Molecular and morphological characterization of Austrodiplostomum ostrowskiae Dronen, 2009 (Digenea: Diplostomatidae), a parasite of cormorants in the Americas

AbstractThe diplostomid genus Austrodiplostomum currently contains two species, i.e. A. mordax and A. ostrowskiae. Adults of these species inhabit the intestine of cormorants of the genus Nannopterum, whereas larval forms (metacercariae) are found in the eyes (vitreous humor) of freshwater fishes. Records of both species have been established across a wide geographic range in the Americas. Diplostomid adults and metacercariae were collected from a wide geographical range that spans from south-eastern Mexico, southwards to Central and South America. Even though the diplostomid has been largely reported in Mexico as Diplostomum (Austrodiplostomum) compactum, our specimens were identified morphologically as A. ostrowskiae. Sequences of the mitochondrial gene cytochrome c oxidase (cox 1) were obtained for 86 individuals, including 15 adults recovered from the intestine of Nannopterum brasilianus, in five localities, and 71 metacercariae from the eyes of ten fish species, in 13 localities. Sequences were used to evaluate the genetic diversity, and to test conspecificity of these specimens with the available sequence of A. ostrowskiae. Sequences were aligned with another 12 taxa representing five genera of Diplostomatidae, forming a dataset of 104 taxa with 478 nucleotides. The genetic divergence estimated among the 86 sequenced individuals, and that of A. ostrowskiae from the double-crested cormorant, Nannopterum auritus, in the USA, was very low, ranging from 0 to 0.8%. The maximum likelihood (ML) and Bayesian consensus trees showed that all sequences nested within a monophyletic lineage, with strong bootstrap and Bayesian posterior probability support values (100/1.0). In conclusion, a link between the metacercariae in fish and the adults in cormorants was established, indicating also that a single species is found in the distribution range comprising southern USA, southwards to Venezuela. Previous records of this species, particularly from Mexico, need to be corrected.

The genus Diplodinium includes 31 species of ophryoscolecid ciliates with two retractable ciliary zones in the anterior end of the body, with no skeletal plates, inclined macronucleus and two contractile vacuoles. They have a broad geographical distribution and low host specificity. The genus has several taxonomic inconsistencies, mainly due to polymorphism of most species. Diplodinium anisacanthum is a case of taxonomic inconsistency in the genus. It presents polymorphism in posterior end of the body, so it is considered a polymorphic species or seven distinct species. This study presents the morphological characterization of six D. anisacanthum morphotypes based on general morphology, morphometric analysis and oral infraciliature characterization and taxonomic notes on the genus Diplodinium. The morphological and morphometric characterization of D. anisacanthum showed great similiarities among morphotypes suggesting that these ciliates constitute a single species with polymorphisms. A brief review conducted highlights the inconsistencies in the taxonomy of the genus Diplodinium and demonstrates the need for recent studies with representatives of the genus. There is no information on how polymorphism of Diplodinium anisacanthum morphotypes correlates to molecular data, since there are only two sequences of this species available in the GenBank database.

The new species Diabrotica wartensis Cabrera & Sosa-Gómez was found feeding on roots and leaves of sunflower and soybean in the north of the state of Paraná. A set of characters of diagnostic value: mouthparts, hind wing venation, binding patch, metendosternite, and details of male and female genitalia previously overlooked in the genus was provided. In addition to the morphological characterization, the molecular analyses of the COI mitochondrial gene and the ITS-1 rDNA nuclear region were included. The analysis of the ITS-1 nuclear region alone suggests proximity of D. wartensis to the Diabrotica fucata group, whereas molecular evidence based on combined analysis of COI and ITS-1 region suggest that this new species is distantly related to the Diabroticina with known sequences deposited in the GenBank.

Bumblebees (genus Bombus Latreille) are pollinator insects of great ecological and economic importance, which commercial use for pollination has increased since the 80s. However, the introduction of foreign Bombus terrestris (Linnaeus) has resulted in a decline of native bumblebee populations in Japan, Chile or Argentina among others. To study the potential introgression of commercial B. terrestris into the Iberian endemic subspecies Bombus terrestris lusitanicus Krüger, it is necessary to find a precise molecular marker that differentiates both subspecies. For this purpose, comparative analyses were carried out between B. t. lusitanicus and B. t. terrestris (Linnaeus) from Spain and from Belgium by sequencing the nuclear genes elongation factor 1-α and arginine kinase and the mitochondrial gene 16S ribosomal RNA, and genotyping with eleven microsatellite loci. No differentiation was observed at the nuclear level, but haplotypes found within the 16S sequence correlated with the morphological characterization of the subspecies. In a case study including individuals sampled before the establishment of bumblebee rearing companies and others from recent samplings, we detected hybrid individuals (those with non-matching morphological subspecies and 16S haplotype) more frequently in the south supporting the naturalization of commercial B. t. terrestris and introgression events between both subspecies. This marker should be used in Iberian populations with the aim to support management and conservation actions in endemic populations of B. t. lusitanicus.

