Diarrhea is one of the most common causes for thousands of deaths every year. Therefore, identification of new source of antidiarrheal drugs becomes one of the most prominent focuses in modern research. Our aim was to investigate the antidiarrheal and cytotoxic activities of methanolic extract ofMaranta arundinacealinn. (MEMA) leaves in rats and brine shrimp, respectively. Antidiarrheal effect was evaluated by using castor oil-induced diarrhea, enteropooling, and gastrointestinal motility tests at 200 mg/kg and 400 mg/kg body weight in rats where the cytotoxic activity was justified using brine shrimp lethality bioassay at different concentrations of MEMA. The extract showed considerable antidiarrheal effect by inhibiting 42.67% and 57.75% of diarrheal episode at the doses of 200 and 400 mg/kg, respectively. MEMA also significantlyp<0.01reduced the castor oil-induced intestinal volume (2.14 ± 0.16 to 1.61 ± 0.12 mL) in enteropooling test as well as intestinal transit (33.00 to 43.36%) in GI motility test, compared to their respective control. These observed effects are comparable to that of standard drug loperamide (5 mg/kg). On the other hand, in brine shrimp lethality test after 24 h, surviving brine shrimp larvae were counted and LD50was assessed. Result showed that MEMA was potent against brine shrimp with LD50value of 420 µg/mL. So the highest dose of 400 µg/mL of MEMA was not toxic to mice. So these results indicate that bioactive compounds are present in methanolic extract ofMaranta arundinacealeaves including significant antidiarrheal activity and could be accounted for pharmacological effects.

The present study was designed to investigate the thrombolytic activity and cytotoxic potential of the methanolic extract of bark of Terminalia bellerica Roxb. The cytotoxicity was assessed with the brine shrimp lethality bioassay and thrombolytic effect with human blood. The brine shrimp lethality bioassay was used to evaluate cytotoxicity (LC50 = 3.21 ?g/ml) compared to vincristine of sulphate (LC50 = 0.512 ?g /ml). It was also evaluated as thrombolytic agent as compared to streptokinase. It has significant thrombolytic activity (32.95%) compared to standard streptokinase (70%). This investigation is made upon the plant Cuscuta reflexa, the flowers of it, to find out its Cytotoxicity property. The anti-oxidant property of this plant part was investigated using methanol extraction. Methanolic extract of Cuscuta reflexa.Was used to evaluate its cytotoxicity in Brine shrimp lethality bioassay where vincristine sulphate was used as standard drug. In Brine shrimp lethality bioassay, LC50 value of the extract was 36.72?g/ml and vincristine sulphate served as the positive control showed LC50 value 10.51?g/ml. So, compared to vincristine sulphate, it is evident that the methanol extract of flowers of Cuscuta reflexa was cytotoxic. In case of anti-oxidant the scavenging power (IC50) of DPPH radical was 29.26, 17.07, 18.29, 19.55 and 54.87?g/ml respectively.Asian J. Med. Biol. Res. June 2015, 1(2): 285-291

Cryptocoryne ciliata belonging to the Araceae family has been investigated for isolation of its secondary metabolites and evaluation of biological activities of the crude extractives with special emphasis to the antioxidant activity and brine shrimp lethality bioassay. The whole plant was extracted with methanol and concentrated extract was partitioned using petroleum ether, carbon tetrachloride and ethyl acetate. Aqueous soluble fraction of the methanolic extract showed the highest antioxidant activity. The carbon tetrachloride soluble fraction of the methanolic extract and the ethyl acetate soluble fraction of the methanolic extract showed moderate antioxidant activity as compared to free antioxidant activity of tert-butyl-1-hydroxytolunene. In the brine shrimp lethality bioassay, among all extracts of whole plant of C. ciliata, the carbon tetrachloride soluble fraction of the methanolic extract showed strong cytotoxic activity. Aqueous soluble fraction of the methanolic extract, methanolic crude and ethyl acetate soluble fraction of the methanolic extract showed mild cytotoxity as compared to that of vincristine sulphate. The study confirms the mild to moderate antioxidant and moderate potent cytotoxic activities of C. ciliata plant extract as compared to reference standards and therefore demands the isolation of active principles and thorough bioassay.DOI: http://dx.doi.org/10.3329/icpj.v2i2.13196 International Current Pharmaceutical Journal 2013, 2(2): 38-41

