Abstract Objective: Cooking oils (edible oils) are vegetable oils which are extensively used for cooking in India. These edible oils are rich in triglycerides, sterol, tocoferol, carotenes and pigments. Upon frying, edible oils gives rise to formation of free radicals and other harmful agents. The degraded products of oils generated while cooking produce unfavorable effects. The present study was designed to evaluate the effects of reused edible oils (sunflower oil and palm oil) on vital organs of experimental animals. Materials and Methods: Wistar rats were fed fresh and reused edible oils - sunflower oil and palm oil (15 gm oil/100gm of feed consumed) and control group received normal food and water for a period of 8 weeks. The parameters studied include changes in physicochemical properties of reused oils, change in body weight of animals, plasma lipid profile, biochemical parameters and histopathological examination. Results: The result showed the change in physicochemical characteristics of reused oils. The animals fed with fresh and reused palm oil significant increase the body weight while reused sunflower oil fed group showed decrease significantly. The biochemical parameters, SGPT (serum glutamate–pyruvate transaminase), SGOT (serum glutamate- oxaloacetate transaminase), and ALP (alkaline phoshphatase) were increased in reused oils fed groups. The histopathological study showed the change in size of liver, heart, kidney and testes cells in reused oil groups. Conclusions: From the present study, it can be concluded that reused sunflower oil and palm oil, can be toxic and can cause considerable damage to the vital organs of the experimental animals.

To assess the effect of methanolic leaf extract of Caladium bicolor on the histomorphology of gastrointestinal tissues of experimental animals. Twenty four Wistar rats (weighing between 175-190 g) were randomly and equally divided into four groups which include one control group (CG) and three treatment groups (TG I, TG II and TG III). The CG was administered with distilled water [2 ml/kg body weight (b.w.)] while TGs I, II and III were administered with 100 ml/kg, 200 ml/kg and 300 ml/kg (b.w.) of C. bicolor extract respectively. All administrations were done orally and once daily for a period of thirty days. The body weight of all animals was recorded at the beginning and end of study. After the period of study, gastric and small intestinal tissues of experimental animals were harvested, processed, converted to tissue blocks and sectioned. Tissue sections were stained using Haematoxylin and Eosin (H&E) technique. Thereafter, stained sections microscopically examined for observable histopathological changes within study tissues. The results of this study showed that exposure to C. bicolor extract causes significant (p < 0.05) body weight loss in TGs I-III compared to CG. In addition, prominent histopathological changes were observed in gastrointestinal tissues of experimental animals in TGs I-III including gastric mucosal surface erosion and intestinal villi degeneration compared to normal gastrointestinal histomorphology of CG animals. These histopathological changes may be associated with toxic effect of phytochemicals constituents of the extract. Therefore, its application for therapeutic purposes needs to be thoroughly re-validated or perhaps disallowed where alternative therapeutic agents with minimal toxic potential exist.

The aim of this study was to evaluate the influence of ligature-induced periodontal disease in pregnant rats on their newborn's health parameters. Twenty-four female adult Wistar rats were divided into two groups: the control group (G1) and the group that was submitted to dental ligatures around second upper molars (G2). After the four week period of development of periodontitis, the female animals were mated with male adult Wistar rats. There were no differences in the body weight of females between the two groups during mating and pregnancy. No differences were observed among the groups in relation to the viable newborn index. However, there were differences in newborn birth weight, explained by the diverse size of the litters. In this study, ligature-induced periodontal disease did not promote changes during pregnancy that resulted in low birth weight in newborn Wistar rats.

Background. Herbal medication is a worldwide and ancient practice, mostly in developing countries, where a large part of the population is involved in this practice. Hence, studies must be conducted to evaluate their safety and efficiency to avoid or prevent toxicological risks due to their usage. In Togo, Carissa spinarum is a medicinal plant belonging to Apocynaceae family, used as an aphrodisiac or to heal some ailments including malaria, sickle cell anemia, hypertension, pain, and asthma. Notwithstanding its several ethnomedicinal benefits, just a few toxicological data associated with its chronic use are available. Objective. Therefore, this study aims to assess the toxicity of an ethanolic root extract of Carissa spinarum in Wistar rats. Methods. The 90-day oral toxicity process following OECD TG 408 guidelines is used. Male Wistar rats received Carissa spinarum root hydroethanolic extract at 500 and 1000 mg/kg for 90 days by oral gavage. Body weight changes, hematological and blood biochemical parameters, organ weight changes, malondialdehyde as a lipoperoxidation marker expressed according to tissue proteins, and histopathology of vital organs were assessed. Results. No signs of toxicity or mortality were observed during the 90 days experiment. Hematological parameters have not shown any treatment-related abnormalities. According to biochemical parameters, an increase in the chloride ion level was observed at 1000 mg/kg p < 0.01 . There was no significant difference between the treated groups and the control group concerning the malondialdehyde concentration, body weight, and organ relative weight. No changes in necropsy and histopathology of vital organs associated with extract treatment were observed. Conclusion. The results indicated that an ethanolic root extract of Carissa spinarum does not cause adverse effects, which can lead to Wistar rats’ death after 90-day oral administration at 500 and 1000 mg.

