Abstract Heat stress (HS) compromises efficient production of pigs, in part by increasing oxidative stress and inflammation. Therefore the aim of this experiment was to investigate if supplementation of the anti-oxidants selenium or superoxide dismutase (SOD) could ameliorate HS in the pig. Sixty-four female Large White x Landrace pigs (av. 27.75) were acclimated one of the following diets: control (standard grower diet), selenium (Se, + 0.3 ppm organic Se), recombinant SOD (rSOD, 50 IU/d) and Se plus rSOD (SS) for 14 days under thermoneutral conditions (TN, 200C). All rSOD was administered top dressed in liquid form. Pigs were then moved to climate rooms and housed under TN or heat-stress (8 h 350C, 16h 280C/d) for 3 days and blood, urine and tissue collected for analysis terminally on d3. All data was analysed using an ANOVA with Tukeys post-hoc tests. Heat stress increased respiration rate (RR), reduced blood pCO2 and HCO3-, which coupled with a lower base excess indicated that the pigs were experiencing respiratory alkalosis. Furthermore HS increased rectal and skin temperatures, and reduced thyroid hormone (T4) concentrations, however HSP 27 and 70 concentrations were not influenced. Heat stress increased plasma adiponectin and IL-1b, reduced SOD and TNF-a, while no changes in IL-6 and leptin were observed. The effects of the anti-oxidant diets were that Se and SS increased RR, however parameters of respiratory alkalosis were not negatively influenced. The Se diet reduced T4 independently of HS. No improvement in plasma SOD was observed with the rSOD diets, but rSOD reduced adiponectin in HS pigs and both rSOD and SS reduced IL-6 overall. In conclusion a reduction in inflammatory responses were observed with the antioxidants Se and rSOD, including during HS and therefore may be a useful addition to existing amelioration strategies to ameliorate seasonal heat stress in pigs.

Heat stress stimulates the production of reactive oxygen species (ROS), which cause oxidative damage in the kidney. This study clarifies the mechanism by which saikosaponin-d (SSd), which is extracted from the roots of Bupleurum falcatum L, protects heat-stressed pig kidney proximal tubular (LLC-PK1) cells against oxidative damage. SSd alone is not cytotoxic at concentrations of 1 or 3 μg/mL as demonstrated by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. To assess the effects of SSd on heat stress-induced cellular damage, LLC-PK1 cells were pretreated with various concentrations of SSd, heat stressed at 42°C for 1 h, and then returned to 37°C for 9 h. DNA ladder and MTT assays demonstrated that SSd helped to prevent heat stress-induced cellular damage when compared to untreated cells. Additionally, pretreatment with SSd increased the activity of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) but decreased the concentration of malondialdehyde (MDA) in a dose-dependent manner when compared to controls. Furthermore, real-time PCR and Western blot analysis demonstrated that SSd significantly increased the expression of copper and zinc superoxide dismutase (SOD-1), CAT, GPx-1 and heat shock protein 72 (HSP72) at both the mRNA and protein levels. In conclusion, these results are the first to demonstrate that SSd ameliorates heat stress-induced oxidative damage by modulating the activity of anti-oxidant enzymes and HSP72 in LLC-PK1 cells.

Heat stress (HS) compromises productivity of pork production, in part as a result of increased oxidative stress and inflammatory responses, particularly within the gastrointestinal tract. This study aimed to investigate whether plant-derived betaine and isoquinoline alkaloids could ameliorate HS in pigs. Fifty female Large White × Landrace grower pigs, which were acclimated to control (CON), control plus betaine (BET), or control plus isoquinoline alkaloids (IQA) diets for 14 days were then exposed to heat stress or thermoneutral condition. Both BET and IQA partially ameliorated increases in respiration rate (p = 0.013) and rectal temperature (p = 0.001) associated with HS conditions. Heat stress increased salivary cortisol concentrations and reduced plasma creatinine, lactate, and thyroid hormone concentrations. Heat stress increased colon FD4 permeability, which was reduced by IQA (p = 0.030). Heat stress increased inflammation in the jejunum and ileum, as indicated by elevated interleukin-1β (p = 0.022) in the jejunum and interleukin-1β (p = 0.004) and interleukin-8 (p = 0.001) in the ileum. No differences in plasma total antioxidant capacity (TAC) were observed with HS, but betaine increased plasma TAC compared to IQA. Dietary BET increased betaine concentrations in the jejunum, ileum (p < 0.001 for both), plasma, liver, kidney (p < 0.010 for all), urine (p = 0.002) and tended to be higher in muscle (p = 0.084). Betaine concentration was not influenced by HS, but it tended to be higher in plasma and accumulated in the liver. These data suggest that betaine and isoquinoline alkaloids supplementation ameliorated consequences of heat stress in grower pigs and protected against HS induced increases in colonic permeability.

