In folk medicine in the Baikal region, an infusion from the aerial part of the plant Bupleurum scorzonerifolium (Willd) is widely used as a choleretic and anti-inflammatory agent. The choleretic effect of herbal remedies is mainly due to the content of phenylpropanoids and flavonoids. Using high performance liquid chromatography with a diode-matrix detector and 3Q electrospray ionization detector, we studied the metabolic profile of the phenylpropanoids and flavonoids of the aboveground organs of B. scorzonerifolium (Willd.), which grows in the Baikal region. In the aboveground organs of B. scorzonerifolium, 7 phenylpropanoids and 17 flavonoids were identified, of which 15 compounds for the species were identified for the first time. Quantitative determination of individual components was carried out in the aboveground organs of B. scorzonerifolium and dry extract by microcolumn HPLC with UV detection. The predominant components in the aerial part of the plant are 5-O-caffeic quinic acid, 3,5-di-O-caffeic quinic acid, isoquercitrin, isorhamnetin-3-O-glucoside, kaempferol-3-O-rhamnoside and rutin. The dry extract obtained from the aerial part of B. scorzonerifolium contains the entire complex of phenylpropanoids and flavonoids, which are identified in the plant. The content of active ingredients in the extract is 2.5–3.0 times higher than in the aboveground organs. The aboveground organs and dry extract of B. scorzonerifolium can be used as a source of phenylpropanoids and flavonoids.

The article is devoted to investigate the changes in the composition of biologically active substances (essential oils, lipid fraction, phenolic compounds) of the aerial part of the Bupleurum scorzonerifolium Willd. in depending on the phase of plant development. The greatest variety of compounds included in the composition of essential oils is characterized by a flowering phase. Fifteen terpene compounds – p-cymol, β-myrcene, trans-β-ocymene, limonene, (E,E)-α-farnesene, α-copaene, β-elemene, humulene, germacrene D, caryophyllene, γ-muurolene, bicyclogermacrene, Δ-cadinene, t-muurolol, spatulenol – are constant components of essential oils, regardless of phenophase. The quantitative content of these components varies from trace to major at different phenological phases. The coincidence of the main components of the lipid fraction was established. The main saturated acid is 16:0. Its content changes depending on the phase of vegetation – it is approximately at the same level determined in the phases of vegetation and flowering phases (26.78% and 27.26%, respectively) and there is a decrease in the fruit phase (18.17%). The main monounsaturated fatty acid is 18:1n9. Its greatest content was found in the fruiting phase, the smallest – flowering. The predominant polyunsaturated fatty acid is 18:2n9,12. Its greatest content was found in the vegetation phase (26.78%), the lowest - flowering (24.23%). However, the total content of unsaturated fatty acids exceeds the total content of saturated fatty acids in all phases of plant development. Although, their ratio varies from phenophase. There is a general pattern of highest content of phenolic compounds in the flowering phase for all studied samples. It was shown that during the flowering phase, the maximum accumulation of essential oils, lipids and phenolic compounds is observed.

Background: Chromatographic techniques such as TLC basically and, HPLC, GC, HPTLC equipped with various detectors are most frequently used for the qualitative and quantitative examination of herbals. Method: An overview of the recent literature concerning the usage of HPTLC for the analysis of medicinal plants has been reviewed. Results: During the last decade/s, HPTLC, a modern, sophisticated and automatized TLC technique with better and advanced separation efficiency, detection limit, data acquisition and processing, has been used for the analysis of herbal materials and preparations since the rapid development of technology in chromatography world. HPTLC with various detectors is a powerful analytical tool especially for the phytochemical applications such as herbal drug quantification and fingerprint analysis. Conclusion: In this review, a latest perspective has been established and some of the previous studies were summarized for the usage of HPTLC in the analysis of herbal remedies, dietary supplements and nutraceuticals.

The genus Scorzonera comprises nearly 200 species, naturally occurring in Europe, Asia, and northern parts of Africa. Plants belonging to the Scorzonera genus have been a significant part of folk medicine in Asia, especially China, Mongolia, and Turkey for centuries. Therefore, they have become the subject of research regarding their phytochemical composition and biological activity. The aim of this review is to present and assess the phytochemical composition, and bioactive potential of species within the genus Scorzonera. Studies have shown the presence of many bioactive compounds like triterpenoids, sesquiterpenoids, flavonoids, or caffeic acid and quinic acid derivatives in extracts obtained from aerial and subaerial parts of the plants. The antioxidant and cytotoxic properties have been evaluated, together with the mechanism of anti-inflammatory, analgesic, and hepatoprotective activity. Scorzonera species have also been investigated for their activity against several bacteria and fungi strains. Despite mild cytotoxicity against cancer cell lines in vitro, the bioactive properties in wound healing therapy and the treatment of microbial infections might, in perspective, be the starting point for the research on Scorzonera species as active agents in medical products designed for miscellaneous skin conditions.

