Determination of mitochondrial DNA haplotypes from sting remnants of the honeybee Apis mellifera (Hymenoptera:Apidae)

AbstractAfricanized honeybees are presently expanding their geographic range within the USA and are considered undesirable due to their substantial defensive behaviour. Africanized honeybees can be differentiated from honeybees of European ancestry using discriminant analysis of morphological characters, based on a minimum of ten intact specimens per colony. In this paper we report a PCR-based method suitable for the identification of African or European mtDNA from sting remnants, such as typically remain on victims following a stinging incident. We experimentally simulated collection and shipment conditions with dried and alcohol preservation of stings, and also report that the method was suitable for sting remnants stored with a victim sample for over one year in alcohol. The determination of mtDNA haplotypes from stinger remnants must be tempered by the constraints inherent in interpretation of mtDNA haplotype data. Such data do not provide any information regarding the genetic contribution of the paternal lineage. However, in geographic areas where baseline information regarding European haplotype frequencies is known, the detection of a different mtDNA haplotype in stingers from a victim, especially when associated with observations of extreme defensive behaviour, would certainly be suggestive of Africanization.

Abstract. We used a large dataset of greater flamingo chicks banded and measured at Camargue, France, to verify the applicability of discriminant function analysis to sex this species. Males and females sexed genetically differed significantly in all of the morphological characters measured (body mass, tarsus and wing length), with males being significantly larger than females. Although the discriminant rate varied substantially from one year to another, we found that it increased with the sample size of genetically sexed individuals. Our results suggest that discriminant function analysis (DFA) does not provide an efficient tool to sex greater flamingo chicks as these relationship are highly variable across years, requiring the genetic determination of sex on a large number of individuals every year for calibrating the DFA and still providing an overall low accuracy in sex determination. Indeed, conditions at breeding seasons can vary between years and can be considered proximate causes affecting the correct discriminant rate. Like previous studies, we recommend caution in dealing with discriminant equations computed from small datasets, and our simulation suggests that 325 genetically sexed individuals are needed to obtain 80 % correctly classified greater flamingo chicks.

To find out the ability of Orobanche seeds to germinate immediately after seed set, seeds were germinated periodically at an interval of three months for one year in GR24. Some Orobanche seeds were capable of germination immediately after seed set but most required about nine months as after ripening or incubation period to be able to germinate. The phenomenon of after ripening in Orobanche seeds could be taken as an ecological measure to dormant over following unfavorable wet summer season. The growth hormone studies on Orobanche seed germination have shown that GA3 at a concentration of 100 ppm substantially enhanced seed germination when applied during pre-conditioning period. NAA showed some stimulatory effect at 0.5 - 1.0 ppm when applied during post-conditioning period but the hormone if applied during pre-conditioning period inhibited the germination. Kinetin failed to stimulate the germination at all the concentrations tested. Key words: Germination, root-parasite, hormone. Ecoprint Vol.11(1) 2004.

