Abstract Background Aging has detrimental effects on the ovary, such as a progressive reduction in fertility and decreased hormone production, that greatly reduce the quality of life of women. Thus, the current study was undertaken to investigate whether human placenta-derived mesenchymal stem cell (hPD-MSC) treatment can restore the decreases in folliculogenesis and ovarian function that occur with aging. Methods Acclimatized 52-week-old female SD rats were randomly divided into four groups: single hPD-MSC (5 × 105) therapy, multiple (three times, 10-day intervals) hPD-MSC therapy, control (PBS), and non-treated groups. hPD-MSC therapy was conducted by tail vein injection into aged rats. The rats were sacrificed 1, 2, 3, and 5 weeks after the last injection. hPD-MSC tracking and follicle numbers were histologically confirmed. The serum levels of sex hormones and circulating miRNAs were detected by ELISA and qRT-PCR, respectively. TGF-β superfamily proteins and SMAD proteins in the ovary were detected by Western blot analysis. Results We observed that multiple transplantations of hPD-MSCs more effectively promoted primordial follicle activation and ovarian hormone (E2 and AMH) production than a single injection. After hPD-MSC therapy, the levels of miR-21-5p, miR-132-3p, and miR-212-3p, miRNAs associated with the ovarian reserve, were increased in the serum. Moreover, miRNAs (miR-16-5p, miR-34a-5p, and miR-191-5p) with known adverse effects on folliculogenesis were markedly suppressed. Importantly, the level of miR-145-5p was reduced after single- or multiple-injection hPD-MSC therapy, and we confirmed that miR-145-5p targets Bmpr2 but not Tgfbr2. Interestingly, downregulation of miR-145-5p led to an increase in BMPR2, and activation of SMAD signaling concurrently increased primordial follicle development and the number of primary and antral follicles. Conclusions Our study verified that multiple intravenous injections of hPD-MSCs led to improved ovarian function via miR-145-5p and BMP-SMAD signaling and proposed the future therapeutic potential of hPD-MSCs to promote ovarian function in women at advanced age to improve their quality of life during climacterium.

This study aimed to evaluate the effects of treatment with Peruíbe Black Mud (PBM) on the clinical parameters and quality of life of patients with knee osteoarthritis and to compare the effects of PBM samples simply matured in seawater and PBM sterilized by gamma radiation. A controlled, double-blind trial was conducted with 41 patients divided into two treatment groups composed of 20 and 21 patients: one group was treated with matured PBM and the other with sterilized PBM. Evaluations were done using the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and the Medical Outcomes Study Short Form 36 (SF-36) questionnaires, the Kellgren and Lawrence (KL) radiographic scale, and the quantification of the serum levels of inflammatory biomarkers. An improvement in pain, physical functions, and quality of life was observed in all of the patients who underwent treatment with both simply matured and sterilized PBM. Nine patients showed remission in the KL radiographic scale, but no statistically significant differences were observed in the serum levels of inflammatory mediators before or after treatment. Peruíbe Black Mud proves to be a useful tool as an adjuvant treatment for knee osteoarthritis (OA), as shown by the results of the WOMAC and SF-36 questionnaires and by the remission of the radiographic grade of some patients on the Kellgren and Lawrence scale.

Hidradenitis suppurativa (HS) is a relapsing, inflammatory disease characterized by painful nodules, abscesses, sinuses track formation and scarring. HS has a great impact on patients’ quality of life and its treatment may be really challenging. Adalimumab provides a new therapeutic option for HS. Our aim was to assess the therapeutic potential of adalimumab on patients with HS based on the data from the daily clinical practice of an HS Outpatient Clinic. 19 patients with clinically evident moderate to severe HS, under adalimumab treatment for at least 24 week, participated in this observational, retrospective study. The Hidradenitis Suppurativa Physician’s Global Assessment scale, Modified Santorius scale and Dermatology Life Quality Index (DLQI) at baseline, week 4, week 12 and week 24 were retrieved from the records. Both Modified Santorius score and DLQI were significantly decreased during the weeks of evaluation (Friedman’s test; P < 0.001). The proportion of patients who achieved clinical response was 10.5% (n = 2) at week 4, 42.1% (n = 8) at week 12 and 63.2% (n = 12) at week 24. Treatment with adalimumab was linked with both clinical remission of HS and improvement of patients’ quality of life.

