AbstractOxidative stress, associated with an imbalance between the oxidants (reactive oxygen species) and the antioxidants in the body, contributes to the development of many diseases. The body’s fight against reactive oxygen species is supported by antioxidants. Nowadays, there are too many analytical methods, but there is no one universal technique for assessing antioxidant properties. Moreover, the applied different ways of expressing the results lead to their incompatibility and unreasonable interpretation. The paper is a literature review concerning the most frequent ways of antioxidant activities expression and for an easy and universal method of the obtained results discussion. This paper is an attempt to point out their disadvantages and advantages. The manuscript can support the searching interpretation of the obtained results which will be a good tool for the development of a number of fields, especially medicine what can help in the future detection and treatment of many serious diseases. Berberis lyceum Royle has such pronounced medicinal values that it is used as cure of many diseases and has exhibited great therapeutic effects among the local communities throughout the world. The present research was carried out to evaluate the quantitative ethnobotanical status and phytochemical analysis of B. lyceum. Regular field trips were arranged to the study area (Shangla District) in August 2017 to October 2019 and interviews with 100 residents (age range: 30 to 50 years) were conducted. The approach adopted for ethnobotanical data was semi-scientific as the inhabitants were not aware about the modern names of some diseases and therefore physician prescriptions were also consulted. Ethnobotanical data were examined using relative frequency of citation and % use value. The % use value of B. lyceum indicates that the people of District Shangla mostly used it for curing of different diseases. In spite of tremendous uses the plant still survived in this area mainly due to the non-accessibility for humans of the mountain tops; otherwise, increase in anthropogenic activities even in these hilly areas poses a threat of the eradication of this plant. To correlate the folkloric uses with its phytochemical composition, HPLC (high performance liquid chromatography) analysis was performed and a total of six phenolic compounds (quercetin, chlorogenic acid, berberine, rutin, mandellic acid, and hydroxy benzoic acid) were identified in its root. As most of the health complications are correlated to oxidative stress therefore in vitro antioxidant activity were also performed using DPPH (2, 2- diphenyl-1-picrylhydrazyl) and ABTS (2, 2′-azinobis-3-ethyl benzo thiazoline -6- sulfonic acid) assays. The observed antioxidant potential may most probably be due to berberine and chlorogenic acid that were present in highest concentration in the analyzed extract. The effectiveness of the selected plant as remedy for a number of diseases (that were pointed out by the local community) may be due to its phytochemical composition especially berberine and chlorogenic acid as oxidative stress is the root cause of many diseases. The plant extract exhibited high antioxidant potential (DPPH IC50 = 165µg/mL; ABTSIC50 = 110µg/mL) in relation to the detected concentration of berberine and chlorogenic acid. It can be inferred from experimental results that the ethnopharmacological efficacy of this endangered species may be due to its phytochemical composition and antioxidant activities. This case study helps to revive the importance of B. lyceum in local communities and emphasizes the need for its conservation.

Alzheimer’s disease (AD) is strongly associated with oxidative stress which can damage neural cells. Silibinin has shown potential antioxidative effects. However, due to its low solubility in water, silibinin provides low biological activity and bioavailability. Therefore, to increase its pharmacological effects, silibilin was encapsulated into human serum albumin (HSA) nanoparticles and well-characterized by DLS and TEM techniques. The antioxidant activity of silibinin-HSA nanoparticles was evaluated on LPS-induced oxidative stress in neuron-like cells (SH-SY5Y) through MTT, antioxidant activity and apoptotic assay. It was shown that the mean diameter of HSA and silibinin-HSA nanoparticles were 88 and 105 nm, respectively with a drug loading of 24.08%, drug encapsulation rate of 94.72%, and the yield of silibinin-HSA nanoparticles of around 83.41% and the HSA nano-formulation released silibinin for 15 h. The results displayed that cell viability was reduced by LPS (10 μg/mL), who’s also determined to stimulate oxidative stress and apoptosis. However, co-incubation of cells with silibinin (50 μg/mL) or silibinin-HSA nanoparticles led to the recovery of cell viability, activation of SOD and CAT, increase of GSH content, and reduction of ROS level, Caspase-3 activity and fragmentation of DNA. It was also indicated that the neuroprotective and antioxidant activities of silibinin-HAS nanoparticles was greater than free silibinin, indicating that using albumin can be a potential formulation approach for improving the antioxidant efficacy of silibinin.

Morchella conica Pers. (M. conica) has been used both as a medical and edible mushroom and possesses antimicrobial properties and antioxidant activities. However, the antioxidant properties of polysaccharides purified from M. conica have not been studied. The aim of this study was to investigate the in vitro antioxidant properties of a polysaccharide NMCP-2 (neutral M. conica polysaccharides-2) purified from M. conica, as determined by radical scavenging assay and H2O2-induced oxidative stress in HEK 293T cells. Results showed that NMCP-2 with an average molecular weight of 48.3 kDa possessed a much stronger chelating ability on ferrous ions and a higher ability to scavenge radical scavenging 2,2-diphenyl-1-picrylhydrazyl (DPPH) than the other purified fraction of NMCP-1 from M. conica. Moreover, 3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetra-zolium bromide (MTT) assay showed that NMCP-2 dose-dependently preserved cell viability of H2O2-induced cells. The NMCP-2 pretreated group reduced the generation of reactive oxygen species (ROS) content and increased the mitochondria membrane potential (MMP) levels. In addition, Hoechst 33342 staining revealed cells treated with NMCP-2 declined nuclear condensation. Ultrastructural observation revealed that NMCP-2 pretreatment alleviated the ruptured mitochondria when exposed to H2O2. Furthermore, western blot analysis showed that NMCP-2 prevented significant downregulation of the protein expression of Bax, cleaved caspases 3, and upregulated Bcl-2 levels. These results suggest the protective effects of NMCP-2 against H2O2-induced injury in HEK 293T cells. NMCP-2 could be used as a natural antioxidant of functional foods and natural drugs.
