Abstract In the present study, we assessed oxidative stress in patients with dilated cardiomyopathy of ischemic or idiopathic etiology. For this reason we measured whole blood reduced glutathione, erythrocyte superoxide dismutase, susceptibility of erythrocyte membranes and erythrocytes to peroxidation, and SH content of erythrocyte membranes in 12 patients (8 men and 4 women, ages 31 to 66 years) with idiopathic dilated cardiomyopathy, in 11 patients (8 men and 3 women, ages 32 to 65 years) with ischemic dilated cardiomyopathy, and in 21 healthy volunteers (12 men and 9 women, ages 25 to 67 years). There was no statistically significant difference between the two patient groups for the indicators studied (P >0.05). Blood glutathione, erythrocyte superoxide dismutase, and membrane SH content of both groups of patients was decreased compared with controls (P <0.05), whereas erythrocyte and membrane susceptibility to peroxidation were increased (P <0.05). We conclude that patients with idiopathic or ischemic dilated cardiomyopathy exhibit abnormalities of a range of markers of increased oxidative stress. These abnormalities may contribute to contractile dysfunction, increased incidence of fatal arrhythmias, and sudden death.

Background: The aim of this study was to determine the levels of lipid peroxidation (MDA) and antioxidants such as reduced glutathione (GSH), catalase (CAT) and superoxide dismutase (SOD) in the blood serum of patients with cirrhosis and liver transplantation. Materials and Methods: In this study, serum malondialdehyde acid (MDA) levels, superoxide dismutase (SOD), reduced glutathione (GSH), and catalase (CAT) activities were measured spectrophotometrically and compared to the results of the healthy control group. Results: SOD, CAT and GSH activities were significantly decreased in the patient groups compared to the healthy control group (p<0.05). MDA levels were significantly higher in the patient group compared to the healthy control group (p <0.05). Conclusion: In conclusion, this study demonstrated that oxidative stress may play an important role in the development of liver cirrhosis and in liver transplantation. This study is the first one to show how MDA, SOD, CAT and GSH levels change in liver cirrhosis and liver transplantation, while further studies are essential to investigate antioxidant enzymes and oxidative stress status in patients with cirrhosis and liver transplantation.

Abstract Diabetes mellitus is a metabolic disorder characterised by hyperglycemia and oxidative stress. The aim of the present study is to explore the antioxidant effect of Spirulina maxima in rat model along with the histopathological observations. Diabetes was induced by feeding 10% fructose solution orally to Wistar rats (n = 6) for 30 days, analysed for plasma blood glucose and the markers of the oxidative stress [catalase (CAT), superoxide dismutase (SOD), reduced glutathione (GSH) and thiobarbituric acid reactive substances (TBARS)]. These biochemical studies were associated with histopathological examination of liver and kidney sections. The microalga Spirulina maxima being rich in proteins and other essential nutrients is widely used as a food supplement. S. maxima at a dose of 5 and 10% per kg and the metformin (500 mg/kg) as reference drug were given orally for 30 days to the diabetic rats. Diabetic rats showed significant (p < 0.001) elevations in plasma blood glucose, thiobarbituric acid-reactive substances and significant reduction in catalase, superoxide dismutase and reduced glutathione activity. Oral administration of 5 and 10% aqueous extract of S. maxima for 30 days restored not only of blood glucose levels but also markers of oxidative stress. Histopathological observations of tissues manifested that the S. maxima administration had the protective and therapeutic effects against fructose-induced abnormalities in diabetic rats. It is concluded that S. maxima is effective in reinstating the antioxidant activity in addition to its antidiabetic effect in type 2 diabetic rats.