Abstract Background Balancing the intensity and duration of antiplatelet therapy according to thrombotic risk is a fundamental need in order to optimize therapy effectiveness and safety. Incorporation of new predictors in thrombotic risk stratification is therefore of a crucial importance for antiplatelet therapy net clinical benefit. Purpose The present analysis aimed to evaluate the relation of miR-126-3p and miR-223-3p, new markers of platelet activation, in order to facilitate prediction of recurrent thrombotic events after acute myocardial infarction (AMI). Method The analysis included 598 patients (age median 62 years, men 77.8%) randomized in the Prague-18 study (ticagrelor vs. prasugrel in AIM treated with primary PCI). During the study follow up, 40.6% of patients switched to clopidogrel. Determination of miR was evaluated 24 hours after admission; miR-126-3p and miR-223-3p were normalized by miR-423-3p and miR-150-5p. Quantitative determination of selected miRNAs was performed with a novel microRNA immunoassay method. Selected miRNAs were compared with key efficacy endpoints (cardiovascular death, nonfatal MI and stroke), stent thrombosis and all hemorrhagic events, and analysed using univariate and multivariate logistic regressions. Results Increased values of miR-223-3p were significantly related to the occurrence of combined ischemic endpoint within 30 days [OR (95% CI) 15.739 (2.066; 119.932) p=0.008] and within one year [3.175 (1.40; 7.186) p=0.006]. Decreased ratio of miR-126-3P/miR-223-3p was significantly related to the occurrence of combined ischemic endpoint within 30 days [0.137 (0.031; 0.609) p=0.009] and one year [0.372 (0.169; 0.819) p=0.014]. MiRNAs were identified as independent predictors even after adjustment for confounding clinical predictors (Study arm, Switch to Clopidogrel, Age, Men, BMI, Smoking, History of Hyperlipidemia, Hypertension, DM, MI, PCI, CABG, Chronic heart failure, Chronic renal failure, Peripheral arterial disease, LBBB, RBBB, TIMI <3 after PCI, Number of diseased vessels >1, Stem disease, Suboptimal of failure of PCI, Time to hospital). Adjusted ORs (95% CI) are 11.828 (1.472; 98.011), p=0.022 and 2.394 (1.021; 5.610), p=0.045 for increased value of miR-223-3p and the occurrence of combined ischemic endpoint within 30 days and one year respectively; 0.151 (0.030; 0.757), p=0.022 and 0.407 (0.179; 0.925), p=0.032 for decreased ratio of miR-126–3P/miR-223-3p and the occurrence of combined ischemic endpoint within 30 days and one year respectively. No association between miRNA and bleeding complications was identified. Conclusion The miR-223-3p and miR-126-3p to miR-223-3p ratio are strong independent predictors of thrombotic ischemic events and can be used to stratify patients post AMI.

AbstractRelation between morphological traits of the root system and yield related traits is an important issue concerning efforts aiming at improving of ideotype of cultivated plants species, including pea. In this paper, to analyse the dependency between traits describing the root system morphology and yield potential, Person’s andSpearman's_correlations as well as canonical correlations were used.Root system was analyzed in 14 and 21 day-old seedlings growing in blotting-paper cylinders. Yield potential of pea was analysed in a field experiment. Results of Person’s and Spearman's_correlations revealed that number of lateral roots and lateral roots density were correlated witch yield related traits. Correlation between root length and shoot length was observed only for 14 day-old seedlings. The result of canonical correlations revealed that number of lateral roots and lateral roots density had the largest effect on yield related traits. This work highlights, that in order to improve the yield of pea it might become necessary to understand genetic determination of morphological traits of the root system, especially number of lateral roots.

AbstractIn this paper, a long-term equilibrium model of a local market is developed. Subject to minor qualifications, the model is arbitrage-free. The variables modelled are the prices of risk-free zero-coupon bonds – both index-linked and conventional – and of equities, as well as the inflation rate. The model is developed in discrete (nominally annual) time, but allowance is made for processes in continuous time subject to continuous rebalancing. It is based on a model of the market portfolio comprising all the above-mentioned asset categories. The risk-free asset is taken to be the one-year index-linked bond. It is assumed that, conditionally upon information at the beginning of a year, market participants have homogeneous expectations with regard to the forthcoming year and make their decisions in mean-variance space. For the purposes of illustration, a descriptive version of the model is developed with reference to UK data. The parameters produced by that process may be used to inform the determination of those required for the use of the model as a predictive model. Illustrative results of simulations of the model are given.

Serum/plasma fructosamine (SeFa) concentration is a reliable indicator used in human diabetic control. Tests for monitoring the carbohydrate/energy metabolism of (farm) animals are less commonly performed in veterinary laboratories, since most of the reliable determinations, both automated and manual, are relatively expensive. The aim of this study was to develop a precise, money- (and time-) saving automated micro method for measuring SeFa. ELISA microplates (20 µL samples and 200 µL reagents) and an automatic microplate autoreader were used. The classical nitroblue tetrazolium (NBT) stain reagent solution of Johnson et al. (1982) was modified using a SIGMA reagent to render it stable for up to one year. SeFa concentrations measured by the new method in 30 human blood plasma samples were compared with values obtained by the standard (generally used) LaRoche kit procedure. Fifteen cow, 13 dog and 18 chicken plasma samples were assayed by the new automated ‘micro’ method as well as by the manual test tube ‘macro’ method commonly used earlier. The modified reagent was applied for both methods. The coefficient of correlation (r) between the results obtained by the two methods was consistently between 0.94 and 0.98 (p < 0.001).

