Cryogenic fluorescent light microscopy of flash-frozen cells stands out by artifact-free fixation and very little photobleaching of the fluorophores used. To attain the highest level of resolution, aberration-free immersion objectives with accurately matched immersion media are required, but both do not exist for imaging below the glass-transition temperature of water. Here, we resolve this challenge by combining a cryoimmersion medium, HFE-7200, which matches the refractive index of room-temperature water, with a technological concept in which the body of the objective and the front lens are not in thermal equilibrium. We implemented this concept by replacing the metallic front-lens mount of a standard bioimaging water immersion objective with an insulating ceramic mount heated around its perimeter. In this way, the objective metal housing can be maintained at room temperature, while creating a thermally shielded cold microenvironment around the sample and front lens. To demonstrate the range of potential applications, we show that our method can provide superior contrast in Escherichia coli and yeast cells expressing fluorescent proteins and resolve submicrometer structures in multicolor immunolabeled human bone osteosarcoma epithelial (U2OS) cells at −140°C.

Drinking ice-cold water is prohibited in Avicenna’s “The Canon of Medicine” book in which he emphasized that ice-cold water drinking was hazardous for the body organs such as the liver. Little information can be found regarding the effects of ice-cold fluid drinks on liver and its probable sequels on this vital organ. Accordingly, we investigated the effects of long-term ice-cold water drink on the rat liver function and histology. Eighteen male Wistar rats, weighing 180±20 g, were randomly divided into three groups of six as two months ice-cold water drink, CW2M; three months ice-cold water drink, CW3M; and three months room temperature water drink; control group. Upon completion of the care period, a blood sample has been taken for liver enzymes and lipid profile assessment. Liver tissue has also been used for histological studies of H&E staining and microscopic examination. Histological findings showed hepatocellular micro-vesicle formation, necrosis and derangement of the cellular cords and infiltration of Kupffer cells in ice-cold water taken animals. Serum TGs, VLDL-C and ALP significantly increased with sound decrease in FBS and LDL-C in ice-cold water taken animals. It seems that long-term ice-cold water has deleterious functional and structural effects on the liver.

We explored the putative existence of crossmodal correspondences between sound attributes and beverage temperature. An online pre-study was conducted first, in order to determine whether people would associate the auditory parameters of pitch and tempo with different imagined beverage temperatures. The same melody was manipulated to create a matrix of 25 variants with five different levels of both pitch and tempo. The participants were instructed to imagine consuming hot, room-temperature, or cold water, then to choose the melody that best matched the imagined drinking experience. The results revealed that imagining drinking cold water was associated with a significantly higher pitch than drinking both room-temperature and hot water, and with significantly faster tempo than room-temperature water. Next, the online study was replicated with participants in the lab tasting samples of hot, room-temperature, and cold water while choosing a melody that best matched the actual tasting experience. The results confirmed that, compared to room-temperature and hot water, the experience of cold water was associated with both significantly higher pitch and fast tempo. Possible mechanisms and potential applications of these results are discussed.

Abstract Composite materials are known to have many advantages and they can be used in the manufacture of a wide variety of products which make them commercially valuable. However, they may come into contact with various conditions that might lead to weathering. Weathering might lead to discoloration, decrease in durability, decline in mechanical properties and decrease in mass in composites. Hence, it is paramount to study the effects of weathering on composites before they are used in service. For this study, wood polyester composite (WPC), jute polyester composite (JPC) and coir polyester composite (CPC) were fabricated and subjected to four weathering tests - water ageing test at room temperature, water immersion test at 100°C, exposure to external environment and exposure to UV rays. It was seen that CPC samples subjected to water ageing at room temperature and immersion at 100°C showed comparatively higher water absorption rate than the WPC and JPC samples as coir fibres in CPC are hollow at the centremost region. JPC and CPC also turned whitish after 24 hours of water ageing. However, exposing the samples to the environment for a period of 144 hours did not show any significant change in the sample. It was also found that the tensile strength of all the composites decreased after being exposed to UV rays in a QUV spray weathering tester. Additionally, the mechanical properties of the composites were also modelled with analytical techniques (Halphin Tsai method) and FEM analysis and the results were found to be analogous.

