Change in tissue concentrations of lipid hydroperoxides, vitamin C and vitamin E in rats with streptozotocin-induced diabetes

The tissue concentration of lipid hydroperoxides, which was determined by a specific method involving chemical derivatization and HPLC, increased significantly in the heart, liver, kidney and muscle of diabetic rats 8 weeks after the intraperitoneal injection of streptozotocin compared with that of the control group. These results demonstrate that an enhanced oxidative stress is caused in these tissues by diabetes. Vitamin C concentrations of the brain, heart, lung, liver, kidney and plasma of the diabetic rats decreased significantly after 8 weeks compared with those of the control group. Vitamin E concentrations of the brain, heart, liver, kidney, muscle and plasma of the diabetic rats increased significantly after 4 weeks compared with the control group. After 8 weeks, an elevation in vitamin E concentration was observed in the heart, liver, muscle and plasma of the diabetic rats.

The effect of Protein isolate from fermented melon seeds (Ogiri Protei Isolates; OPI) of Cucumeropsis manii on blood glucose, lipid profile, and antioxidant enzyme activities in streptozotocin (STZ)-induced diabetic rats was investigated. Thirty Male Wistar rats were divided into five equal groups. GThe first control group with no exposure. The second group of rats with Streptozotocin-induced non-treated diabetes. The 3rd and 4th groups of rats with Streptozotocin-induced diabetes supplemented with Ogiri protein isolates (200, 600 mg/kg in diet). And the 5th group of rats with Streptozotocin-induced diabetes administered glibenclamide in a dose 500 ug/kg in diet [17]. The OPI was administered for 6 weeks. The administration of OPI reduced the blood glucose concentration of the STZ-induced diabetic rats. Sera and hepatic superoxide dismutase, activities of the STZ-induced diabetic rats were significantly (P< 0.05) increased in comparison with the diabetic control rats. Lipid peroxidation of the supplemented OPI diabetic rats was significantly (P< 0.05) decreased in comparison with the diabetic control rats as the administration of OPI to the STZ-induced diabetic rats significantly increased the enzymes’ activities. The concentration of low-density lipoproteins in the OPI supplemented rats was significantly elevated. These data demonstrate that OPI supplements might be beneficial for correcting hyperglycemia but the consumption of OPI can modulate some tissue lipids in a direction not beneficial for CVD risk in patients with diabetes.

Free radical that are released in cigarette smoke, believed to play an important role in male infertility. The free radicalproduces Reactive Oxygen Species (ROS) that is found increased in male smokers. ROS is one of the free radicalscomponent can be neutralized by vitamin C and E as an antioxidant. This study using those agents to the fifteen minuteshalf piece cigarette to expose mice and count their sperm as the fertilility parameter. The research design used was posttest-only control group with twenty mice were divided into five groups. A group of no smoke exposed was categorizedas a baseline control, the exposed groups were consisted of one group without antioxidant, one group with vitamin Ctreatment, a group of vitamin E treatment and at last the combination of vitamin C and E. All groups were treatedfor fourteen days. The results were analyzed using oneway ANOVA with p<0.05. It is obviously proven that thecombination of vitamin C and vitamin E treatment on the usual dose gave the highest sperm count compared to othergroups.

A 30-day experiment involving 720 White Leghorn layer hens was carried out to evaluate the effects of vitamins C and E on leukocyte and heterophil/lymphocytes ratios of the layer hens during the natural summer conditions. The birds were divided into 4 groups, one control with basal diet and three experimental with diet supplemented with vitamin C, vitamin E, or combination of both vitamins. Each group was divided into 4 subgroups. Exposure of layer hens to 31 ± 3 °C and 33 ± 0 °C ambient temperature and 84.6% and 81.5% relative humidity during the experiment caused an increase in the value of temperature humidity index of 15.5 above the threshold value of 70 established for this species. Total white blood cells, lymphocytes, and eosinophil levels were significantly (p < 0.001) higher in all treated groups compared to control. Similarly, heterophil/lymphocyte ratios of 0.51, 0.52, and 0.50 for groups with diet supplemented with vitamin C, vitamin E and vitamins C + E, respectively, were significantly (p < 0.001) lower compared to 0.59 recorded in the control group. However, heterophil, basophil and monocyte values were significantly (p < 0.001) higher in the control group compared to all treated groups. The results of the present study showed that dietary supplementation of vitamins C and E, or their combination, reduces the harmful effect of high ambient temperature and humidity on the leukocyte indicators of exposed layer hens, thereby alleviating the stress imposed on their thermoregulatory mechanism and physiological status.

