Definition of Blood Serum Antioxidant Activity of Patients with Liver Pathology by Two Chemiluminescent Methods

Introduction. The overwhelming number of publications contains only data on the content of individual antioxidants, but not on the overall antioxidant activity of the blood in patients with vibration disease.The aim of the study was to determine the total antioxidant activity of blood serum in patients with vibration disease.Materials and methods. Th e main group consisted of 30 people diagnosed with “Vibration disease” of 1 degree (n=21) and 2 degrees (n=9). Th e control group consisted of 30 clinically healthy men, comparable in age with the main group (p=0.66). Th e total activity of antioxidant systems of blood plasma was evaluated photometrically using the test system “Total antioxidant status-Novo” (“Vector-best”, Russia).Results. The indicator of the total antioxidant status (TAS) was 1,038±0.232 mmol/l in the examined main group, against 1,456±0.225 mmol/l in the examined control group (p<0.000001). Th e coefficient of variation (CV) in patients with vibration disease was 22.35%, 1.45 times higher than in the control group (15.45%). In the main group there was a positive correlation between age and TAS (R=0.525), in the control group there was no such relationship (R=0.095). Th e degree of decrease depended on the severity of vibration disease.Conclusions. 1. The development of vibration disease is accompanied by a decrease in the antioxidant status of blood serum. 2. Th e degree of decrease in the antioxidant status of blood serum correlates with the severity of vibration disease. 3. Reduction of TAS can serve as a pathogenetic justification of the need to include drugs and/or biologically active additives with antioxidant activity in therapy

Investigation results are reported on immunobiochemical indices in carassius carassius at a chronic exposure to sub-lethal Zink ions concentrations. In blood serum and liver, the following characteristics were investigated: antimicrobial properties, proportion of immune- deficient animals, content of non specific immune complexes, common lipids level, content of lipids peroxidation products and antioxidant activity. The relation between magnitudes of indices under investigation in fishes and the duration of their presence under experimental conditions is shown.

The effects of chitosan on various metabolic and digestive processes in the animal body are an important to study because of the production quality for animal nutrition, healthcare, etc. The major known changes in the biochemical parameters of the animal liquids (by chitosan usage as feed additive) are the following: ratio of volatile and fatty acids, content of fat, mineral composition, etc. The aim of this study was to evaluate the total amount of water-soluble antioxidants (TAWSA) of feed additives based on chitosan and high protein concentrate of microbiological synthesis and to assess the effect of these feed additives on TAWSA values of sheep blood serum by amperometric method. Firstly, the antioxidant activity of feed components based on chitosan or/and a high-protein microbiological synthesis concentrate was studied in this work. Second, the experiments are carried out on 6 fistula animals to confirm the results of laboratory studies. Third, the determination of the total antioxidant activity of sheep blood by using chitosan additives (as the feed components) by amperometric method was carried out. A particular difference in antioxidant activity of the control and experimental samples of the sheep blood was established for the first time.

The influence of fattening conditions on the digestive and metabolic processes in animals is actively studied to increase the production of animal food. The aim of the study was to estimate the total amount of water-soluble antioxidants in the blood serum of Duroc boars depending on the duration of fattening. Experiments were conducted on 102 animals to confirm the results of laboratory studies, as well as determination of the total antioxidant activity of pig blood serum by amperometric method. For the first time, a significant difference in antioxidant activity was found in blood samples of Duroc boars with the duration of their fattening from 81 to 101 days.

Objective: definition of indicators of system prooksidanty-antioxidants at active and passive smoking during pregnancy. Material and methods. On the basis of city clinic for women 39 pregnant women on the term of a gestation of 37 weeks are examined. Questioning, rapid test for identification of a kotinin in urine, determination of level of a malon dialdehyde (MDA) and also activities superoxide dismutases (SOD) and catalases is carried out to bloods of surveyed. Patients are divided into 3 groups: I-of 11 smoking pregnant women subject to II-13 to passive smoking at pregnancy, III-control group of 15 women. Results. At patients of I and II groups substantial increase of level MDA in blood serum is noted. Reliable decrease of the activity of SOD of erythrocytes in the I group and insignificant decrease of the activity in II is taped. The indicator of catalase/SOD was statistically higher at active smoking. Conclusion. Active and passive smoking at pregnancy leads to change in prooxidatic and antioxidatic systems.

