adsorbent is used aerosils of brand of А-300 (a production is Germany) from the calculation of 3 ± 0.1mg/sm3 culture; the supernatants was recovered by concentration of liquid after inactivation. The microbial mass of vaccine is 3.5 ± 0.3×1093. The tests of vaccine of “ЕМКАРВАК” showed that preparation was sterile, inactivated, harmless (a bioassay is on guinea-pigs), owns high antigen (the level of antibodies for the instilled animals presented 1:828 ± 257 on a 21th day after inoculation) and immunogenicity activity. The field tests of vaccine of “ЕМКАРВАК” showed  that a vaccine owns high antigenicity, providing even in 12 months after her inoculation  high level of specific antibodies (the level for the instilled animals presented 1:32 ± 6.0 against 1:11 ± 3.6 for control animals). The productive tests of the productive-experienced series of vaccine of  “ЕМКАРВАК’ conducted in the conditions of 22 unhappy economies of the Volyn’ area on young cattle. Rejections of general (oppression, increase of temperature, loss of appetite and others like that) or changes of local character (slight swelling in place inoculation, increase of regional lymphatic knots) it was not educed. Thus, the vaccine of “ЕМКАРВАК” in the conditions of the field test appeared harmless and immunogenicity, able to support tense immunity at the receptive contingent of cattle during 9 months (term of supervision).

In a field experiment it has been found that the biological activity of the soil under agriculture, as an indicator of its ecological health, depends much on the weather. In extreme excessive humidity and temperatures exceeding average 2–5°C for periods compared to the same hot, but arid periods the number of total microbial mass increases in soil, synthetic processes are intensified and carbon emissions are activated. Accordingly, the yield of green mass of corn is raised, though grain yield reduces significantly. In the year of high temperature and lack of providing moisture harvest of green mass reduces significantly, but the crop is formed at the level of optimal under the weather conditions of the year, due to relative drought-resistance of the crop.

In this work the influence of experimental forms of biological product from Streptomyces albus UN44 on the growth processes of agricultural plants (wheat, peas, corn) was studied. The advantages of the preparation are demonstrated, which contains not only antibiotic and enzymatic complexes, synthesized by the producer, but also its biomass for the stimulation of seed germination processes, growth and development of the said plants seedlings. The proposed process description of biological product Streptofungin-Phyto production in two forms is the basis for engineering development and operating regulations. The preparation can be developed as a suspension with a cell titer of 109 CFU/cm3 in PET-containers (2 – 5 l) or as a dried mass in plastic bags (1 – 2 kg). The biological product can be recommended for use on the stage of pre-planting treatment of seeds by soaking, as well as deep root watering and irrigation. The remedy is safe for humans, animals and plants and can be used together with other microbial mass-based biofertilizers.

Application to land is the preferred method for the treatment of wastewaters in New Zealand. For land treatment to be effective, it is essential that the soils can accept the volumes of wastewater applied and degrade or store the constituents in the wastewater. We report on 14 soil chemical, biochemical and physical characteristics of soils (0–10 cm depth) used for wastewater treatment at the Fonterra dairy factories at Hautapu, Lichfield and Edgecumbe in the North Island of New Zealand. The soils are under grazed pasture for dairying and receive wastewater by spray irrigation. The soils were monitored approximately every 2 years between 1995 and 2005 and at the end of monitoring had been under irrigation for 10–26 years. Matched, non-irrigated pasture soils on adjacent dairy farms were sampled for comparison. The wastewater composition from the three factories differed, reflecting the products manufactured. Loadings were greatest at the Hautapu factory, which also had the longest history of irrigation (26 years). At all three sites, the physical characteristics of irrigated soils were very similar to their non-irrigated comparisons. A consistent trend was for microbial mass and activity, and particularly nitrogen (N) turnover, to be markedly greater on the irrigated soils. The C (carbon) : N ratios of irrigated and non-irrigated soils at Lichfield and Edgecumbe were similar, but at Hautapu the C : N ratio of irrigated soil was 8.3 and significantly (P < 0.05) lower than non-irrigated soil (11.1), suggesting little further capacity to store additional N as organic matter. Irrigation tended to increase the soil pH at all sites to above neutral even though the wastewater was acidic. We consider that the characteristics of irrigated soils at Edgecumbe and Lichfield factories are generally satisfactory. Fonterra is continuing to reduce loadings in both composition and volumes of wastewater irrigated.
