Plaque-Forming Cells and Humoral Antibody in Rainbow Trout (Salmo gairdneri) Induced by Immersion in a Yersinia ruckeri O-Antigen Preparation

Rainbow trout (Salmo gairdneri) were exposed to the O-antigen of Yersinia ruckeri by various immunization regimens. The passive hemolytic plaque assay was used to show specific splenic plaque-forming cells (PFC) and passive hemagglutination demonstrated humoral antibody titers in fish injected with or immersed in the antigen preparations. Preceding antigen immersion with a 2-min immersion in either a 5.2 or a 2.6% NaCl solution did not affect the numbers of PFC or levels of antibody. Fish held for 2 min in ethyl m-aminobenzoate methanesulfonate (MS-222®) before antigen immersion showed no significant difference from the control fish. Dosage studies showed that the minimal concentration of antigen for induction of PFC by immersion was 5.0 μg/mL. Rainbow trout (Salmo gairdneri) produced antibodies specific for Aeromonas hydrophila. Two parenteral routes yielded no measurable difference in rate of antibody synthesis. Method of bacterin suspension indicated adjuvant suspension was superior in maintenance of humoral antibody and challenge protection when compared to saline suspension of the organism.Antibody synthesis from adjuvant-suspended bacterin lagged approximately 30 days behind the rate of synthesis by fish receiving saline suspended bacteria. The highest measurable antibody titer reached by an individual was 1:64. Individuals receiving saline suspended bacterin showed evidence of loss of titer approximately 2 months after the last antigen inoculation. Definite decrease in titer was noted 5 months after the last antigen inoculation and the return to a probable nonimmune class by approximately one-half of the individuals 7 months after the last inoculation. All individuals receiving adjuvant-suspended bacterin remained in the probable immune class for as long as 6 months following the last bacterin inoculation, but only approximately one-half of the individuals remained in the probable immune class 7 months postinoculation.All fish receiving a weighed quantity of killed A. hydrophila orally each day for 9 consecutive months showed presence of humoral A. hydrophila antibody. This immunization procedure produced 50% of the individuals with humoral antibody titers considered to be in the probable immune class.Fish receiving no immunization remained with negative antibody titers throughout the experimental period.Definite protection was given fish which had been immunized by parenteral inoculation.Similar protection was given after immunization by either intramuscular or intraperitoneal routes. Fish receiving either saline or adjuvant-suspended bacterin parenterally showed analogous protection when challenged by the specific bacteria. Fish receiving oral bacterin received questionable protection against parenteral challenge of the specific organism.Phagocytosis was found to occur to a greater extent in immunized rainbow trout than in nonimmunized rainbow trout. An average phagocytic index of parenterally immunized rainbow trout was 1.08. Nonimmunized rainbow trout of the same age and egg source had an average phagocytic index of 0.30. The opsonic index was 3.6.

Preferred temperatures of underyearling rainbow trout (Salmo gairdneri) were determined in both vertical and horizontal temperature gradients. No statistically significant difference was found between the preferred temperatures by the two different methods. This suggests that the nature of the gradient plays a lesser role than generally believed in laboratory investigations of temperature preference.

In this study, effects of Yersinia ruckeri antigens on the immune mechanisms of rainbow trout (Oncorhynchus mykiss) were examined. The weight of the 120 fish used in this study was 20–30 g. After injecting 1 mg of formalin-inactivated whole cells (FKC) and O-antigen (Ag-O) intraperitoneally, blood was taken from the caudal vein of anaesthetized fish and metabolic activity of leukocytes (Nitroblue tetrazolium (NBT) activities), phagocytic activity (PA), phagocytic index (PI), serum protein and serum total immunoglobulin (TIg) levels were determined on day 30 after the first immunization. The same procedure was conducted in the control group. In all the experimental groups, considerable increases in the immune indicators were found and significant differences detected between the control and experimental groups (p < 0.05). Metabolic activity of leukocytes decreased significantly (p < 0.05) during the following treatment with antigens compared to the control fish group.

