Background: Traditional knowledge has been a legacy of the past to the present. Barks of Ficus hispida Linn. and leaves of Ficus pomifera Wall. (Moraceae) have been used traditionally for the treatment of diabetes in North-east India and many other places. As many drugs have been developed from traditional plants, the authors have taken up the plants for the study of hypoglycemic activity. Objective: To investigate the hypoglycemic activities of the triterpenoids isolated from the plants and their antioxidant activities. Methods: The bioactive compounds were determined by biochemical analysis, antioxidant activity using DPPH method. Hypoglycemic activity was detected using glucose tolerance test in normal rats and alloxan induced diabetic rats with Gliclazide as standard. Results: The biochemicals and trace elements were present in appreciable amounts. Triterpenoids, (1-5), from F. pomifera and 19-hydroxyphlogacantholide (6), 3-O-[ß-D-glucopyranosyl-(1’→2’)-α- L-rhamnopyranosyl-phlogacanthoside] (7) and galanolactone (8) along with stigmasterol (9), stigmasta- 5,22-dien-7-on-3ß-ol (10), 5-(decahydro-1,1,4a-trimethyl-6-methylene-5-yl)-3-methylpent-2- enal (11), stigmasterol glucoside (12) and stigmast-4-en-3-one (13) from F. hispida Linn., respectively, were isolated. The different extracts of the barks and leaves of these plants along with the isolated compounds had antioxidant and hyploglycemic activities. Conclusions: The five triterpenoids (1-5) were isolated from the methanol extract of the leaves of F. pomifera, and compounds (6-13) were isolated from the chloroform extract of the barks of F. hispida. Methanol extract of the leaves of F. pomifera and the chloroform extract of the barks of F. hispida; compounds (1-13) isolated from these two plants reduced DPPH free radicals in a concentrationdependent manner. It was also observed that the methanol and chloroform extracts of the plants, F. pomifera and F. hispida respectively, and the compounds (1, 6 & 7) exhibited anti-diabetic properties and also caused a highly significant reduction in the blood glucose levels of normal rats.

Propolis is one of the natural products produced by kelulut bees and is still not widely used. The type of stingless bee that is the prima donna in the community is Heterotrigona itama. This study aims to determine the phytochemical content of the n-hexane fraction of Heterotrigona itama bee propolis collected from Kutai Kartanegara, East Kalimantan. The n-hexane fraction was obtained from the methanol extract of H. itama propolis by the liquid-liquid partition method. After obtaining the n-hexane fraction, the research continued with a qualitative phytochemical test to identify the compound and determine total phenolic. Antibacterial activity was determined by the agar well diffusion method with a serial concentration in Escherichia coli bacteria. Qualitative phytochemical analysis in the form of color changes showed that the n-hexane fraction of H. itama propolis contained flavonoids, alkaloids, saponins, and tannins. Based on the results, the total phenolic content of the n-hexane fraction sample was 490 mgGAE/100 g. It caused the n-hexane fraction to have lower phenolic content than the methanol extract (792 mg GAE100 g). Furthermore, this result indicated that the non-polar fraction was not substantial enough to extracted phenolic compounds. It correlated to the antibacterial activity of the n-hexane fraction, which was very weak (2  mm ± 1.5) at  200µg/mL concentration.

