Evaluation of efficacy of Bombax ceiba extract and its major constituent, mangiferin in streptozotocin (STZ)-induced diabetic rats

AbstractObjectivesBased on the ethno-medicinal use of Bombax ceiba leaf, in the treatment of diabetes, the present study is aimed at evaluation of antidiabetic potential of leaf extract and its major constituent mangiferin.MethodsEfficacy of hydroalcoholic extract of Bombax ceiba leaf (BCL, 200 and 400 mg/kg body wt.) and mangiferin (MF, 20 mg/kg body wt) was studied in streptozotocin (STZ)-induced diabetic rats and associated complications visually, retinopathy, cardiopathy and nephropathy. After 20 days, serum glucose, lipid profiles, glycol-hemoglobin % (HbA1c%), liver enzymes activity and glycogen content, and histopathology of the pancreas were corroborated. The study was coxswained for development of validated RP-HPLC method for the estimation of MF in BCL.ResultsThe results demonstrated significant reductions in the levels of glucose (p<0.001), glycated hemoglobin (HbA1c%, p<0.001), cholesterol, triglycerides and low-density lipoproteins, and concurrent elevation of high density lipoproteins level in the groups administered BCL and MF relative to the controls. It significantly reversed most of the altered metabolic and oxidative stress parameters and histopathological changes. Mangiferin content in BCL was found to be 0.04%w/w.ConclusionsThe anti-diabetic effects of BCL may be attributed to its ability to enhance insulin release, antioxidant and hypolipidemic potential.

INTRODUCTION – Diabetes Mellitus is an emerging endocrine and metabolic disorder which has affected millions of people globally. Homeopathy a science of ultra-molecular doses with a holistic approach is used in treatment of Diabetes Mellitus. Homeopathic medicines are prepared from plant, mineral, sarcodes, nosodes and animal parts. Insulinum, Pancreatinum and Uranium nitricum is used in homeopathy for treatment of Diabetes Mellitus. However no any preclinical studies have been investigated for the anti-diabetic effect and its safety. METHODS – Homeopathic medicines Insulinum, Pancreatinum and Uranium nitricum in 6C (1012) dilution factor were used to examine antihyperglycemic effects in streptozotocin induced diabetic rats. Caesalpinia bonduc has been used in herbal medicines for the treatment of a wide range of diseases from decades. The present study has explored the remedial potential and underlying mechanism of polyphenol extract of Caesalpinia bonduc in alloxanized diabetic rats. HPLC/MS analysis confirmed the presence of phenolics in considerable concentrations in Caesalpinia bonduc extract. Administration of different doses (250 and 500 mg/kg) of CPP extract to hyperglycemic rats for 8 weeks restored blood and serum glucose, insulin, glycosylated hemoglobin, leptin, amylin, and carbohydrate metabolizing enzymes level towards normal compared to alloxanized diabetic group. The effect of CPP extract on various genes such as Pdx-1, Ins-1, ngn-3, GLUT-4, and IRS-1 in insulin signaling pathway and Traf-4, Traf-6, and Mapk-8 in MAPK downstream JNK cascade was examined through qRT-PCR to access the core molecular mechanism involved in CPP-induced recovery of diabetes. Results have revealed that CPP extract reduced oxidative stress in pancreatic β cells by restoring free radical scavenging potential, reducing the mRNA expression of Mapk-8, Traf-4, and Traf-6, and increasing the Pdx-1, Ins-1, ngn-3, GLUT-4, and IRS-1 expression ensuing regeneration of β cells and subsequent insulin release from pancreas. The results obtained in this study recommend that CPP extract may be a promising therapeutic restorative agent in the treatment of diabetes mellitus.

The present study was carried out to investigate the antidiabetic and antihyperlipidemic properties of hydroalcoholic extract of Peganum harmala in streptozotocin-induced diabetic male rats. In an experimental study, 64 normal Wistar albino male rats (200–230 g) were randomly divided into 8 groups. Control and diabetic rats were treated with normal saline and three different doses (30, 60, and 120 mg/kg) of hydroalcoholic extract of Peganum harmala seeds for 4 weeks orally. At the end of treatment, blood samples were taken and glucose, triglycerides, total cholesterol, LDL-c, HDL-c, malondialdehyde (MDA), total antioxidant capacity (TCA), ALT, AST, GGT, bilirubin, and glycosylated hemoglobin (HbA1C) were determined. STZ-induced diabetic rats showed significant changes in the values of glucose, triglycerides, total cholesterol, LDL-c, MDA, TAC, ALT, AST, GGT, bilirubin, and HbA1C in comparison with normal rats. Administration of the extract to diabetic rats resulted in a remarkable decrease in glucose, lipid profiles, MDA, ALT, AST, GGT, bilirubin, and HbA1C levels and increase in TAC relative to diabetic group. The results of this study indicated that hydroalcoholic extract of Peganum harmala seeds possesses antidiabetic and hypolipidemic activities and could be useful in treatment of diabetes.

