In the past decade, progress has been made in sex determination mechanism in Vitis. However, genes responsible for sexual differentiation and its mechanism in V. amurensis remain unknown. Here, we identify a sex determination candidate gene coding adenine phosphoribosyl transferase 3 (VaAPRT3) in V. amurensis. Cloning and sequencing of the VaAPRT3 gene allowed us to develop a molecular marker able to discriminate female individuals from males or hermaphrodites based on a 22-bp InDel. Gene expression and endogenous cytokinin content analysis revealed that the VaAPRT3 gene is involved in sex determination or, to be precise, in female organ differentiation, through regulating cytokinin metabolism in V. amurensis. This study enlarged the understanding of sex determination mechanism in the genus Vitis, and the sex marker could be used as a helpful tool for sexual identification in breeding programs as well as in investigation and collection of V. amurensis germplasms.

Aiming at the problems of practical Byzantine fault tolerance (PBFT) algorithm, such as high communication complexity, frequent switching views because of Byzantine node become primary nodes and random selection of primary node, HR-PBFT algorithm is proposed. First, the HR-PBFT algorithm uses a hash ring to group nodes, which ensures the randomness and fairness of the grouping. Then, a dual-view mechanism is used in the consensus process, where the first layer node maintains the primary view and the second layer node maintains the secondary view to ensure the proper operation of the algorithm. Finally, the Byzantine node determination mechanism is introduced to evaluate the node status according to the node behavior in the consensus process, improve the reliability of primary node selection, and reduce the frequency of view changes. The experimental results show that the optimized HR-PBFT algorithm can effectively improve the problem of the sharp increase in the number of communications caused by the increase in the number of nodes in the network and prevent frequent view changes.

The snakeskin gourami (Trichopodus pectoralis) has a high meat yield and is one of the top five aquaculture freshwater fishes in Thailand. The species is not externally sexually dimorphic, and its sex determination system is unknown. Understanding the sex determination system of this species will contribute to its full-scale commercialization. In this study, a cytogenetic analysis did not reveal any between-sex differences in chromosomal patterns. However, we used genotyping-by-sequencing to identify 4 male-linked loci and 1 female-linked locus, indicating that the snakeskin gourami tends to exhibit an XX/XY sex determination system. However, we did not find any male-specific loci after filtering the loci for a ratio of 100:0 ratio of males:females. This suggests that the putative Y chromosome is young and that the sex determination region is cryptic. This approach provides solid information that can help identify the sex determination mechanism and potential sex determination regions in the snakeskin gourami, allowing further investigation of genetic improvements in the species.

Abstract We utilize a Generalized Exact Affine Stone Index system to evaluate the structure of residential water demand that recognizes demand interrelationship between residential and bottled water in the United States, allowing for precommitted consumption. Further, we address expenditure and price endogeneity by accounting for the supply side of the price determination mechanism. A significant substitutability relationship between residential and bottled water is found, while substantial precommitments are established in both residential and bottled water consumption. Residential demand becomes price-elastic once the precommitted level is reached. Finally, ignoring substitutability, precommitments, or endogeneity distorts the demand structure, resulting in erroneous policy implications.

1AbstractSexual reproduction is almost universal in the animal kingdom; therefore, each species must have a mechanism for designating sex as male or female. Fish especially have a wide range of sex determining systems. While incompatible systems are thought to increase reproductive isolation, interspecific hybridization is common among groups such as cyprinid minnows, thus, studies such as this can provide insight into hybridization and evolutionary diversification of this clade. In the present study, we aimed to identify a genetic basis for sex determination in the common creek chub (Semotilus atromaculatus) using genotyping-by-sequencing (GBS) data. No sex-associated markers were found by RADSex or a GWAS using GEMMA, however, our Weir and Cockerham locus-specific FST analysis and discriminant analysis of principal components revealed some genetic differentiation between the sexes at several loci. While no explicit sex determination mechanism has been yet discovered in creek chub, these loci are potential candidates for future studies. This study also highlights technical challenges involved in studying sex determination in species with extremely variable mechanisms.

