AbstractObjectivesThe aim of the present study is to evaluate the diagnostic powers of fractional magnesium, sodium and potassium as markers of renal tubular damage in normotensive type 2 diabetes mellitus (T2DM) patients with respect to microalbuminuria and estimated glomerular filtration rate (eGFR).Materials and methodsForty healthy volunteers and 91 normotensive T2DM patients were included in the study. Patient group was divided into two according to albuminuria level; 49 were normoalbuminuric and 42 were microalbuminuric. In addition to albumin in urine, urine and serum Na, K, Mg and creatinine values were measured to calculate fractional electrolyte excretion rates.ResultsIn normoalbuminuric and microalbuminuric groups, fractional excretion of magnesium (FEMg) values were found to be significantly higher than the control group (p < 0.05). There was a moderate correlation between FEMg and albümin to cratinin ratio (ACR) (Spearman r = 0.3215, p < 0.05). In the ROC analysis for eGFR and FEMg based on microalbuminuria, the areas under the curve were 0.625 and 0.732, respectively (diagnostic sensitivity 59.52% and 66.67%; specificity 70.79% and 77.53%, p < 0.05).ConclusionFor renal tubular damage predicted by microalbuminuria, FEMg could be accepted as a candidate biochemical marker with diagnostic and prognostic value.

Abstract: Kidney damage can be caused by many things, such as hypovolemia, sepsis, acute glomeluronefritis, rhabdomyolysis and drugs (gentamicin and NSAID). Gentamicin is an broad spectrum antimicrobial with high toxicity. Gentamicin is an aminoglycoside antibiotic known to be toxic to the kidneys and the side effects that gentamicin cause are renal tubular damage. There are many kinds of herbal plant in Indonesia with benefit, one of them is the binahong plants. Binahong (Anredera cordifoli (Ten) Steenis), has a high antioxidant that is equal to 9.614% of the compound flavonoid.5 Antioxidant contained in binahong leaves can be said as nefroprotective on kidney function. This study aims to reveal the renal histopathologic of wistar rat that have administered binahong extract after gentamicin administration. This study was an experimental study using 25 wistar rats that were divided into 5 groups. Group A is the negative control group (terminated the 7th day), while group B, C, D and E (the treatment group) were administered gentamicin 0.3 ml/day for 6 days. After administration of gentamicin, group B immediately terminated (day 7), group C were administered 50 mg/day of binahong extratct for 3 days (terminated on day 10), group D were administered 100 mg/day of binahong extract for 3 days (terminated on day 10), group E were administered gentamicin for 6 days (terminated on day 10). The results showed that the renal histopathological of wistar rats group that were administered gentamicin for 6 days, showed swelling and necrotic tubular epithelial cells. Wistar rats group that were administered of binahong showed regeneration of renal tubular epithelial cells. Conclusion: The administration of Gentamicin with toxical dose or 0.3 ml/day for 6 days showed acute tubular necrosis. Regeneration of renal tubular epithelial cells is better in the group that were administered binahong extract than the group that were administered pellets. Renal histopathologic of wistar rats at administration of binahong extract with dose of 100 mg are better compared to 50 mg. Keywords: gentamicin, binahong extract, histopathologogical image of wistar rat’s kidney Abstrak: Kerusakan ginjal dapat disebabkan oleh berbagai hal, seperti hipovolemia, sepsis, glomeluronefritis akut, rabdomiolisis dan obat-obatan (Gentamisin dan NSAID). Abstract Jianpi Qushi Heluo Formula (JQHF) is an empirical traditional Chinese medicine prescription for treating membranous nephropathy (MN) clinically in China. The therapeutic effect of JQHF has been reported in our previous studies. However, the exact mechanism is still unknown. In this study, by establishing an experimental rat model of MN induced by Sheep anti-rat Fx1A serum, we evaluated the effects of JQHF and Tetrandrine(TET), and Benazepril was used as a positive control group. As an autophagy agonist, TET is one of the most active components in JQHF. After 4 weeks, significant kidney damage was observed in the rats in the Model group; comparatively, JQHF markedly decreased 24 hour urinary protein, total cholesterol (TC) and increased serum albumin (ALB). Histology showed that JQHF significant improvements of glomerular hyperplasia, renal tubular damage, IgG immune complex deposition and the ultrastructure of mitochondria in MN rats. Flow cytometry analysis showed that treatment with JQHF decreasing the level of Reactive Oxygen Species and apoptosis rate, upregulating the level of mitochondrial membrane potential. Western blot analysis demonstrated that JQHF could protect against mitochondrial dysfunction and apoptosis by upregulating the expression of Parkin and LC3II, and downregulating the expression of Cytochrome c and Cleaved caspase-3 in the kidneys of MN rats. Similarly, TET treatment significantly upregulates the mitochondrial autophagy and decrease the apoptosis of rats after 4 weeks compared with the Model group. Notably, combined with the above results, the ability to alleviates renal damages of JQHF was significantly better than those of Benazepril and TET. It was demonstrated that JQHF could delay pathology damage to the kidney and hold back from the progression of MN by inhibiting apoptosis and upregulating the mitochondrial autophagy by Parkin pathways.

