This study intends to present the value of use as an eco-friendly, bioactive functional material by utilizing the undervalued tapiocaunhulled barley dried distiller's grains with solubles (TBDDGS). The physiological activity of TBDDGS presented through whitening activity, antioxidant activity, and anti-wrinkle effect. As a result of measuring the mushroom tyrosinase inhibitory activity in the hot water extracts of TBDDGS, the extracts showed 1.21% at 20 mg/mL concentration, 7.39% at 50 mg/mL concentration, and 25.78% at 100 mg/mL concentration, depending on the concentration of the extracts. The radical scavenging ability of DPPH was 28.7% at 10 mg/mL concentration, 38.0% at 20 mg/mL concentration, 60.9% at 50 mg/mL concentration, and 80.1% at 100 mg/mL concentration. The collagenase inhibitory activity of the extracts was 92.8% at a 6 mg/mL concentration. Elastase inhibitory activity was 97.8% at 100 mg/mL concentration. From the above results, the collagenase inhibitory activity of the extracts was 92.8% at a 6 mg/mL concentration. The elastase inhibitory activity was 97.8% at 100 mg/mL concentration. Although TBDDGS in this study has lower physiological activity compared to the control group, it was considered to have industrial value as a functional cosmetic raw material in consideration of the advantages of stable raw material supply and price competitiveness as an eco-friendly cosmetic raw material.

Edible bird’s nest (EBN) is an unusual mucin glycoprotein. In China, it is popular among consumers due to its skin whitening activity. However, the relationship between protein, sialic acid, and the whitening activity of EBN after digestion is still unclear. In the present work, the whitening activity (antioxidant activity and tyrosinase inhibitory activity) of digested EBN were studied by HepG2 and B16 cell models. The dissolution rate of protein and sialic acid was 49.59% and 46.45% after the simulated digestion, respectively. The contents of free sialic acid and glycan sialic acid in EBN digesta were 17.82% and 12.24%, respectively. HepG2 cell experiment showed that the digested EBN had significant antioxidant activity, with EC50 of 1.84 mg/mL, and had a protective effect on H2O2-induced oxidative damage cells. The results of H2O2-induced oxidative damage showed that the cell survival rate increased from 40% to 57.37% when the concentration of digested EBN was 1 mg/mL. The results of the B16 cell experiment showed that the digested EBN had a significant inhibitory effect on tyrosinase activity, and the EC50 value of tyrosinase activity was 7.22 mg/mL. Cell experiments showed that free sialic acid had stronger antioxidant activity and tyrosinase inhibitory activity than glycan sialic acid. The contribution rate analysis showed that protein component was the main antioxidant component in digestive products, and the contribution rate was 85.87%; free sialic acid was the main component that inhibited tyrosinase activity, accounting for 63.43%. The products of the complete digestion of EBN are suitable for the development of a new generation of whitening health products.

Hydroquinone has been used in cosmetics because of its whitening activity. In previous studies, B-carotene in Moringa plants was also known as an inhibitor of the tyrosinase enzyme. It is necessary to know how the interaction mechanism of B-carotene with tyrosinase (5M8N) and which compounds between hydroquinone and B-carotene provide computationally better activity as whitening. Tyrosinase was prepared using Discovery Studio Visualizer. Ligands were prepared using Autodock 4.2. Autodock-Vina is used for ligand docking between proteins. The result is the binding affinity (kcal/mol) of the ligand to protein. Visualization of docking between ligands and proteins using the Ligplot + Program with a 1 year license. Media used for the docking process is a computer with an Intel Core i7-3770 CPU with a speed of 3.40 GHz 8 cores, 1920x1080p resolution, VGA NVIDIA GeForce GTX 750, 8 GB RAM, Windows 8 64-bit. The docking results showed that the binding affinity of B-carotene to tyrosinase was -11.2 while hydroquinone with tyrosinase was -5.4 with RMSD 0. The results of visualization showed that B-carotene binds more amino acid receptors than hydroquinone. B-carotene in moringa has been shown to be active not only in wet laboratories, but also in dry laboratories.

