A survey to estimate the prevalence of verotoxigenic E. coli (VTEC) or enterohaemorrhagic E. coli (EHEC) in rectal swabs from healthy dairy cattle aged three weeks, three months and one year was conducted in three herds from the Czech Republic. Screening for the presence of the stx1, stx2 and eaeA genes in faecal swab cultures was performed by PCR, and in positive samples, isolated colonies were examined. Immunomagnetic separation was used for the isolation of the VTEC serogroup O157 from samples. VTEC were detected in animals from all three herds under study. In the group of 3-week-old calves, VTEC were only detected in samples collected in the summer months. However, in the other age-groups, VTEC were detected in both the summer and winter months. EHEC shedding was observed in 30 to 100% of the total samples collected from cattle aged three months and one year in the summer months, and in 30 to 60% of samples taken in the winter months. EHEC strains of serogroup O157 were detected in two herds. The range of verotoxins shed by VTEC isolates of serogroup O157 differed between herds. Besides serogroup O157, additional EHEC belonging to the antigen groups O26, O103, O128 and O153 have been identified, and in some of them, no somatic antigen was detected.

ABSTRACTThe study was performed in the Czech Republic during 2007 to 2009. OfEscherichia coliisolates from 275 children aged 6 weeks, 36% (n= 177) were resistant to 1 to 7 antibiotics. Of isolates from 253 children aged 6 to 17 years, 24% (n= 205) were resistant to 1 to 5 antibiotics. There was no significant difference in the prevalences of antibiotic-resistantE. coliisolates between these groups of children, even though the consumptions of antibiotics were quite different.

Abstract Background: Avian pathogenic Escherichia coli (APEC) can cause various extraintestinal infections in poultry, resulting in massive economic losses in poultry industry. In addition, some avian E. coli strains may have zoonotic potential, making poultry a possible source of infection for humans. Due to its extreme genetic diversity, this pathotype remains poorly defined. This study aimed to investigate the diversity of colibacillosis-associated E. coli isolates from Central European countries with a focus on the Czech Republic. Results: Of 95 clinical isolates subjected to preliminary characterization, 32 were selected for whole-genome sequencing. A multi resistant phenotype was detected in a majority of the sequenced strains with the predominant resistance to β -lactams and quinolones being associated with TEM-type beta-lactamase genes and chromosomal gyrA mutations respectively. The phylogenetic analysis confirmed a great diversity of isolates, that were derived from nearly all phylogenetic groups, with predominace of B2, B1 and C phylogroups. Clusters of closely related isolates within ST23 (phylogroup C) and ST429 (phylogroup B2) indicated a possible local spread of these clones. Besides, the ST429 cluster carried blaCMY-2, -59 genes for AmpC beta-lactamase and isolates of both clusters were generally well-equipped with virulence-associated genes, with considerable differences in distribution of certain virulence-associated genes between phylogenetically distant lineages. Other important and potentially zoonotic APEC STs were detected, incl. ST117, ST354 and ST95, showing several molecular features typical for human ExPEC.Conclusions: The results support the concept of local spread of virulent APEC clones, as well as of zoonotic potential of specific poultry-associated lineages, and highlight the need to investigate the possible source of these pathogenic strains.

More than 4 000 E. coli strains isolated from diarrhoeic piglets in 111 pig herds in the Czech Republic during the period 1995–2000 were examined for serogroup and virulence factors. Gene typing of the K88 marker by polymerase chain reaction (PCR) was used for the examination of 283 enterotoxigenic strains (ETEC) which agglutinated with antisera against K88 or adhered to intestinal brush borders. The K88 gene was detected in 237 strains; among them 232 strains possesed the K88 variant. Genotype K88ab was found in two strains of the serogroup O8 from one herd and the gene K88ad was detected in three strains of the serogroup O8 originating from another herd. The results show that the type K88ac is predominant in ETEC strains with colonisation factors K88 in pig herds in the Czech Republic.