Oxidative stress is believed to be a pathogenic factor in the development of diabetic complications. In the present study, we aimed to evaluate the effects of different fractions of heart wood of Pterocarpus marsupium on antioxidant enzyme like protein thiols and also check the efficacy of the extract for the protection of the renal function in alloxan induced diabetic rats. The present study also investigates the levels of liver glycogen which are considered as the best biomarker for assaying the hypoglycemic activity of any drug. Diabetes was induced by administering alloxan dissolved in saline, while the normal control group was given propylene glycol. Diabetes induced animals were randomly assigned into different groups. Blood samples were collected from all the experimental and control groups. Estimation of urea, uric acid and creatinine along with protein thiols was made on day 30 only. At the end, all the animals were sacrificed to collect liver tissue to analyze glycogen content. The 30 days treatment with various extracts (75 mg/kg body wt) significantly lowered protein thiol levels, which probably represents increased utilization for neutralizing free radicals. There was no significant increase in the levels of renal parameters in the extract treated groups which revealed that the employed dose of the extract is nontoxic to the kidney. There was also a significant decrease in the glycogen content in insulin and alcohol-extract treated groups and should be encouraging for the treatment of diabetes mellitus. The extract showed a promising antioxidant effect, as well as hypoglycemic activity, and should be encouraged for the treatment of diabetes.

Zingiber officinale (ZO), commonly known as ginger, has been traditionally used in the treatment of diabetes mellitus. Several studies have reported the hypoglycaemic properties of ginger in animal models. The present study evaluated the antihyperglycaemic effect of its aqueous extract administered orally (daily) in three different doses (100, 300, 500 mg/kg body weight) for a period of 30 d to streptozotocin (STZ)-induced diabetic rats. A dose-dependent antihyperglycaemic effect revealed a decrease of plasma glucose levels by 38 and 68 % on the 15th and 30th day, respectively, after the rats were given 500 mg/kg. The 500 mg/kg ZO significantly (P < 0·05) decreased kidney weight (% body weight) in ZO-treated diabetic rats v. control rats, although the decrease in liver weight (% body weight) was not statistically significant. Kidney glycogen content increased significantly (P < 0·05) while liver and skeletal muscle glycogen content decreased significantly (P < 0·05) in diabetic controls v. normal controls. ZO (500 mg/kg) also significantly decreased kidney glycogen (P < 0·05) and increased liver and skeletal muscle glycogen in STZ-diabetic rats when compared to diabetic controls. Activities of glucokinase, phosphofructokinase and pyruvate kinase in diabetic controls were decreased by 94, 53 and 61 %, respectively, when compared to normal controls; and ZO significantly increased (P < 0·05) those enzymes' activities in STZ-diabetic rats. Therefore, the present study showed that ginger is a potential phytomedicine for the treatment of diabetes through its effects on the activities of glycolytic enzymes.

One group was untreated and fed with Nrp, four groups were fed with 25 %, 50 %, 75 % or 100 % Xs, one group was fed with 100 % Xs and administered with glibenclamide, while a 7th group was fed with Nrp and administered with glibenclamide.ResultsThis study shows that treatment of diabetes with corm ofConclusionscorm can be recommended for inclusion in diets of diabetics without causing further deterioration of health of the diabetic patients.

Objective: The comparative effects of food restriction (FR), Diabecon treatment (DT) and their combined therapy (FR+DT) were studied in the regulation of alloxan-induced diabetes mellitus, alterations in thyroid hormones (TH) and oxidative stress. Methods: Young diabetic rats were either kept on 50% FR and/or DT (2 gm/Kg body weight) for two months and then alterations in serum glucose, insulin, TH concentrations, hepatic glycogen and pancreatic antioxidants along with oxidative stress markers were evaluated. Results: Significantly increased serum glucose, tissue stress markers with decreased TH, hepatic glycogen (P<0.0001 for all) and pancreatic antioxidants (P<0.05-0.001) were observed in diabetic rats. Rats kept on different therapies exhibited significant (P<0.05) improvements than diabetic rats in all studied parameters. FR+DT group showed a significantly more decrease in serum glucose (P<0.05) that FR or DT group, while in other parameters improvement was found to be more or less equally improved in all treated groups. Conclusion: FR appeared to mimic the effects of Diabecon in most of the indices. However, FR+DT appears to be more effective. Possibly both therapies ameliorate diabetes and oxidative stress following some common metabolic pathways.
