Use of a long-term rumen simulation technique (Rusitec) to provide micro-organisms for in vitro digestibility assays

SUMMARYFour experiments involving the two-stage in vitro digestibility technique of Tilley & Terry (1963) were undertaken to investigate whether cow rumen liquor could be replaced with effluent or extract from feed residue containing micro-organisms derived from the long-term rumen simulation technique (Rusitec) of Czerkawski & Breckenridge (1977).In Expts 1 and 2, the dry matter digestibility (DMD) of hay was determined using either cow rumen liquor or Rusitec liquid effluent or extract from feed after 48 h of digestion in a Rusitec apparatus using various volumes of liquor and artificial saliva. Digestibilities determined with rumen liquor were c. 70% (c.v. 1·2%) while those determined with Rusitec effluent or extract were c. 62% (c.v. 1·8%). In treatments common to Expts 1 and 2, Rusitec effluent digestibilities were more repeatable than extract or rumen liquor digestibilities.In Expts 3 and 4, Rusitec effluent was compared with cow rumen liquor for determining DMD of seven forages with DMD ranging from 33 to 70%. For a given feed there was good agreement between experiments in DMD values determined with Rusitec effluent, and for ten paired values there was a high correlation (r2 = 0·976) between DMD determined by the two methods. DMD determined using rumen liquor (y) was accurately predicted by Rusitec liquid effluent digestibility (x) from the equation y = 2·663 + 1·06477.x; R.S.D. 2·433.The results show that Rusitec liquid effluent can be used instead of rumen liquor as a source of micro-organisms for the two-stage in vitro digestibility method. Routine use of Rusitec would obviate the need for using fistulated animals.

Previous studies (El Shaer, Omed and Axford, 1987; Akhter, Owen, Fall, O'Donovan and Theodorou, 1994) with the two-stage in vitro procedure of Tilley and Terry (1963) have shown a high correlation between digestibilities of forages as determined using either sheep rumen liquor, sheep faeces or cow faeces as the microbial inoculum. In the first study of the of the present investigation one objective was to examine the repeatability of these digestibility measurements when made on different occasions. A second objective was to assess whether the correlations between faecal and rumen fluid based inocula could be improved if microorganisms were obtained from pairs rather than individual animals. The objective in the second study using forages of known in vivo digestibility, was to investigate the effect of freezing or freeze-drying of faeces on the repeatability of digestibilities of forages determined in vitro using micro-organisms from cow faeces.

A two-stage cellulase-pepsin technique (CS) was compared with the two-stage rumen inoculum-pepsin (IVDMD) method to predict in vivo dry matter digestibility (in vivo DMD) in grasses (n = 30) and legumes (n = 25). Comparisons of CS with IVDMD in whole-plant corn (n = 186) were also conducted. There were significant correlations (P < 0.01) between CS and in vivo DMD and between CS and IVDMD in all forages. The coefficients of determination (r2) and residual standard deviations for the prediction of IVDMD by CS varied between 0.76 and 0.90 and 2.5 and 3.4, respectively. The apparent dry matter digestibility of diets for rabbits was measured in vivo (ADMDvv) and in vitro with the Ankom DaisyII Incubator. Four diets were tested: low fiber (LF), LF + 5% of pregerminated fenugreek seeds (PGFS) (LF5), LF + 10% PGFS (LF10), and high fiber (HF). For the ADMDvv, feces samples were collected from 56 White New-Zealand × Californian rabbits fed the 4 diets; animals were randomly allocated into 4 groups and housed in individual cages. For the in vitro trial, 3 methods were tested: fecal inoculum (FA) with Kansans State buffer; fecal inoculum with artificial saliva (FB); and multienzyme (ENZ). Fecal inocula were collected at slaughtering from the distal colon of rabbits fed ad libitum the LF diet. For FA and FB methods, the digestibility was measured at 36 and 48 h. The in vitro methods ranked the apparent dry matter digestibility of diets in the same order as in vivo, but ENZ values were always higher than FA and FB at 36 and 48 h. The prediction equations of in vivo digestibility with the ENZ method showed higher coefficient of determination (R2 = 0.69) and lower SE (0.190) than FA and FB; also, reproducibility was higher with ENZ (CV = 3.1%). In conclusion, different methods can be applied to the Ankom DaisyII Incubator to study the digestibility in rabbits. In our trial, the better reproducibility was observed with the multienzyme method than FA and FB were probably related to the variability of inocula.