Sunflower oil contain high concentrations of essential polyunsaturated fat with antioxidant vitamin E. The study was carried out on Swiss Albino healthy mice to evaluate the effects of additional supplementation of sunflower oil in feed on body weight, hematological parameters (total erythrocyte count and hemoglobin concentration) and biochemical parameters (total serum cholesterol, triglycerides, high density lipoproteins, low density lipoprotein and serum creatine). A total of 20, three-weeks old mice were randomly divided into two equal groups ( n=10) as A and B. Group A was considered as control (standard poultry pellet) and group B was supplemented with sunflower oil (5 mL/kg feed) respectively in addition to standard poultry pellet for 50 days. At the end of experimental trial, the mice were sacrificed for analysis of hemato-biochemical parameters. Body weight was significantly (P<0.05) higher in sunflower oil supplemented group compared to control. The total erythrocyte count and hemoglobin concentration were increased significantly (P<0.05) in group B compared to control group A. The total serum cholesterol, HDL and LDL values increased significantly (P<0.05) whereas the triglycerides concentration decreased significantly (P<0.05) in sunflower oil treated group compared to control group. On the other hand serum creatinine value showed non-significant trend to increase (P<0.2). These results suggest that sunflower oil may have beneficial effects to maintain healthy life style. Background: With the increased trend of urbanization of our society, the incidence of various stress related diseases are increasing day by day. People under chronic stress often tends to seek relief through drugs, which may have side effects. But some natural supplements called adaptogen, can be more beneficial in reducing symptoms of stress. Objective: The present animal model experimental study was designed to observe the effects of restraint stress on body weight, serum ALT, AST, Glucose, Cholesterol and their modifications by Ocimum sanctum Linn (tulsi) pretreatment which is an Ayurvedic adaptogen and has a long history of therapeutic use. Study design: For this purpose, 30 albino rats aged 90 to 120 days were included in this study. Twenty rats of experimental group were further subdivided into two groups. One group consisted of 10 rats exposed to one hour restraint stress daily for 7 days and 10 rats of the second group were pretreated with tulsi for 7 days before exposure to stress in the same way. 10 non stressed , non pretreated rats were taken into control group.This study was undertaken in the laboratory of the Department of Physiology,BSMMU during the period from July 2003 to June 2004. Methods: The body weight of all control rats were recorded daily in the morning. Body weight of the experimental rats were recorded just before exposure to stress. After completing the experiment , the animals were sacrificed and blood was collected . Serum glucose, cholesterol and ALT,ASTwere determined by standard laboratory technique. Data were compared among the groups and the results were statistically analyzed using unpaired student t' test. Results : The body weight in untreated stressed group was significantly lower (p < 0.001) than those of the control group and tulsi pretreated group. Serum levels of glucose, cholesterol, aminotrasferases (ALT and AST) were significantly higher (p < 0.001) in stressed group than those of control . Again in Tulsi treated group all these biochemical parameters were significantly lower (p < 0.001) than those of stressed group. This restraint stress-induced changes in body weight and biochemical parameters may be due to hypophagia, altered secretion of various metabolic hormones and neurotransmitters, changes in membrane permeability & hypovolaemia resulting from stress induced secretion of corticosterone and epinephrine through hypothalamo-pituitary adrenal axis activation. Conclusion: Prevention of stress induced changes in biochemical parameter by tulsi pre treatment indicates its anti stressor effect. The study was carried out to know the effects of probiotics (Proetxin® Boost, Novartis Bangladesh Ltd.) and antibiotic (oxytetracycline-Renamycin®, Renata Animal Health) on growth performances and haemato-biochemical parameters of "Shaver Star Bro" broiler chickens during September and October 2003. A total of 20, day old broilers were randomly selected and assigned into four equal groups n = 5) as A, B, C and D. Group A was considered as control group fed with commercial ration while groups B, C and D were fed with commercial ration with the addition of 200 mg probiotics / liter drinking water, 100 mg probiotics and 50 mg antibiotic (oxytetracycline- Renamycin®) / liter drinking water and 100 mg antibiotic (oxytetracycline- Renamycin®) / liter drinking water respectively up to 35 days of age. The results showed that the body weight gains differed significantly (p < 0.05) at the 2nd, 4th and 5th weeks of age in different treatment groups. The meat yield not differed significantly (p > 0.05). The drumstick, wing differed significantly (p < 0.01) and spleen weight differed at p < 0.05 among different groups. The mean haemato-biochemical values of Hb, ESR, PCV, heterophil, eosinophil, basophil, triglyceride, HDL, LDL, SGPT and SGOT were differed significantly (p < 0.01) in different groups. The present findings suggest that supplementation of probiotics has significant effect on growth performance and certain haemato-biochemical parameters of broiler chickens as compared to antibiotic supplementation.

