Blood sampling from the cranial vena cava in the Norway rat (Rattus norvegicus)

This paper describes blood sampling from the cranial vena cava (CVC) in the Norway rat. In order to limit stress, the blood sampling should be done under short-term inhalation anaesthesia, for example, an oxygen/isoflurane mixture. The injection site is just cranial to the first rib, 0.3–0.8 cm lateral to the manubrium when the animal is in dorsal recumbency. The needle, attached to a syringe, is inserted at 30° in the direction of the opposite femoral head. After penetration of the skin, negative pressure is developed in the syringe and the insertion of the needle is continued for another 0.2–1 cm in the given direction until blood begins to flow. The amount of blood sampled ranges from 0.8 to 2.5 mL depending on the body weight of the patient. A trial on 50 rats aged 5–24 months included 25 rats sampled once, eight rats sampled twice with an interval of seven days, 11 rats sampled twice with an interval of three weeks and four rats sampled four times with intervals of four weeks – a total of 87 blood samplings. The serious complications quoted in association with blood sampling from the CVC in other experimental animals (vascular lacerations, heart puncture, serious haemorrhage, tracheal and throat trauma) were not observed in our study. There were only four blood samplings (4.5%) with mild haemorrhage from the injection site, due to erroneous sampling from the jugular vein.

At present, the amount of people suffering from various types of eating disorders is steadily increasing all over the world. A large proportion is occupied by people with manifestations of anorexia. Primarily this disorder affects female representatives at the age of 13-20 years. Taking into account the severity of the consequences that anorexia leads to, cases of death are increasingly being recorded, including adolescents. That’s why the question of providing effective assistance to such patients is especially relevant. However, without establishing the true causes, the results achieved during the treatment may be only short-term. In this connection, the author made an attempt to look at the problem more deeply. Thus, the article is devoted to the consideration of the causes of anorexia of adolescent girls, lying in the field of the unconscious, in particular, in a deep psychological trauma received in childhood. According to the author, the reasons of this trauma are the perception as a humiliation of the manifestations of the brother’s admiration or friends’ son from the girl’s parents, as opposed to the lack of attention, warmth and care towards her. The arising misunderstanding of the reason for such a different attitude contributes to the formation of a girl’s confidence that it is better to be a boy and, as a result, an unwillingness to be a woman. This, in turn, leads to anorexic behavior, as anorexia can inhibit the transformation of the body into a woman. The given assessment was confirmed in a conducted study among 128 girls aged 13-18 years with various eating disorders, including 46 with manifestations of anorexia nervosa. The substantiation of the hypothesis put forward at the beginning of the study was checked by means of a statistical method - the Fisher-φ test.

The aim of the study was to determine the development of ramie plants (Boehmeria nivea L. Gaud) and the effect of using ramie leaves on feed on the body weight gain of Wonosobo Sheep (Dombos). Research on the development of ramie plants using survey methods in the area of ramie plant development in Wonosobo Regency. While the research on the use of ramie leaves for fattening was carried out in Butuh Village, Kalikajar District, Wonosobo Regency in 2018. 21 male Dombos were divided into 3 feed treatments with forage proportions of 70%, 50% and 30 ramie leaves respectively. %. The results showed that currently ramie plants were being developed in Wonosobo Regency by CV. Ramindo Berkah Persada Sejahtera in Gandok Village, Kalikajar District, Wonosobo Regency, Central Java. Until now the area of the crop has reached 13 ha. Of this area will produce ramie leaves 195,000 kg / year. If one sheep needs 4 kg of ramie / tail / day leaves, then the potential capacity of sheep is 135 heads / year, if the given one is 50% then the Jurnal Litbang Provinsi Jawa Tengah, Volume 16 202 Nomor 2 – Desember 2018potential capacity is 270 heads / year and if it is reduced again to 25% of ramie leaves then the potential capacity 440 heads / year. The use of ramie leaves as a feed for Wonosobo Sheep fattening can be given as much as 30% in fresh form.

Gridhrasi (Sciatica) is a disorder in which low back pain is found, that spreads through the hip, to the back of the thigh and down the inside of the leg. Mechanical low back pain (LBP) remains the second most common symptom related reason for seeing a physician. 85% of total population will experience an episode of mechanical LBP at some point during their lifetime. Fortunately, the LBP resolves for the vast majority within 2-4 weeks. There are many causes for low back pain, however true sciatica is a symptom of inflammation or compression of the sciatica nerve. The sciatica nerve carries impulses between nerve roots in the lower back and the muscles and nerve of the buttocks, thighs and lower legs. Compression of a nerve root often occurs as a result of damage to one of the discs between the vertebrae. In some cases, sciatic pain radiate from other nerves in the body. This is called referred pain. Pain associated with sciatica often is severe, sharp and shooting. It may be accompanied by other symptom, such as numbness, tingling, weakness and sensitivity to touch. There is only conservative treatment giving short term relief in pain or surgical intervention with side effect. But these are not successful and therefore those who are suffering from this are always in search of result oriented remedy. Walking distance and SLR test were taken for assessment parameter, VAS score was adopted for pain. Before treatment patient was not able to walk even 4 to 5 steps due to severe pain, was brought on stretcher and his SLR was 30° of right side. After 22 days of treatment he was able to walk up to 500 meters without any difficulty, SLR was changed to 60° and patient had got 80 % relief in pain. This case report showed that Ayurvedic protocol is potent and safe in the treatment of Gridhrasi.