  THE CORRELATION BETWEEN VITAMIN D SERUM LEVELS WITH QUALITY OF LIFE IN EPILEPSY PATIENTABSTRACTIntroduction: Epilepsy affects overall health status and decreases the life quality of epilepsy patient. Seizure frequency, seizure type, daily activity disorder, depression and anxiety also affect the quality of life epilepsy patient. Vitamin D is considered as a neurosteroid modulator of nerve excitability and seizure susceptibility. Studies of vitamin D direct role in epilepsy are limited. Nevertheless, some studies show the role of vitamin D as an anticonvulsant that reduces the incidence of seizures.Aim: To determine the correlation between vitamin D serum levels with quality of life in epilepsy patient.Method: A cross sectional observational study on people with epilepsy in neurology clinic at Dr. Soeselo Hospital, Slawi, Dr Kariadi Hospital, and Tugurejo Hospital, Semarang in July 2017-January 2018. The quality of life was assessed with Quality of Life in Epilepsy 31 (QOLIE-31). Vitamin D serum levels measured using enzyme-linked immunosorbent assay (ELISA) method. Data were analyzed using Spearman test. Results were considered significant if p<0.05.Results: There was no correlation between vitamin D serum levels with quality of life in epilepsy. There was significant correlation between anxiety with quality of life and there were difference between age group with quality of life. Discussion: There was no correlation between serum vitamin D levels and  quality of life in epilepsy patient.Keywords: Epilepsy,quality of life epilepsy, vitamin DABSTRAKPendahuluan: Epilepsi mempengaruhi status kesehatan secara keseluruhan dan menurunkan kualitas hidup pasien epilepsi. Namun beberapa penelitian menunjukkan peran vitamin D sebagai antikonvulsan yang mengurangi kejadian bangkitan.Tujuan: Mengetahui hubungan kadar vitamin D serum dengan kualitas hidup pada pasien epilepsi.Metode: Penelitian potong lintang terhadap pasien epilepsi yang berobat ke Poliklinik Saraf RSUD Dr. Soeselo, Slawi, RSUP Dr. Kariadi, Semarang, dan RS Tugurejo, Semarang pada bulan Juli 2017-Januari 2018. Kualitas hidup dinilai menggunakan kuesioner Quality of Life in Epilepsy 31 (QOLIE-31), pengukuran kadar vitamin D serum menggunakan metode enzyme-linked immunosorbent assay (ELISA). Data dianalisis dengan uji Spearman, hasil dianggap bermakna jika p<0,05.Hasil: Tidak didapatkan hubungan antara kadar vitamin D serum dengan kualitas hidup serta domainnya pada pasien epilepsi. Didapatkan hubungan yang bermakna antara ansietas dengan kualitas hidup dan antara kelompok usia dengan kualitas hidup.Diskusi: Tidak terdapat hubungan bermakna antara kadar vitamin D serum dengan kualitas hidup pada pasien epilepsi.Kata kunci: Epilepsi, kualitas hidup, vitamin D

