The presence of microorganisms in dental structures with experimentally induced necrosis was evaluated. The materials were tested to evaluate their antimicrobial activity and tissue repair efficacy. Four dogs were used in this experiment, with a total of 64 roots of premolar teeth, divided into three groups. The root canals of Group I were filled with gutta-percha and zinc oxide/eugenol cement; Group II were filled with calcium hydroxide, and Group III were not filled. All animals were clinically and radiographically examined 15 days after surgery andthen again every subsequent 15 days until 120 days, when the teeth were extracted en bloc.Histopathological analysis showed inflammatory infiltration, cement and bone resorption andnecrotic tissue in the apical delta in different proportions. Histomicrobiological analysis showedthe presence of microorganisms inside the teeth structures, with different concentrationsaccording to the treatment used. There was statistical significance between the groups(p>0.05). Gutta-percha with zinc oxide/eugenol demonstrated good antimicrobial activity;calcium hydroxide was not efficient. The conclusion of this study is that gutta-percha withzinc oxide/eugenol is the better protocol for filling root canals in dogs.

The antimicrobial activity of filling materials was studied to improve the quality of treatment of complicated caries in vitro. Resorcin-formalin cement Resodont (“Latus”, Ukraine) - № 1; Endocort zinc oxide-eugenol cement (“Latus”, Ukraine) - No. 2; materials based on zinc oxide eugenol cement with dexamethasone: Endomethasone ivory (Septodont, France) - No. 3 and Endofil ("Produit Dentaires SA", Switzerland) - No. 4. To assess the antimicrobial activity of the studied materials, test microorganism strains were used: Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 6783, Candida albicans ATCC 885-653. As a result of the study, it was established that filling materials have different antimicrobial activity. Resodont has the greatest antimicrobial activity on the test culture. Endomethason is active in relation to the test cultures studied, inferior to Resodont in its antimicrobial properties. Endocort and Endofil have less antimicrobial activity than Resodont and Endomethason, but have approximately equal growth retardation rates. It should be noted that with respect to Staphylococcus aureus, Endocort was found to be almost 1.5 times more active than Endofil. The conducted studies confirm that the materials for filling of root canals by Resodont and Endocort (Latus, Ukraine) have a pronounced antibacterial effect. They are competitive with foreign sealers and are the materials of choice, having a significantly lower cost.

Objectives: In this study, chitosan was introduced and used as a substitute for pulpectomy obturation against conventional materials: zinc oxide eugenol (ZOE) and iodoform-calcium hydroxide (Ca(OH)2) compounds. Also, efficacies of rotary versus manual instrumentations were compared. Materials and Methods: This preliminary in-vitro study was performed on 152 intact non-resorbed root canals of primary molars divided into rotary (n=78) versus hand-instrumentation (n=74) and also into ZOE (n=53), iodoform-Ca(OH)2 (n=50), and 3% nano-chitosan (n=49). Canals were cleaned/shaped using hand or rotary files. Canal spaces were measured using spiral computed tomography (CT). Canals were then obturated using the three materials. The percentages of obturation volume (POV) were estimated. Rotary and manual instrumentations were compared in terms of canal spaces before and after obturation. Three obturation materials were compared in terms of canal spaces after obturation (α=0.05). Results: Average POVs of materials were 96.54% (ZOE), 97.87% (Metapex), and 74.74% (nano-chitosan; P=0.000). POV of chitosan differed from the other two (P=0.000) but the other two were similar (P=0.896). Average POVs were 91.46% (manual) and 88.51% (rotary); the difference was not significant (P=0.322). Pre-obturation spaces of canals for different methods were 3.89 mm3 (manual) and 3.26 mm3 (rotary); the difference was significant (P=0.013). Two-way ANCOVA showed a significant effect of materials (P=0.000) but not root length (P=0.585) or shaping methods (P=0.362) on POVs. Conclusions: Nano-chitosan showed a considerable success rate but it still needs reformulation as it was weaker than the extremely successful commercial competitors. Rotary instrumentation can provide results similar to hand-filing in terms of POV although it might yield smaller canals.

This study evaluated microscopically the effects of root surface treatment with three different solutions in delayed rat teeth replantation. Central incisors from 30 rats (Rattus norvegicus, albinus Wistar) were extracted and left on a bench for 6 h. The pulps were extirpated and root canals were irrigated with 1% sodium hypochlorite. After endodontic treatment, the root surfaces of all teeth were submitted to a 10-min treatment with 1% sodium hypochlorite, changed every 5 min. The teeth were then rinsed with saline for 10 min and assigned to 3 groups with ten specimens each. Groups I, II and III were treated, respectively, with 2% acidulated-phosphate sodium fluoride, vitamin C solution and effervescent vitamin C (2 g, Redoxon®). After root surface treatment, the teeth were filled with calcium hydroxide and replanted. The animals were sacrificed after 10 and 60 days. Group I (fluoride) presented the largest areas of replacement resorption and ankylosis. Comparing both vitamin C groups, Group III (effervescent vitamin C) yielded better results, showing more areas of ankylosis and replacement resorption than areas of inflammatory resorption.