Digesting forages with a prepared cellulase solution (PCS) has been shown to be a rapid method of obtaining dry matter digestibility (DMD) estimates in many species. This procedure has not been previously used to obtain DMD estimates of broadleaf birdsfoot trefoil (Lotus corniculatus L.). A broad based population consisting of 19 accessions, representing 11 countries of origin, three germplasm pools, and two domesticated cultivars of birdsfoot trefoil was evaluated. The PCS was produced from culturing Trichoderma reesei Simmons culture number QM9414. Fiber and PCS determinations were compared to an in vitro rumen fermentation (IVDMD) procedure. The fiber components measured were; neutral detergent fiber (NDF), acid detergent fiber (ADF), and hemicellulose (HEMI). The PCS procedures involved digestion using a PCS without any pre- or post-treatment (CS), acid-pepsin digestion followed by a single-stage digestion in PCS (APCS), acid detergent solution (ADS) followed by a two-stage digestion in PCS (ADSCS), and neutral detergent solution (NDS) followed by a two-stage digestion in PCS (NDSCS). All of the fiber and PCS procedures were significantly (P ≤ 0.001) correlated with IVDMD except HEMI. The CS procedure underestimated IVDMD by 199 g kg−1; however, it had the best regression coefficient (b = 0.99), had the second best correlation coefficient (r = 0.86), and required less time than the other PCS procedures. The APCS procedure provided the best estimate of IVDMD. It overestimated the IVDMD mean of birdsfoot trefoil by only 30 g kg−1 and was the most accurate (residual standard deviation (RSD) = 22 g kg−1) approach. The CS procedure is recommended when a ranking of samples is desired and actual values of DMD are not of primary interest.Key words: Fiber digestion, germplasm, in vitro dry matter digestibility, Lotus corniculatus L., Trichodenna reesei Simmons

Abstract Background The study highlighted the potential of three common and under-utilized tropical leguminous seeds (Tomentosa nilotica, Dioclea reflexa and Monodora myristica) to be used as supplementary feed to ruminant livestock. These seeds (their plants inclusive) are valuable sources of food and medicine for the prevention of illness and maintenance of human health. The medicinal properties of these seeds include antimicrobial, anti-inflammatory, anti-oxidant and immuno-stimulant. Trypsin inhibitors, which are common anti-nutritional factors in legumes and for monogastric animals, do not exert adverse effects in ruminants because they are degraded in the rumen. Hence, the crux of this study is to examine the effect of processing methods on the nutritional composition (proximate, fibre fractions, minerals, anti-nutrients) and in vitro digestibility of Tomentosa nilotica, Dioclea reflexa and Monodora myristica seeds and their suitability as feedstuff (protein sources) in small ruminant feed, particularly during off-season. Results From the results, raw Tomentosa nilotica and Monodora myristica have the highest crude protein (30.35% CP) and fat (22.40% EE), respectively. It is noteworthy that roasting best improve the mineral and significantly reduce the anti-nutrients observed in this study better compared to boiling and soaking methods. The highest organic matter digestibility, short-chain fatty acids, metabolizable energy and in vitro dry matter digestibility values were obtained in Dioclea reflexa compared to other test seeds. Roasting best improved the nutritive values, while Dioclea reflexa seed was rated highest for all the nutritional attributes and in vitro digestibility. Conclusions Dioclea reflexa could be incorporated in ruminants’ diet as protein source, particularly during the off-season, for improved ruminant production in Nigeria. However, in vivo study is therefore recommended to validate this report.

