AbstractThe Anemone genus is a tuberous geophyte which undergoes a dormancy period during unfavorable environmental conditions for growth. Five species of the Anemone genus naturally grow in several regions of Iran. The diverse uses of Anemone in gardens for landscaping, cut flowers, and potted plants indicate its high ornamental potential. Its dormancy and flowering are influenced by various factors. The present paper was conducted to explore the flowering behavior of Anemone accessions in response to different pre-treatments. For this purpose, tubers of 18 Anemone accessions (A. coronaria and A. biflora) were collected from natural regions of six provinces in Iran. These tubers were subjected to different conditions of non-chilling (20 °C, 90 days), chilling (4 °C, 90 days), GA3 (150 mgL-1; 24 h), and 5-azaCitidine (5-azaC; 40 µM; 24 h) prior to the cultivation. Most of the accessions were able to enter the flowering stage without chilling. The shortest period for the sprouting of tubers (16.89 ± 7.83 days) belonged to 5-azaC pre-treatment. In addition, this treatment accelerated the flowering time (about 30 days earlier) and diameter of the stem, bud, and flower. Morphological characteristics, such as stem height, number of leaves, bud, and petal and the longevity of flowers on the plant were significantly affected by GA3 pre-treatment. Our results indicated a positive correlation between flower length, stem height, and stem diameter with flower longevity under different pre-treatment conditions. The present study demonstrated that accessions Anm3, Anm12, and Anm18 had ornamental values higher than the population mean across four conditions.

Relevance. Morphological features of 10 sorts of tulips in the conditions of early-spring forcing explored. An assessment of the influence of tulip bulb preplanting on the rate of plant growth and development as well as final product yield (cut flowers) taken. Material and methods. 10 varieties of tulips of early and middle-early flowering period: Algarve (DHT), Аvocado (TT), Crispion Sweet (FT), Davenport (FT), Destination (TT), Double Price (DVT), Luba (TT), Surrender (TT), Largo (DVT) и Snowboard (TT) were selected as objects of study. Country of origin - Netherlands. Before planting one part of tulip bulbs were completely peeled of cover scale and the other part were left unpeeled. Each variant of bulb preparation for bedding were evaluated by growth and development rate of plants, as well as the number of cut flowers on the plot. During the growing season, we studied the following morphological characteristics: plant height, flower diameter, flower height, number of leaves. Results. Studies conducted have found that all varieties under research are suitable to early spring forcing and they differ in decorativeness and fully correspond to saleable quality standards. It was found out that in case of complete removing of cover scale, the tulip bulbs root faster, start to bloom faster and give much higher percentage of flowering plants in the plots in comparison with unpeeled samples.

In this study, we aimed to evaluate half-sibling progenies of Myrocarpus frondosus based on morphological characteristics and to select seedlings with high growth vigor and quality. Seedlings were evaluated for shoot height and diameter, shoot height/diameter ratio, and number of leaves. The shoot height showed the highest estimations of linear correlation with the other characteristics. The linear correlation of shoot height between two consecutive evaluations was high from 60 days of cultivation, indicating that selection should be applied after this period. Selection of 101 seedlings within the best progenies resulted in a genetic gain of 15.13% for shoot height. The greatest indirect gain from selection was for the shoot height/diameter ratio, whereas the lowest indirect gain was for the stem diameter. Myrocarpus frondosus progenies can be selected for shoot height at 60 days of nursery cultivation, resulting in high direct gain for growth and indirect gain for stem height/diameter ratio.

The first commercial use of 1-methylcyclopropene (1-MCP) was with cut flowers and potted plants, and treatment with this compound is still the preferred strategy for protection of most ethylene-sensitive crops. Research is continuing into optimum treatment conditions and strategies for efficient application in commerce. In studies using carnation (Dianthus caryophyllus L. ‘White Sim’) petals to determine the optimal conditions for commercial treatment, we noted some aspects of the inhibition response that were not consistent with the current competitive inhibition model of 1-MCP action. Our data are better explained by an alternative model in which 1-MCP binds to a site that is exposed during the allosteric changes that accompany the enzymatic activities of the binding site in the absence of ethylene.