Mycobacteriophage R1 was originally isolated from a lysogenic culture of M. butyricum. The virus was propagated on a leucine-requiring derivative of M. smegmatis, 607 leu−, isolated by nitrosoguanidine mutagenesis of typestrain ATCC 607. Growth was accomplished in a minimal medium containing glycerol and glucose as carbon source and enriched by the addition of 80 μg/ ml L-leucine. Bacteria in early logarithmic growth phase were infected with virus at a multiplicity of 5, and incubated with aeration for 8 hours. The partially lysed suspension was diluted 1:10 in growth medium and incubated for a further 8 hours. This permitted stationary phase cells to re-enter logarithmic growth and resulted in complete lysis of the culture.

Religious institutions in the USA, under the First Amendment, exhibit great strength in employment termination, given freedom by the Supreme Court to conduct their labour and employment practices with limited scrutiny. This article examines ways in which a Presbyterian seminary board report, justifying its decision not to renew a professor's contract, demonstrates discrimination in its use of the 'good family' ideal prominent within conservative Christianity. Focusing on intertextuality and representation of the professor's wife, a disabled woman, analysis presents evidence of an overall strategy of exclusion. The report consistently demonstrates support for negative witness statements about the professor and his wife while undermining the professor's accounts. The report's characterization of the professor's wife subsumes her identity under her husband's and assumes moral reasons for her disability and chronic illness, consistent with a nouthetic counselling ethos. Findings support the discriminatory potential of the 'good family' ideal, underscoring employees' unique vulnerability within religious higher education institutions.

The different integrated production systems can directly interfere with its bacterial community. The present study aimed to assess density, bacterial diversity and the influence of dry and rainy season in different integrated and an exclusive production system. The fallow and a native forest area was assessed to. Samples were collected in 2012 March and September. The isolation were carried out into Petri dishes containing DYGS medium. The number of colony forming units (CFU) was counted after 48 hours and. The bacterial density ranged between 106 and 107 CFU g-1 soil. The crop system affected the dynamics of the bacterial community only in the rainy season. The rainy season showed greater density of total bacteria when compared to the dry period regardless of the cropping system. The dendrograms with 80 % similarity showed thirteen and fourteen groups in the rainy and dry seasons. Isolates with the capacity to solubilize phosphate in vitro were obtained from all areas in the two seasons, but this feature has been prevalent in bacteria isolated during the rainy season

Background: Endophytic fungi live in plants’ tissue and can produce the same bioactive compounds as its host plant produces. Syzygiumpolyanthum leaves have known to be one of the antibacterial compound producers. Aim and Objective: This study aimed to characterize morphologically, microscopically, and molecularly the antibacterial-producing endophytic fungi of Syzygiumpolyanthum leaves. Methods: The isolation of endophytic fungi was done by fragment planting method on PDA medium. The antibacterial screening was performed using the antagonistic test as the first screening followed by the disc diffusion test method. The morphological characterization was based on isolate’s mycelia color, growth pattern, margin, and surface texture of the colony, while the microscopic characterization was based on its hyphae characteristics. The molecular characterization of the isolate was done by nitrogen base sequence analysis method on nucleotide constituent of ITS rDNA genes of the isolate. Results: The results found that isolate DF1 has antibacterial activity against E.coli, S.aureus, P.acne, and P.aeruginosa, with the greatest inhibition at 10% concentration of broth fermentation extract on S.aureus with a diameter of inhibition of 13.77 mm. Conclusion: Based on macroscopic, microscopic, and molecular characterization, DF1 isolate is similar to Ceriporialacerate.

In the current work, the characterization of novel chitosan/silica nanoparticle/nisin films with the addition of nisin as an antimicrobial technique for blueberry preservation during storage is investigated. Chitosan/Silica Nanoparticle/N (CH-SN-N) films presented a stable suspension as the surface loads (45.9 mV) and the distribution was considered broad (0.62). The result shows that the pH value was increased gradually with the addition of nisin to 4.12, while the turbidity was the highest at 0.39. The content of the insoluble matter and contact angle were the highest for the Chitosan/Silica Nanoparticle (CH-SN) film at 5.68%. The use of nano-materials in chitosan films decreased the material ductility, reduced the tensile strength and elongation-at-break of the membrane. The coated blueberries with Chitosan/Silica Nanoparticle/N films reported the lowest microbial contamination counts at 2.82 log CFU/g followed by Chitosan/Silica Nanoparticle at 3.73 and 3.58 log CFU/g for the aerobic bacteria, molds, and yeasts population, respectively. It was observed that (CH) film extracted 94 regions with an average size of 449.10, at the same time (CH-SN) film extracted 169 regions with an average size of 130.53. The (CH-SN-N) film presented the best result at 5.19%. It could be observed that the size of the total region of the fruit for the (CH) case was the smallest (1663 pixels), which implied that the fruit lost moisture content. As a conclusion, (CH-SN-N) film is recommended for blueberry preservation to prolong the shelf-life during storage.
