Studies of Mineral Reserves in Pea (Pisum sativum) Cotyledons Using Low-Water-Content Procedures

Most of the phytin in pea (Pisum sativum) cotyledons is water soluble. In order to determine where K and P are located it was necessary to use anhydrous or low water content tissue preparation procedures to obtain samples suitable for energy dispersive X-ray analysis studies using a transmission electron microscope. While some protein bodies do contain electron-dense globoid crystals, most do not. Globoid crystals are more prevalent in the abaxial part of the cotyledon where the provascular network is located. When present, globoid crystals contain considerable Mg, and/or Ca along with P and K. Protein bodies that lack globoid crystals still contain considerable P and K with lesser amounts of elements such as S, Cl and Mg. This is consistent with these protein bodies containing K-phytate in the proteinaceous matrix. While there is a lot of K inside the protein bodies, K is widespread in pea cotyledon tissue and could be detected in starch grains, cell walls and the cytoplasmic matrix.

Biosolids (Bs) for use in agriculture are an important way for introducing and transferring TiO2 nanoparticles (NPs) to plants and food chain. Roots of Pisum sativum L. plants grown in Bs-amended soils spiked with TiO2 800 mg/kg as rutile NPs, anatase NPs, mixture of both NPs and submicron particles (SMPs) were investigated by Transmission Electron Microscopy (TEM), synchrotron radiation based micro X-ray Fluorescence and micro X-ray Absorption Near-Edge Structure (µXRF/µXANES) and Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES). TEM analysis showed damages in cells ultrastructure of all treated samples, although a more evident effect was observed with single anatase or rutile NPs treatments. Micro-XRF and TEM evidenced the presence of nano and SMPs mainly in the cortex cells near the rhizodermis. Micro-XRF/micro-XANES analysis revealed anatase, rutile, and ilmenite as the main TiO2 polymorphs in the original soil and Bs, and the preferential anatase uptake by the roots. For all treatments Ti concentration in the roots increased by 38–56%, however plants translocation factor (TF) increased mostly with NPs treatment (261–315%) and less with SMPs (about 85%), with respect to control. In addition, all samples showed a limited transfer of TiO2 to the shoots (very low TF value). These findings evidenced a potential toxicity of TiO2 NPs present in Bs and accumulating in soil, suggesting the necessity of appropriate regulations for the occurrence of NPs in Bs used in agriculture.

In Russia, tetramethylthiuram disulfide (TMTD) is a fungicide widely used in the cultivation of legumes, including the pea (Pisum sativum). Application of TMTD can negatively affect nodulation; nevertheless, its effect on the histological and ultrastructural organization of nodules has not previously been investigated. In this study, the effect of TMTD at three concentrations (0.4, 4, and 8 g/kg) on nodule development in three pea genotypes (laboratory lines Sprint-2 and SGE, and cultivar ‘Finale’) was examined. In SGE, TMTD at 0.4 g/kg reduced the nodule number and shoot and root fresh weights. Treatment with TMTD at 8 g/kg changed the nodule color from pink to green, indicative of nodule senescence. Light and transmission electron microscopy analyses revealed negative effects of TMTD on nodule structure in each genotype. ‘Finale’ was the most sensitive cultivar to TMTD and Sprint-2 was the most tolerant. The negative effects of TMTD on nodules included the appearance of a senescence zone, starch accumulation, swelling of cell walls accompanied by a loss of electron density, thickening of the infection thread walls, symbiosome fusion, and bacteroid degradation. These results demonstrate how TMTD adversely affects nodules in the pea and will be useful for developing strategies to optimize fungicide use on legume crops.

Chitosan-ionic liquid conjugation (CILC), which was prepared through the reaction of 1-(4-bromobutyl)-3-methylimidazolium bromide (BBMIB) with chitosan, was firstly used to prepare functionalized graphene composite via the chemical reduction of graphene oxide (GO). The obtained water soluble graphene-based composite was characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), ultraviolet-visible (UV–Vis) spectroscopy and so on. CILC-RGO showed excellent dispersion stability in water at the concentration of 2.0 mg/mL, which was stable for several months without any precipitate. This may be ascribed to the electrostatic attraction and [Formula: see text]–[Formula: see text] interaction between CILC and graphene.

Hexagonal phase NaYF4:Yb3+, Er3+ nanoclusters were prepared using a facile solvothermal method. The structures and morphologies of the products were investigated using an X-ray powder diffractometer and a transmission electron microscope. Modified by the capping agent of poly(acrylic acid), the as-prepared NaYF4 NCs show excellent water solubility. Under 980-nm excitation, the aqueous solution of the sample displayed a bright green upconversion (UC) fluorescence. The luminescence dynamics were further studied based on the UC excitation pathways and the emission processes. Such water soluble UC samples may find application in the field of biological imaging.