Abstract Background and Aims Hyperprolactinemia is a hormonal disorder associated with many diseases, including chronic kidney disease (CKD). Some studies have shown that serum prolactin (PRL) levels increase as CKD progresses, due to decreased renal clearance and increased production. This prolactin retention leads to inhibition of gonadotropic hormone production, and testosterone deficiency in male patients with CKD. Thus, this study aimed to evaluate PRL levels in male patients with different stages of CKD and its association with clinical and laboratory characteristics, strength, sexual dysfunction, and quality of life. Method This is a transversal study conducted in a tertiary hospital in southern Brazil. Patients with CKD stage IV and V were included and divided into three groups: 1) stage IV; 2) non-dialysis-dependent stage V and; 3) dialysis-dependent stage V (VD). Patients with prescribed hormone therapy were excluded. PRL, creatinine, testosterone, estradiol, albumin, SHBG, LH, and FSH levels were measured. Free testosterone was estimated by the Vermeulen equation. Patients were also evaluated for strength and musculoskeletal function through the “handgrip strength” test. For evaluation of the quality of life and sexual dysfunction, validated questionnaires were applied: AMS (Aging Male's Symptoms Scale), ADAM (Androgen Deficiency in the Aging Male) and SF-36 Health. Results In this study, 164 patients were included (IV=75; V=41; VD=48). The mean age was 64 [55-71] years and BMI 26,9 [23,3-30,04] kg/m2. The dialysis-dependent group had higher prevalence of hyperprolactinemia (22,6% vs. 49% vs. 66%, p&lt;0,001), higher PRL (13,4 vs. 19,43 vs. 26,13, p&lt;0,001) and estradiol (25,6 vs. 24,4 vs. 30,5, p=0,041). PRL levels were positive correlated with CKD stage (rs=0,482, p&lt;0,001) and creatinine (rs=0,467, p&lt;0,001) and negative correlated with testosterone (rs=-0,286, p&lt;0,001) and free testosterone levels (rs=-0,307, p&lt;0,001). There was no difference in PRL levels between hemodialysis and peritoneal dialysis (p=1,000). Analyzes related to the association of PRL levels with strength, quality of life, and sexual dysfunction are still being analyzed. Conclusion Hyperprolactinemia is associated with the severity of CKD, with a higher prevalence in dialysis patients. The relation of PRL levels with strength, quality of life, and sexual dysfunction are being evaluated.

This is a prospective, controlled, randomized clinical trial in canine patients with overt Dilated Cardiomyopathy (DCM). We hypothesized that the addition of the third generation beta-blocker carvedilol to the standard treatment of diuretic (furosemide), inotropic support (digoxin) and ACE-inhibitor (benazepril) would have beneficial effects over cardiac function and quality of life (QoL) that would be measurable 90 days post-treatment. 16 dogs diagnosed with overt DCM were recruited. They underwent clinical examination, electrocardiography, echocardiography and neurohormonal profiling (NT-proBNP and NO). Dogs were divided in two groups, receiving standard therapy or standard therapy plus carvedilol and subjected to re-evaluation on 90 days post-treatment. Our results indicated that the addition of carvedilol to the standard therapy improved echocardiographic indices of systolic function (FS and EF), reduced NT-proBNP and NO serum levels and quality of life within the group but did not showed the significant improvement over standard therapy. This suggested that the addition of carvedilol to the standard therapy in canine patients with overt DCM might have beneficial effects in cardiac function and quality of life in the treatment was continued for longer period.

Functional dyspepsia is of high prevalence with little treatment options. The aim of this study was to develop a new treatment method using self-management transcutaneous electroacupuncture (TEA) for functional dyspepsia (FD). Twenty-eight patients with FD were enrolled and underwent a crossover clinical trial with 2-week TEA at ST36 and PC6 and 2-week sham-TEA at nonacupuncture sham-points. Questionnaires were used to assess symptoms of dyspepsia and quality of life. Physiological testing included gastric emptying and electrogastrography. It was found that (1) TEA but not sham-TEA significantly improved dyspeptic symptoms and 4 domains in quality of life; improvement was also noted in self-rated anxiety and depression scores; (2) gastric emptying was significantly and substantially increased with 2-week TEA but not sham-TEA; and (3) gastric accommodation was also improved with TEA but not sham-TEA, reflected as increased ingested nutrient volumes at the levels of satiety and maximum tolerance. These findings suggest a therapeutic potential of self-administrated TEA method for functional dyspepsia, possibly attributed to improvement in gastric motility.
