EFFECT OF CALCITONIN ON THE REGENERATION OF A CIRCUMSCRIBED BONE DEFECT (BORED HOLE IN THE RAT TIBIA)

ABSTRACT In 20 intact and 20 parathyroidectomized female Sprague-Dawley rats, holes with a diameter of 1.8 mm were bored into the proximal third of the tibia. Half the animals served as controls and received the vehicle, the remainder received daily subcutaneous injections of 100 MRCmU calcitonin in 5 % gelatin. Tetracycline labelling was performed weekly. 5 animals from each group were sacrificed after 3 weeks, the other 5 after 6 weeks. Nondecalcified ground sections through the center of the holes were evaluated with an integration ocular; the surface of the newly formed bone was compared with the original size of the defect. The course of the regeneration for the control animals was in agreement with previous data in the literature. Calcitonin treatment produced a distinct acceleration of the healing process. Presumably, the hormone does not only act via inhibition of bone resorption, but also by stimulating the activity of osteoblasts and by increasing the mineralization of the osteoid seams.

Background: Carbonate hydroxyapatite (CHA) is used as a scaffold to repair bone resorption. Alveolar bone resorption in periodontitis caused by an infection requires the presence of an antibacterial to support bone regeneration. Human β-defensin 3 (HBD3) is an antimicrobial peptide. The local application of the HBD3 antimicrobial is beneficial to inhibiting drug resistance and protecting tissue regeneration against invasive bacteria. Purpose: This study aims to investigate the effect of the administration of a combination of   CHA with HBD3 on the collagen density of periodontitis rats (Sprague Dawley). Methods: This study was a true experimental study with a post-test control group design. Thirty-two Sprague Dawley animal models were randomly blind selected placed under anaesthetic, then a 2-mm silk ligature was attached as a ligation to the mandibular incisors for 14 days in order to generate periodontitis. The study subjects were divided into two groups, the group with CHA and CHA loaded HBD3 (CHA + HBD3) implantation. On days 7, 14, 21 and 28, four rats were taken randomly from each group for decapitation, followed by histological processing and examination with trichome Mallory staining. The data was analysed using the Kruskal Wallis test (p<0.05). Results: An increase in collagen density during the healing process was found. There was a significant difference between CHA and CHA+HBD (p=0.004 and p=0.008; p<0.05) in collagen density between the groups. Conclusion: The combination of CHA and HBD3 can enhance the collagen density in periodontitis Sprague Dawley rats, compared to CHA only groups.

The aim of the study was to evaluate if a pre-incisional N-acetylcysteine (NAC) treatment altered the process of wound healing in a rat model. The dorsal skin of 24 Sprague-Dawley rats was incised in six locations. Before the incisions were made, skin was injected either with lidocaine and epinephrine (one side) or with these agents supplemented with 0.015%, 0.03%, or 0.045% NAC (contralaterally). Photographic documentation of the wound healing process was made at 11 time points. Rats were sacrificed 3, 7, 14, or 60 days after incision to excise scars for histological analysis. They included: Abramov scale scoring, histomorphometry analysis, and collagen fiber arrangement assessment. Skin pretreated with 0.03% NAC produced the shortest scars at all analyzed time points, though this result was statistically insignificant. At this NAC concentration the scars had smaller areas on the third day and were narrower on the day 4 compared with all the other groups (p < 0.05). On day 7, at the same concentration of NAC, the scars had a higher superficial concentration index (p = 0.03) and larger dermal proliferation area (p = 0.04). NAC addition to pre-incisional anesthetic solution decreased wound size and width at an early stage of scar formation at all concentrations; however, with optimal results at 0.03% concentration.

Abstract Background Diffusion tensor imaging (DTI) is mainly used for detecting white matter fiber in the brain. From this, DTI has been applied to assess fiber in liver disorders by prior studies. But non-sufficient data has been obtained if DTI could be used for exactly staging chronic hepatitis. This study is to assess the value of DTI for staging of liver fibrosis (F), necroinflammatory activity (A), and steatosis (S) of chronic hepatitis in rats. Methods Seventy male Sprague-Dawley rats were divided into control group(n = 10) and experimental group(n = 60). The rat models of chronic hepatitis were established by abdominal subcutaneous injections of 40% CCl4. All rats underwent 3.0T MRI. ROIs were placed on DTI to estimate MR parameters (rADC value and FA value). Histopathology was the reference standard. Multiple linear regression was used to analyze the association between MR parameters and pathology. The differences in rADC value and FA value among pathological stages were evaluated by MANOVA or ANOVA. LSD was used to test the differences between each two groups. ROC analysis was performed. Results The numbers of each pathology were as follows: F0(n = 15), F1(n = 11), F2(n = 6), F3(n = 9), F4(n = 6); A0(n = 8), A1(n = 16), A2(n = 16), A3(n = 7); S0(n = 10), S1(n = 7), S2(n = 3), S3(n = 11), S4(n = 16). The rADC value had a negative correlation with liver fibrosis (r=-0.392, P = 0.008) and inflammation (r=-0.359, P = 0.015). FA value had a positive correlation with fibrosis (r = 0.409, P = 0.005). Significant differences were found in FA value between F4 and F0 ~ F3 (P = 0.03), while no significant differences among F0 ~ F3 were found (P > 0.05). AUC of FA value in differentiating F4 from F0 ~ F3 was 0.909(p < 0.001) with 83.3% Sensitivity, 85.4% specificity when the FA value was at the cut-off of 588.089(× 10− 6mm2/s). Conclusion FA value for DTI can distinguish early cirrhosis from normal, mild and moderate liver fibrosis.