Iris adriatica is indigenous to Mediterranean part of Croatia. It is an attractive plant and has opportunities for introduction into horticulture and the ornamental plant market. Research was conducted in the experimental garden of the Faculty of Agriculture, University of Zagreb, to assess the possibility of growing this species as a pot plant. Research included 360 potted plants placed as randomized block design experiment. The goal of this research was to determine influence of chosen substrates and fertilizers, as well as to explore growth and development dynamic of the species cultivated outdoors from June till August 2010. The following characteristics were measured: plants height, number of leaves, and number of rosettes. Biometric analysis showed that the development of Iris adriatica was significantly influenced only by substrate, while fertilization and interaction of substrate and fertilization showed no significant impact on examined plants characteristics.

Aim: To study the effect of shade net on the growth characteristics of two medicinal plants viz., Tulsi and Indian Borage in terms of plant height, number of leaves, leaf area and flowering in relation to open condition. Place and Duration of Study: Lawspet, Puducherry, India between June 2018 and October 2018. Methodology: Tulsi and Indian Borage plants were grown in similar type of soil. The potted plants were kept in open, 35% and 75% shade net conditions and studied for 3 months. The plant height, number of leaves, and leaf area were measured including number of inflorescence branches for Tulsi. Results: The average air temperature was lower under shade net. Relative humidity was more in shade net conditions with maximum in 75% shade and least in open conditions. The number of leaves of Tulsi and Indian Borage recorded was maximum when grown under 35% shade and least in open conditions. Plant height was maximum in 75% shade for Tulsi and 35% shade in Indian Borage and least in open conditions on both the cases. Leaf area was maximum for both Tulsi and Indian Borage in 75% shade. Conclusion: Growth of the two medicinal plants was influenced by the microclimate conditions. The present study shows that the medicinal plants can be grown in shade net conditions to increase the yield of medicinally useful parts.

Eremophila spp. (Myoporaceae family), endemic to Australia, are evergreen shrubs or small trees occurring in arid, semi-arid, tropical, or temperate regions. In Europe, Eremophila spp. are grown for their horticultural appeal. During 2009 and 2010, extensive wilting was observed on 2-month to 1-year-old potted plants of Eremophila laanii F. Muell., E. glabra subsp. carnosa Chinnock, and E. maculata (Ker Gawl.) F. Muell. grown in a commercial nursery near Catania (southern Italy). Internally, symptomatic plants had conspicuous vascular discoloration from the crown to the canopy. Diseased crown and stem tissues were surface disinfested for 30 s in 1% NaOCl, rinsed in sterile water, plated on potato dextrose agar (PDA) amended with 100 mg/liter of streptomycin sulfate, and incubated at 25°C. A Fusarium sp. was consistently isolated from affected plant tissues. Colonies with purple mycelia and violet reverse colors developed after 9 days. On carnation leaf agar, single-spore isolates produced microconidia on short monophialides, macroconidia that were three to five septate with a pedicellate base, and solitary and double-celled or aggregated chlamydospores. A PCR assay was conducted on two representative isolates (ITEM 12591 and ITEM 12592) by analyzing sequences of the partial CaM gene (coding calmodulin protein) and benA (coding beta-tubulin protein) using the primers as reported by O'Donnell et al. (1). Calmodulin sequences of ITEM 12951 and ITEM 12952 isolates (GenBank Nos. FR671157 and FR671158) exhibited 99.8 and 99.5% identity with Fusarium oxysporum strain ITEM 2367 (GenBank No. AJ560774), respectively, and had 99.5% homology between them. BenA gene sequences of ITEM 12951 (GenBank No. FR671426) exhibited an identity of 100% to F. oxysporum f. sp. vasinfectum strain CC-612-3 (GenBank No. AY714092.1), and benA gene sequences of ITEM 12952 (GenBank No. FR671427) exhibited an identity of 100% to F. oxysporum f. sp. vasinfectum strain LA 140 (GenBank No. FJ466740.1), whereas the homology between the two strains is 99.5%. Morphological characteristics, as well as CaM and benA sequences, identified the isolates as F. oxysporum Schlechtend:Fr. Pathogenicity tests were performed by placing 1-cm2 plugs of PDA from 9-day-old mycelial cultures near the crown on potted, healthy, 3-month-old cuttings of E. laanii, E. glabra subsp. carnosa, and E. maculata. Twenty plants for each species were inoculated with each isolate. The same number of plants served as noninoculated controls. All plants were enclosed for 4 days in plastic bags and placed in a growth chamber at 24 ± 1°C. Plants were then moved to a greenhouse where temperatures ranged from 23 to 27°C. Symptoms identical to those observed in the nursery developed 20 days after inoculation with both strains. Crown and stem discoloration was detected in all inoculated plants after 45 days. Wilting was detected on 15% of plants. Control plants remained symptomless. F. oxysporum was consistently reisolated from symptomatic tissues and identified as previously above. To our knowledge, this is the first report of F. oxysporum causing disease of Eremophila spp. worldwide. Reference: (1) K. O'Donnell et al. Mycoscience 41:61, 2000.

