ABSTRACT Cladophora glomerata, a macrophytic green alga, is commonly found in the Great Lakes, and significant accumulations occur along shorelines during the summer months. Recently, Cladophora has been shown to harbor high densities of the fecal indicator bacteria Escherichia coli and enterococci. Cladophora may also harbor human pathogens; however, until now, no studies to address this question have been performed. In the present study, we determined whether attached Cladophora, obtained from the Lake Michigan and Burns Ditch (Little Calumet River, Indiana) sides of a breakwater during the summers of 2004 and 2005, harbored the bacterial pathogens Shiga toxin-producing Escherichia coli (STEC), Salmonella, Shigella, and Campylobacter. The presence of potential pathogens and numbers of organisms were determined by using cultural methods and by using conventional PCR, most-probable-number PCR (MPN-PCR), and quantitative PCR (QPCR) performed with genus- and toxin-specific primers and probes. While Shigella and STEC were detected in 100% and 25%, respectively, of the algal samples obtained near Burns Ditch in 2004, the same pathogens were not detected in samples collected in 2005. MPN-PCR and QPCR allowed enumeration of Salmonella in 40 to 80% of the ditch- and lakeside samples, respectively, and the densities were up to 1.6 × 103 cells per g Cladophora. Similarly, these PCR methods allowed enumeration of up to 5.4 × 102 Campylobacter cells/g Cladophora in 60 to 100% of lake- and ditchside samples. The Campylobacter densities were significantly higher (P < 0.05) in the lakeside Cladophora samples than in the ditchside Cladophora samples. DNA fingerprint analyses indicated that genotypically identical Salmonella isolates were associated with geographically and temporally distinct Cladophora samples. However, Campylobacter isolates were genetically diverse. Since animal hosts are thought to be the primary habitat for Campylobacter and Salmonella species, our results suggest that Cladophora is a likely secondary habitat for pathogenic bacteria in Lake Michigan and that the association of these bacteria with Cladophora warrants additional studies to assess the potential health impact on beach users.

Northern Nigeria represents the most populous region in Nigeria and adequate water supply and sanitation typically leaves a lot to be desired in the region in order to prevent water related diseases to the vulnerable communities of the region. This research work was carried out in Kano state with specific emphasis on Tomas dam which is one of the largest dams established for multipurpose use in Nigeria. The research was aimed at evaluating Physico-chemical and Bacteriological quality of Tomas Dam in Kano State. 240 water samples from 5 sampling stations within eight months were analyzed for bacteriological and physico-chemical quality. The physico-chemical parameters indicated Biochemical Oxygen Demand (BOD) and Water Current Speed results were not significantly different (P>0.05), while other physico-chemical parameters analyzed varied significantly (P<0.05). The study also revealed that Dissolved Oxygen (DO), BOD, Turbidity, and Nitrates recorded higher values than WHO and Federal Environmental Protection Agency, Nigeria (FEPA) limits. Total aerobic bacterial counts (TBC) and total coliform counts (TCC) were determined using pour plate and Most Probable Number (MPN) techniques. TBC and TCC were high and exceeded acceptable limits. Faecal coliforms, faecal Streptococciand Klebsiella species constituted 25% of the indicator organisms identified.The observed parameters indicated the Dam was contaminated with pathogenic bacteria. Variations in the weather conditions, animal and anthropogenic interferences were all directly or indirectly related to faecal contamination in the dam. Preventing indiscriminate faecal discharge close to the Dam and sufficient water treatment before consumption are of prime importance to public health as the dam was established for agricultural purposes. The occurrence of pathogenic bacteria in an aquatic system might increase waterborne infections including salmonellosis. This might raise a concern related to the increasing pathogenic bacteria, including the multidrug-resistant Salmonella spp. in the two major rivers, namely Lamnyong and Krueng Aceh crossing the city of Banda Aceh, Indonesia. The objectives of the present study were to isolate and enumerate Salmonella spp. recovered from aquatic environment in Banda Aceh and to determine their antibiotic susceptibility. Bacterial enumeration was performed using most probable number (MPN) method followed by bacterial isolations using selective media Salmonella-Shigella Agar (SSA); whereas, antibiotic susceptibility test was conducted using a procedure as recommended by the National Committee for Clinical Laboratory Standards. There was no significant Salmonella spp. densities/mL of both locations (p = 0.200) although, on average there was about 441.67 Salmonella spp. MPN/mL in Krueng Aceh as opposed to 20.47 MPN/mL of Lamnyong river. There were 57 Salmonella spp. isolates recovered in the sampling sites, i.e. 38 isolates of Lamnyong and 19 isolates of Krueng Aceh. The results of antibiotic susceptibility tests showed that Salmonella spp. isolates recovered from Lamnyong and Krueng Aceh showed antibiotic resistance to azithromycin, tetracycline, and streptomycin. Moreover, Salmonella spp. isolates also showed high resistance to ofloxacin, ciprofloxacin, dan ceftriaxone. All Salmonella spp. isolates were classified as MDR with the number of isolates resistant to 4-6 antibiotics was 33 (57.9%), whereas those that were resistant to 7-9 antibiotics and over 10 antibiotics were 23 (40.4%%) and 5 (8.8%) respectively. The prevalence of MDR Salmonella spp. in the current study suggest that river remain the major reservoirs for the transmission of the pathogen in aquatic systems. An appropriate assessment and monitoring are necessary to reduce the potential health risks and to ensure water quality in the aquatic environment.