ABSTRACT Aim The purpose of this study is to compare the efficacy of calcium hydroxide and zinc oxide eugenol as a root canal filling material in primary teeth using endodontic pressure syringe system. Materials and methods A total of 40 primary mandibular molars were selected and divided into two groups-group I (calcium hydroxide was used as root canal filling material) and group II (zinc oxide eugenol used as canal filling material). Clinical review was undertaken at 1, 3, 6 and 9 months interval for evaluating pain, tenderness, mobility and any other sign of pathology. Radiographic examination was done at interval of 3, 6 and 9 months for the evaluation of any change in the radiolucency around the apices and inter-radicular space. Resorption of root and filling material was also assessed. Results Reduction in the clinical symptoms were noted in both groups (p < 0.01). All the 20 cases in group I demonstrated a decrease in size of radiolucency. However in group II, an increase in radiolucency was observed in 2 teeth at 3 months radiographic examination. Further no decrease in the size of radiolucency was noticed at subsequent follow-up visits. Complete resorption of overfilled material was observed at the 3 months radiographic examination for group I whereas partial resorption of overfilled material was seen at 6 months radiographic examination for group II. Conclusion The findings of the present study indicate that calcium hydroxide could be used exclusively or as an alternative to zinc oxide eugenol as a root canal filling material for the primary teeth.

Abstract Introduction Nanoparticles and associations to calcium hydroxide can be used to increase antimicrobial activity. Objective To evaluate antibacterial activity against Enterococcus faecalis and pH of intracanal medications (IM) based on calcium hydroxide (CH) and zinc oxide (ZnO) micro- or nanoparticles, and their association with 0.4% chlorhexidine (CHX). Material and method Root canals from single-rooted human teeth were inoculated and incubated for 21 days. After sample (S1), the root canals were filled with the medications for 7 days and samples were collected immediately after medication (S2) and 7 days later (S3). Counting of CFU mL-1 was performed. Polyethylene tubes filled with the medications were used for the pH evaluation after 3, 7, 14 and 28 days. Data were submitted to ANOVA and Tukey tests (p<0.05). Result All IM promoted elimination of E. faecalis immediately after medication (S2). All the pastes promoted a similar pH increase. Conclusion CH/ZnO micro- or nanoparticles associated with CHX promoted greater bacterial reduction in the root canals and similar pH.

One of the primary objectives of endodontic treatment of teeth with pulp necrosis is the elimination of microorganisms from the root canal system, as effectively as possible, especially in cases with chronic periapical lesions. AIM: The purpose of this study was to analyze the response of the periapical tissue of dogs' teeth with chronic periapical lesions to endodontic treatment performed with utilization of metronidazole, calcium hydroxide, and an association of both as root canal dressings. METHODOLOGY: Forty root canals were submitted to pulpectomy and the root canals were kept exposed to the oral environment for 6 months. Then, they were submitted to biomechanical preparation and divided into 4 study groups with 10 specimens: group I - no root canal dressing; group II - calcium hydroxide; group III - metronidazole; group IV - calcium hydroxide associated to metronidazole. After 15 days, the root canals were filled with Fill Canal sealer. After 90 days, the animals were killed and the especimens processed for histological analysis. RESULTS: Calcium hydroxide dressing provided a significantly better outcome compared to other experimental groups (alpha = 0.01). Also, the results of the association of metronidazole and calcium hydroxide were similar to those observed for the metronidazole group. The worst results were obtained by the no root canal dressing group. CONCLUSION: The use of metronidazole alone or associated with Calcium hydroxide, did not improve periapical healing when compared to Calcium hydroxide dressing.

PURPOSE: To study morphologic and histochemical alterations arising at the ileocystoplasty site. METHODS: Sixteen Wistar female rats were subjected to ileocystoplasty and sacrificed after eight weeks. Material collected was divided into four groups for histological and histochemical studies: Group I (control) - isolated ileum segment removed during ileocystoplasty; Group II - ileoileal anastomosis; Group III - ileovesical anastomosis and Group IV - ileal segment from the neobladder. Histological and histochemical study assessed dysplasia, metaplasia, acute and chronic inflammation, fibrosis, atrophy, hypertrophy, total mucins, sialomucins and sulfomucins. The non-parametric Wilcoxon and Mann-Whitney tests were employed in statistical analysis. RESULTS: None of the groups presented dysplasia. Acute inflammation and atrophy occurred in Groups II, III and IV, not reaching statistical significance. Metaplasia was significant only in Group III (p=0.012). Chronic inflammation, fibrosis and hypertrophy were significant in Groups II, III and IV. There was a significant increase in total mucin content in Group IV (p=0.014) and a reduction in Group III (p=0.016). Increases in sialomucins were observed in samples for Groups III (p=0.003) and IV (p=0.002) along with reduced sulfomucins in samples from Groups III (p=0.013) and IV (p=0.008). CONCLUSION: Ileocystoplasty in female rats caused squamous metaplasia, chronic inflammatory infiltration, fibrosis, hypertrophy, increase in sialomucin content, reduction in sulfomucins, and alterations in total mucin content with statistical significance, as well acute inflammatory infiltration and muscular atrophy with less intensity.