Background: The purpose of work was development of a fast and reproduced procedure for measurement of the total complement activity (ТСА) in human or animal blood serum. Materials and methods: Steady at storage liposomes preparations, which surface sensitized 2,4-DNP haptens, and the internal volume contains calceine or sulforhodamine 101 are obtained. Complement-dependent immune lysis of liposomes at presence of the anti-2,4-DNP immunoglobulines and complement preparations from animals are investigated. Results: It is shown that the degree of liposomes immune lysis depends on complement concentration in a wide range that can be used for definition of TCA level. Research of blood sera from patients has revealed correlation (r =0,793) between data received with the help of liposome immunolytic systems, and the data of nephelometric analysis with application of suspension sheep erythrocytes. Conclusion: The method allows to define total complement activity in blood serum in 15 minutes without separation of reaction components. This might be useful for measurement ТСА level at patients with various diseases and realization of scientific researches.

Objective: to determine the clinical efficiency of antioxidant therapy in patients with liver cirrhosis and acute blood loss.Material and methods. The study included 286 patients with liver cirrhosis and acute blood loss of varying degrees of severity. The patients were divided into two series of studies. In the first series, the patients received the therapy according to the clinical diagnostic and treatment protocols, in the second series 1 according to the same protocols in combination with an intravenous administration of the antioxidant complexes “Reamberin” and “Fluimucil”. After 48 hours from admission, the antioxidant status of blood serum, the blebbing index of lymphocytes, the concentration of uric acid, and the level of hemoglobin were analyzed. The indicators of the clinical efficiency of the treatment were determined: risk of absolute mortality, risk of relative mortality, the number needed to treat.Results. The blood serum of the patients from the first series of studies was characterized by pronounced prooxidant activity, high values of the lymphocyte blebbing index, and a small amount of uric acid, which was indicative of the development of oxidative stress. The mortality rate in this series of studies was 26.0 %. The patients of the second series of studies were characterized by pronounced antioxidant activity of blood serum, low values of the lymphocyte blebbing index, and high concentrations of uric acid, which indicated a decrease in the activity of systemic oxidation processes. The mortality rate in this series of studies was 18.2 %. The duration of the treatment decreased from 16.5 days (first series) to 13.0 days (second series).Conclusion. The combined use of the “Reamberin” and “Fluimucil” complexes as part of the therapy according to the clinical diagnostic and treatment protocols leads to a relief of oxidative stress in patients with liver cirrhosis complicated by acute blood loss. The blood serum of these patients is characterized by pronounced antioxidant activity, low values of the lymphocyte blebbing index, and a high concentration of uric acid, which reduces the risk for the development of multiple organ failure syndrome. This therapy decreases the mortality rate to 18.2 % and the duration of the treatment to 13.0 days.

Chronic renal failure (CRF) is often diagnosed in cats and dogs of old age. CRF is a consequence of metabolic deviations because of diseases (for example, generalized atherosclerosis, hypertensive disease, diabetes and others) and incorrect feeding. CRF is also a result of the delayed diagnosis of diseases of the urinary system and/or inadequate treatment. Aim of research. To study the biochemical parameters of protein metabolism that are important for determination of CRF stages and sub-stages in old small animals during correction of metabolic deviations. Materials and methods. There were analyzed biochemical indices (creatinine concentration in the blood serum; the ratio of protein/creatinine (UP/C) in the urine) and measured arterial blood pressure in 5 cats and 5 dogs with chronic renal failure selected from patients of the clinic according to medical history, clinical examination and special analysis. Results. It was analyzed CRF stages and sub-stages classification according to the International Renal Interest Society (IRIS). According to the date of creatinine concentration in the blood serum, CRF of the second stage was observed in 5 cats (140-250 μmol/L) and 3 dogs (125-180 μmol/L), and of the third stage - in 2 dogs (181-440 μmol/L). The CRF sub-stage as «without proteinuria» (UP/C < 0.2) was identified in 1 cat, «marginal proteinuria» (UP/C 0.2-0.4) – in 1 cat and «proteinuria» (UP/C > 0.4) - in 3 cats, and as «proteinuria» (UP/C > 0.5) - in 5 dogs. Conclusions. An important step of the successful treatment of CRF is the definition of the stages and sub-stages according to IRIS based mostly on the measurements of biochemical indices in the blood and urine of sick animals ‒ indicators of creatininemia and proteinuria, a change of which was caused mainly by metabolic processes of proteins under influence of exogenous factors (the traditional treatment of CRF plus enalapril and diet nutrition).

Although the term 'antioxidant' is used very frequently, there are problems with the definition of antioxidants and estimation of antioxidant activity. The distinction between antioxidant and antiradical activities is not always obvious. This minireview discusses critically the principles, advantages and limitations of the most frequently used methods of estimation of antiradical and antioxidant activities.