ABSTRACT Twenty newly diagnosed, but as yet untreated patients of both sexes with classical juvenile diabetes were investigated by determining the mean plasma immunoreactive growth hormone (IRHGH) and urinary IRHGH for a 24 hour period before and during initial insulin treatment. The plasma IRHGH was significantly higher (0.05 > P > 0.01) before than during initial insulin treatment. During initial insulin treatment, the mean plasma IRHGH was significantly higher (0.01 > P > 0.001) than in a control group. The urinary IRHGH was significantly higher (0.01 > P > 0.001) before than during insulin treatment. The increased urinary IRHGH observed before insulin treatment is thought to be partly due to a defective renal tubular reabsorption of growth hormone. No significant correlation was found between the mean blood sugar and plasma or urinary IRHGH either before or during insulin treatment.

AbstractAbnormal renin-angiotensin system (RAS) activation plays a critical role in the initiation and progression of chronic kidney disease (CKD) by directly mediating renal tubular cell apoptosis. Our previous study showed that necroptosis may play a more important role than apoptosis in mediating renal tubular cell loss in chronic renal injury rats, but the mechanism involved remains unknown. Here, we investigate whether blocking the angiotensin II type 1 receptor (AT1R) and/or angiotensin II type 2 receptor (AT2R) beneficially alleviates renal tubular cell necroptosis and chronic kidney injury. In an angiotensin II (Ang II)-induced renal injury mouse model, we found that blocking AT1R and AT2R effectively mitigates Ang II-induced increases in necroptotic tubular epithelial cell percentages, necroptosis-related RIP3 and MLKL protein expression, serum creatinine and blood urea nitrogen levels, and tubular damage scores. Furthermore, inhibition of AT1R and AT2R diminishes Ang II-induced necroptosis in HK-2 cells and the AT2 agonist CGP42112A increases the percentage of necroptotic HK-2 cells. In addition, the current study also demonstrates that Losartan and PD123319 effectively mitigated the Ang II-induced increases in Fas and FasL signaling molecule expression. Importantly, disruption of FasL significantly suppressed Ang II-induced increases in necroptotic HK-2 cell percentages, and necroptosis-related proteins. These results suggest that Fas and FasL, as subsequent signaling molecules of AT1R and AT2R, might involve in Ang II-induced necroptosis. Taken together, our results suggest that Ang II-induced necroptosis of renal tubular cell might be involved both AT1R and AT2R and the subsequent expression of Fas, FasL signaling. Thus, AT1R and AT2R might function as critical mediators.

ABSTRACT: The present study evaluated the efficacy of a single oral dose of doramectin in the control of Psoroptes ovis and Leporacarus gibbus in naturally infested rabbits. Sixteen adult rabbits were selected and distributed in two experimental groups. The treated group received 200 μg/Kg of oral doramectin and the control group received the same volume of saline solution. The diagnosis of the mites was made with a stereoscopic microscope. Hairs from the dorsal part of the neck, lumbar right, lumbar left, ventral side of the tail and ventral abdomen were evaluated for L. gibbus, and ear wax evaluated for P. ovis. The evaluation of the efficiency and the clinical assessment of the lesions was made in days 0, +3, +7, +14, +21, +28, and +35 after treatment. An efficacy of 75% and 87,5% was observed for L. gibbus in days +3 and +7 after treatment, an efficacy of 100% was observed in days +14, + 21, +28 e +35. An efficacy of 100% for the control of P. ovis was observed following day +7. The clinical lesion score of the control group remained unaltered, except for one animal which conditions worsened during experimentation. In the treated group animals, regression of the lesions was observed following day +3, and on day +21 no signal of infestation by P. ovis was present. None of the animals from the treated group presented secondary collateral effects caused by the doramectin, which proved itself as an optimal alternative for mite control in naturally infested rabbits.

Crumb rubber, as a recycled material used in asphalt mixture, has gained more attention in recent years due to environmental benefits and the advantages of its pavement, such as excellent resistance to cracking, improved durability, less road maintenance, lower road noise, etc. However, the high-temperature performance of mixture with crumb rubber does not perform well. In order to improve the performance, this paper examined the effect of additives on the laboratory performance of asphalt rubber Stone Matrix Asphalt (AR-SMA) with additives. Three groups of AR-SMA: no additives, Styrene–Butadiene–Styrene (SBS) and Granular Polymer Durable additive (GPDa) were included, with no additives as a control group. Each group was investigated at three asphalt rubber content (ARC): 6.4%, 6.9%, 7.4% with regard to high-temperature and fatigue properties. The results show that with increasing ARC, the high-temperature performance of mixture without additive decreases, and the high-temperature performance increases first and then decreases for SBS and GPDa. Moreover, the rutting resistance of AR-SMA with GPDa at 6.9% ARC performs best. Under the condition of mixtures with appropriate ARC, AR-SMA with GPDa has higher fatigue life and sensitivity to fatigue cracking than the control group. Simultaneously, the fatigue performance of AR-SMA with GPDa is not as significant as that without additive with increasing ARC. In a word, GPDa is a good choice to improve the performance of AR-SMA. However, it should be noted that optimal asphalt content of AR-SMA mixtures with GPDa is higher than that of traditional mixtures.

Reading ability seems to involve analytic-sequential processing of selected letters and a holistic-simultaneous perception of the salient features of the entire word. The dyslexic child, being normal in intelligence, is most likely deficient in either one of the two processes while being normal in the other. To test this hypothesis, 28 reading disabled children, on the basis of the nature of errors made in a writing from dictation task, were divided into two groups: analytic-sequential deficient and holistic-simultaneous deficient. Further testing showed that the first group was poor in processing a sequence of digits but normal on holistic-simultaneous memory tasks. The opposite pattern of performance was shown by the second group. A control group of 14 normal readers did not show such an imbalance.
