Digestibility of amino acids in maize, wheat and barley meals measured in pigs with ileo-rectal anastomosis and isolation of the large intestine

Evaluation was made of the extent to which amino acids in maize, wheat and barley meals were digested by pigs.Apparent Digestibility (AD) of an individual amino acid was determined as the difference between amino acid intake and amino acid in the ileal digesta, expressed as a percentage of the intake. AD was adjusted to True Digestibility (TD) by correcting for the endogenous amino acid in the digesta, measured after feeding protein-free diets. An attempt was made to account for the possible influence of dietary fibre on digestility determinations by using protein-free diets which provided similar intakes of crude fibre as that achieved with the cereals.4 littermate male pigs (22 kg live weight) were each surgically modified to create an ileo-rectal anastomosis. The large intestine was sealed at both ends and a cannula exteriorized from its lumen to enable fermenting residues to escape. Pigs were housed in individual metabolism crates. Digesta by-passed the large intestine, were expelled via the anus and collected in trays. The spurious influence of bacteria in the large intestine, on digestibility measurement, was thus avoided.

1. A study was made of factors affecting the plasma concentrations of free amino acids (PAA) and urea (PU) in calves receiving approximately equal daily amounts of concentrates (flaked maize and protein supplements) and straw, the former at 10.00 and 17.00 hours, the latter at 17.00 hours only.2. For calves receiving a diet containing 20 g nitrogen/kg dry matter in which the protein supplement was decorticated, extracted groundnut meal (DCGM) (diet A) there were marked increases in PAA and PU about 1–2 h after a morning feed, then a fall in these values 2 h later to a level which was maintained for the next 3 h. No similar changes occurred after the evening feed. Samples taken 3 h after the morning feed were used in subsequent comparative experiments. There was much more variation between animals than within animals in total PAA, PU and the concentrations of most individual amino acids in these samples.3. Total PAA and most individual amino acid concentrations were not appreciably affected when the DCGM intake was reduced to give 10 g N/kg dry matter in the diet (diet C), but PU was halved. When maize gluten replaced DCGM as the protein supplement at the higher N intake (diet B) then PU doubled, but again total PAA and most individual amino acid concentrations were little affected. Exceptions were arginine, which was halved, and leucine, which was doubled.4. Infusions of more than 4·4 g L-methionine/d into the abomasums of calves (110–160 kg live weight) receiving diet A led to a marked increase in plasma methionine concentration. This was considered to correspond with the point at which methionine requirements were met. Using a chromic oxide marker to estimate flows of methionine and cystine from the rumen to the duodenum, it was calculated that under these conditions the methionine requirement was 9·8 g/d, with a cystine flow of 4·9 g/d. Similar calculations showed the corresponding value to be 7·5 g/d with a cystine flow of 2·8 g/d for calves receiving diet C.5. Infusion of increasing levels of L-lysine into the abomasums of calves (110–160 kg live weight) receiving diet B led to a progressive increase in plasma lysine concentration. There was no consistent change in the rate of increase with increasing amounts infused. Estimated lysine requirement appeared therefore to be less than the flow of lysine from the rumen to the duodenum under these conditions (18·8 g/d).

1. Digesta were collected from seventeen pigs initially of 30 kg live weight fitted with single re-entrant cannulas in either the duodenum, jejunum or ileum. A further twenty-four pigs were used in a conventional digestibility trial.2. The pigs received three types of diet containing: barley, fine wheat offal, white fish meal, minerals and vitamins (diet BWF); starch, sucrose, maize oil, cellulose, minerals and vitamins and either groundnut (diet SSG) or casein (diet SSC).3. Amino acids were measured in samples representative of the digesta flow in 24 h periods and in the faeces collected in 5 d periods.4. For each diet the total flow in 24 h periods in the duodenum for aspartic acid, threonine, serine and glycine exceeded or equalled intake, while the amounts of the other amino acids were usually rather less than intake.5. For each diet in the jejunum, the amounts of glycine and cystine exceeded intake in 24 h periods, while methionine, arginine and tyrosine were the most rapidly absorbed amino acids anterior to the cannula site. On average 0.22, 0.25 and 0.31 of the dietary amino acids were absorbed anterior to the cannula site for diets BWF, SSG and SSC, respectively.6. For each diet in the ileum, the least apparently absorbed dietary amino acids were glycine and cystine. On average 0.81, 0.83 and 0.95 of the dietary amino acids were absorbed anterior to the cannula site for diets BWF, SSG and SSC, respectively.7. There was net disappearance of most amino acids in the large intestine, but some net accumulation occurred in this region.8. The results are discussed in relation to the amino acid composition of endogenous secretions (particularly glycine in bile), protease and peptidase specificity, free amino acid absorption and the role of the microflora in the large intestine.