Objective: The present study aimed to evaluate and compare six different materials commonly used for filling the root canals of primary teeth for antimicrobial efficacy against some of the microorganisms commonly found in infected root canals. Study design: In this experimental in vitro study six root canal filling materials were tested for antimicrobial efficacy against eight microbial strains using the agar diffusion method. Results: Zinc oxide eugenol paste exhibited the strongest antimicrobial potential followed by Endoflas™, zinc oxide-calcium hydroxide-sodium fluoride mixture, zinc oxide-calcium hydroxide mixture and calcium hydroxide paste (Apexcal™). The addition of sodium fluoride to the zinc oxide-calcium hydroxide mixture enhanced the antimicrobial efficacy. Metapex™ demonstrated minimal inhibition and Vaseline™ was non-inhibitory. Conclusions: All the test filling materials demonstrated varying antimicrobial activity against the microorganisms tested. Zinc oxide eugenol paste and materials containing zinc oxide were found to be more effective against the microorganisms compared to materials without zinc oxide.

All root canal sealers should have antimicrobial activity so that they could act against any remaining bacteria after root canal obturation. The aim of this study was to evaluate the antimicrobial activity of five root canal sealers: AH26, a resin based paste; Apexit, calcium hydroxide based paste; Endomethasone and Tubliseal, zinc oxide eugenol based materials and Ketac Endo Aplicap, glass ionomer based sealer. Matherials And Methods: Antimicrobial activity was tested against S. mutans 70C and L.casei ATCC 27773 using ADT (agar diffusion inhibitory test) on TYC SB, blood and MRS agars. Freshly prepared paste or cement was placed in grooves of 4 mm in diameter in agar plates. Prepared agar plates were incubated at 37?C in GAS PAC system for 24hours. The antimicrobial effect was determined according to the diameter of growth inhibition zone around tested materials. Three measurements were done for each material and values were recorded in millimeters. Statistical analysis was done using Mann Whitney test. The results confirmed that epoxy resin and zinc oxide eugenol based sealers had the greatest antimicrobial effect. Calcium hydroxide and glass ionomer based sealers showed significantly lower antimicrobial activity compared to AH26, Endomethasone and Tubliseal. Conclusion: The greatest antimicrobial activity was found for epoxy resin based sealer (AH26) for both tested microorganisms.

ABSTRACT Aim and objective To compare the fracture resistance of teeth prepared with rotary system and filled with single cone guttapercha followed by lateral condensation with different sealers like zinc oxide eugenol and resin based (AH26) to that of resilon. Materials and methods A total number of 70 extracted intact human permanent maxillary incisors were selected. All prepared samples were divided into one control group (n = 10) and three experimental groups (n = 20 per group). Group 1 control. This group received no obturation; the root canal opening was sealed with a temporary filling material (Cavit, Premier Dental Products, Plymouth Meeting, PA) Group 2: Gutta-Percha and zinc oxide Eugenol sealer. Group 3: Gutta-Percha and AH26 sealer. (DiaDent, Korea) dipped in AH26 sealer. Group 4: Resilon cones and RealSeal Resin Sealer. Obturation was accomplished using a 0.06 taper size 40 gutta-percha master point. All the root samples were stored in 100% humidity at 37°C for 2 weeks to allow the sealer to set completely. The root samples were then prepared for mechanical testing and the data was recorded and analyzed statistically. Results One-way ANOVA and Post hoc test (Duncan Multiple range test) were employed to determine possible statistical variation among the groups tested in this study. The force for group 2 was significantly greater than that for the control group 1 (no obturation). The force for group 3 was significantly greater than that for group 2. The force for group 4 was significantly greater than that for group 3. All other groupwise comparisons were not significant at 5% level. Group 4 seemed to have the greatest force among the three groups of interest in the study. Conclusion Root canals filled with Resilon increased the in vitro resistance of single canal extracted teeth compared to other experimental groups. The mean fracture resistance value for the experimental groups in ascending order was as follows: Root canals instrumented but not filled, filled with gutta-percha and zinc oxide eugenol sealer, filled with gutta-percha and AH26 sealer and filled with Resilon.
