Polar vineyard pruning extracts increase the activity of the main ligninolytic enzymes in Lentinula edodes cultures

Lentinula edodes is considered an alternative recycling agent for agricultural wastes, and there have been several studies to understand the relationship between its growth and ligninolytic activity. We tested the effect of wood from viticulture pruning, extracted with solvents of differing polarity, on the biomass production and activity pattern of ligninolytic enzymes. The analysis was done by measuring the mycelial dry mass and enzyme activity of liquid growth medium during the culture of L. edodes, adding either single extracts or a combination of extracts. Polar extracts enhanced mycelial production, and the activity patterns of lignin peroxidase, manganese peroxidase, aryl alcohol oxidase, and laccase were comparable to their activities predicted by ligninolysis models proposed for other fungi. We conclude that the polar extracts could be useful for enhancing fungal biomass production and for modifying lignin degradation because the regulation of ligninolytic enzyme activity is differentially influenced by the polarity of the extract.

Ligninolytic enzymes are known as extracellular enzymes produced by the white rot fungi class of basidiomycetes. One of the most well-known fungi of the white rot fungus isPleurotus ostreatus. The aim of this study to calculate the activity of ligninolytic enzymes in the growth media of Pleurotus ostreatusand their application in decolorization of dye colour. The ligninolytic enzyme extract obtained was used to decolorize bluedyes (MethyleneBlue)and red dyes(Congo Red). The highest laccase enzyme activity was in the first month of 0.35 U/mL with E1 media composition; the highest manganese peroxidase (MnP) enzyme activity was in the fourth month at 31.818 U / mL with E4 media composition; and the highest lignin peroxidase (LiP) enzyme activity was in the fifth month at 0.269 U / mL with E1 media composition. The enzyme extract obtained was then applied to decolorize red and blue dyes. Decolorization of dyes was measured using spectrophotometry with a blue wavelength of 470 nm and red 685 nm. The highest reduction in decolorization of blue dye and red dye was 12 hours with concentration of enzyme addition of 0.5%. Penelitian ini bertujuan menghitung aktivitas enzim ligninolitik pada media pertumbuhan jamur tiram  (Pleurotus ostreatus) dan aplikasinya dalam dekolorisasi zat warna.  Ekstrak enzim ligninolitik yang didapatkan kemudian dimanfaatkan untuk dekolorisasi zat warna biru(Methylene Blue)dan merah (Congo Red). Aktivitas enzim lakase tertinggi ada pada bulan pertama sebesar 0,35 U/mL dengan komposisi media E1; aktivitas enzim mangan peroksidase (MnP) tertinggi ada pada bulan keempat sebesar 31,818 U/mL dengan komposisi media E4; dan aktivitas enzim lignin peroksidase (LiP) tertinggi ada pada bulan kelima sebesar 0,269 U/mL dengan komposisi media E1. Ekstrak enzim yang didapat kemudian diaplikasikan untuk dekolorisasi zat warna merah dan biru. Dekolorisasi zat warna diukur menggunakan spektrofotometri dengan panjang gelombang biru pada 470 nm dan merah pada 685 nm. Penurunan dekolorisasi zat warna birudan zat warna merahtertinggi selama 12jam dengan konsentraasi penambahan enzim sebesar 0,5%.Berdasarkan hasil tersebut, enzim ligninolitik sangat potensial untuk dikembangkan sebagai agen bioaktif untuk deterjen.[Kata kunci: dekolorisasi, lakase, mangan peroksidase, lignin peroksidase,  spektrofotometri]

Lentinula edodes 3565 and Ganoderma lucidum 9621 were compared for their ability to produce lignocellulolytic enzymes in submerged (SM) and surface liquid (SL) fermentation of hydrolysed colza straw lignin waste that remained after the production of furfural and bioethanol (CS lignin). Application of cultivated mushrooms to dispose of pretreated colza straw agricultural waste is an approach to decrease the quantity of residual lignin while simultaneously obtaining active substances, e.g., the ligninolytic enzyme complex from mycelium. The effect of adding CS lignin to culture media on the yield of L. edodes and G. lucidum mycelium and extracellular laccase activity was studied. It was revealed that the mycelial growth of G. lucidum on solid media was significantly improved by adding CS lignin. Laccase activity during SL cultivation of L. edodes on medium with CS lignin gradually increased over the experiment starting on day 21 and peaked at 520 U/mL on day 28. G. lucidum expressed the maximum laccase activity, 540 U/mL, during the first 14 days of mycelium SM cultivation. Extracellular laccase activity was enhanced about 35- to 40-fold at cultivation of L. edodes and about 10- to 15-fold in the case of G. lucidum by supplementing liquid culture media with CS lignin.