Background: The prevalence of anabolic steroids abuse in athletes and non-athletes is associated with the risk of injury to various organs, but there are limited studies of oxidative changes in kidney tissue following nandrolone (N) administration. Objectives: the aim of this study was to investigate the effect of N treatment with and without resistance training (RT) on superoxide dismutase (SOD) concentration and tissue pathology of kidney tissue in rats. Methods: In this experimental study, 20 male Wistar rats were randomly divided into four groups of five rats including 1) control (C), 2) sham (normal saline) (Sh), 3) N, and 4) N + RT. Groups 3 and 4 received 10 mg/kg N peritoneally, and the N + RT group performed 1 m ladder climbing for eight weeks and three sessions per week. SOD levels of kidney tissue were measured by ELISA and radioimmunoassay. Hematoxylin-eosin (H&E) staining was used to evaluate oxidative stress levels in kidney tissue. One-way ANOVA with Bonferroni’s post- hoc tests were used for analysis of research findings in SPSS version 22 (P ≤ 0.05). Results: SOD levels in the C group were higher than the Sh (P = 0.001), N (P = 0.001), and N + RT (P = 0.001) groups. SOD levels were lower in the Sh group than in the N (P = 0.049) and N + RT (P = 0.001) groups. However, there was no significant difference in SOD levels in the N + RT group and N group (P = 0.28). Also, oxidative stress levels were normal in tissue studies in all groups. Conclusions: It seems that Ntreatment with and without RT reduces SOD activity in kidney tissue, but more studies are needed in this regard given the normality of tissue oxidative stress results.

Introduction: Frailty syndrome (FS), as a health problem in elderly, can interfere with the quality of life. Many factors play roles in its occurrence, one of which is oxidative stress. Superoxide dismutase (SOD), in which one of its components is zinc (Zn), is one of the endogenous antioxidants that plays a role in preventing oxidative stress. This study was aimed to know the correlation between serum zinc level (sZn) and erythrocyte SOD (eSOD) activity in non-frail and frail geriatric patients.Methods: This was cross-sectional study with consecutive sampling method conducted in the Geriatric Clinic Cipto Mangunkusumo hospital, from August through September 2014, on 60 patients (30 patients for each group of non-frail and frail). Data were collected from interviews, physical measurements, and venous blood sampling.Results: Among 90% of the subjects had low Zn intake, at the same time, the mean of sZn was 8.41 ± 1.36mol/l and 93.3% of the subjects had experienced Zn deficiency. The mean of eSOD activity was 1526 ± 508U/gHb, and there was no significant difference between the groups. There were no significant correlation between sZn and eSOD activity both in the overall subject (r = -0.04; p = 0.076), as well as the non-frail group (r = -0.099; p=0.604) and frail  group (r = 0.01; p = 0.957).Conclusion: Serum zinc level did not have any significant correlation with the eSOD activity, neither on overall or both non-frail and frail groups.

Objective: The production of reactive oxygen species in exercise causes oxidative stress which disturbs the balance of oxidants and antioxidants, causing destructive effects on cells. The present study aims to investigate the effect of three types of massage (Swedish, Russian, Thai) on serum levels of Malondialdehyde (MDA), Glutathione Peroxidase (GPX) and Superoxide Dismutase (SOD) following one session of exhaustive exercise. Methods: This quasi-experimental study was conducted on 48 female futsal players aged 17-22 years in Zahedan, Iran who were selected using a purposive sampling method, and randomly divided into four groups of Swedish massage (Long strokes with pressing and tapping using hands), Russian massage (Medium to high pressure), Thai massage (Pressure to certain parts of the body) and Control. The exercise program was based on Bruce protocol. Serum levels of MDA, GPX and SOD were measured by before and immediately after exercise and after massage. Data analysis was performed using repeated measures ANOVA, considering a significance level of P≤0.05. Results: In all three types of massage, there was a significant decrease in serum level of MDA (0.22±0.08), and a significant increase in GPX (1.84±0.46) and SOD (10.02±2.86) levels after exhaustive (P=0.001). No significant difference was observed in the control group. Conclusion: It seems that Russian, Thai, and Swedish types of massage can affect the serum levels of the MDA (as an oxidative stress marker) and the antioxidant enzymes of GPX and SOD during the post-exercise recovery period.

