AbstractMost of the maize (Zea mays L.) varieties in developing countries have low content of micronutrients including vitamin A. As a result, people who are largely dependent on cereal-based diets suffer from health challenges due to micronutrient deficiencies. Marker assisted recurrent selection (MARS), which increases the frequency of favorable alleles with advances in selection cycle, could be used to enhance the provitamin A (PVA) content of maize. This study was carried out to determine changes in levels of PVA carotenoids and genetic diversity in two maize synthetics that were subjected to two cycles of MARS. The two populations, known as HGA and HGB, and their advanced selection cycles (C1 and C2) were evaluated at Ibadan in Nigeria. Selection increased the concentrations of β-carotene, PVA and total carotenoids across cycles in HGA, while in HGB only α-carotene increased with advances in selection cycle. β-cryptoxanthine increased at C1 but decreased at C2 in HGB. The levels of β-carotene, PVA, and total carotenoids increased by 40%, 30% and 36% respectively, in HGA after two cycles of selection. α-carotene and β-cryptoxanthine content improved by 20% and 5%, respectively after two cycles of selection in HGB. MARS caused changes in genetic diversity over selection cycles. Number of effective alleles and observed heterozygosity decreased with selection cycles, while expected heterozygosity increased at C1 and decreased at C2 in HGA. In HGB, number of effective alleles, observed and expected heterozygosity increased at C1 and decreased at C2. In both populations, fixation index increased after two cycle of selections. The greatest part of the genetic variability resides within the population accounting for 86% of the total genetic variance. In general, MARS effectively improved PVA carotenoid content. However, genetic diversity in the two synthetics declined after two cycles of selection.

Carrot (Daucus carota L.) is ranked among vegetables as the most consumed and the best provitamin A provider. Moreover, carrot also contains vitamins, phenolic compounds, and other antioxidant micronutrients. The influence of carrot genetic background on the content of several micronutrients was investigated. Carotenoids and vitamins (C and E) were analyzed by HPLC in 20 varieties of carrot, and antioxidant activity of carrots was investigated with colorimetric methods (ORAC and Folin-Ciocalteu). There were large differences among cultivars in carotenoid content (0.32 to 17 mg/100 g of fresh weight). In yellow and purple carrots, lutein represents nearly half of the total carotenoids. By contrast, in orange carrots, β-carotene represents the major carotenoid (65%). The concentration of vitamin E ranged from 191 to 703 μg/100 g of fresh weight, whereas the concentration in ascorbic acid ranged from 1.4 to 5.8 mg/100 g. For all these components, dark-orange carrots exhibited the highest values. Significant differences among these 20 varieties were also recorded for mineral and total phenolic compound concentrations. Purple and dark-orange carrots could be preferred to usual carrot varieties to benefit from their specific micronutrients (anthocyanins, carotenoids, or vitamin E). ORAC is a complex reflection of phytomicronutrients but is not tightly linked to vitamin C levels, as shown for white carrots, which are rich in this vitamin.

Abstract The present study aimed to assess population structure and phylogenetic relationships of nine subspecies of Brassica rapa L. represented with thirty-five accessions cover a wide range of species distribution area using isozyme analysis in order to select more diverse accessions as supplementary resources that can be utilized for improvement of B. napus. Enzyme analysis resulted in detecting 14 putative polymorphic loci with 27 alleles. Mean allele frequency 0.04 (rare alleles) was observed in Cat4A and Cat4B in sub species Oleifera accession CR 2204/79 and in subspecies trilocularis accessions CR 2215/88 and CR 2244/88. The highest genetic diversity measures were observed in subspecies dichotoma, accession CR 1585/96 (the highest average of observed (H0) and expected heterozygosity (He), and number of alleles per locus (Ae)). These observations make this accession valuable genetic resource to be included in breeding programs for the improvement of oilseed B. napus. The average fixation index (F) is significantly higher than zero for the analysis accessions indicating a significant deficiency of heteozygosity. The divergence among subspecies indicated very great genetic differentiation (FST = 0.8972) which means that about 90% of genetic diversity is distributed among subspecies, while 10% of the diversity is distributed within subspecies. This coincides with low value of gene flow (Nm = 0.0287). B. rapa ssp. oleifera (turnip rape) and B. rapa ssp. trilocularis (sarson) were grouped under one cluster which coincides with the morphological classification.