Background: Some Bupleurum species, such as the Bupleurum chinense DC. or the Bupleurum scorzonerifolium Willd have been extensively studied (especially their roots) for the treatment of inflammation. In contrast, only compounds extracted from the aerial parts of Bupleurum rotundifolium have been studied and showed anti-inflammatory or antiproliferative activities. This study was conducted to investigate the antioxidant, anti-inflammatory, and immunomodulatory effects of Bupleurum rotundifolium roots. Methods: To tackle the various aspects of inflammation, we studied in vitro a methanolic extract from the roots of Bupleurum rotundifolium on peripheral blood mononuclear cells (PBMCs), polymorphonuclear neutrophils (PMNs), and the monocytic cells THP-1. Its antioxidant capacities and iron-chelating activity were assessed. The extract was tested on THP-1 differentiation, reactive oxygen species (ROS) production by leukocytes, neutrophils chemotaxis, cytokines, PGE2 production, and NF-κB activation in PBMCs. Results: The extract showed a decreased ROS production in stimulated cells. It increased PBMC chemokine secretion and up-regulated the differentiation of THP-1 monocytes into macrophage-like cells, indicating a potential interest of the extract in the resolution of acute inflammation. In addition, the analysis of cytokine production suggests that Bupleurum rotundifolium has immunomodulatory properties. Conclusions: Cytokines secretion, especially IL-1β and IL-12p70, provided us with a set of indicators suggesting that the extract might be able to drive the polarization of macrophages and lymphocytes toward a Th2 anti-inflammatory profile in excessive inflammation.

Sea cucumbers are an important ingredient of traditional folk medicine in many Asian countries, which are well-known for their medicinal, nutraceutical, and food values due to producing an impressive range of distinctive natural bioactive compounds. Triterpene glycosides are the most abundant and prime secondary metabolites reported in this species. They possess numerous biological activities ranging from anti-tumour, wound healing, hypolipidemia, pain relieving, the improvement of nonalcoholic fatty livers, anti-hyperuricemia, the induction of bone marrow hematopoiesis, anti-hypertension, and cosmetics and anti-ageing properties. This study was designed to purify and elucidate the structure of saponin contents of the body wall of sea cucumber Holothuria lessoni and to compare the distribution of saponins of the body wall with that of the viscera. The body wall was extracted with 70% ethanol, and purified by a liquid-liquid partition chromatography, followed by isobutanol extraction. A high-performance centrifugal partition chromatography (HPCPC) was conducted on the saponin-enriched mixture to obtain saponins with a high purity. The resultant purified saponins were analyzed using MALDI-MS/MS and ESI-MS/MS. The integrated and hyphenated MS and HPCPC analyses revealed the presence of 89 saponin congeners, including 35 new and 54 known saponins, in the body wall in which the majority of glycosides are of the holostane type. As a result, and in conjunction with existing literature, the structure of four novel acetylated saponins, namely lessoniosides H, I, J, and K were characterized. The identified triterpene glycosides showed potent antifungal activities against tested fungi, but had no antibacterial effects on the bacterium Staphylococcus aureus. The presence of a wide range of saponins with potential applications is promising for cosmeceutical, medicinal, and pharmaceutical products to improve human health.

Diabetes mellitus has been treated orally with herbal remedies based on folk medicine since ancient times.Embelia ribes burm(Myrsinaceae), known commonly as vidanga, was used in Ayurveda for its anthelmintic activity. Ayurveda describes vidanga as pungent, causes increase in digestive fire, and cures flatulence and colic. A single study reported the antihyperglycemic activity of decoction ofE. ribesin glucose-induced hyperglycemic albino rabbits. In the present study, the lipid-lowering and antioxidant potential of ethanolic extract ofE. ribes burmwas investigated in streptozotocin (40 mg/kg, IV, single injection)-induced diabetes in rats. Twenty days of orally feeding the extract (200 mg/kg) to diabetic rats resulted in significant (P< 0.01) decrease in blood glucose, serum total cholesterol, and triglycerides, and increase in HDLcholesterol levels when compared to pathogenic diabetic rats. Further, the extract also lowered the liver and pancreas thiobarbituric acid–reactive substances (TBARSs) values (P< 0.01) when compared to TBARS values of liver and pancreas of pathogenic diabetic rats. The results of test drug were comparable to gliclazide (25 mg/kg, orally), a standard antihyperglycemic agent. This is the first pilot study to provide biochemical evidence of potential ofE. ribesin diabetic dyslipidemia.