A major reason for resistance of <I>Enterobacteriaceae</I> to beta-lactam antibiotics is production of ESBLs and AmpC beta-lactamases. As their more detailed description in poultry is unavailable in the Czech Republic, the presented study aimed at assessing their occurrence and molecular characteristics. A total of 154 composite samples from broilers and 150 cloacal swabs from turkeys were examined. Production of ESBLs was detected in seven <I>Escherichia coli</I> isolates and AmpC enzymes in two <I>E. coli</I> isolates. The most frequent ESBL types were CTX-M-1 and SHV-12 and the most common AmpC enzymes were the CMY-2 types.

The aim of this work was to determine if Escherichia coli isolates carrying the virulence genes eae and eltB and exhibiting the Ehly phenotype are present in feces and milk samples from healthy dairy cattle on farms. Isolates from calves showed a statistically higher prevalence of eae and eltB compared with isolates from older animals. The other factors tested (stx1, stx2, and Ehly) were not statistically different between the two groups. Two isolates originating from calf feces were identified as serotype O157:H7; one of these isolates carried stx1 and eae, the other stx2 and eae. E. coli isolated from milk contained stx1, stx2, and eltB. The results show that feces or milk from healthy dairy cattle may contain E. coli pathotypes that express virulence genes, indicating that these materials have zoonotic potential. The results also reinforce the idea that host age can influence the dynamics of virulence genes in E. coli from cattle.

The persistence of Escherichia coli O157[ratio ]H7 in cattle and the farm environment was investigated on eight Ontario dairy farms positive for E. coli O157[ratio ]H7 in a longitudinal study commenced one year previously. Faecal samples from cows, calves, humans, cats, rodents, wild birds, a composite fly sample and numerous composite and individual environmental samples were cultured and tested for verotoxin-producing E. coli (VTEC). VTEC isolates were serotyped and E. coli O157[ratio ]H7 isolates were phage typed. E. coli O157[ratio ]H7 phage type 34 was isolated from one calf on each of two farms. The same phage type had been isolated on one of these farms 12 months earlier. Most E. coli O157[ratio ]H7-positive animals and farms became culture-negative within 2 and 3 months, respectively. E. coli O157[ratio ]H7 was not isolated from any environmental samples, although evidence of VTEC was found in composite samples from calf feeders (19·1%), calf barn surfaces (18%), cow feeders (14·9%), flies (12·5%), cow barn surfaces (11·3%), and individual milk filters (12·5%). VTEC belonging to 21 non-O157 serotypes were isolated from 24 cows (8·2%), 21 calves (18·3%), 2 cow feeder samples (3·0%), and 1 calf feeder sample (4·8%). Shedding of E. coli O157[ratio ]H7 by infected dairy cattle appears to be transient and persistence of E. coli O157[ratio ]H7 was not demonstrated from the farm environment sites tested.

Resistance of bacteria to antibiotics is a global medical problem requiring close cooperation between veterinary and human physicians. Raw materials and foods of animal origin may be not only a source of pathogenic bacteria causing alimentary tract infections but also a source of bacteria with a dangerous extent of resistance to antibiotics, potentially entering the human food chain. This article presents results of the first study in the Czech Republic detecting the presence of Enterobacteriaceae-producing extended-spectrum β-lactamases (ESBLs) in swabs collected in slaughterhouses from surfaces of healthy animal carcasses. In 2012, swabs taken from pig (n = 166) and cattle (n = 140) carcass surfaces were analyzed. In 17% of 53 studied slaughterhouses, ESBL-producing Escherichia coli strains were isolated. ESBLs were found in 11 and 4% of porcine and bovine samples, respectively. Swabs collected from pigs yielded 18 ESBL-producing E. coli strains. The bla genes were found to encode production of CTX-M-1 group enzymes in 16 strains, SHV in one case, and both CTX-M-1–like and TEM in another case. In swabs taken from cattle, five ESBL-producing E. coli strains were isolated. In three cases, the bla genes for CTX-M-1–like production were identified; in two cases, genes for both CTX-M-1–like and TEM production were found. The similarity/identity of ESBL-positive isolates was compared by pulsed-field gel electrophoresis. This is the first report and characterization of the presence and nature of ESBL-producing E. coli in swabs collected from surfaces of healthy pig and cattle carcasses in slaughterhouses in the Czech Republic.

