In vitro and in vivo fungicidal effects of γ-irradiation synthesized silver nanoparticles against Phytophthora capsici causing the foot rot disease on pepper plant

Stem rot disease caused by Phytophthora capsici is an important disease on chilli. Eight endophytic fungi that had been isolated and screened based on pathogenecity test were further tested for their potential as the biological control agent of the stem rot disease of chilli. The endophytic fungi suspension was applied twice during the trial. The first application was on 100 seed lot, by soaking them in 100 mL of suspension. The second application was on the 3 weeks-old chili seedlings by drenching them with 10 mL suspension per plant. The concentration of endophytic fungi  in the suspension was 2.8 × 106 cfu mL-1. The disease intensity and AUDPC value were measured for 4 weeks after the pathogen inoculation. The growth inhibition test of P. capsici  was performed in vitro and the colonization abilities of endophytic fungi were observed at 4 weeks-old chilli seedlings. Eight endophytic fungi  inhibited the growth of the P. capsici, and two of those isolates namely Penicillium strain MAG1 and Penicillium strain PAB2 showed antibiosis mechanism. Endophytic fungi has the ability more to colonize at the root (26–60%) than in the stem (20–40%). Fusarium strain MAGR1 has the highest level of endophytic colonization i.e. 60% compared to others.  Based on in vivo assay, six endophytic fungi isolates, i.e. Fusarium strain MAGR1, Penicillium strain MAG1, Penicillium strain PAB2, sterile hyphae HAJ1, sterile hyphae HAJ2, and  sterile hyphae PBG7, showed the potency to control stem rot disease with inhibition level of 25.5–35.5%

Foot rot disease epidemic on black pepper in variety of environmental conditions. The foot rot disease on black pepper caused by Phytophthora capsici Leonian, is often destructive to farmers of black pepper. This research aims to study the infection rate and the pattern of development of the black pepper foot rot disease in various condition of the environment. The research was conducted in the areas of black pepper cultivation in the Ultisol and Entisol soils. In each location plots were set with few (<25%) and abundant (>75%) weeds. Variable observed was disease incidence. The data were used to analyze model of development of the black pepper foot rot disease and infection rate. Results of the research showed that the model of development of the black pepper foot rot disease was not affected by soil type, but the type of soil affected the infection rate. At the pepper plantation with abundance of weeds, the disease development followes Gompertz model. At the plantation with few weeds, however the disease developed in a logistic model. The rate of disease infection on the black pepper plant with abundant weeds lower compared with those with few weeds.

Vietnam is the leading black pepper export country in the world. However, the production of pepper may be affected by natural disasters, pests such as Phytophthora capsici. The Phytophthora capsica disease has caused a significant decline in pepper yields. The disease is characterized with a high mortality rate (up to 100%) and rapid outbreak and thus is very challenging to control. In the present study, eight strains of P. capsici were isolated from 100 samples of leaf, stem, root and soil that were collected from pepper-growing areas of Dong Nai, Binh Duong and Ba Ria - Vung Tau provinces. These isolates' straits had capacity to cause foot rot on pepper leaf after two days of inoculation. Among them, the isolates with the highest pathogenicity are BR-L1, DN-D1 and DN-D2. In order to determine the resistance level to P. capsici of commonly grown pepper varieties (Vinh Linh, Se, Xanh, Trau and Kuching), the pepper leaves were in vitro infected with P. capsici BR-L1 spores in laboratory and greenhouse conditions. Trau variety showed the highest resistance level to P. capsici with the as indicated by (P < 0.01) disease ratio (74.1%) and disease index (73.7%) after 6 days in laboratory condition and the corresponding numbers of 17.3% and 15.8% after 12 days in greenhouse condition.

Silver nanoparticles (AgNPs) have been imposed as an excellent antimicrobial agent being able to combat bacteria in vitro and in vivo causing infections. The antibacterial capacity of AgNPs covers Gram-negative and Gram-positive bacteria, including multidrug resistant strains. AgNPs exhibit multiple and simultaneous mechanisms of action and in combination with antibacterial agents as organic compounds or antibiotics it has shown synergistic effect against pathogens bacteria such as Escherichia coli and Staphylococcus aureus. The characteristics of silver nanoparticles make them suitable for their application in medical and healthcare products where they may treat infections or prevent them efficiently. With the urgent need for new efficient antibacterial agents, this review aims to establish factors affecting antibacterial and cytotoxic effects of silver nanoparticles, as well as to expose the advantages of using AgNPs as new antibacterial agents in combination with antibiotic, which will reduce the dosage needed and prevent secondary effects associated to both.

Rhizoctonia root-rot disease causes severe economic losses in a wide range of crops, including Vicia faba worldwide. Currently, biosynthesized nanoparticles have become super-growth promoters as well as antifungal agents. In this study, biosynthesized selenium nanoparticles (Se-NPs) have been examined as growth promoters as well as antifungal agents against Rhizoctonia solani RCMB 031001 in vitro and in vivo. Se-NPs were synthesized biologically by Bacillus megaterium ATCC 55000 and characterized by using UV-Vis spectroscopy, XRD, dynamic light scattering (DLS), and transmission electron microscopy (TEM) imaging. TEM and DLS images showed that Se-NPs are mono-dispersed spheres with a mean diameter of 41.2 nm. Se-NPs improved healthy Vicia faba cv. Giza 716 seed germination, morphological, metabolic indicators, and yield. Furthermore, Se-NPs exhibited influential antifungal activity against R. solani in vitro as well as in vivo. Results revealed that minimum inhibition and minimum fungicidal concentrations of Se-NPs were 0.0625 and 1 mM, respectively. Moreover, Se-NPs were able to decrease the pre-and post-emergence of R. solani damping-off and minimize the severity of root rot disease. The most effective treatment method is found when soaking and spraying were used with each other followed by spraying and then soaking individually. Likewise, Se-NPs improve morphological and metabolic indicators and yield significantly compared with infected control. In conclusion, biosynthesized Se-NPs by B. megaterium ATCC 55000 are a promising and effective agent against R. solani damping-off and root rot diseases in Vicia faba as well as plant growth inducer.