School children snacks are food that is found and routinely consumed by children in the school environment.Good and healthy food must be free from hazardous and toxic materials, such as microbial contamination,chemicals and other materials. The consumed food must meet the microbial contamination requirementsbased on SNI (Standar Nasional Indonesia) safety testing. Pathogenic bacteria still a serious problem invarious countries including Indonesia, since they are capable of causing foodborne diseases. Sapeken Islandis located in Sapeken Village, Sapeken District, Sumenep Regency, East Java Province. The purpose of thestudy was to determine the presence of pathogenic bacteria in samples of snacks for school children onSapeken Island. The method used was the Most Probable Number (MPN) for Escherichia coli, Test Numberof Bacillus sp., Test Number of Staphyllococcus sp., Identification of Salmonella, and Total Plate Count(TPC). The test results obtained in the MPN of Escherichia coli included SD M (9.9 ± 3.8) MPN / g, SD 5(8.68 ± 2.73) MPN / g, SD IT (5.08 ± 0.66) MPN / g, and SD 4 (4.26 ± 2.44) MPN / g exceed the SNI limit.JURNAL BIOLOGI UDAYANA 23(2): 60-71 P ISSN: 1410-5292 E ISSN: 2599-285661The results of JPT E. coli in snack empek-empek at SD I and SD 2, and JPT E. coli ojek in all elementaryschools is <3 MPN / g. The conclusion of the study is snack forschool children (empek-empek) and ojek on Sapeken Island does not contain contamination of pathogenicbacteria Escherichia coli, Bacillus sp., Staphylococcus sp., Salmonella identification, and TPC, except forthe JPT E. coli test on empek empek in SD M, SD 5, SD IT and SD 4 that exceed SNI limits.

Water is the main source for life and also the most severe substance caused by pollution. The mandatory parameters for determining microbiological quality of drinking water are total non-fecal Coliform bacteria and Coliform fecal (Escherichia coli). Coliform bacteria are a group of microorganisms commonly used as indicators, where these bacteria can be a signal to determine whether a water source has been contaminated by bacteria or not, while fecal Coliform bacteria are indicator bacteria polluting pathogenic bacteria originating from human feces and warm-blooded animals (mammals) . The water inspection method in this study uses the MPN (Most Probable Number) method which consists of 3 tests, namely, the presumption test, the affirmation test, and the reinforcement test. The results showed that of 15 drinking water samples 8 samples were tested positive for Coliform bacteria with the highest total bacterial value of sample number 1, 15 (210/100 ml), while 7 other samples were negative. From 8 positive Coliform samples only 1 sample was stated to be negative fecal Coliform bacteria and 7 other samples were positive for Coliform fecal bacteria with the highest total bacterial value of sample number 1 (210/100 ml).

Between November 1992 and February 1993, a large outbreak of Escherichia coli O157[ratio ]H7 infections occurred in the western USA and was associated with eating ground beef patties at restaurants of one fast-food chain. Restaurants that were epidemiologically linked with cases served patties produced on two consecutive dates; cultures of recalled ground beef patties produced on those dates yielded E. coli O157[ratio ]H7 strains indistinguishable from those isolated from patients, confirming the vehicle of illness. Seventy-six ground beef patty samples were cultured quantitatively for E. coli O157[ratio ]H7. The median most probable number of organisms was 1·5 per gram (range, <0·3–15) or 67·5 organisms per patty (range, <13·5–675). Correlation of the presence of E. coli O157[ratio ]H7 with other bacterial indicators yielded a significant association between coliform count and the presence of E. coli O157[ratio ]H7 (P=0·04). A meat traceback to investigate possible sources of contamination revealed cattle were probably initially colonized with E. coli O157[ratio ]H7, and that their slaughter caused surface contamination of meat, which once combined with meat from other sources, resulted in a large number of contaminated ground beef patties. Microbiological testing of meat from lots consumed by persons who became ill was suggestive of an infectious dose for E. coli O157[ratio ]H7 of fewer than 700 organisms. These findings present a strong argument for enforcing zero tolerance for this organism in processed food and for markedly decreasing contamination of raw ground beef. Process controls that incorporate microbiological testing of meat may assist these efforts.