With increases in the frequency, intensity and duration of heat waves forecast plus expansion of tropical agriculture, heat stress (HS) is both a current and an emerging problem. As cinnamon has been shown to increase insulin sensitivity, which is part of the adaptive response to HS, the aim of this experiment was to determine if cinnamon could improve insulin sensitivity and ameliorate HS in grower pigs. In a 2 × 2 factorial design, 36 female Large White × Landrace pigs were fed control (0%) vs. cinnamon (1.5%) diets and housed for 7 day under thermoneutral (20 °C, TN) vs. HS conditions (8 h 35 °C/16 h 28 °C, 35% relative humidity). At the completion of the challenge, insulin sensitivity was assessed by an intravenous glucose tolerance test (IVGTT). Heat stress increased parameters such as respiration rate and rectal temperature. Furthermore, biochemical changes in blood and urine indicated the pigs were experiencing respiratory alkalosis. Minimal modelling of parameters of insulin sensitivity showed that HS pigs had a lower insulin response to the IVGTT and improved insulin sensitivity. Cinnamon had additive effects with heat stress, reflected in lowering the insulin area under curve (AUC) and elevated insulin sensitivity compared to TN. However, this apparent improvement in insulin sensitivity did not ameliorate any of the other physiological symptoms of HS.

Abstract Pregnant sows, especially during late-gestation, may be susceptible to heat stress due to increased metabolic heat production and body mass. Therefore, the study objective was to determine the thermoregulatory and physiological responses of sows exposed to increasing ambient temperature (TA) at 3 reproductive stages. In 3 repetitions, 27 multiparous sows (parity 3.22±0.89) were individually housed and had jugular catheters placed 5.0±1.0 d prior to the experiment. To differentiate between reproductive stages, sows were categorized as open (not pregnant, n=9), mid-gestation (59.7±9.6 days pregnant, n=9), or late-gestation (99.0±4.8 days pregnant, n=9). During the experiment, sows were exposed to 6 consecutive 1 h periods of increasing TA (period 1, 14.39±2.14°C; period 2, 16.20±1.39°C; period 3, 22.09±1.87°C; period 4, 26.34±1.39°C; period 5, 30.56±0.81°C; period 6, 35.07±0.96°C), with 1 h transition phases in between each period. Respiration rate (RR), heart rate (HR), skin temperature, and vaginal temperature (TV) were measured every 20 min and the mean was calculated for each period. At the end of each period, blood gases, leukocytes, and red blood cell counts were measured. Overall, RR and HR were greater (P≤0.04; 45.6% and 12.9%, respectively) in late-gestation versus mid-gestation sows. Compared to mid-gestation and open sows, TV tended to be greater (P=0.06) during period 4 (0.18°C and 0.29°C, respectively) and period 5 (0.14°C and 0.18°C, respectively) in late-gestation sows. Blood O2 increased (P&lt; 0.01; 18.1%) for all sows with advancing period, regardless of reproductive stage. Late-gestation sows had reduced (P=0.02; 16.1%) blood CO2 compared to mid-gestation sows, regardless of period. In summary, late-gestation sows appear to be more sensitive to increasing TA as indicated by increased RR, HR, TV, and blood O2, and reduced blood CO2 when compared to mid-gestation or open sows. This change in O2 and CO2, due to increasing RR and heat stress sensitivity of late-gestation sows, may suggest an alteration to the acid-base balance, leading to respiratory alkalosis.