Abstract Objectives 1) to estimate the probability of inadequate amino acid intake among infants 9–10 months of age in rural Malawi 2) to evaluate whether dietary amino acid intake or protein quality are associated with length gain from 6 to 12 months of age Methods We assessed total amino acid intake from breast milk and complementary foods in 285 infants. Breast milk intake and complementary foods were estimated using dose-to-mother deuterium oxide dilution method and repeat 4-pass interactive 24-hour recall interviews, respectively. Amino acid composition values were taken from FAO human milk profile, Tanzania Food Composition table and International Minilist. Protein quality was estimated using Digestible Indispensable Amino Acid Score (DIAAS). Probability of intake below Estimated Average Requirement (EAR) for each amino acid was estimated using National Cancer Institute (NCI) method. We estimated protein quality of complementary food using median DIAAS. We assumed a DIAAS of ≥0.75 to represent a diet or food with good protein quality. Relationships between amino acid intake or protein quality with length gain were assessed using regression models. Length was measured at 6 and 12 months of age and length for age z-score (LAZ) velocity was calculated (ΔLAZ/months). Results The probability of inadequate amino acid intake from breast milk and complementary food that included a lipid-based nutrient supplement (LNS) was 3% for lysine, 0% for tryptophan, threonine, valine, histidine, isoleucine, leucine, sulfur containing amino acids (SAA), and aromatic amino acids (AAA). Without LNS, the probability was 7% for lysine and 0–2% for the other amino acids. The median (interquartile range) DIAAS for complementary food with and without LNS was 0.70 (0.28) and 0.64 (0.32), respectively. Dietary amino acid intake and protein quality were not significantly associated with length gain velocity from 6 to 12 months even after adjusting for confounding factors. Conclusions The prevalence of inadequate amino acid intake in 9–10 months old infants in rural Malawi is very low. However, in conditions of frequent clinical or sub-clinical infections this situation may be different. Linear growth at 6–12 months does not appear to be limited by dietary amino acid intake or protein quality in this setting. Funding Sources The Bill & Melinda Gates Foundation.

Multidrug resistance-associated protein 3 (MRP3), unlike other MRPs, transports taurocholate (TC). The difference in TC transport activity between rat MRP2 and MRP3 was studied, focusing on the cationic amino acids in the transmembrane domains. For analysis, transport into membrane vesicles from Sf9 cells expressing wild-type and mutated MRP2 was examined. Substitution of Arg at position 586 with Leu and Ile and substitution of Arg at position 1096 with Lys, Leu, and Met resulted in the acquisition of TC transport activity, while retaining transport activity for glutathione and glucuronide conjugates. Substitution of Leu at position 1084 of rat MRP3 (which corresponds to Arg-1096 in rat MRP2) with Lys, but not with Val or Met, resulted in the loss of transport activity for TC and glucuronide conjugates. These results suggest that the presence of the cationic charge at Arg-586 and Arg-1096 in rat MRP2 prevents the transport of TC, whereas the presence of neutral amino acids at the corresponding position of rat MRP3 is required for the transport of substrates.