Whitening cosmetics market has a bright future, and pure natural whitening products of traditional Chinese medicine have always been a research hotspot. In this research, the whitening active ingredient of Chinese medicine Trichosanthes pulp was isolated and purified for the first time, and its whitening mechanism was clarified. Chromatographic methods such as silica gel, ODS, and HPLC were used to isolate and purify them. B16 cells were used to measure the antioxidant activity, tyrosinase activity, and melanin removal activity. A total of 20 compounds were isolated, including p-hydroxybenzaldehyde (1), salicylic acid (2), vanillic acid (3), isovanillic acid (4), protocatechuate (5), trans-cinnamic acid (6), 4-coumaric acid (7), trans-ferulic acid (8), drechslerol-B (9), cyclotucanol 3-palmitate (10), 5-acetoxymethyl-2-furaldehyde (11), 5-hydroxymethylfurfural (12), diosmetin (13), apigenin (14), chrysoeriol (15), luteolin (16), 4′-hydroxyscutellarin (17), quercetin (18), 3′,5-dihydroxy-7-(β-D-glucopyranosyloxy)-4′-methoxyflavone (19), and cofloxacin-7-O-β-D-glucoside (20). Among them, compounds 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, and 20 have good antioxidant repairing effects; compounds 3, 4, 5, 6, and 7 have high black inhibition; compounds 1, 2, 3, 4, 5, 6, 7, 8, 11, 12, 13, 14, 15, 16, 17, 18, 19, and 20 have obvious tyrosine acidase inhibitory activity. The results laid foundation for the further development and utilization of Trichosanthes pulp resources and also provide a basis for the development of natural whitening cosmetics.

The development of functional cosmetics with skin improvement effects from natural sources is necessary. In this study, the antioxidant, antiwrinkling, moisturizing, and whitening effects of Gardeniae fructus extract (GF) were investigated in keratinocytes, melanocytes, and fibroblast cells. Antioxidant activity was determined by a DPPH free radical scavenging assay. MMP-1, MMP-9, HAS1, and filaggrin mRNA levels were measured by RT-PCR in keratinocytes and fibroblast cells. MITF and tyrosinase protein levels were evaluated by blotting analysis in melanocytes. DPPH free radical activity was investigated to determine whether GF showed dose-dependent inhibitory activity. GF induced the upregulation of HAS1 and filaggrin mRNA expression in keratinocytes and fibroblast cells. GF led to the downregulation of MMP mRNA levels in keratinocytes and fibroblast cells. Western blotting was performed to confirm the whitening-related protein (MITF and tyrosinase) levels induced by GF in melanocytes, and the inhibitory activity was superior to that of the α-MSH used for the comparison test. GF showed marked antioxidant, antiwrinkling, skin moisturizing, and whitening activity in keratinocytes, melanocytes, and fibroblast cells. Through the results of these experiments, the applicability of GF as a natural and functional cosmetic material was verified.

This study showed the use of Centella asiatica as a good source of compounds for skin therapy and antioxidant activation cosmetics. The goal of this study was to provide cosmeceutical activity data about the extraction yield and total phenol content of C. asiatica by optimizing the focused high ultrasound (INEFU) and ultrasound extraction (UE) conditions. Under optimal extraction conditions, that is, 1800 W for 45 minutes and 500 W for 45 minutes, 28.73% and 30.21% (w/w) of the highest extraction yields and polyphenol contents were obtained, respectively. For the skin whitening activity, tyrosinase inhibitory activity was observed at 61.52% in INEFU extracts, which was circa 20.3% higher than that of UE extracts (51.11%). To reduce melanin production in Clone M-3 cells, 86.9% melanin production was observed following the addition of control, water extraction relative to the control without a sample, and 79.6% and 74.5% were found in UE and INEFU conditions, respectively. In conclusion, the highest total phenol secretion was obtained under optimal conditions and resulted in a significant improvement in the cosmetic activities of C. asiatica extracts.