Abstract Blood sample collection from the caudal vena cava at the site of uterine–ovarian drainage provides a more exact evaluation of the concentration and pattern of secretion of uterine or ovarian secreted products for studies of reproductive processes in cyclic and pregnant cattle compared with samples collected from general circulation. This paper describes a thorough and updated procedure for cannulating the coccygeal vein into the caudal vena cava for the collection of serial blood samples at or near the site of uterine–ovarian drainage. Concentrations of progesterone were quantified in cows of different reproductive tract sizes with an active corpus luteum to assess the distance for proper catheter placement compared with circulating concentrations collected from the jugular vein. This procedure has a low risk for side effects, can be used effectively in pregnant animals with no major consequence to the viability of the pregnancy, and provides means for frequent collections up to 12 d.

Abstract Background Bedside ultrasound helps to estimate volume status in critically ill patients and has traditionally relied on diameter, respiratory variation, and collapsibility of the inferior vena cava (IVC) to reflect fluid status. We evaluated collapsibility of the internal jugular vein (IJ) with ultrasound and correlated it with concomitant right heart catheterization (RHC) measurements in patients with presumed pulmonary hypertension. Methods and results We studied 71 patients undergoing RHC for evaluation of pulmonary hypertension. Using two-dimensional ultrasound (Sonosite, Washington, USA), we measured the diameter of the IJ at rest, during respiratory variation, and during manual compression. Collapsibility index during respiration (respiratory CI) and during manual compression (compression CI) was calculated. We correlated mean right atrial pressure (mRAP) and pulmonary artery occlusion pressure (PAOP) defined by RHC measurements with respiratory and compression CI. A secondary goal was examining correlations between CI calculations and B-type natriuretic peptide (BNP) levels. Baseline characteristics demonstrated female predominance (n = 51; 71.8%), mean age 59.5 years, and BMI 27.3. There were significant correlations between decrease in compression CI and increase in both mRAP (Spearman: − 0.43; p value = 0.0002) and PAOP (Spearman: − 0.35; p value = 0.0027). In contrast, there was no significant correlation between respiratory CI and either mRAP (Spearman: − 0.14; p value = 0.35) or PAOP (Spearman:− 0.12; p value = 0.31). We also observed significant negative correlation between compression CI and BNP (Spearman: − 0.31; p value = 0.01) but not between respiratory CI and BNP (Spearman: − 0.12; p value = 0.35). Conclusion Increasing use of ultrasound has led to innovative techniques for estimating volume status. While prior ultrasound studies have used clinical parameters to estimate fluid status, our study used RHC measurements and demonstrated that compression CI potentially reflects directly measured mRAP and PAOP.

Hereditary tyrosinemia type 1 (HT1) is an inherited condition in which the body is unable to break down the amino acid tyrosine due to mutations in the fumarylacetoacetate hydrolase (FAH) gene, coding for the final enzyme of the tyrosine degradation pathway. As a consequence, HT1 patients accumulate toxic tyrosine derivatives causing severe liver damage. Since its introduction, the drug nitisinone (NTBC) has offered a life-saving treatment that inhibits the upstream enzyme 4-hydroxyphenylpyruvate dioxygenase (HPD), thereby preventing production of downstream toxic metabolites. However, HT1 patients under NTBC therapy remain unable to degrade tyrosine. To control the disease and side-effects of the drug, HT1 patients need to take NTBC as an adjunct to a lifelong tyrosine and phenylalanine restricted diet. As a consequence of this strict therapeutic regime, drug compliance issues can arise with significant influence on patient health. In this study, we investigated the molecular impact of short-term NTBC therapy discontinuation on liver tissue of Fah-deficient mice. We found that after seven days of NTBC withdrawal, molecular pathways related to oxidative stress, glutathione metabolism, and liver regeneration were mostly affected. More specifically, NRF2-mediated oxidative stress response and several toxicological gene classes related to reactive oxygen species metabolism were significantly modulated. We observed that the expression of several key glutathione metabolism related genes including Slc7a11 and Ggt1 was highly increased after short-term NTBC therapy deprivation. This stress response was associated with the transcriptional activation of several markers of liver progenitor cells including Atf3, Cyr61, Ddr1, Epcam, Elovl7, and Glis3, indicating a concreted activation of liver regeneration early after NTBC withdrawal.

In a former communication we incidentally mentioned that in a rabbit killed by the injection of cobra-poison into the jugular vein we had observed the pulmonary vein pulsating after all motion had ceased in the cavities of the heart. We have since observed the same phenomenon three or four times under conditions which show that this pulsation is not due to the action of the cobra-poison with which the animal in which we first observed it had been killed. The following example will show the changes in rhythm observed in these pulsations. A cat was chloroformed, and the vagi exposed and irritated by an interrupted current. Artificial respiration was kept up by air containing chloroform vapour, and the thorax was then opened, and a solution of atropia injected directly into the heart by means of a Wood’s syringe. The vagi were again irritated, but without any effect being produced on the heart, the inhibitory apparatus in it being evidently paralyzed by the atropia. A solution of glycerine extract of physostigma was now injected into the heart in a similar way. The vagi were now irritated again, and the heart stood still, the effect of the atropia having been counteracted by the physostigma. After the irritation ceased the heart again commenced to pulsate.