Экспериментальные исследования кормовых добавок Абиотоник и Чиктоник проводили на базе вивария кафедры эпизоотологии и организации ветеринарного дела, а ветеринарносанитарную экспертизу продуктов убоя цыплятбройлеров проводили на кафедре паразитологии и ветеринарносанитарной экспертизы ФГБОУ ВО МГАВМиБ МВА имени К.И. Скрябина и ФГБНУ ФНЦ ВИЭВ РАН, аминокислотный состав мяса перепелов в Государственном бюджетном учреждении Краснодарского края Кропоткинская краевая ветеринарная лаборатория . Из цыплят в суточном возрасте кросса Кобб500 было сформировано три группы по 10 голов в каждой две опытные и контрольная опытным группам выпаивали кормовые добавки из расчета 1 мл/кг живой массы птицы до 50 суток выращивания, убой птицы проводили на 56 сутки. Тушки птицы после 24х часов созревания в холодильной камере при температуре 4 С подвергали исследованиям по общепринятым методикам: ГОСТ Р 519442002. Мясо птицы. Методы органолептических показателей, температуры и массы ГОСТ 314702012. Мясо птицы, субпродукты и полуфабрикаты из мяса птицы. Методы органолептических и физикохимических исследований аминокислотный состав мяса исследовали согласно М 0438 2009. Корма, комбикорма и сырье для их производства. Методика измерений массовой доли аминокислот методом капиллярного электрофореза с использованием системы капиллярного электрофореза Капель. Применение кормовых добавок в дозе 1 мл/кг живой массы способствовало увеличению живой массы птицы при применении Абиотоника на 54,23, а при применении Чиктоника на 37,70 по отношению к контролю. Во всех исследуемых пробах количество ЛЖК находится в пределах нормы и составило: в 1й опытной 1,390,03 мг КОН, во 2й опытной 1,420,04 мг КОН и в контрольной группе 1,810,06 мг КОН, что говорит о свежести и доброкачественности мяса. Значение рН мяса цыплятбройлеров находилось в трех группах в пределах нормы и не превышало 6,0. When isolated segments of rabbit proximal straight tubules were perfused under oil at 37 degrees C, we observed that droplets of absorbate formed on the peritubular surface. Volume absorption under these conditions was the same as with customary aqueous bathing solutions when calculated either from the rate of absorbate formation (0.39 +/- 0.03 nl X min-1 X mm-1) or from the difference in measured perfusion and collection rates (0.37 +/- 0.04 nl X min-1 X mm-1). Absorbate formation continued at a steady rate for at least 2 h but was inhibited by 71% at 28 degrees C. The absorbate was found to have a composition that differed from the perfusate, as would be expected in the presence of preferential absorption of glucose, amino acids, and HCO-3. The Cl- concentration in the absorbate was 11.2 +/- 1.8 mM less than in the perfusate. The glucose concentration in the absorbate was 4.5 mM compared with 0.9 mM in the perfusate. Finally, the nonmetabolizable amino acid cycloleucine, which was added to the perfusate at 0.35 mM, had a concentration of 2.9 mM in the epithelial cells compared with 1.6 mM in the absorbate. These data establish the usefulness of this technique for examining solute and water absorption in the proximal nephron and show that the absorbate can differ considerably in solute composition from the luminal perfusate.