Pendahuluan: Pencabutan gigi merupakan prosedur yang sering dilakukan di kedokteran gigi. Setelah pencabutan gigi akan dihasilkan suatu perlukaan. Proses penyembuhan luka dapat dipercepat pada kondisi tertentu. Salah satu bahan alami yang dapat membantu proses penyembuhan luka adalah getah tanaman betadine (Jatropha multifida L.). Penelitian bertujuan untuk menganalisis efektivitas getah tanaman betadine terhadap penyembuhan luka dan terhadap tanda-tanda infeksi pasca pencabutan gigi Metode: Desain penelitian studi eksperimental laboratorium dengan rancangan penelitian post-test only control group design menggunakan 30 ekor tikus Sprague-Dawley. Teknik pengambilan sampel yaitu convenience sampling. Sampel dibagi menjadi 2 kelompok yaitu kelompok perlakuan dan kelompok kontrol. Kelompok perlakuan diberikan getah tanaman betadine dan kelompok kontrol diberikan asam traneksamat secara oral menggunakan sonde lambung. Tunggu selama 4 jam setelah pemberian getah tanaman betadine dan asam traneksamat. Anestesi pada tikus menggunakan ketamin 50 mg/kg berat badan secara intramuskular kemudian dilakukan pencabutan pada gigi tikus. Pengamatan dilakukan dengan melihat kriteria klinis pada hari 1,3,7 dan diperhatikan sampai luka sembuh serta lihat tanda-tanda infeksi. Analisis data dilakukan dengan uji normalitasShapiro Wilik. Hasil penelitian menunjukkan bahwa data berdistribusi tidak normal. Analisis data dilanjutkan menggunakan uji statistik mann whitney. Hasil: Terdapat perbedaan signifikan penyembuhan luka soket pasca pencabutan gigi setelah diberikan getah betadine dan asam traneksamat dengan nilai p=0,037 (p<0,005). Simpulan: Pemberian getah tanaman betadine terbukti lebih efektif terhadap proses penyembuhan luka soket pasca pencabutan gigi dibandingkan dengan pemberian asam traneksamat. Kata kunci: tikus Sprague-Dawley; penyembuhan luka; pencabutan gigi; getah batang betadine ABSTRACTIntroduction: Tooth extraction is a procedure often performed in dentistry. Tooth extraction will always cause injuries. However, the wound healing process can be accelerated under certain conditions. One of the natural ingredients that can accelerate the wound healing process is betadine (Jatropha multifida L.) plant sap. The study was aimed to analyzed the effect of betadine plant sap on wound healing and signs of infection after tooth extraction. Methods: Experimental laboratory study design with post-test only control group design was conducted towards 30 Sprague-Dawley rats. The sampling technique was convenience sampling. The sample was divided into two groups, the treatment group and the control group. The treatment group was administered with betadine plant sap, and the control group was administered with tranexamic acid orally using a gastric probe, then waited 4 hours after. The anaesthesia was then performed using 50 mg/kg body weight of ketamine intramuscularly. The extraction was performed after. Observations was conducted at the clinical criteria on days 1, 3, and 7 and continue to be monitored until the wound heals. Then, the signs of infection were observed. Data analysis was carried out using the Shapiro Wilk normality test. The results showed that the data was not normally distributed. Thus, data analysis was continued using the Mann Whitney statistical test. Results: The results showed a significant difference in the healing of socket wounds after tooth extraction after being administered with betadine sap and tranexamic acid with a value of p=0.037 (p<0.005). Conclusions: Administration of betadine plant sap is proven to be more effective in accelerating the healing process of socket wounds after tooth extraction than tranexamic acid. Keywords: Sprague-Dawley rats; wound healing; pencabutan gigi; getah batang betadine