In this work, the following genuses of mycelial fungi, capable of producing ligninolytic enzymes of various actions, were considered:Penicillium, Aspergillus, Fusariumand Altermaria. Fungi of the genus Aspergilluswere capable of producing laccase, manganese peroxidase and lignin peroxidase in the medium. Penicillium mostly produced laccase. Fusariumproduced laccase, aryl alcohol oxidase, manganesedependent peroxidase, manganese-independent peroxidase and lignin peroxidase. Alternariaproduced laccase, lignin peroxidase and manganese peroxidase. The results demonstrated the possibility of using specific substrates in the study of enzyme activity, as well as the influence of some factors introduced into the medium on the synthesis of enzymes. The auxiliary influence of these fungi on the synthesis of ligninolytic enzymes in symbiosis with otherswas considered. Abstract. Stress-related stimuli may be presented outside of awareness and may ultimately influence health by causing repetitive increases in physiological parameters, such as blood pressure (BP). In this study, we aimed to corroborate previous studies that demonstrated BP effects of subliminally presented stress-related stimuli. This would add evidence to the hypothesis that unconscious manifestations of stress can affect somatic health. Additionally, we suggest that these findings may be extended by measuring affective changes relating to these physiological changes, using measures for self-reported and implicit positive and negative affectivity. Using a repeated measures between-subject design, we presented either the prime word “angry” ( n = 26) or “relax” ( n = 28) subliminally (17 ms) for 100 trials to a student sample and measured systolic and diastolic BP, heart rate (HR), and affect. The “angry” prime, compared to the “relax” prime, did not affect any of the outcome variables. During the priming task, a higher level of implicit negative affect (INA) was associated with a lower systolic BP and diastolic BP. No association was found with HR. Self-reported affect and implicit positive affect were not related to the cardiovascular (CV) activity. In sum, anger and relax primes elicited similar CV activity patterns, but implicit measures of affect may provide a new method to examine the relationship between (unconscious) stress and health.

Abstract Introduction Few studies have examined circadian phase after job loss, an event that upends daily routine. It is common that a daily routine begins with the consumption of breakfast, and breakfast behavior may contribute to health status in adults. Therefore, we sought to examine whether a later midpoint of sleep was associated with breakfast skipping among adults whose schedules were no longer dictated by employment. Methods Data were obtained from the Assessing Daily Activity Patterns Through Occupational Transitions (ADAPT) study. The sample of 155 participants had involuntarily lost their jobs in the last 90 days. Both cross-sectional and 18-month longitudinal analyses assessed the relationship between sleep midpoint after job loss and current and later breakfast skipping. Assessment periods were 14 days. Sleep was measured via actigraphy, and breakfast skipping was measured via daily diary (1 = had breakfast; 0 = did not have breakfast). The midpoint of sleep was calculated as the circular center based on actigraphy sleep onset and offset times. Results The midpoint of sleep at baseline was negatively associated with breakfast consumption at baseline (B = -.09, SE = .02, p = .000). Also, a later midpoint was associated with breakfast skipping over the next 18 months (estimate = -.08; SE = .02; p = .000). Prospective findings remained significant when adjusting for gender, ethnicity, age, perceived stress, body mass index (BMI), education, and reemployment over time. Education (estimate = 14.26, SE = 6.23, p < .05) and BMI (estimate = -.51, SE = .25, p < .05) were the only significant covariates. No other sleep indices predicted breakfast behavior cross-sectionally or prospectively. Conclusion Consistent with research in adolescents, unemployed adults with a later circadian phase are more likely to skip breakfast more often. Breakfast skipping was also associated with higher BMI. Taken together, these findings provide support for the future testing of sleep/wake scheduling interventions to modify breakfast skipping and potentially mitigate weight gain after job loss. An investigation was undertaken into the relationship between the enzyme activity of cells harvested from raw milk and time taken for age gelation (TAG) to occur in the milk after ultra-high-temperature processing. It was shown that there was no relationship between the TAG and the bacterial counts on milk agar at 30°C or 7°C nor was there a relationship between the counts and the level of enzyme activity of the harvested cells. There was, however, a significant correlation between the level of enzyme activity of the harvested cells and the TAG. When extra bovine leucocytes were added to raw milk before processing, the TAG was increased. This suggested that there was an inhibitory action of leucocytes in development of age gelation.

Abstract In 424 African-American and 75 white subjects, we found that the −5 (TPI 592 A→G), −8 (TPI 589 G→A), and −24 (TPI 573 T→G) variants in the triosephosphate isomerase (TPI) gene occurred frequently (41.0%) in the African-American subjects but did not occur in the whites. These data suggest that this set of polymorphisms may turn out to be one of the higher-incidence molecular markers of African lineage, a surprising finding because others had reported that these nucleotide substitutions were restricted to a small subset of African Americans who had been characterized as TPI-deficiency heterozygotes. Additionally, we investigated the relationship of these variants to TPI-enzyme activity. Although the variant substitutions (occurring in three haplotypes: −5 alone, −5 −8, and −5 −8 −24) were associated with moderate reduction in enzyme activity, severe-deficiency heterozygotes could not be identified with certainty, and none of the haplotypes were restricted to subjects with marked reduction of enzyme activity. Three subjects were homozygous for the −5 −8 haplotype, a finding inconsistent with the putative role of this haplotype as the cause of a null variant incompatible with life in homozygotes. Despite these findings, the possibility remains that the −5 −8 or −5 −8 −24 haplotypes may in some instances contribute to compound heterozygosity and clinical TPI deficiency. © 1998 by The American Society of Hematology.