SummaryLong-term heat stress (HS) induced by testicular insulation generates oxidative stress (OS) on the testicular environment; consequently activating antioxidant enzymes such as superoxide dismutase (SOD), glutathione reductase (GR) and glutathione peroxidase (GPx). The aim of this work was to immunolocalize antioxidant enzymes present in different cells within the seminiferous tubule when rams were submitted to HS. Rams were divided into control (n = 6) and treated group (n = 6), comprising rams subjected to testicular insulation for 240 h. After the testicular insulation period, rams were subjected to orchiectomy. Testicular fragments were submitted to immunohistochemistry for staining against SOD, GR and GPx enzymes. We observed immunolocalization of GPx in more cell types of the testis after HS and when compared with other enzymes. In conclusion, GPx is the main antioxidant enzyme identified in testicular cells in an attempt to maintain oxidative balance when HS occurs.

Immunohistochemical methods were applied in the present study to investigate the expression of stress proteins such as metallothioneins (MT), which are metal-binding proteins, and heat shock proteins (Hsp70), as well as an antioxidant enzyme (superoxide dismutase, SOD), in the freshwater crustacean copepod Hemidiaptomus roubaui (Richard, 1888) exposed to cadmium or heat stress. The results show a tissue-specific distribution of MT-like protein after cadmium exposure in the brain and in the nerve cord. Cadmium stress did not provoke inducible Hsp70 or SOD expression. Unlike cadmium, heat stress induced the expression of Hsp70 and SOD in the shell glands, a structure involved in the reproductive function, and more particularly in the formation of the diapause egg envelope. MT expression is not induced in animals exposed to heat stress.

Heat stress induces the hypothalamic-pituitary-adrenal (HPA) axis activation, influences biological responses, and reduces energy metabolism. Geongangbuja-tang (GBT) and its components, Zingiberis Rhizoma (ZOR) and Aconiti Lateralis Radix Preparata (ALRP) have been used to induce energy metabolism; however, the effects of GBT and its ingredients on heat-induced inflammatory responses have not yet been investigated. In this study, we performed an open-field test to evaluate locomotor activity in mice. To assess the effects of GBT and its ingredients on inflammation, the protein levels of c-fos, pro-inflammatory cytokines, and cortisol were measured in the mouse hypothalamus and serum. The results showed that GBT alleviated locomotive activity and reduced c-fos levels in a dose-dependent manner under the heat exposure. After investigating the active constituent of GBT, we found that compared to GBT and ZOR, ALRP significantly suppressed c-fos expression under heat stress. Subsequently, ALRP decreased the expression of pro-inflammatory cytokines, such as interleukin-9 and -13 and prostaglandin, under the heat stress in the mouse hypothalamus. Moreover, treatment with ALRP inhibited cortisol secretion in the mouse serum following heat exposure. These results indicate that GBT and its active component, ALRP, could be the thermoregulatory agents that regulate the HPA axis.

Five adult male dik-dik (Madoqua kirkii) were exposed in a climatic chamber to an air temperature of 45 degrees C. Measurements were made of rectal temperature (Tre) and respiratory frequency (f) and arterial blood samples taken before and during heat exposure were analyzed for pH, PCO2 and PO2. During exposure, Tre and f increased in all animals. In the first 80 min dik-dik displayed thermal tachypnea and minor changes in blood gases. Continued exposure lead to hyperpnea accompanied by a fall in PaCO2 and a rise in pH. PaCO2 at first fell and then increased toward or above control levels. The dik-dik did not display second phase breathing. This observation confirms that second phase breathing is not essential to the development of respiratory alkalosis. The main conclusion of the study is that the dik-dik, unlike another heat-adapted antelope, the wildebeest (Taylor, Robertshaw, and Hoffmann. Am. J. Physiol. 217:907–910, 1969), is unable to resist alkalosis during heat stress.