This study aims to determine the effects of open and closed formulated standard diets supplied from different sources on growth performance and internal organ development of laboratory rats. Five-week-old 32 Wistar rats were used. A special control group diet was produced in accordance with the criteria determined by the National Research Council (NRC) (1995). Three different most preferred commercial open and closed-formula diets produced by international and local companies were used as trial groups’ diets. The experiment was carried out for 12 weeks. Weekly feed consumption, body weight change, internal organ weight, intestinal organ weigths and lengths, intestinal villi heigth and crypt depth were measured in groups. The body weight values of the control group and the first group fed with open-formula diet were found at the highest level (P <0.05). The control group diet had a positive effect on small intestine villi heigth and crypt depth (P <0.05). The nutrient contents and energy values of the diets of experimental groups were determined as different from the commercial firm notifications. As a result of the research, it is concluded that the diets prepared with open-formula give more reliable results in the growth performance and development of internal organs of Wistar rats.

PURPOSE: To evaluate the effect of soluble fiber or fructooligosaccharide (FOS) supplementation upon trinitrobenzene sulphonic acid (TNBS)-induced colitis in rats. METHODS: 64 Wistar rats were given water, soluble fiber or FOS intragastrically during 14 days prior to colitis induction with TNBS (n=48) or rectal enema with water (n=16; control group). On the 7th or 14th day following colitis induction the rats were weighed and euthanized in order to determine the colon weight/length ratio and macroscopic and microscopic scores. RESULTS: On the 7th day following colitis induction the body weight had decreased significantly, the colon weight/length ratio had increased and macroscopic and microscopic colon lesions were observed. On the 14th day following colitis induction no difference in body weight was observed, in spite of the persistence of macroscopic and microscopic lesions and increased colon weight/length ratio. Supplementation with soluble fiber or FOS did not revert colon lesions or any of the study parameters. Supplementation with FOS, but not with fiber, was associated with increased colon weight/length ratio on the 14th day. CONCLUSION: Supplementation with soluble fiber or FOS produced no significant impact on TNBS-induced colitis in rats.

AbstractHyperhomocysteinemia (HHC), both basal and after methionine load, may occur due to genetic disorders or deficiencies of nutrients that affect the remethylation or trans-sulphuration pathways during methionine metabolism. HHC is involved in the pathogenesis of many illnesses as a result of its prooxidative effect and its impairment of antioxidative protection. The aim was to examine the effects of subchronic methionine overload on the body weight and standard biochemical parameters in rat serum and to examine whether simultaneous subchronic intraperotoneal administration of methionine alone or together with L-cysteine or N-acetyl-cysteine resulted in a change in the body weight and biochemical parameters in the rat serum. The research was conducted during a three-week period (male Wistar albino rats, n=36, body weight of approximately 160 g, age of 15-20 days), and the animals were divided into a control group and three experimental groups of 8-10 animals each: a) control group (0.9% sodium chloride 0.1-0.2 ml/day); b) methionine (0.8 mmol/kg/bw/day) (MET group); c) methionine (0.8 mmol/kg/bw/day) + L-cysteine (7 mg/kg/bw/day) (L-cys+MET group); and d) methionine (0.8 mmol/kg/bw/day) + N-acetyl-L-cysteine (50 mg/kg/bw/day) (NAC+MET group). In addition to the body weight monitoring, the levels of total homocysteine and the standard biochemical parameters in blood samples (plasma or serum) were determined. The results indicated that monitoring the homocysteine levels and standard biochemical parameters in blood could be used for analysis and could provide an excellent guideline for distinguishing between toxic and non-toxic doses of methionine intake, which may be meaningful for clinical applications.

Physiological stress for a long time can lead to impaired kidney organ function. Snakehead fish meat are known to contain important nutrients such as albumin, glutamine, glycine, cysteine, and various minerals that can improve kidney function. This study was conducted to examine and analyze the effect of snakehead fish supplementation in feed towards body weight, kidney weight, diameter and number of glomerulus in Wistar rats with physiological stress. This study used 20 male Wistar rats divided into 5 groups consisted of 1 control group (P0) and 4 treatment groups (P1: 5%, P2: 10%, P3: 15%, P4: 20%). The treatments were performed with 4 times repetition. This study used Completely Randomized Design (CRD). The variables measured were body weight, kidney weight, diameter and number of renal glomerulus. The data obtained were analyzed using Analysis of Variance (ANOVA) at 5% significance level and continued with Duncan test with 5% significance level. The results of this study were: the supplementation of snakehead fish with 20% of concentration can increase the body weight; concentration 10; 15; and 20% can increase the weight of the kidney; concentrations of 5%, 10%, 15%, and 20% can increase the glomerular diameter in Wistar rats with physiological stress. The supplementation of snakehead fish with concentration of 5% to 20% did not affect the number of glomerulus of Wistar rats with physiological stress. Results of this study is useful as information for people who study the mechanism of kidney repair due to physiological stress.