In this study, the effect of dietary antioxidants, such as astaxanthin and Flavangenol®, and a combination of both, in counteracting oxidative stress in streptozotocin-induced diabetes was investigated. Streptozotocin-induced diabetic rats were divided into four groups: control, astaxanthin, Flavangenol, and combined astaxanthin and Flavangenol (mix group). Each group other than the control group was fed with an astaxanthin diet (0.1 g/kg), Flavangenol diet (2.0 g/kg), or an astaxanthin (0.1 g/kg)-Flavangenol (2.0 g/kg) mixture diet, respectively. After 12 weeks of feeding, the results showed that the lipid peroxide levels of plasma and lens and the plasma triglyceride (TG) level in the mix group were significantly decreased by 44%, 20%, and 20%, respectively, compared with the control group. In the mix group, lipid peroxidation was also significantly reduced by 70% in the liver and 20% in the kidney compared with the control group. Furthermore, the level of urinary 8-hydroxy-2'-deoxyguanosine (8-OHdG) in the mix group was significantly lower, 36%, than the control group. The α-tocopherol concentrations in the plasma, liver, and kidney in the astaxanthin and mix groups were significantly higher, 3-9 times, than in the control group. The degree of cataract formation in the Flavangenol and mix groups tended to be lower than the control group. These results indicate that the combination of astaxanthin with Flvangenol has an improved protective effect on oxidative stress associated with streptozotocin-induced diabetes than either agent used alone. Thus, this combination may be beneficial in preventing the progression of diabetic complications.

Background: Infectious disease remains a health problem for the elderly. The elderly are susceptible to infection,because the body's ability to fight infection decreases immunity. The immune status can be improved through the effortsof immunization, and nutrition. However the elderly are not sufficient to meet the nutritional needs of food, so probioticcould support nutritional adequacy.Objective: Analyze the effect of probiotic Lactobacillus helveticus Rosell-52 and Lactobacillus rhamnosus Rosell-11 onlevels of lymphocytes in the elderly in Tresna Elderly Social Institution Puspakarma Mataram, West Nusa Tenggara.Methods: This was quasi-experimental study, randomized pre-post control group design. The population is elderly aged60-75 years, involving 45 subjects were divided randomly into two groups, namely the treatment groups givenprobiotics and the control group was given a placebo. Probiotics were given for 4 weeks, with the provision of onecapsule a day. Immunity status in this study was lymphocytes. Data examined included the intake of nutrients (protein,vitamin A, vitamin C, vitamin E, Fe, Zn), health status, quality of sleep, and levels of lymphocytes in the elderly. Datawere analyzed using bivariate with different test Independent sample t-test, Mann-Whitney U test, Spearman, Pearson,and Paired t-test.Results: Increasing number of lymphocytes were significantly in the treatment group (χ ± SD: 1.84 ± 1.85) and in thecontrol group (0.95 ± 5.0). There is no relationship between intake (protein, vitamin A, vitamin C, vitamin E, Fe, Zink),physical health status, and quality of sleep with levels of lymphocytes.Conclusion: Probiotics in 4 weeks is proven to increase the number of lymphocytes in the elderly.

Aim: Antioxidants, such as vitamin C and vitamin E, is widely used as supplements. The aim of this study is to analyze the profile of blood glucose, serum insulin, and  HOMA in active teenagers after vitamin C and vitamin E supplementation.Methods: Subjects (14-16 y.o) consisted of 12 boys and 5 girls, divided into 3 groups: control (4 boys, 2 girls), ‘moderate dose’ of vitamin C and vitamin E combination group (5 boys, 1 girls), and ‘high dose’ of vitamin C and vitamin E combination group (3 boys, 2 girls). The treatment was given for 5 days. Vitamin C and vitamin E for ‘moderate dose’ was 500mg;  200IU, and for ‘high dose’ was 1000mg; 400IU. Fasting Blood Glucose (FGB) and 1 hour BG (1hr_BG), fasting serum insulin (FSI) and 1 hour SI (1hr_SI) was collected after treatment. We also calculated the HOMA-IR and HOMA-β.Result: There was no significant difference on FBG, 1hr_BG, FSI, 1hr_SI, HOMA-IR, and HOMA-β (p≥ 0.05). However, mean FBG and 1hr_BG tended to be higher on the treatment groups. The control group had the lowest HOMA-IR and the highest HOMA-β.Conclusions: We suggest that the supplementation of vitamin C and vitamin E in active teenagers is not essential on glucose homeostasis.