Abstract Actinodaphne angustifolia Nees (Family: Lauraceae) is commonly used in folk medicine against urinary disorder and diabetes. The objective of the present study was to evaluate the antioxidant, cytotoxic, thrombolytic, and antidiarrheal activities of carbon tetrachloride (CCl4) fraction of leaves of A. angustifolia (CTFAA) in different experimental models. Antioxidant activity was evaluated by using qualitative and quantitative assays, while antidiarrheal effects assessed with castor oil-induced diarrheal models in mice. The clot lysis and brine shrimp lethality bioassay were used to investigate the thrombolytic and cytotoxic activities, respectively. CTFAA showed antioxidant effects in all qualitative and quantitative procedures. The fraction produced dose-dependent and significant (P&lt;0.05 and P&lt;0.01) activities in castor oil-induced diarrheal models. Moreover, CTFAA significantly (P&lt;0.05) demonstrated a 15.29% clot lysis effect in the thrombolytic test, and the brine shrimp lethality assay LC50 value was 424.16 μg/ml bioassay. In conclusion, the current study showed CTFAA has significant antidiarrheal effects along with modest antioxidant and thrombolytic effects, and these data warrant further experiment to justify and include CTFAA as a supplement to mitigate the onset of diarrheal and cardiovascular disease.

The present study was conducted to test for in vivo Brine Shrimp Lethality Assay (BSLA) of the Aqueous and ethanolic extracts of Annona reticulata with Allium fistolisum and Brassica oleraceaeand correlate cytotoxicity results with known pharmacological activities of the plants. Cytotoxicity was evaluated in terms of LC50 (lethality concentration). Ten nauplii were added into three replicates of each concentration of the plant extract. After 24 h the surviving brine shrimp larvae were counted and LC50 was assessed. Results showed that the extracts of Annona reticulata with Allium fistolisumand Brassica oleraceaewere potent against the brine shrimp when compared alone with combined extracts. It indicated that bioactive components are present in these plants that could be accounted for its pharmacological effects. Thus, the results support the uses of these plant species in traditional medicine.

Aims: Tinospora cordifolia is a herb used in the traditional folk medicine due to their health benefits. Studies reported the biochemical composition and some biological properties of the plant stem. The aim of the current study was to investigate the antimicrobial and cytotoxic properties from the extract of Tinospora cordifolia stem. Place and Duration of Study: The study was accomplished by eleven months in the Department of Pharmacy, University of Science and Technology Chittagong (USTC), Bangladesh.  Methodology: Presence of phytochemical constituents such as alkaloids, flavonoids and tannins, saponins were evaluated. Antimicrobial activity of the stem extract was assayed separately using an agar diffusion method against some gram negative and gram positive bacteria. The cytotoxic potential of the stem extract was evaluated by the Brine-Shrimp Lethality Assay mathod.  Results: The zone of inhibition formed by the methanolic extract in the antimicrobial assays was significant. In addition, cytotoxicity of the extract was found to be significant as compared to the standard gallic acid in Brine-Shrimp lethality assay. Conclusion: The methanolic extract of Tinospora cordifolia may possess significant antimicrobial property which requires more studies to isolate the specific bioactive compound for developing a new antimicrobial agent from this plant.

On the basis of the ‘one strain, many compounds’ (OSMAC) strategy, chemical investigation of the marine-derived fungus Trichothecium roseum resulted in the isolation of trichomide cyclodepsipeptides (compounds 1–4) from PDB medium, and destruxin cyclodepsipeptides (compounds 5–7) and cyclonerodiol sesquiterpenes (compounds 8–10) from rice medium. The structures and absolute configurations of novel (compounds 1, 8, and 9) and known compounds were elucidated by extensive spectroscopic analyses, X-ray crystallographic analysis, and ECD calculations. All isolated compounds were evaluated for cytotoxic, nematicidal, and antifungal activities, as well as brine shrimp lethality. The novel compound 1 exhibited significant cytotoxic activities against the human cancer cell lines MCF-7, SW480, and HL-60, with IC50 values of 0.079, 0.107, and 0.149 μM, respectively. In addition, it also showed significant brine shrimp lethality, with an LD50 value of 0.48 μM, and moderate nematicidal activity against Heterodera avenae, with an LC50 value of 94.9 μg/mL. This study constitutes the first report on the cytotoxic and nematicidal potential of trichomide cyclodepsipeptides.