ABSTRACT This study assessed the levels of two key pathogens, Salmonella and Campylobacter, along with the indicator organism Escherichia coli in aerosols within and outside poultry sheds. The study ranged over a 3-year period on four poultry farms and consisted of six trials across the boiler production cycle of around 55 days. Weekly testing of litter and aerosols was carried out through the cycle. A key point that emerged is that the levels of airborne bacteria are linked to the levels of these bacteria in litter. This hypothesis was demonstrated by E. coli. The typical levels of E. coli in litter were ∼108 CFU g−1 and, as a consequence, were in the range of 102 to 104 CFU m−3 in aerosols, both inside and outside the shed. The external levels were always lower than the internal levels. Salmonella was only present intermittently in litter and at lower levels (103 to 105 most probable number [MPN] g−1) and consequently present only intermittently and at low levels in air inside (range of 0.65 to 4.4 MPN m−3) and once outside (2.3 MPN m−3). The Salmonella serovars isolated in litter were generally also isolated from aerosols and dust, with the Salmonella serovars Chester and Sofia being the dominant serovars across these interfaces. Campylobacter was detected late in the production cycle, in litter at levels of around 107 MPN g−1. Campylobacter was detected only once inside the shed and then at low levels of 2.2 MPN m−3. Thus, the public health risk from these organisms in poultry environments via the aerosol pathway is minimal.

The consumption of lamb meat in Brazil has increased in the last years but little information about the microbiological quality of this product is available. To evaluate the hygienic-sanitary conditions of lamb carcasses, the quantification of microorganism populations indicators (mesophiles and psychrotrophs; total and thermotolerant coliforms; Escherichia coli; moulds and yeasts) and the pathogenic microorganisms indentification (Salmonella sp. and Listeria spp.) were performed. A total of 60 lamb carcasses were sampled from one abattoir in São Paulo. Swab samples were collected from three points (forequarter, back and hindquarter) on the muscle surface after carcasses final washing. Statistical analysis consisted of descriptive evaluation of the results whose counts were grouped by intervals of microorganism populations. Counts ranged from 1.0 x 10¹ to 8.0 x 10(4) colony-forming unit cm-2 (CFU cm-2) for mesophiles; 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for psychrotrophs; < 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for moulds and yeasts; < 0.3 to > 32.0 most probable number/cm² (MPN cm-2) for total and thermotolerant coliforms and Escherichia coli. Salmonella sp. and Listeria spp. were not found in any of the carcasses. Most carcasses presented low counts for all microorganisms. Overall results may be explained by the small size of the industry where the study was taken. Results suggest that good microbiological quality lamb meat is possible to be obtained, but improvement in hygienic-sanitary conditions is still required.

Objective: The present study was aimed at the biological synthesis of magnetic iron nanoparticles by using the plant extract of Tridax procumbens and also to study their antimicrobial property against gram-negative bacteria (Escherichia coli). Methods: The synthesis of magnetic iron nanoparticles was carried out by the co-precipitation method using biological methods like plant extract as reducing agent and capping agents are biocompatible and non-hazardous. These nanoparticles were characterized by UV-Visible spectroscopy, XRD (X-Ray Diffraction), and SEM (Scanning Electron Microscope). As well as antibacterial activity of the nanoparticles was carried out by agar well diffusion method and Most Probable Number (MPN) method against gram-negative E. coli (Escherichia coli) bacteria. Results: The average crystallite size of Magnetic Nanoparticles (MNPs) was found to be 72 nm by X-ray diffraction. The optical absorption band at wavelengths of 240 nm and 402 nm was obtained from the UV Visible spectrum. Spherical shape morphology was observed in SEM studies. The antibacterial assay clearly expressed that E. coli showed a maximum zone of inhibition (15±0.15 mm) at 2 mg/ml and 1 mg/ml concentration was found for Magnetic Nanoparticles. In the Most Probable Number (MPN) test it is seen that the bacterial count is reduced after adding synthesized NPs into the water sample. Conclusion: The results of the present study conclude that the Magnetic Nanoparticles synthesized using Tridax procumbens leaf extracts is found to be stable and show good antibacterial activity against gram-negative (Escherichia coli) bacteria.

Air merupakan komponen yang sangat penting dalam kehidupan, seperti penggunanaan air sebagai air minum. Air minum juga digunakan oleh pedagang makanan. Tingginya kasus diare di Kelurahan Jati dan Jati Baru Kota Padang dapat dipicu penggunaan air minum yang tidak bersih. Tujuan penelitian ini adalah menentukan adanya kontaminasi oleh bakteri Escherichia coli dalam air minum pada rumah makan dan cafe di Kelurahan Jati dan Jati baru kota Padang. Penelitian ini bersifat deskriptif dengan perlakuan eksperimental terhadap 16 sampel air minum di Kelurahan Jati dan Jati Baru Kota Padang. Pengambilan sampel secara langsung dengan botol steril. Analisis data dengan menggunakan tabel (Most Probable Number) MPN 5-1-1 dan ditemukannya koloni bakteri Escherichia coli pada sampel air minum. Hasil penelitian ini didapatkan 13 sampel adanya kontaminasi air terhadap bakteri Coliform dengan indeks MPN didapatkan 3 sampel menunjukkan nilai indeks MPN yang tinggi yaitu: 2 sampel dengan indeks MPN 240/100 ml dan 1 sampel dengan indeks MPN 98/100 ml. Berdasarkan pemeriksaan Mac Conkey Agar ditemukan nya 13 sampel terdapat koloni bakteri Escherichia coli.
