Effect of Nerve Training Technology on Apoptosis of Cartilage and Osteoblasts and Expression of Aggrecan Protein in Osteoporotic Arthritis

Arthritis and osteoporosis are two common disorders in the world, especially for the elder, but the current treatments have limited efficacy. Herein, we aimed to determine whether the novel technique, neurological training can alleviate osteoporosis complicated with arthritis in rat model. Thirty rats were assigned into normal group, model group, and treatment group (treated with forsythin and neurological training) (n = 10) followed by assessment of chondrocytes and osteoblasts using Mankin score, apoptosis by TUNEL and flow cytometry, and IL-1β, TNF-α, and Aggrecan levels. Apoptotic chondrocytes of treatment group (27.43±1.34) was lower than model group (p < 0.05), whereas amount of osteoblast was increased upon forsythin and neurological training, with lower Mankin’s score (6.38±0.76). Besides, the content of IL-1β and TNF-α of treatment group was significantly lower but Aggrecan mRNA and protein expression was significantly higher. In conclusion, neurological training could protect and alleviate osteoporosis complicated with arthritis.

Objective: To investigate the protective effects and mechanisms of esculetin(Esc) on H9C2 cells injury induced by doxorubicin (DOX).  Methods: H9C2 cells were cultured and divided into control group, model (DOX) group and intervention (ESC + DOX) group. Flow cytometry was used to detect apoptosis of H9C2 cells and reactive oxygen (ROS) level in H9C2 cells; Western blotting to detect the cleaved Caspase-3, cleaved PARP, bid and Bcl-2 protein expression in H9C2 cells.  Results: The number of apoptosis and ROS level of H9C2 cells in the model group, the expression of cleaved Caspase-3, cleaved PARP and Bid protein in the model group were obviously higher than control group; and the Bcl-2 protein expression was obviously lower (P < 0.05).The number of apoptosis and ROS level, cleaved Caspase-3, cleaved PARP and Bid protein expression in H9C2 cells in intervention group were obviously lower than model group;the Bcl-2 protein expression was obviously higher (P< 0.05).  Conclusion: Esculetin can reduce the cardiotoxicity induced by doxorubicin by reducing apoptosis and ROS level.

The incidence of knee arthritis is high and treatment effect is not satisfactory. Alendronate (ALN) can treat metabolic bone diseases. But its role and mechanism in knee arthritis remains unclear. SD rats were randomly assigned into control group; model group; and alendronate group followed by analysis of the pathological changes by HE staining and Masson staining, bone mineral density by dual energy bone densitometry, expression of glycosaminoglycan and MMP-3 by immunohistochemistry, BMP-2 and BMP-4 expression by Real time PCR, IL-1β, IL-6 secretion by ELISA as well as ERK signaling protein level by Western blot. In model group, the pathological changes of joint inflammation were obvious with fibrosis on the cartilage surface, significantly increased synovial hyperplasia score, inflammatory infiltration score and Mankin score, decreased bone density, expression of glycosaminoglycan, MMP-3, BMP-2 and BMP-4 as well as increased IL-1β, IL-6 secretion and ERK1/2 expression (P <0.05). Alendronate treatment can significantly improve joint inflammation, reduce inflammatory infiltration score and Mankin score, increase bone density, expression of glycosaminoglycan, MMP-3, BMP-2 and BMP-4, decrease the secretion of IL-1β, IL-6 and expression of ERK1/2 in a rat model of knee arthritis (P <0.05). Alendronate can promote glycosaminoglycans and MMP-3 expression, and synthesis of BMP-2 and BMP-4 by inhibiting ERK pathway, inhibiting the secretion of inflammatory factors, thus ameliorating knee arthritis.

This study was aimed to investigate the effects of Wenyangbushen formula on the mRNA and protein expression of VEGF, OPG, RANK, and RANKL in the steroid- induced avascular necrosis of the rabbit femoral head (SANFH) and to further explore the potential mechanism of this formula on the treatment of SANFH. One hundred and thirty six New Zealand rabbits were randomly divided into 5 groups (n=24 rabbits in each group): normal group, model group, Traditional Chinese Medicine (TCM) Wenyangbushen decoction at low, moderate and high dose group. The normal group and positive control group were intragastrically (i.g.) administered with saline. The TCM group was treated with Wenyangbushen decoction at the indicated dosage. After treated for eight weeks, the mRNA and protein expression of VEGF, OPG, RANK, and RANKL in the femoral head tissues was determined by RT-PCR and Western blotting, respectively. Wenyangbushen decoction treatment can effectively promote bone cells, osteoblasts and chondrocytes growth, as well as prevent the cell apoptosis in SANFH. The mRNA and protein expression of OPG and VEGF was increased while the levels of RANK and RANKL were reduced in the necrotic tissue of model group compared with those in the normal rabbits. And Wenyangbushen treatment prevented those changes, as manifested by up-regulation of VEGF and OPG, while down-regulation of RANK and RANKL levels in a dose-dependent manner. Wenyangbushen Formula treatment can alleviate necrosis of femoral head induced by steroid. It can promote bone cells, osteoblasts and chondrocytes growth, as well as prevent the cell apoptosis. Meanwhile, it up-regulates OPG and VEGF while inhibits RANK and RANKL expression. This may be one of the mechanisms for the effective treatment of SANFH.