ABSTRACT Sweetpotato (Ipomoea batatas) is a root crop grown in many countries. This tuberous root is a source of energy, nutrients, and phytochemicals. In this study, bioactive compounds and physical and physicochemical qualities of sweetpotato genotypes were evaluated. Eight new genotypes of sweetpotato produced by Embrapa Hortaliças (orange-fleshed: MD09026-OF and MD09024-OF; cream-fleshed: MD09011-CF, MD09004-CF, MD10039-CF, and MD10004-CF; yellow-fleshed: MD09017-YF and MD12002-YF) and two cultivars used as controls (Beauregard and Brazlândia Roxa) were evaluated for color, soluble solids, dry matter, phenolic compounds, total carotenoids and β-carotene. Hue angles differed even between those sweetpotatoes with the same flesh color. The orange-fleshed genotypes MD09024-OF, MD09026-OF, and Beauregard, had the lowest L*, showing to be darker than the others. These sweetpotatoes also had the brightest flesh colors with higher C*. The orange-fleshed genotypes MD09026-OF and MD09024-OF were sweeter (10.55oBrix and 9.23oBrix) than Beauregard (5.12oBrix). Brazlândia Roxa had the highest dry matter content (38.05%), followed by the genotypes MD10004-CF, MD09017-YF, MD09026-OF MD10039-CF, and MD09011-CF, which showed similarity, ranging from 32.33% to 29.12%. The highest contents of total carotenoids were found for the orange-fleshed genotypes MD09026-OF (80.06 mg g-1) and MD09024-OF (70.56 mg g-1) and Beauregard (73.12 mg g-1). These same genotypes showed the highest total phenolic compounds (0.815 mg g-1 and 0.686 mg g-1, respectively). MD09026-OF showed the highest content of β-carotene (46.47 mg g-1). MD09026-OF was the most prominent genotype among those evaluated, as it showed the highest total carotenoid, β-carotene, phenolic compounds, and soluble solids content, in addition to a high dry matter content.

Aims: The aim of this study was to evaluate the nutritional and functional properties of powder from 2 Cameroonians squash pulp species (Cucurbita moschata Yellow and Orange pulp, and Cucurbita pepo orange pulp) and squash pulp-base biscuit to promote the integration of squash in the diet to help fight against micronutrient deficiencies and non-communicable diseases Methodology: The squash were cleaned and the pulp was blanched in boiling water for 3 minutes and dried at 60 °C for 24 h. The dried pulp was then finely crushed and sieved to obtain the powder. The proximal composition, the levels of macronutrients, vitamin C, carotenoids and minerals were determined as well as the functional properties. The powder sample with the highest carotenoid content was substituted by wheat flour in the proportions 0%, 5%, 10%, 15% and 20% for the biscuit formulation. Sensory properties (color, taste and overall acceptability) and total carotenoids content of the biscuit were then evaluated. Results: The nutrient contents per 100g of powder were 1.65g, 6.38-23.36 mg, 15.70-20.54 mg, 102.56-119.65 mg respectively for crude fibers, total carotenoids, vitamin C and potassium. Water absorption capacity (WAC), Water holding capacity (WHC) and oil holding capacity (OHC) varied from 7.50-8.25, 3.35-6.05 and 1.02-2.04 respectively. Values of swelling capacity (SC) and water solubility index (WSI) varied from 119-140 and 15-17.63, 314-348 and 22.33-24.9, 388-459 and 35.08-38.75 at room temperature, 65°C and 95°C respectively. Sensory analysis of the biscuits showed that the biscuit made with 10% of squash powder was the most appreciated and contained 2.29 mg/100g of total carotenoids. Conclusion: Regular consumption of these Cameroonians squash pulp powder or foods formulated with them could help to combat nutritional disease.