Germination of Euonymous americanus in the laboratory has previously been unsuccessful. Ability to germinate Euonymous americanus. commonly known as the american strawberry bush, is important in that it represents a valuable food source for the white-tailed deer. Utilizing the knowledge that its seeds spend a period of time in the rumin fluid of deer during their dormant stage, we were successful in initiating germination. After a three month drying period, the seeds were placed in 25 ml of buffered rumin fluid, pH 8 at 40°C for 48 hrs anaerobically. They were then allowed to dry at room temperature for 24 hrs, placed on moistened filter paper and enclosed within an environmental chamber. Approximately four weeks later germination was detected and verified by scanning electron microscopy; light microscopy provided inadequate resolution. An important point to note in this procedure is that scarification, which was thought to be vital for germination, proved to be unnecessary for successful germination to occur. It is believed that germination was propagated by the secretion of enzymes or prescence of acids produced by microorganisms found in the rumin fluid since sterilized rumin failed to bring about germination.

Background: Natural preservatives are compounds produced by natural ingredients that can suppress bacterial growth and development. Natural preservatives are carried out because most of the preservatives circulating are chemicals and unsafe for the body. One of the natural preservatives is by using garlic extract (Allium sativum L).  Objective: This study is aimed to determine the effectiveness of the antibacterial garlic (Allium sativum L) as a natural preservative in fresh African catfish (Clarias gariepinus).  Method: This research used the Pour Plate iroculation method. African catfish (Clarias gariepinus) which is soaked with garlic (Allium sativum L) with a concentration of 7%, 14% and 21% for 30 minutes, then the fish will be kept at room temperature with a storage period of 24 hours and 48 hours and calculated growth in bacterial numbers with the Colony counter.  Result: Based on the research result, it was found that garlic extract (Allium sativum L) can obstruct the effectiveness of antibacterial in African catfish (Clarias gariepinus) at a concentration of 14%.

<div>Persistent room-temperature phosphorescence (p-RTP) from pure organics is attractive </div><div>due to its fundamental importance and potential applications in molecular imaging, </div><div>sensing, encryption, anticounterfeiting, etc.1-4 Recently, efforts have been also made in </div><div>obtaining color-tunable p-RTP in aromatic phosphors5 and nonconjugated polymers6,7. </div><div>The origin of color-tunable p-RTP and the rational design of such luminogens, </div><div>particularly those with explicit structure and molecular packing, remain challenging. </div><div>Noteworthily, nonconventional luminophores without significant conjugations generally </div><div>possess excitation-dependent photoluminescence (PL) because of the coexistence of </div><div>diverse clustered chromophores6,8, which strongly implicates the possibility to achieve </div><div>color-tunable p-RTP from their molecular crystals assisted by effective intermolecular </div><div>interactions. Here, inspirited by the highly stable double-helix structure and multiple </div><div>hydrogen bonds in DNA, we reported a series of nonconventional luminophores based on </div><div>hydantoin (HA), which demonstrate excitation-dependent PL and color-tunable p-RTP </div><div>from sky-blue to yellowish-green, accompanying unprecedentedly high PL and p-RTP </div><div>efficiencies of up to 87.5% and 21.8%, respectively. Meanwhile, the p-RTP emissions are </div><div>resistant to vigorous mechanical grinding, with lifetimes of up to 1.74 s. Such robust, </div><div>color-tunable and highly efficient p-RTP render the luminophores promising for varying </div><div>applications. These findings provide mechanism insights into the origin of color-tunable </div><div>p-RTP, and surely advance the exploitation of efficient nonconventional luminophores.</div>