Aquaculture systems are widely recognised as hotspots for horizontal gene transfer, and the need for screening for bacteria carrying antimicrobial resistance genes in aquaculture systems is becoming more important. In this study, we characterised seventeen bacterial strains (Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii, and A. nosocomialis) resistant to colistin originating from retailed aquaculture products imported from Vietnam to the Czech Republic. The mcr-1.1 gene was found located on plasmid types IncHI2, IncI2, and IncX4, as well as on the rarely described plasmid types IncFIB-FIC and IncFIB(K), phage-like plasmid p0111, and on the chromosome of E. coli. One E. coli strain carried the mcr-3.5 gene on IncFII(pCoo) plasmid in addition to the mcr-1.1 gene located on IncHI2 plasmid. K. pneumoniae was found to carry the mcr-1.1 and mcr-8.2 genes on IncFIA(HI1) plasmid. The mcr-4.3 gene was found on similar untypeable plasmids of A. baumannii and A. nosocomialis strains, pointing to the possible interspecies transfer of plasmids carrying the mcr-4 gene. Our results highlight that some aquaculture products of Asian origin can represent an important source of variable plasmids carrying mcr genes. The results showed an involvement of phages in the incorporation of the mcr-1 gene into plasmids or the chromosome in E. coli strains from aquaculture. The detection of E. coli with the mcr-1 gene in the chromosome points to the risks associated with the stabilisation of the mcr genes in the bacterial chromosome.

To investigate if there is a reservoir of Escherichia coli O157 in Norwegian cattle, faecal samples from 197 cattle herds were screened for E. coli O157 by the use of immunomagnetic separation (IMS) and PCR during the 1995 grazing season. Six E. coli O157[ratio ]H-isolates were detected in two herds, one isolate in one and five in the other. The isolates carried the stx1, stx2, and eae genes, and a 90 MDa virulence plasmid. They were toxinogenic in a Vero cell assay. From 57 other herds, 137 faecal samples were positive for stx1 and/or stx2 genes detected by PCR run directly on IMS-isolated material. Among these samples, stx2 were the most widely distributed toxin encoding genes. No difference was found among milking cows and heifers in the rate of stx1 and/or stx2 in positive samples.

Unemployment affects individuals socially, personally, and economically. The impact of being jobless can be long-lasting. Five different generations participate in the workplace today. As countries throughout the world went into lockdown to combat the spread of Covid-19, unemployment numbers rose rapidly. This study aimed to examine the effects of unemployment in three-generation groups in V4 and Austria in the pre-Covid-19 era and during Covid-19. Descriptive statistics were used to present the collected data. OECD data were used for the analysis. Based on the data collected, unemployment decreases with age. The group aged 15-24 shows significantly higher unemployment than the other two groups. A gender difference in unemployment was confirmed only in the Czech Republic and Slovakia. Unemployment has increased during the Covid-19 pandemic. The unemployment gap for females before and during Covid-19 was not confirmed. The T-Test confirmed the difference in unemployment before and during the crisis in the age categories 15-24 and 25-54. In Hungary, Poland, and Slovakia, the number of people with a duration of employment of up to one year differs in all age categories. In the Czech Republic, there is a significant difference only between the youngest group and the other two. In all countries, the largest number of people with the employment of up to one year is in the age group 25-54. In none of the examined countries was a gender unemployment gap proved before Covid-19.