Dorsal aorta blood samples were taken from cannulated rainbow trout (Salmo gairdneri) exposed to pH 4.0 (acid group) or pH 7.0 (controls) in normocapnic conditions at 10 °C. Over a 5-d period there was a significant gradual decrease in pH and total CO2 in the acid group but no significant difference in pO2 and lactate compared to the controls. After uncannulated rainbow trout were exposed to the same conditions for 12 d there were significant increases in hemoglobin, hematocrit, and erythrocyte levels in caudal vein samples from the acid group. The results show that rainbow trout exposed to acid without hypercapnia develop acidaemia which is not a result of anaerobic respiration. The increase in erythrocyte concentration probably offsets the effects of acidaemia upon blood oxygen carrying capacity. Differences in ambient pCO2 and/or species differences could account for varying acid-base values in acid exposed fish reported by different workers. We assessed the influence of hypoxia, which is likely to reduce the productivity of fish culture, on the digestibility of nutrients in rainbow trout (Salmo gairdneri). Rainbow trout (seven to each diet) were kept for 8 d in water with an oxygen saturation level of 40.0 ± 4.0%, while control fish were kept in 89.4 ± 4.2% oxygen-saturated water. Three isonitrogenous and isocaloric diets composed of different sources of protein (animal, plant, and mixed) were used. The apparent digestibility coefficients (ADC) were calculated by the indirect method (chromic oxide) after the automatic collection of feces. The experimental hypoxia used did not change the ADC for protein (total nitrogen), gross energy, or dry matter. The amino acids only showed a tendency for better absorption: proline, glycine, alanine, and tryptophan had a significantly higher ADC (P < 0.05) in experimental fish than in control fish when the animal protein based diet was used. Finally, we noted that feed intake was diet dependent. In contrast with what we observed with the commercial (mixed protein sources) and plant protein based diets, the fish receiving the animal protein based diet maintained a normal (or returning to normal) feed intake when they were subjected to hypoxia.

Implicit in biotelemetry studies is the assumption that transmitter attachment does not affect fish behavior or performance. We conducted experiments to determine effects of external, surgical, and stomach tag attachments on the swimming performance and behavior of rainbow trout (Salmo gairdneri) and white perch (Morone americana). Only one rainbow trout changed dominance rank after dummy tag attachment. Subordinate fish had significantly lower weights than subdominant and dominant fish, but there were no significant differences in exhaustion times. Externally tagged trout had significantly lower exhaustion times than other tagged groups and controls. There was no significant difference in exhaustion times among tagged white perch and controls. Externally and surgically tagged perch contracted serious fungal infections during a 45-d survival study; however, few disease and no survival problems among tagged and untagged rainbow trout were noted up to 21 d. With all factors taken into account, it appears that stomach tagging is the best method of transmitter attachment, except when regurgitation and/or stomach atrophy are likely to be encountered.

1. Duplicate groups of rainbow trout (Salmo gairdneri) were each given partially purified diets which were either adequate or depleted in selenium for 40 weeks.2. Although there was no significant difference in weight gain, liver Se concentration was significantly lower in fish given the deficient diet.3. Glutathione (GSH) peroxidase (EC 1. 11. 1. 9) activity was significantly reduced in liver of Se-deficient fish but a differential assay did not indicate the presence of a non-Se-dependent GSH peroxidase activity, although liver GSH S-transferase (EC 2. 5. 1. 18) was significantly increased.4. Perfusion of livers from trout given Se-adequate diets with t-butyl hydroperoxide (BuOOH) or hydrogen peroxide caused an increase in the rate of release of glutathione disulphide (GSSG) into the perfusate.5. Perfusion of livers from Se-deficient trout with BuOOH or H2O2 did not result in any change in rate of release of GSSG into the perfusate.6. These findings confirm the absence of any compensatory non-Se-dependent peroxidase activity in Se-depleted trout.

Exposure of rainbow trout (Salmo gairdneri) to 5.7 × 10−10 M (5 × 10−5 mg/L) morpholine during smoking period did not influence the subsequent neural response to this substance, when tested immediately and 12 mo after smoking. No significant difference in the morpholine-induced responses was obtained between the exposed and unexposed fish. Using N-ethylmaleimide, a sulfhydryl reagent, it was further confirmed that the morpholine-effect is caused by a mechanism not directly associated with the normal olfactory function. Perfusion of the olfactory organs with 1.0 × 10−1 and 1.1 × 10−1 M (1%) morpholine for 10 min irreversibly inhibited the olfactory responsiveness to odorants. Sensitization of the olfactory bulbar electrical response is discussed in connection with homing of morpholine-imprinted salmonid fishes.