The leaf of Osyris quadripartita is traditionally used for the management of diarrhea in different parts of Ethiopia. However, its use has not been scientifically validated for its efficacy. The aim of this study was to investigate antidiarrheal activity of hydroalcoholic leaf extract of O. quadripartita in mice models. Different doses of the methanolic leaf extract of O. quadripartita (100, 200, and 400 mg/kg) were tested for antidiarrheal activity using castor oil–induced diarrhea, enteropooling, and gastrointestinal motility models in Swiss Albino mice. The activities of the extract at different doses were compared with standard drugs and negative control groups of mice. The extract at all tested doses resulted in significant reduction ( P < .01) in number of wet feces, whereas significant reduction ( P < .01) in frequency of defecation in castor oil–induced diarrhea was seen at a dose of 400 mg/kg. It also showed a dose-dependent and significant reduction of volume of intestinal content in the enteropooling model at all tested doses and the observed results in 200 and 400 mg/kg were better than the standard drug, loperamide. However, significant antimotility effect was not observed at any of the tested doses. From these results we can conclude that methanolic leaf extract of O. quadripartita showed antidiarrheal activity.

The study reports the in vivo antidiarrheal and in vitro anthelmintic, cytotoxic, and thrombolytic activity of methanol extract of Hedychium coccineum rhizomes (MEHC). The antidiarrheal activity was determined using Castor oil-induced diarrhea and Gastrointestinal motility test in mice at the doses of 200 and 400 mg/kg body weight, whereas an aquarium worm, Tubifex tubifex, was used to determine the anthelmintic activity. The cytotoxic and thrombolytic activity of MEHC was performed by Brine shrimp lethality bioassay and clot lysis method respectively. In antidiarrheal, castor oil-induced diarrhea and gastrointestinal motility exhibited a significant reduction in diarrhea and defecation and an extremely significant inhibition in intestinal motility and peristalsis index by 200 and 400 mg/kg of MEHC. The MEHC (5, 10, and 20 mg/mL) showed a significant dose-dependent manner paralysis time and times to death in multiple comparisons to the different levamisole concentrations (0.5, 0.8, and 1 mg/mL) at in vitro anthelmintic activity. The brine shrimp lethality bioassay exhibited a weak LC50 (681.95 µg/mL; R² = 0.951) while in thrombolytic a significant percentage of clot lysis (32.70%, P < 0.05) demonstrated. The findings demonstrate that H. coccineum rhizomes could be potential sources for biological activity.

Bangladesh possesses a rich flora of medicinal plant. Out of the estimated 5000 species of phanerogams and pteridophytes growing in this country, more than a thousand are regarded as having medicinal properties. The aim of this study was to evaluate the in vitro antimicrobial activity and brine shrimp lethality of the whole plant extracts of Spilanthes paniculata, a plant belonging to the family Asteraceae. The crude extract, n-Hexane, chloroform and ethyl acetate soluble fraction of crude extract showed significant to moderate antimicrobial activity against three Gram positive and three Gram negative microorganisms. The zones of inhibition produced by the crude extract (methanolic extract), n-hexane, choloform and ethyl acetate soluble fractions were found to be 14.89 mm-19.40 mm, 14 mm-19.40 mm, 10.66 mm-13.50 mm and 9.50 mm-13.26 mm, consecutively at a concentration of 30 µg/disc. N-Hexane, chloroform and ethyl acetate soluble fractions of methanolic extract of Spilanthes paniculata were screened for antitumor properties using brine shrimp lethality bioassay. From the results of the brine shrimp lethality bioassay, it can be well predicted that n-hexane, chloroform and ethyl acetate soluble fractions of methanolic extract possess mild cytotoxicity on shrimp naupalii. The median lethal concentration (LC50, the concentration at which 50% mortality of brine shrimp nauplii occurred) of n-hexane, chloroform and ethyl acetate were 48.978 mg/ml, 92.61 mg/ml and 216.770 mg/ml, respectively, comparison with positive control vincristine sulphate with 0.839 mg/ml.