Objective: The objective was to quantify the morphological diversity of Calophyllum brasiliense Cambes from four municipalities of the State of Tocantins employing morphological characteristics, to strengthen information on conservation and future breeding of the species. Methodology: Seeds were collected from four municipalities in the State of Tocantins and one in the State of São Paulo. The following were evaluated: plant height, stem diameter, root length, number of leaves, leaf area, root dry mass, shoot dry mass, total dry mass, and Dickson´s quality index. The data were subjected to univariate analysis of variance, Tocher grouping method, and UPGMA, obtaining a dendrogram through the generalized Mahalanobis distance. Results: The results showed a statistical difference of 1 and 5% probability. Dueré stood out in stem diameter(5.52 mm), Sandolandia in height (34.84 cm) and root length (42.13 cm). Formoso stood out in the number of leaves (34 leaves). Lagoa da Confusão in leaf area (856.28 cm 2 ) and São Paulo in root dry mass (16.20 g), shoot dry mass (12.38 g), total dry mass (16.20 g), and Dickson´s quality index (1.57). Implications: Variations in morphological characteristics can be used as a tool for genetic studies of guanandi progeny accordingto their similarity and/or differences. Conclusions: The morphological divergence evidenced that among the five studied areas it is possible to direct the collection of seeds to subsidize conservation strategies and future breeding of the species.

In this study, microcrystalline cellulose (MCC) was extracted from empty fruit bunch (EFB) with alkaline treatment, bleaching and acid hydrolysis treatment and its properties were compared with commercial MCC. Two conditions were optimized in this study which are fiber consistency and sodium hydroxide (NaOH) concentration in alkaline pretreatment. The obtained optimum consistency and alkaline concentration are 5% and 20%respectively. The physicochemical and morphological characteristics, elemental composition and size distribution of the obtained alpha cellulose and MCC were analyzed in this work. Fourier transform infrared spectroscopy (FTIR) analysis provided clear evidence that the characteristic peak of lignin and hemicellulose were absent in the spectrum of the alpha cellulose and MCC. The difference in surface morphology and aggregation between alpha cellulose, MCC and commercialized MCC were observed by scanning electron microscopy (SEM). The mean length of approximately 251.3 μm, 41.4 μm and 138.6 μm were measured for alpha cellulose, MCC and commercialized MCC respectively. Thus, the isolated MCC from EFB has a good potential to be utilized as reinforcing agent in green composites and may be a precursor for future roselle derived nanocellulose, and thus a promising subject in nanocomposite research.