An energy dispersive X-ray (EDX) analytical method was used to study the freeze-dried powder of seeds of field peas (Pisum sativum L.) with good and poor cooking quality. EDX analysis of the electron-dense particles in the freeze-dried powder revealed the presence of high concentrations of Mg, P, and K, suggesting that the particles were protein bodies. Seeds with different cooking quality were compared with respect to the ratios of these elements in the dense particles. Statistical analysis indicated a significant correlation between these ratios and cooking quality.Key words: Pisum sativum, protein bodies, elemental analysis

Spheroid hydroxyapatite nanoparticles were successfully prepared by titrating Ca(NO3)2•4H2O and (NH4)2HPO4 aqueous solution in the reverse microemulsion, which consists of mixed OP-10(surfactant), cyclohexane(oil phase), and isobutanol (cosurfactant).The structure and morphology of the prepared powders were characterized by means of X-ray diffraction (XRD) and transmission electron microscope (TEM). The optimum composition was investigated via the analysis of the aqueous solution conductivity, and the mechanism of aqueous reaction. The effect of the pH value and the amount of surfactant on the particle size were studied. The results indicate that the best conditions are which the concentration of OP-10 and isobutanol both are 0.2 M, when the water content is between 130 ml-250 ml,it results in pure nano-hydroxyapatite spheroid-like powders, and the water content is about 200 ml, whose diameters are 50 nm-80 nm, and good particle diameter distribution, the particle microstructure is homogeneous, good crystal structure and higher crystallinity.

Phytate, the main mineral nutrient storage compound in seeds, is stored inside protein bodies. Phytate is usually concentrated in dense bodies called globoid crystals. In 1985, Lott and co-workers proposed that the balance of (Mg + Ca):K may be important in controlling globoid crystal formation and provided some experimental evidence to support this proposal. To test this hypothesis further, developing Cucurbita fruits were injected with sterile K salt solutions. Squash cotyledons generally have large globoid crystals and a relatively high (Mg + Ca):K ratio. We hypothesized that experimental reduction of the ratio by the addition of K would result in the alteration of the size and number of globoid crystals. Developing seeds were remarkably resistant to attempts to alter the elements taken up for storage. The elemental content of embryo tissues was measured quantitatively with neutron activation analysis. Controls plus those few samples showing a distinct shift in the ratio owing to elevated K content were analyzed further. Energy dispersive X-ray analysis of cryogenically prepared samples was used to study the elemental content of globoid crystals and transmission electron microscopy was used to study the ultrastructure of the protein bodies. Abstract Proccessible FePt3 alloy nanoparticles with sizes smaller than 50 nm open the avenue to novel magnetic sensor, catalytic and biomedical applications. Our research objective was to establish a highly scalable synthesis technique for production of single-crystalline FePt3 alloy nanoparticles. We have elaborated a one-pot thermal decomposition technique for the synthesis of superparamagnetic FePt3 nanoparticles (FePt3 NPs) with mean sizes of 10 nm. Subsequent tiron coating provided water solubility of the FePt3 NPs and further processibility as bidental ligands enable binding to catalyst surfaces, smart substrates or biosensors. The chemical composition, structure, morphology, magnetic, optical and crystallographic properties of the FePt3 NPs were examined using high resolution transmission electron microscopy, high-angle annular dark field-scanning transmission electron microscopy, scanning transmission electron microscopy-energy-dispersive X-ray spectroscopy mapping, Fourier transform infrared-attenuated total reflection, X-ray powder diffraction, X-ray photoelectron spectroscopy, vibrating sample magnetometry and UV–Vis absorption spectroscopy.

In this work, water-soluble and blue-emitting carbon nanodots (CDs) were synthesized from apple peels for the first time via one-step hydrothermal method. The synthetic route is facile, green, economical and viable. The as-prepared CDs were characterized thoroughly by transmission electron microscopy (TEM), X-ray diffraction (XRD), Raman, Fourier transform infrared (FT-IR), X-ray photoelectron (XPS), fluorescence and UV–Vis absorption spectroscopy in terms of their morphology, surface functional groups and optical properties. The results show that these CDs possessed ultrasmall size, good dispersivity, and high tolerance to pH, ionic strength and continuous UV irradiation. Significantly, the CDs had fast and reversible response towards temperature, and the accurate linear relationship between fluorescence intensity and temperature was used to design a novel nanothermometer in a broad temperature range from 5 to 65[Formula: see text]C facilely. In addition, the fluorescence intensity of CDs was observed to be quenched immediately by Cr(VI) ions based on the inner filter effect. A low-cost Cr(VI) ions sensor was proposed employing CDs as fluorescent probe, and it displayed a wide linear range from 0.5 to 200[Formula: see text][Formula: see text]M with a detection limit of 0.73[Formula: see text][Formula: see text]M. The practicability of the developed Cr(VI) sensor for real water sample assay was also validated with satisfactory recoveries.