Efforts to increase β-carotene in cassava have been successful, but the ability of high-β-carotene cassava to prevent vitamin A deficiency has not been determined. Two studies investigated the bioefficacy of provitamin A in cassava and compared the effects of carotenoid content and variety on vitamin A status in vitamin A-depleted Mongolian gerbils (Meriones unguiculatus). Gerbils were fed a vitamin A-free diet 4 weeks prior to treatment. In Expt 1, treatments (ten gerbils per group) included 45 % high-β-carotene cassava, β-carotene and vitamin A supplements (intake matched to high-β-carotene cassava group), and oil control. In Expt 2, gerbils were fed cassava feeds with 1·8 or 4·3 nmol provitamin A/g prepared with two varieties. Gerbils were killed after 4 weeks. For Expt 1, liver vitamin A was higher (P < 0·05) in the vitamin A (1·45 (sd 0·23) μmol/liver), lower in the control (0·43 (sd 0·10) μmol/liver), but did not differ from the β-carotene group (0·77 (sd 0·12) μmol/liver) when compared with the high-β-carotene cassava group (0·69 (sd 0·20) μmol/liver). The bioconversion factor was 3·7 μg β-carotene to 1 μg retinol (2 mol:1 mol), despite 48 % cis-β-carotene [(Z)-β-carotene] composition in cassava. In Expt 2, cassava feed with 4·3 nmol provitamin A/g maintained vitamin A status. No effect of cassava variety was observed. Serum retinol concentrations did not differ. β-Carotene was detected in livers of gerbils receiving cassava and supplements, but the cis-to-trans ratio in liver differed from intake. Biofortified cassava adequately maintained vitamin A status and was as efficacious as β-carotene supplementation in the gerbil model.

Apricots are well known for the high content of bioactive compounds such as carotenoids, polyphenols, vitamins and minerals. Several studies have pointed out the chemical composition or the biological effects of apricots, but limited information are available regarding the stability of active compounds during storage or processing. The aim of this study was to determine the stability of major carotenoids in commercial dried apricots during storage.Carotenoids were extracted monthly from dried apricots kept in a dark environment, at room temperature, for twelve months. Total carotenoids were determined using the spectrophotometric method while the most relevant carotenoids were analyzed by high-performance liquid chromatography-photodiode array detection (HPLC-PDA) on a C30 column and using a gradient elution system.Initial carotenoid content of dried fruits was 6.72 mg/100g, while after six months of storage it decreased to 2.46 mg/100g. After twelve months of storage the total carotenoid content was 0.82 mg/100g, representing 20.35 % of the initial concentration. The major carotenoids identified in apricots were: all trans β-carotene, its geometrical isomers (9-cis-β-carotene; 13-cis-β-carotene; 9,13-di-cis β-carotene); β-carotene-5,8-epoxide; β-cryptoxanthin and β-cryptoxanthin palmitate. Significant decreases were observed for all pigments but all trans β-carotene appears to be the most sensitive pigment, with 15.7 % residual concentration. Although the concentrations of β-cryptoxanthin palmitate is small, it has shown significant increased stability compared to carotenes.

Vitamin A deficiency (VAD) is a serious problem in sub-Saharan Africa (SSA) and other parts of the world. Understanding the effect of marker-based improvement (MARS) of two maize synthetics (HGA and HGB) representing different heterotic groups on their agronomic performance, carotenoid content, and combining abilities could help identify suitable sources to develop divergent inbred lines for optimizing heterosis. This study involved three selection cycles each of the two synthetics and their nine varietal-cross hybrids together with a released check variety was conducted across four diverse locations in Nigeria in 2018 and 2019. Environment and hybrid effects were significant on grain yield and other agronomic traits as well as provitamin A content and other carotenoids. Genetic improvement per cycle of MARS in the parental synthetics was 15% for provitamin A, 25% for β-carotene and 26% for lutein in HGA and 4% for grain yield, 3% for zeaxanthin and 5% for α-carotene in HGB. Grain yield and agronomic traits of the two maize synthetics were controlled by additive and non-additive gene effects, while provitamin A content and other carotenoids were mainly controlled by additive gene effects. Some selection cycles which were high in grain yield and provitamin A content were identified as potential sources of new and divergent maize inbred lines in maize breeding programs. Some varietal-cross hybrids expressed significant mid-parent heterosis for grain yield and moderate mid-parent heterosis for provitamin A, β-carotene and xanthophylls. These hybrids could be commercialized at reasonable prices to small-scale farmers in rural areas that are most affected by vitamin A deficiency.