The main objective of this study was to evaluate the ability of both antagonistic bacteria Bacillus amyloliquefaciens (SF14) and Alcaligenes faecalis (ACBC1) used in combination with salicylic acid (SA) to effectively control brown rot disease caused by Monilinia fructigena. Four concentrations of salicylic acid (0.5%, 2%, 3.5%, and 5%) were tested under in vitro and in vivo conditions. Furthermore, the impact of biological treatments on nectarine fruit parameters’ quality, in particular, weight loss, titratable acidity, and soluble solids content, was evaluated. Regardless of the bacterium, the results indicated that all combined treatments displayed a strong inhibitory effect on the mycelial growth of M. fructigena and disease severity. Interestingly, all SA concentrations significantly improved the biocontrol activity of each antagonist. The mycelial growth inhibition rate ranged from 9.79% to 88.02% with the highest reduction rate recorded for bacterial antagonists in combination with SA at both concentrations of 0.5% and 3.5%. The in vivo results confirmed the in vitro results with a disease severity varying from 0.00% to 51.91%. A significant biocontrol improvement was obtained with both antagonistic bacteria when used in combination with SA at concentrations of 0.5% and 2%. The lowest disease severity observed with ACBC1 compared with SF14 is likely due to a rapid adaptation and increase of antagonistic bacteria population in wounded sites. The impact of all biological treatments revealed moderate significant changes in the fruit quality parameters with weight loss for several treatments. These results suggest that the improved disease control of both antagonistic bacteria was more likely directly linked to both the inhibitory effects of SA on pathogen growth and induced fruit resistance.

Despite the wide adoption of mefenoxam (Ridomil Gold EC) for vegetables in North Carolina, the incidence of Phytophthora blight on pepper (Capsicum annuum) and squash (Cucurbita pepo) is high. Seventy-five isolates of Phytophthora capsici were collected in five pepper and one squash field in order to assess mefenoxam sensitivity. The relative fitness of resistant and sensitive isolates was contrasted in vitro by their respective rates of colony growth and their ability to produce sporangia in unamended V8 juice agar medium. In in vivo experiments, the aggressiveness of isolates on pepper was evaluated. The frequency of resistant isolates in North Carolina populations was 63%, considerably higher than resistance levels in areas where mefenoxam is not widely adopted. Resistant isolates grew on amended media at rates >80 to 90% and >100% of the nonamended control at 100 μg ml-1 and 5 μg ml-1, respectively. Sensitive isolates did not growth at 5 or 100 μg ml-1. All isolates from three fields, including two pepper and a squash field, were resistant to mefenoxam. Populations from other fields were composed of either mixes of sensitive and resistant isolates or only sensitive isolates. Response to mefenoxam remained stable during the course of in vitro and in planta experiments. Occurrence of a mefenoxam-resistant population of P. capsici on squash is reported here for the first time in North Carolina. When measured by rate of colony growth, sporulation in vitro, or aggressiveness in planta, fitness of resistant isolates was not reduced. Mefenoxam-resistant isolates from squash were as aggressive on pepper as sensitive or resistant pepper isolates. These results suggest that mefenoxam-resistant populations of P. capsici are as virulent and fit as sensitive populations.

ABSTRACTThe abilities of endophytic bacteria from potato roots and tubers to suppress soft rot disease (Erwinia carotovora pv. carotovora) in potato tuberSoft rot disease caused by Erwinia carotovora pv. carotovora is one of limiting factors in cultivation and post harvest of potato. The eco-friendly control measure that can be developed for controlling the diseases is biological control. Microbes that are potential as biological control agents include endophytic bacteria. This paper discussed the results of study examining the potential of endophytic bacteria isolated from roots and tubers of potato to inhibit the growth of E. carotovora pv. carotovora in vitro and suppress soft rot disease in potato tuber. The results showed that among 24 isolates examined, four isolates of endophytic bacteria (one isolate from potato tuber and three isolates from potato roots) inhibited the growth of E. carotovora pv. carotovora in vitro with inhibition zone 3.5-6.8 mm. In the in vivo test, the isolates inhibited the soft rot disease in potato tuber by 71.5-86.4%. Kelompok mikroba yang berpotensi sebagi agens pengendali biologi adalah bakteri endofit. Artikel ini mendiskusikan potensi isolat bakteri endofit yang berasal dari ubi dan akar kentang untuk menghambat pertumbuhan bakteri E. carotovora pv. carotovora secara in vitro dan menekan perkembangan penyakit busuk lunak pada ubi kentang. Hasil percobaan menunjukkan bahwa diantara 24 isolat bakteri yang diuji, terdapat empat isolat bakteri endofit (satu isolat dari ubi kentang dan tiga isolat dari akar kentang) yang dapat menghambat pertumbuhan bakteri E. carotovora pv. carotovora secara in vitro dengan zona penghambatan sebesar 3,5-6,8 mm. Pada pengujian secara in vivo, isolat-isolat tersebut dapat menekan perkembangan penyakit busuk lunak pada ubi kentang sebesar 71,5-86,4%. Isolat yang cenderung menunjukkan penghambatan relatif lebih baik secara in vitro dan in vivo adalah isolat bakteri endofit asal ubi kentang yaitu isolat CK U3 (Lysinibacillus sp.).Kata Kunci: Pengendalian biologi, Bakteri endofit, Pascapanen, Kentang, Penyakit busuk basah