Akhter et al.(1994) demonstrated bovine faeces to have considerable potential as a source of microorganisms to replace rumen liquor in the in vitro digestibility method of Tilley and Terry (1963) for evaluating temperate gramminaceous forages, under UK laboratory conditions. In view of the difficulties and costs of maintaining fistulated animals in tropical, less-developed countries, the present study was undertaken in Tanzania to investigate the potential of bovine faeces as a substitute for rumen liquor in the Tilley and Terry technique for evaluating local, gramminaceous forages. Use of equine faeces was also investigated in view of the prevalence of equines in some tropical countries. It was also hypothesised that equines, being hind-gut fermentors, would yield faeces containing more micro-organisms than faeces from bovines, with a consequent improvement in digestibility.

A comparison of two near infrared (NIRA) calibrations (C1 and C2) for the prediction of in vitro dry matter digestibility (IVDM) in whole-plant corn (WPC) was conducted. C1 consisted of 40 WPC samples collected from four locations across Ontario (Brucefield, London, Guelph and Elora). C2 consisted of 90 samples and included the above locations plus Pakenham and Winchester. Nine wavelengths were used in both equations but only three were common in C1 and C2 equations. These wavelengths were 2139 nm, 2100 nm, and 1445 nm, respectively. The predictions of IVDM utilizing both C1 and C2 were good. Coefficients of determination (r2) and standard error of the estimate (SEE) for calibration and prediction sets were 0.91, 1.7; 0.85, 1.7 for C1 and 0.88, 1.6; 0.77, 1.6 for C2 respectively. Regression analysis within location, however, showed low r2 values for the prediction of IVDM for Pakenham and Winchester in both calibrations. The more mature stage of harvest at these locations might be the cause of the poorer predictions. Key words: In vitro digestibility, whole-plant corn, near infrared reflectance

The in vitro dry matter digestibility (I.V.D.) of the stem and total leaf fractions of four orchardgrass clones was similar at the preheading stage. Even at 100% head emergence, differences in I.V.D. between stems and living leaves were not great with the early-maturing clones. However, wide differences existed between the values for stems and living leaves in mature plants. Heads were only slightly higher than stems in digestibility.Leaf digestibility was lower for those leaves dead or even partially dead. Nevertheless, in mature plants, dead leaves were higher in I.V.D. than heads or stems. Large differences occurred among clones in stem digestibility. However, the variability among clones in leaf digestibility was small.

Abstract The objective of this study was to evaluate the effect of supplementing 10% of sorghum grain (SG), soybean meal (SBM) or dried citrus pulp (DCP) upon dry matter digestibility (DMD), organic matter digestibility (OMD), and metabolizable energy content (ME) of diets based on dried leaves of two shrub species (Acacia farnesiana and Acacia rigidula) and one grass (Cenchrus ciliaris) browsed by small ruminants. Twelve experimental diets were elaborated using three forage sources: C. ciliaris, A. farnesiana and A. rigidula and evaluated either alone (0% supplementation) or supplemented with 10% of SG (9.0% CP, 22.3% NDF), SBM (42.8% CP, 16.3% NDF) or DCP (4.9% CP, 19.1% NDF). In vitro DMD (Daisy incubator, ANKOM) at 48 h, and in vitro gas production (GP, in calibrated 100 mL glass syringes) at 24 h, were determined by incubating samples in an inoculum prepared by using rumen liquor of two rumen fistulated sheep and artificial saliva in a 1:4 ratio. OMD and ME content were calculated from the gas production, crude protein, ash, and ether extract content. Data were evaluated according to a 3 x 4 factorial experiment. There were no significant (P &gt; 0.05) forage source x supplement interaction effects. Leaves of A. farnesiana had higher (P &lt; 0.05) DMD (64%), GP (26 mL), ME (1910 kcal ME/kg DM), and OMD (53%) than other forages (mean 47% DMD, 17 mL GP, 1232 kcal ME/kg DM, and 39% DMO). Supplements increased (P &lt; 0.05) DMD (average +8%), but only DCP increased (P &lt; 0.05) the GP (+23%) of evaluated forages. Leaves of A. farnesiana had higher DMD (P &lt; 0.05; 64%). There were no effects (P &gt;0.05) of the 10% added supplements in ME content or DMO. In conclusion, leaves of A. farnesiana and DCP are valuable sources of nutrients and energy for small ruminants in northeastern Mexico.