Effects of selected agronomical factors on the content of health-promoting compounds in spaghetti squash (Cucurbita pepoL. subsp.pepo)The study examined the effects of the sowing date (5, 15 and 25 May) and plant spacing (1 × 0.6 m, 1 × 0.8 m and 1 × 1 m) on the content of carotenoids and L-ascorbic acid in the fruits of two cultivars of spaghetti squash (‘Makaronowa Warszawska' and ‘Pyza’). The field experiment was carried out in central-eastern Poland. Fully mature spaghetti squash fruits were harvested from the beginning of September to the beginning of October. The content of total carotenoids and β-carotene was the greatest in the fruits of plants sown on 15 May. The sowing date did not affect the content of L-ascorbic acid. Increasing the plant spacing from 1 × 0.6 m to 1 × 1 m resulted in an increase of total carotenoid content, particularly for the spaghetti squash sown on 25 May. Plant spacing did not have a significant effect on the content of β-carotene and L-ascorbic acid. Fruits of the ‘Makaronowa Warszawska’ with light orange flesh contained more total carotenoids and β-carotene than fruits of the ‘Pyza’ with cream-coloured flesh. The content of L-ascorbic acid in the fruits of both cultivars did not differ significantly.

The paper presents data on the assessment of the genetic diversity of five subpopulations of the European bison (Bison bonasus) in Belarus — from the National Park “Belovezhskaya Pushcha”, the National Park “Pripyatsky”, the Osipovichi district (Mogilev area), SEI “Berezinsky Biosphere Reserve” and the Grodno region. In general, the work includes 30 samples of muscle tissue from the collection of Gene bank of wild fauna in SSPA “SPC NAS of Belarus on Bioresources” (Minsk, Belarus). Microsatellites were used as markers to assess genetic diversity, structure, and search for signs of a sharp decline in the size of bison subpopulations in the past. A total of 11 microsatellite markers were used, recommended by the Food and Agriculture Organization of the United Nations for cattle research. The analysis of B. bonasus subpopulation from the NP “Pripyatsky” showed signs of passing through the genetic bottleneck. All studied subpopulations are characterized by a similarly low genetic diversity level in all analyzed indicators (mean number of alleles, allelic diversity, observed and expected heterozygosity). The expected heterozygosity (He) for the three subpopulations from the NP “Belovezhskaya Pushcha”, the NP “Pripyatsky” and from the Osipovichi district ranged from 0.37 to 0.39. For the studied subpopulations, the values of the fixation index were negative. The assessment of the presence of genetic structuring between the subpopulations of bison from the NP “Belovezhskaya Pushcha”, the NP “Pripyatsky” and from the Osipovichi district based on the values of such indexes as Fst and DJost which showed no signs of genetic differentiation, which is also confirmed by principal coordinates analysis (PCoA). The European bison conservation in Belarus has required tremendous efforts in the past. So far, even though the impressively large population size reached in Belarus, B. bonasus status still should not be considered as stable, which is closely linked to aspects of its overall low genetic diversity. Our research confirmed the low genetic variability of Belarusian subpopulations. Therefore, the more extensive research concentrated on identifying genetic diversity is necessary to ensure the beneficial control of gene flow and register a potential correlation of unfavorable gene variants with possible inbreeding depression. These attempts are required to lay the groundwork for the management and protection of the European bison in Belarus.

An assessment was made of the consumer accessibility by income to carotenoids in the eleven major Brazilian urban centers. The consumption data published by the POF (National Household Budget Survey, 1995-1996) and the Brazilian database on food carotenoids provided the basis for the study. The USDA-NCC Carotenoid Database for US foods was used whenever the carotenoid content was not found locally. Prudent individual daily intakes of beta-carotene (3 to 6mg), pro-vitamins A (5.2 to 6mg) and total carotenoids (9 to 18mg) were far from attained by the poorer households in all of the regions studied, but the availability rose as the level of income increased in all regions. The principal foods identified, which significantly contributed to the carotenoid supply were: (β-carotene) carrots, squash, mango and tomato, (lycopene) tomato, tomato sauce, watermelon and papaya, (lutein and zeaxanthin) corn flour, kale, lettuce and orange. The study suggests that consumption of carotenogenic foods in Brazil may have been low at the time, despite the wide natural distribution and abundance in the country. The implications that low consumption of carotenogenic foods may have on public health came to be better known in more recent years, but the data should be useful when comparing with the 2002/2003 POF.