Background. This study examines the impact of one-time direct application of haemostatic agent zeolite–bentonite powder to wounded skin on the healing process in rats. Materials and Methods. 24 male Sprague-Dawley rats were randomly allocated into two groups (n=12): (1) the rats whose wounds were washed only with sterile normal saline (NS-treated) and (2) those treated with zeolite–bentonite compound (ZEO-treated). The wound was circular, full-thickness, and 2 cm in diameter. At the end of the 12th day, six animals from each group were randomly selected and terminated. The remaining rats were terminated after 21 days. Just after scarification, skin samples were excised and sent for stereological evaluation. Results. The results showed a significant difference between the two groups regarding the length density of the blood vessels and diameter of the large and small vessels on the 12th day after the wound was inflicted. Besides, volume density of both the dermis and collagen bundles was reduced by 25% in the ZEO-treated rats in comparison to the NS-treated animals after 21 days. Conclusions. One-time topical usage of zeolite–bentonite haemostatic powder on an animal skin wound might negatively affect the healing process through vasoconstriction and inhibition of neoangiogenesis.

Naltrexone (NLTX) in the nucleus of the solitary tract (NTS) decreases feeding induced by neuropeptide Y (NPY) in the paraventricular nucleus (PVN). We sought to determine the NTS region most sensitive to NLTX blockade of PVN NPY-induced feeding. Male Sprague-Dawley rats were fitted with two cannulas; one in the PVN and one in a hindbrain region: caudal, medial, or rostral NTS or 1 mm outside the NTS. Animals received NLTX (0, 1, 3, 10, and 30 μg in 0.3 μl) into the hindbrain region just prior to PVN NPY (0.5 μg, 0.3 μl) or artificial cerebrospinal fluid (0.3 μl). Food intake was measured at 2 h following injection. PVN NPY stimulated feeding, and NLTX in the medial NTS significantly decreased NPY-induced feeding at 2 h, whereas administration of NLTX in the other hindbrain regions did not significantly influence PVN NPY induced feeding. These data suggest that opioid receptors in the medial NTS are most responsive to feeding signals originating in the PVN after NPY stimulation.

Exposure of white rats to 2 °C for 20 weeks caused an immediate and sustained increase in the excretion of catechol amines as compared to controls kept at 23 °C. Adrenaline excretion increased approximately three to four times while noradrenaline excretion increased about eight times. There were no marked differences between Wistar and Sprague–Dawley rats in the amounts of adrenaline and noradrenaline excreted at 23 °C or 2 °C. 'Old' rats kept in activity cages excreted much more of the catechol amines both at 23 °C and 2 °C than did the other rats and they showed a peak for adrenaline excretion after 1 week and for noradrenaline excretion after 3 to 4 weeks in the cold.

We investigated the airway responses evoked by nasal wood smoke in anesthetized Sprague-Dawley rats. Wood smoke (5 ml, 1.4 ml/s) was delivered into an isolated nasal cavity while animals breathed spontaneously. In study 1, nasal wood smoke triggered either an apneic response ( n = 26) or a sniff-like response ( n = 16) within 1 s after smoke exposure in 42 normal rats. Both airway responses were abolished by trigeminal nerve denervation and by nasal application of a local anesthetic or a hydroxyl radical scavenger, but they were not significantly affected by removal of smoke particulates or nasal application of a saline vehicle. In study 2, nasal wood smoke only triggered a mild apneic response in two rats neonatally treated with capsaicin and had no effect on breathing in the other six; the treatment is known to chronically ablate C fibers and some Aδ fibers. In contrast, nasal wood smoke evoked an apneic response in six rats neonatally treated with the vehicle of capsaicin and elicited a sniff-like response in the other two. These results suggest that the apneic and sniff-like responses evoked by nasal wood smoke result from the stimulation of trigeminal nasal C-fiber and Aδ-fiber afferents by the gas-phase smoke and that hydroxyl radical is the triggering chemical factor.

Changes in neutrophil precursor populations in the bone marrow and their alkaline phosphatase reactivity following an inflammatory stimulus were studied in the rat using an osmiophilic method. Seven groups each of 3 Sprague-Dawley rats received subcutaneous injections of turpentine, and femoral marrow was examined at intervals up to 72 hr. Depletion of mature neutrophils resulted in an increased first in the myeloblast-promyelocyte compartment and at 48-72 hr in the myelocyte-metamyelocyte population. By 72 hr replenishment of the mature neutrophil marrow population had begun. Within 6 hr marked acceleration of cytoplasmic maturation was evident, together with accelerated synthetic activity, manifested by marked dilatation of the rough endoplasmic reticulum and an enlarged Golgi. Together with these changes there was an increase in the number of alkaline phosphatase reactive cells, which was evident first in the myeloblast-promyelocyte population at 2 hr. An absolute increase in the amount of enzyme reaction product associated with individual cells was also seen.