Objective To observe the effect of Xiaochaihu Decoction on cytokines and MAO in model rats with experimental autoimmune prostatitis (EAP) to provide an experimental basis for the use of Chinese herbal prescriptions in the treatment of chronic prostatitis. Methods Thirty male Wistar rats were randomly divided into blank Group, model group and treatment group (Xiaochaihu Decoction). Except the blank group, rats in model and treatment groups were injected subcutaneously in multiple points on days 0 and 30 with prostatic protein extractive solution (60 mg/mL), and intraperitoneally injected with diph-theria-pertussis and tetanus vaccine (DPT vaccine) to establish the EAP model. Model rats were administrated Xiaochaihu Decoction and were sacrificed after 4 weeks. Pathological changes in the prostate gland were observed with HE staining and changes in serum interleukin-6 (IL-6), interleukin-8 (IL-8), and Tumor necrosis factor-α (TNF-α) levels were detected with enzyme-linked immunosorbent assay (ELISA). Quantitative PCR to observe the expression of IL-17, CCL2 and MAOA/B. Results After treatment with Xiaochaihu Decoction, the serum IL-6, IL-8, and TNF-α levels all significantly lower in the treatment group as compared with the model group (P < 0.05). The intervention of Xiaochaihu Decoction can change the state of abnormally elevated IL-17, CCL2 and MAOA expression levels. Conclusions The therapeutic efficacy remarkable from the theory of heating into the sperm chamber to experimental autoimmune prostatitis rats.

Objective: To investigate the expression of TNF-a and NF-kB in the uterus of chronic pelvic inflammatory disease (CPID) model rats. Methods: 40 female rats that adaptive fed for 5 days were randomly divided into normal group, model group, high dose group, medium dose group and low dose group. In addition to the normal group, the rats in each group were made chronic pelvic inflammatory model by mechanical injury combined with implantation of bacteria. The rats in each group were administrated by gavage for 20 days. After the last administration, the level of TNF-a and NF-kB in serum was measured by ELASA method. Results: (1) after the establishment of the model, the uterus of the chronic pelvic inflammatory model rats showed the pathological damage of chronic inflammation; the levels of TNF-a and NF-kB in serum were higher than those in the normal group, the difference was statistically significant (P<0.05). (2) After the drug intervention, the uterine tissue morphology of the rats in the Zhuang Yi Liu Fang Teng Fang Group was basically restored to normal, with only a small amount of inflammatory cell infiltration and other pathological changes; compared with the rats in the model group, the TNF-a in serum of the rats in each treatment group was lower, the difference was statistically significant (P<0.05). The expression of NF-kB in serum of each treatment group was lower than that of the model group (P<0.05). Conclusion: Zhuang Yi Liu Teng Fang can effectively improve the endometrial histomorphology of CPID model rats, and regulate the levels of TNF-a and NF-kB in the uterus of chronic pelvic inflammatory model rats.

AIM: To observe the effect of Sposknikovan on spleen in traumatized mice.METHODS: A total of 60 Kunming mice were randomly divided into three groups: normal group, model group and treatment group. Sposknikovan were administered once by intragastric administration (i.g.), lasting seven days. We stripped each group of mice, weighed spleen and calculated spleen index after an hour when the last administration, then stained by propidium iodide dye. We used FACSAria flow cytometry and ModFit software for cell cycle analysis of spleen cell proliferation index and the proportion of the spleen lymphocyte subsets.RESULTS: A total of 60 Kunming mice were analyzed. ① compared with control group, spleen index of model group decreased significantly, while treatment groups were higher than control and model group. ②Proliferation index of spleen cells in model group was lower than control group; treatment group could increase the proliferation index of mouse spleen cells. ③Compared with control group, the ratio of CD3+ CD4+ /CD3+CD8+ was decreased in model group. The ratio of CD3+CD4+/CD3+CD8+ in treatment group was increased compared with control and model group. CONCLUSION: Sposknikovan retrieved the ratio of CD3+CD4+/CD3+CD8+ and enhanced immune function of spleen in traumatized mice by promoting proliferation of lymphocytes in traumatized mice.

Purpose: To determine the influence of tanshinone on the levels of nitric oxide synthase (NOS) and acetylcholinesterase (AChE) in the brain of an Altzheimer Disease (AD) rat model and on its potential therapeutic mechanism. Methods: 100 Male Sprague Dawley rats were divided into three groups: control group, model group and tanshinone treatment group. 10µg A?1-42 was injected bilaterally into the dorsal lateral region of the dentate gyrus in the hippocampus of rats in the model and tanshinone treatment groups to prepare the AD models. 24h after modeling, tanshinone, 50mg/kg, was administered by gastric perfusion to rats in the tanshinone treatment group. Later, immunohistochemical assay and Western blot analysis were used to detect expression of neuronal NOS (nNOS) and inducible NOS (iNOS) in the rat hippocampus. Activity of AChE in each subregion (CA1~CA4) of rats’ hippocampus was determined by a histochemical technique. Results: Expression of nNOS in the model group was down-regulated whereas iNOS was up-regulated. After A?1-42 injection, the number of AChE positive fibers in each subregion (CA1~CA4) of the hippocampus was decreased compared with controls. With tanshinone administration, the changes were improved to varying degrees. Conclusion: Tanshinone modulates AChE and NOS proteins concentrations in the hippocampus of AD rats. This may have therapeutic potential in AD rats.

AbstractWith the rapid onset of an unprecedented lifestyle due to the new coronavirus COVID-19 the world academic scene was forced to reform and adapt to the novel circumstances. Although online education cannot be regarded as a groundbreaking endeavour anymore in the21st century, its current character of exclusivity calls for deeper understanding of, and a sharper focus on the “end-consumer” thereof as well as more cautious procedures to be exercised while teaching. While millennials are no longer thought of as being born with a silver spoon in their mouth but with an iPad or any sort of device in their hand (irrespective of their social status), adults are more hesitant when coerced to alter course unexpectedly and turn to new methods of attaining their learning goals. This is why proper communicative approaches need to be thoroughly considered by online instructors. This article aims at presenting teachers with a set of strategies to employ when the beneficiaries of online academic education are adult learners.