Utilizing airflow therapy, Schwartz (1976) has claimed an 89% success rate with stutterers following treatment and an 83% success rate at one year follow-up. Such claims have yet to be documented in the scientific literature. The purposes of this study were: (a) to investigate the effectiveness of a modified version of airflow therapy; (b) to examine the relative importance of its two main components—passive airflow and elongation of the first vowel spoken. The speech of two adult male stutterers with a lengthy history of stuttering, was assessed with spontaneous speaking and reading tasks. Results indicated marked improvement in both subjects' speech on the reading task was maintained at follow-up 10 weeks later. For spontaneous speech, results were generally weaker and less durable. Effects of the two treatment components were cumulative and did not allow determination of any differential effectiveness between components. Implications of these findings were considered and directions for future research discussed.

The prestress level is a key factor of prestressed concrete (PSC) beams, affecting their long-term serviceability and safety. Existing monitoring methods, however, are not effective in obtaining the force or stress of embedded tendons. This paper investigates the feasibility of elastomagnetic (EM) sensors, which have been used for external tendons, in monitoring the long-term prestress loss of bonded tendons. The influence of ambient temperature, water, eccentricity ratio, plastic duct, and cement grouts on the test results of EM sensors is experimentally examined. Based on the calibrated EM sensors, prestress loss of a group of PSC beams was monitored for one year. In order to further consider the high randomness in material, environment, and construction, probabilistic analysis of prestress loss is conducted. Finally, the variation range of prestress loss with a certain confidence level is obtained and is compared with the monitored data, which provides a basis for the determination of prestress level in the design of PSC beams.

This on line curatorship presents a selection of 11 works by Latin American artists who incorporate in their creations technologies traditionally linked to surveillance and control processes. By Surveillance Aesthetics we understand a compound of artistic practices, which include the appropriation of dispositifs such as closed circuit video, webcams, satellite images, algorithms and computer vision among others, placing them within new visibility, attention and experience regimes. The term referred to in the title of this exhibition is intended more as a vector of research rather than the determination of a field, as pointed by Arlindo Machado under the term “surveillance culture”. (Machado 1991) In this sense, a Latin America Surveillance Aesthetics exhibition is a way to propose, starting from the works presented here, a myriad of questions. How and to what extent do the destinies of surveillance devices reverberate or are subverted by market, security and media logics in our societies? If, in Europe and in the USA, surveillance is a subject related to the war against terror and border control, what can be said about Latin America? What forces and conflicts are involved? How have artistic practices been creating and acting in relation to these forces and conflicts? Successful panoramas of so called Surveillance Art already take place in Europe and North America for at least three decades, the exhibition “Surveillance”, at the Los Angeles Contemporary Exhibitions being one of the first initiatives in this domain. In Latin America however, art produced in the context of surveillance devices and processes is still seen as an isolated event. Article is dedicated to the determination of key features of the United States participation in the international capital movement on the example of FDI outflows and inflows, and confirmation of its heterogeneous nature. Tax reform that has been implemented in the end of 2017 led to the result when USA was deleted of the list of TOP-20 world investors of 2018 (by UNCTAD). The scientific opinion and fears were connected with the forecasts that the tax reform could change the movement of FDI flows back to the USA from foreign countries, especially, and the countries with the low taxes and the most favorable investment regimes. At the same time, it needs to underline that the analysis of U.S. FDI inflows and outflows showed that the negative indicator of U.S. FDI outflow (export) in 2018-2019 was connected with the repatriation of U.S. holding companies profits, that were doing business in countries with the most favorable tax and investment regimes. The authors tried to investigate the nature of the American holdings role and the integration of U.S. in to the global system of FDI and capital movement.