Sweet potato [Ipomoea batatas (L.) Lam], is an extremely versatile vegetable that possesses high nutritional values. It is also a valuable medicinal plant having anti-cancer, antidiabetic, and anti-inflammatory activities. In July 2020, leaf spot was observed on leaves of sweet potato in Nanchang, China (28°45'51"N, 115°50'52"E), which affected the growth and development of the crop and caused tuberous roots yield losses of 25%. The disease incidence (total number of diseased plants / total number of surveyed plants × 100%) was 57% from a sampled population of 100 plants in the field. Symptomatic plants initially exhibited small, light brown, irregular-shaped spots on the leaves, subsequently coalescing to form large irregular brown lesions and some lesions finally fell off. Fifteen small pieces (each 5 mm2) of symptomatic leaves were excised from the junction of diseased and healthy tissue, surface sterilized in 75% ethanol solution for 30 sec and 0.1% mercuric chloride solution for 2 min, rinsed three times with sterile distilled water and incubated on potato dextrose agar (PDA) plates at 28°C in darkness. A total of seven fungal isolates with similar morphological characteristics were obtained as pure cultures by single-spore isolation. After 5 days of cultivation at 28°C, dark brown or blackish green colonies were observed, which developed brown, thick-walled, simple, or branched, and septate conidiophores. Conidia were 18.28 to 24.91 × 7.46 to 11.69 µm (average 21.27 × 9.48 µm, n = 100) in size, straight or slightly curved, middle cell unequally enlarged, brown to dark brown, apical, and basal cells slightly paler than the middle cells, with three septa. Based on morphological characteristics, the fungal isolates were suspected to be Curvularia plantarum (Raza et al. 2019). To further confirm the identification, three isolates (LGZ1, LGZ4 and LGZ5) were selected for molecular identification. The internal transcribed spacer region (ITS), glyceraldehyde-3-phosphate-dehydrogenase (GAPDH), and translation elongation factor 1-alpha (EF1-α) genes were amplified and sequenced using primers ITS1/ITS4 (Peever et al. 2004), gpd1/gpd2 (Berbee et al. 1999), EF-983F/EF-2218R (Rehner and Buckley 2005), respectively. The sequences of ITS region of the three isolates (accession nos. MW581905, MZ209268, and MZ227555) shared 100% identity with those of C. plantarum (accession nos. MT410571-72, MN044754-55). Their GAPDH gene sequences were identical (accession nos. MZ224017-19) and shared 100% identity with C. plantarum (accession nos. MN264120, MT432926, and MN053037-38). Similarly, EF1-α gene sequences were identical (accession nos. MZ224020-22) and had 100% identity with C. plantarum (accession nos. MT628901, MN263982-83). A maximum likelihood phylogenetic tree was built based on concatenated data from the sequences of ITS, GAPDH, and EF-1α by using MEGA 5. The three isolates LGZ1, LGZ4, and LGZ5 clustered with C. plantarum. The fungus was identified as C. plantarum by combining morphological and molecular characteristics. Pathogenicity tests were conducted by inoculating a conidial suspension (106 conidia/ml) on three healthy potted I. batatas plants (five leaves wounded with sterile needle of each potted plant were inoculated). In addition, fifteen wounded leaves of three potted plants were sprayed with sterile distilled water as a control. All plants were maintained in a climate box (12 h light/dark) at 25°C with 80% relative humidity. All the inoculated leaves started showing light brown flecks after 7 days, whereas the control leaves showed no symptoms. The pathogenicity test was conducted three times. The fungus was reisolated from all infected leaves of potted plants and confirmed as C. plantarum by morphological and molecular identification, fulfilling Koch’s postulates. To our knowledge, this is the first report of C. plantarum causing leaf spot on sweet potato in China. The discovery of this new disease and the identification of the pathogen will contribute to the disease management, provide useful information for reducing economic losses caused by C. plantarum, and lay a foundation for the further research of resistance breeding.