South East Anatolia Region has a large genetic plant diversity due to its physical and different climatic charesteristics. These plants are potential sources of antioxidants that prevent oxidative stress caused by oxygen and photons. In recent years, it has become important to study the antioxidant capacity of many molecules found naturally in foods and biological systems. The reason for this is that it is believed that when the consumption of food rich in antioxidants is increased, the risk of developing different degenerative diseases will be reduced. In this study, it was aimed to measure the antoxidant capacity of Quercus infectoria, G.olivier gal seeds grown in Southeastern Anatolia. Q. infectoria gal seeds from Sanlıurfa province were used in our study. Q. infectoria gal seeds were extracted with water, ethanol and methanol and then antioxidant enzyme activities (catalase and superoxide dismutase) and malondialdehyde levels, which are indicators of oxidative stress were determined by spectrophotometric methods. It was found that the antioxidant capacity (catalase and superoxide dismutase activities) of extracts obtained from ethanol and methanol were higher and their malondialdehyde levels were statistically lower than those obtained from water. However, it was determined that there was no statistically significant difference between the antioxidant capacity and malondialdehyde levels of the extracts obtained from methanol compared to the extracts obtained from ethanol. It has been concluded that Q. infectoria gal seed has a effective antioxidant effect. In addition, it was observed that extracts obtained from ethanol and methanol have higher antioxidant capacity than extracts obtained from water.

Background: Reactive oxygen species (ROS) are controlled by endogenous antioxidant systems that include superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and antioxidant vitamins. Defect in these systems may develop oxidative stress, and different studies have shown the profound effect of oxidative stress in the stroke because of high susceptibility of the brain to ROS-induced damage. Superoxide is dismutated to H 2 O 2 by SOD which is further converted to H 2 O and O 2 by GPx or catalase in the mitochondria and the lysosomes. The present study was aimed to investigate the association of SOD and GPx polymorphisms in the development of stroke. Methods: A total 982 subjects ( 674 patients, 308 controls) were recruited in this study. Genotyping of SOD1 ( rs1041740 ), SOD2 ( rs4880 ), GPx1 ( rs1050450 ) were performed by LightCycler real-time PCR using LightSNiPreagents and FastStartDNAMasterHybProbe while genotyping of GPx4 ( rs713041 ) were done by PCR-RFLP were done by PCR-RFLP. All biochemical parameter were measured in automated clinical chemistry analyzer, at department of laboratory medicine. Results: Our study demonstrated that the CC+CT/TT genotype of rs4880 significantly increased the risk of stroke (AOR=1.57, 95%CI=1.10-2.24) and SAD-stroke (AOR=1.64, 95%CI=1.17-2.41) compared with CC genotype. In GPx polymorphsims, rs1050450 showed no association with stoke, while rs713041 and the combination analysis of rs1050450 with rs713041 demonstrated the significant association for the stroke development. Interestingly, the inter-combined effect of genetic polymorphism of SOD/GPx showed the significant association for the stroke. Most of the genotypes of these SNPs demonstrated significant difference between control and stroke cases on different biochemical parameters. Conclusion: This study suggests a possible role for oxidative stress in the risk of stroke and clearly states that genetic component of stroke is polygenic as altered SOD and GPx genes interaction showed increased risk for stroke development

Present study was conducted to investigate postpartum oxidative stress in 20 Surti goats. Blood samples were collected on 0, 7th, 14th, 21st, 30th and 45th days postpartum and analysed for Superoxide Dismutase (SOD), Glutathione Peroxidase (GPx), lipid peroxidation (LPO), reduced Glutathione (GSH) and uric acid. SOD differed significantly between 0, 14th and 21st day postpartum. GPx was significantly low on 14th day and then increased significantly (P<0.01) up to 45th day. Significant (P<0.01) difference was observed between days except 0 and 21st. LPO increased significantly (P<0.01) from 0 to 14th day and then decreased non-significantly up to 45th day. Reduced glutathione was significantly (P<0.05) higher on 0 day. Uric acid was lowest on 0 day and highest on 45th day however they were non-significantly different on 7th, 14th, 30th and 45th day. It can be summarized that on 14th day post kidding, the values of SOD, GPx and GSH were lowest while LPO was highest. Uric acid was significantly (P<0.01) low on the day of kidding. Thus it may be concluded that in Surti goats the period from 0 day to 14th day postpartum is most stressful and critical care should be taken during this period. GPx, SOD along with LPO and GSH can be used as marker of stress during postpartum period.