We have examined the effect of subchronic methidathion (MD) administration on heart damage, and have evaluated possible ameliorating effects of a combination of vitamins E and C against MD toxicity. The experimental groups were: control group, rats treated with 5 mg/kg MD and rats treated with 5 mg/kg body weight MD plus vitamin E and vitamin C (MD+Vit). The groups were given MD by gavage 5 days a week for four weeks at a dose level of 5 mg/kg/day (MD and MD+Vit) by using corn oil as the vehicle. Vitamin E and vitamin C were injected at doses of 50 mg/kg i.m. and 20 mg/kg i.p., respectively, after the treatment with MD in the MD+Vit group. The levels of malondialdehyde (MDA) were determined in the heart tissue, and the levels of cardiac troponin I (TnI) in serum. An autoanalyser was used to determine the serum activities of cholinesterase (ChE). Histopathological examination was carried out in the heart tissue. MDA significantly increased in the MD group as compared to controls (P <0.01). When MD was given concurrently with vitamins E and C, the increase in MDA was significantly less (P <0.01). ChE activity significantly decreased in the MD group as compared to controls (P <-0.01). When MD was given concurrently with vitamins E and C, the decrease in ChE activity was significantly higher (P <-0.05). The serum TnI levels significantly increased in the MD group as compared to controls (P <-0.01). When MD was given concurrently with vitamins E and C, the increase in the serum TnI was significantly less (P <-0.01). MD caused the diffuse loss of striation and myocytolysis of the cardiomyocytes, whereas the combination of vitamins E and C caused a significant decrease in these effects of MD. In conclusion, subchronic MD administration caused heart damage and, in addition, treatment with a combination of vitamins E and C after the administration of MD reduced heart damage caused by MD.

The aim of this study is to explore the mechanisms by which electroacupuncture (EA) enhances the hypoglycemic effect of exogenous insulin in a streptozotocin- (STZ-) diabetic rats. Animals in the EA group were anesthetized and subjected to the insulin challenge test (ICT) and EA for 60 minutes. In the control group, rats were subjected to the same treatment with the exception of EA stimulation. Blood samples were drawn to measure changes in plasma glucose, free fatty acids (FFA), and insulin levels. Western blot was used to assay proteins involved in insulin signaling. Furthermore, atropine, hemicholinium-3 (HC-3), and Eserine were used to explore the relationship between EA and cholinergic nerve activation during ICT. EA augmented the blood glucose-lowering effects of EA by activating the cholinergic nerves in STZ rats that had been exposed to exogenous insulin. This phenomenon may be related to enhancement of insulin signaling rather than to changes in FFA concentration.

Background Vitiligo is a well-recognized pigmentary disorder of the skin and /or mucous membrane characterized by circumscribed ivory or chalky white macules devoid of identifiable melanocytes. The pathogenesis of vitiligo is complex and still not well understood. According to autocytotoxic hypothesis, oxidative stress has been suggested to be the initial pathogenic event in melanocyte degeneration. The role of free radicals and oxidative damage in the pathophysiology of vitiligo has been documented in recent studies.Objective To evaluate the role of oxidative stress in patients with vitiligo and of healthy controls by measuring levels of the oxidant malondialdehyde (MDA) and antioxidants vitamin C and vitamin E in serum and catalase (CAT) in erythrocytes.Method A total of 80 clinically diagnosed cases of vitiligo and 80 control subjects were included in the study to assess the activity of MDA, vitamin C and vitamin E in serum and CAT in erythrocytes of patients and controls by using the spectrophotometric assay.Result There was statistically significant increase in the levels of MDA in patients with vitiligo compared to the control group (p<0.001). No significant difference was found in the levels of vitamin C (p=0.411) and vitamin E (p=0.771) between the patients with vitiligo and control group. The levels of CAT in the vitiligo patients were found to be significantly lower than those of controls (p<0.001).Conclusion Increased oxidative stress and decreased catalase have been observed in vitiligo patients and the data suggesting that the free radicals may be involved in the destruction of melanocytes or dysregulation of melanogenesis.Kathmandu University Medical Journal Vol.12(2) 2014: 132-136