Background: Micro-organisms are responsible for the transmission of a large number of diseases. It is hard to comprehend the amount of diseases, deaths and economic losses caused by micro-organisms alone. Plants are good sources of eco-friendly and readily available antimicrobial agents. Purpose: The aim of this study was to evaluate the chemical constituents and antimicrobial characteristics of three fractions from ethnomedicinal Icacina trichantha. Oliv. (Icacinaceae).Methods: Methanol extract from Icacina trichantha. Oliv was obtained by maceration and fractionated successively using hexane, and ethyl acetate. The antimicrobial properties of Icacina trichantha. Oliv was assessed using agar cup diffusion method on MRSA, P. aeruginosa, S. typhi, C. krusei, S. dysenteriae, S. pyrogenes, E. coli, K. pneumoniae, C. albicans, and C. tropicalis. Phytochemical screening on fractions was also evaluated using standard methods.Results: Phytochemical screening on fractions revealed the presence of saponins, alkaloids, steroids, tannins, and glycosides. Agar diffusion assay on fractions showed growth inhibitory effect on all the organisms except P. aeruginosa, S. typhi, and C. tropicalis. The MIC revealed that n-hexane fraction was active against MRSA, S. pyrogenes, E. coli, K. pneumonia, C. albicans and C. krusei at 10 mg/mL while S. dysenteriae was active at 5 mg/mL. The ethyl acetate fraction was active against all the organisms at a concentration of 5 mg/mL except P.aeruginosa, S.typhi and C.tropicalis. Methanol fraction showed activity of 5 mg/mL against MRSA, S. pyrogenes, E. coli, S. dysenteriae, C. albicans and C. krusei except for K. pneumoniae with activity at 10 mg/mL. Minimum bactericidal concentration/fungicidal concentration MBC/MFC evaluated on the n-hexane fraction revealed that MRSA, S. pyrogenes, E. coli, K. pneumoniae, S. dysenteriae, C. albicans, and C. krusei were activeat 20 mg/mL, while the ethyl acetate fraction had MBC/MFC of 10 mg/mL against all the organisms except P. aeruginosa, S. typhi, C. tropicalis. Methanol extract had MBC/MFC of 10 mg/mL against MSRA, E.coli and S. dysenteriae whereas S. pyrogenes, K. pneumoniae, C. albicans and C. krusei had MBC/MFC at 20 mg/mL.Conclusion: Icacina trichantha. Oliv. contain constituents with concentration dependent antimicrobial properties based on type of organism. The plant could be useful in the prevention and treatment of multi-resistant disease causing microorganisms.

Dried stem bark powder of Leptadania hastata was subjected to maceration with methanol to afford crude methanol extract, which was partitioned with n-hexane, ethylacetate, chloroform and n- butanol to afford different their respective fractions. Extensive phytochemical screening of the n-hexane fraction using column chromatography resulted to the isolation of a white solid substance. Curcuma aromaticais a plant belonging to genusCurcumaof familyZingiberaceaeand is widely used as supplements in Japan. Rhizomes ofC. aromaticahave curcumin as a major yellow pigment and curdione as a main ingredient of essential oils. In this study, we investigated the affect ofC. aromaticaon CYP3A4 using 1α,25-(OH)2-D3-treated Caco-2 clone cells. Caco-2 cells were treated with methanol extract (0.1 mg ml−1), its hexane soluble fraction (0.1 mg ml−1), curcumin (4 μM) and curdione (20 μM) for 72 hours. Nifedipine was used as a substrate of CYP3A4. Methanol extract, hexane fraction and curdione inhibited the formation of oxidized nifedipine by 50–70%, and curcumin showed no effect. The IC50s of methanol extract, hexane fraction and curdione to oxidized nifedipine formation were 21, 14 and 3.9 μg ml−1(16.9 μM), respectively. The content of curdione in methanol extract was 11.4%. Moreover, all of methanol extract, hexane fraction and curdione decreased CYP3A4 protein expression but had no affect on CYP3A4 mRNA expression. Our results showed that these drugs further decreased the CYP3A4 protein expression level after the protein synthesis was inhibited by cychroheximide. These findings suggest that curdione plays an important role in the CYP3A4 inhibitory activity ofC. aromaticaand curdione might inhibit the activity by accelerating the degradation of CYP3A4.

Objective: Despite advanced treatment options available for colorectal cancer, many reported resistance and unresponsiveness to conventional chemotherapeutic agents. Therefore, it is urgent to discover a novel drug for colon cancer. Sarang Semut (Myrmecodia pendans), an Indonesian native plant, has been studied extensively due to its anti-cancer profiles. This study aimed to evaluate the anti-tumour activity of Sarang Semut in colon cancer cells.Methods: We evaluated cytotoxic activity of methanol extract as well as n-hexane and ethyl acetate fraction towards colon cancer cell lines (Caco-2 and HCT-116 cells) utilizing 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay. The most potent fraction was evaluated further in inhibiting cell survival using MTT assay and cell proliferation using trypan blue exclusion assay as well as a clonogenic assay.Results: Our data showed that the n-hexane fraction of Sarang Semut induces more cell death than the methanol extract and ethyl acetate fraction. Therefore, we analyzed the n-hexane fraction further and found that the inhibitory concentration 50% (IC50) of the n-hexane fraction was 24 and 30 parts per million (ppm) for Caco-2 and HCT-116 cells, respectively. Moreover, it inhibited cell growth as well as cell colony formation, in particular, shown by the plating efficiency (P<0.05) and colony area per seed (P<0.01) of the control group were different to the treatment group.Conclusion: The n-hexane fraction of Sarang Semut demonstrates a high potential antitumor activity in colon cancer cell line.