Pre-implantation embryos can produce and consume amino acids in a manner dependent upon stage of embryonic development (Partridge and Leese 1996 Reprod. Fert. Dev. 8, 945) that may also be predictive of subsequent viability (Houghton et al. 2002 Hum. Reprod. 17, 999). To examine these relationships in the pig, the appearance or depletion of 18 amino acids from a presumptive near-physiological mixture was determined by HPLC in porcine in vitro-produced embryos from the zygote to the blastocyst stage. Cumulus oocyte complexes derived from slaughterhouse prepubertal pig ovaries were matured for 40 h in modified TCM-199 before being fertilized (Day 0) with frozen thawed semen in tris-based medium. After 6 h, presumptive zygotes were denuded and cultured in groups of 20 in NCSU medium modified to contain a physiological mixture of 18 amino acids including 0.1 mM glutamine (NCSUaa). Groups of 2–10 embryos (dependent on stage) were removed on Day 0 (1 cell), Day 1 (2- and 4-cell), Day 4 (compact morula), and Day 6 (blastocyst) and placed in 4 μL NCSUaa for 24 h. After incubation, the embryos were removed and the medium analyzed by HPLC. Each stage was replicated 3–9 times. Since amino acid profiles of 2- and 4-cell embryos were not different, data were combined. Overall, arginine (1.19 ± 0.33), glutamine (0.78 ± 0.34) and threonine (0.05 ± 0.04) were significantly (P < 0.01) depleted from the medium whereas alanine (0.21 ± 0.1), glycine (0.20 ± 0.06), asparagine (0.13 ± 0.5), lysine (0.1 ± 0.03), isoleucine (0.08 ± 0.01), valine (0.05 ± 0.01), leucine (0.04 ± 0.02), phenylalanine (0.03 ± 0.01), and histidine (0.02 ± 0.04) significantly (P < 0.05) accumulated (mean of the 4 sampling timepoints; all values pmol/embryo/h ± SEM). The difference between amino acid accumulation and depletion (balance) was approximately equivalent between Day 0 and the morula stage although turnover (sum of depletion and accumulation) steadily decreased during this period from 3.1 on Day 0 to 1.35 pmol/embryo/h at the morula stage. However, at the blastocyst stage, turnover and balance increased to 6.32 and 2.42 pmol/embryo/h, respectively, i.e. net appearance occurred. Notable changes in amino acid profile during development included decreases in accumulation of asparagine, glutamate, and glycine in the medium and the depletion of glutamine over Days 0, 1, and 4, followed by reversal of these trends by Day 6. These data suggest that pig embryos can alter the accumulation and depletion rates of amino acids in a manner that is dependent on the specific amino acid and the stage of embryonic development. Cardiac myofibrils isolated from trout heart have been demonstrated to have a higher sensitivity for Ca2+ than mammalian cardiac myofibrils. Using cardiac troponin C (cTnC) cloned from trout and mammalian hearts, we have previously demonstrated that this comparatively high Ca2+ sensitivity is due, in part, to trout cTnC (ScTnC) having twice the Ca2+ affinity of mammalian cTnC (McTnC) over a broad range of temperatures. The amino acid sequence of ScTnC is 92% identical to McTnC. To determine the residues responsible for the high Ca2+ affinity, the function of a number of ScTnC and McTnC mutants was characterized by monitoring an intrinsic fluorescent reporter that monitors Ca2+ binding to site II (F27W). The removal of the COOH terminus (amino acids 90–161) from ScTnC and McTnC maintained the difference in Ca2+ affinity between the truncated cTnC isoforms (ScNTnC and McNTnC). The replacement of Gln29 and Asp30 in ScNTnC with the corresponding residues from McNTnC, Leu and Gly, respectively, reduced Ca2+ affinity to that of McNTnC. These results demonstrate that Gln29 and Asp30 in ScTnC are required for the high Ca2+ affinity of site II.

1. Six 45 kg pigs with re-entrant ileal cannulas were used in two 3 × 3 Latin-square design experiments to study the site of absorption of protein and amino acids. Semi-purified diets containing soya-bean meal (SBM), rapeseed meal (RSM) or no protein source (protein-free) were offered at the rate of 1 kg dry matter/d.2. Flow-rates of ileal contents for 24 h collection periods, corrected for recovery of marker, were 3135, 3127 and 1243 ml (SE 390) for SBM, RSM and protein-free diets respectively.3. Amounts of dry matter digested in the small intestine were 730, 669 and 809 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001).4. Nitrogen intakes were 32·6, 29·9 and 5·9 g/d, and amounts digested in the small intestine were 25·7, 20·2 and 1·6 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). Amounts digested in the large intestine were 2·6, 3·7 and 0·7 g/d.5. Total amino acid intakes and amounts collected at the ileum and in the faeces were (g/d): SBM, 177, 24 and 18; RSM, 149, 28 and 22; protein-free 3, 9 and 12. Digestibility in the small intestine was higher for SBM than RSM for seventeen of the eighteen amino acids estimated. Greater quantities of arginine, methionine, cystine and tyrosine were voided in the faeces than passed through the ileal cannulas for pigs receiving the SBM and RSM diets. For those receiving the protein-free diet this was true for each amino acid except proline.6. Significant differences were found between all diets in the concentration of some amino acids in ileal and faecal amino-N, and endogenous protein secretions did not mask the differences between diets.7. Differences in digestibility between SBM and RSM were greater at the ileum than in the faeces. Amino acid fermentation in the large intestine obscured or reduced differences between SBM and RSM.