Sex-specific markers play an important role in revealing sex-determination mechanism. Sea urchin (Mesocentrotus nudus) is an economically important mariculture species in several Asian countries and its gonads are the sole edible parts for people. However, growth rate and immunocompetence differ by sex in this species, sex-specific markers have not been identified, and the sex-determination mechanism of sea urchin remains undetermined. In this study, type IIB endonuclease restriction-site associated DNA sequencing (2b-RAD-seq) and a genome survey of M. nudus were performed, and three female-specific markers and three female heterogametic single nucleotide polymorphism (SNP) loci were identified. We validated these sex-specific markers via PCR amplification in a large number of individuals, including wild and artificially bred populations. Several open reading frames (ORFs) were predicted, although there are no potential genes known for sex determination and sex differentiation within the scaffold in which the sex-specific markers are located. Importantly, the female-specific sequences and female heterozygous SNP loci indicate that a female heterogametic and male homogametic ZW/ZZ sex-determination system should exist in M. nudus. The results provide a solid basis for revealing the sex-determination mechanism of this species, and open up new possibilities for developing sex-control breeding in sea urchin.

A large categories of time series fluctuate dramatically, for example, prices of agriculture produce. Traditional methods in time series and stochastic prediction may not capture such dynamics. This paper tries to use machine learning to tune the model for a real situation by establishing a price determination mechanism on the model of stochastic automata (SA) and evolutionary game (EG). Time series volatility attributed to the chaotic process can be obtained through the learning algorithm of Markov Chain Monte Carlo (MCMC). Using machine learning through the chaotic analysis of stochastic automata and evolutionary games, we find that a more spatially aggregated distribution (smaller entropy) leads to larger time series fluctuations, regardless of the initial distribution of crops. By integrating the factors discovered in this study, we can develop a better learning algorithm in a highly volatile time series in agriculture prices.

Abstract A new waveguide stepped septum-type circular polarizer (SST-CP) was developed to operate in the 500-GHz band for radio astronomical and planetary atmospheric observations. In a previous study, we developed a practical SST-CP for the 230-GHz band. However, several issues prevent this device being easily scaled down to the 500-GHz band, such as manufacturing dimensional errors and waveguide flange position errors. In this study, we developed a new waveguide flange with a high-accuracy position determination mechanism and a very small size of 10 × 10 mm. We also developed a new fabrication technique to obtain very good flatness for the device’s blank materials by high-accuracy polishing using a resin fixture. Using these new methods, the manufactured 500-GHz band SST-CP achieved a cross-polarization talk level of better than – 30 dB at 465–505 GHz, a device surface flatness of within 3 μm, and also the horizontal positioning error of ± 3 μm. These results indicate that the developed 500-GHz band SST-CP has high performance in the high-frequency band, and thus the new manufacturing methods are effective in the 500-GHz band.

Cytogenetic features of the hangingfly Bittacus sinicus Issiki, 1931 were investigated for the first time using C-banding and DAPI (4',6-diamidino-2-phenylindole) staining. The karyotype analyses show that the male B. sinicus possesses the lowest chromosome number (2n = 15) ever observed in Mecoptera, and an almost symmetric karyotype with MCA (Mean Centromeric Asymmetry) of 12.55 and CVCL (Coefficient of Variation of Chromosome Length) of 19.78. The chromosomes are either metacentric or submetacentric with their sizes decreasing gradually. Both the C-banding and DAPI+ patterns detect intermediate heterochromatin on the pachytene bivalents of B. sinicus, definitely different from the heterochromatic segment at one bivalent terminal of other bittacids studied previously. The male meiosis of B. sinicus is chiasmate with two chiasmata in metacentric bivalents and one in the submetacentric bivalent. The sex determination mechanism is X0(♂), which is likely plesiomorphic in Bittacidae. Two alternative scenarios of karyotype origin and evolution in Bittacus Latreille, 1805 are discussed.
