This paper discusses a method of micropropagation of Vaccinium corymbosum L. cv. Blue-Berry. The results showed that WPM supplemented with 1,0 mg/l of zeatin in combination with 0,1 mg/l of indolyl-3-butyric acid was more effective for the multiplication of blueberry axillary shoots. The maximum increase in the number of healthy axillary shoots was observed in the fourth subculture, whereas the phenomenon of hyperhydration (vitrification) began to appear in the fifth subculture. In addition, it was established that the presence of indolyl-3-butyric acid and 1,0 g/l of activated charcoal in the nutrient medium lead to the development of good root system of the Vaccinium corymbosum cv. Blue-Berry. The highest mean number of roots formed per explant was obtained on WPM medium, supplemented with 0,5 mg/l indolyl-3-butyric acid after 10 weeks. Acclimatization of in vitro regenerated plantlets of Vaccinium corymbosum with a developed root system in ex-vitro conditions (pH 3,5-4) showed a 100% survival rate.

For efficient clonal reproduction of Vaccinium corymbosum L. of Blue Berry variety, the surface explant sterilization approaches have been developed - the apical part of the stem and stem segments using “Belizna” commercial bleach solution and nutrient media for explant cultivation have been selected. The maximum survival of the explants (100%) was observed during sterilization with a 15% of “Belizna” bleach solution with an exposure time of 15 minutes. The nutrient media Murasige-Skuga, WPM and Anderson were compared for effective breeding. The highest breeding rate of blueberry was on WPM medium with the addition of zeatin and indolyl-3-butyric acid. The maximum number of axillary shoots per explant was 3.80, with an average length of 3.26 cm, obtained on WPM medium containing 1.0 mg/l of zeatin and 0.1 mg/l of indolyl-3-butyric acid. It has been shown that with an increase in the number of passages, growth rates improve. The maximum increase in the number of healthy axillary shoots is observed at the fourth passage, while the phenomenon of vitrification begins to appear from the fifth passage.

Microshoots of four hazelnut genotypes grown in vitro on Nas and Read medium (NRM) containing various combinations of CuSO4 • 5H2O and myo-inositol were successfully rooted and acclimatized ex vitro without any need of in vitro hardening treatments. Dipping of shoot bases in 1000 ppm indole-3-butyric acid (IBA) solution for 5 or 10 seconds followed by placement of shoots in plant growth regulator free NRM gave rise to formation of roots as early as 8 days. Shoots treated for 5 and 10 seconds rooted similarly, and depending on genotype, 88% to 98% rooting was observed within 15 days after treatment with IBA. Ex vitro survival of shoots three months after in vitro-root induction was 73% when shoots were treated with IBA for 5 seconds and 66% when shoots were treated for 10 seconds. The highest ex vitro survival rate (97%) 3 months after root induction was observed when shoots were treated with IBA solution for 10 seconds, and then cultured directly in peat pellets. Shoots developed good roots, and grew up to 70 cm in height 3 months after root induction. The potential use of rooting and acclimatization protocol for commercial micropropagation of hazelnut is presented.

An efficient protocol was developed for shoot proliferation and plant regeneration of Phlogacanthus thyrsiflorus Nees. (Acanthaceae) - a rare medicinal shrub of Bangladesh, through in vitro culture using shoot tip and nodal explants. Best shoot induction was observed on MS with 1.0 mg/l BAP + 0.5 mg/l NAA, in which 84.2% of nodal explants responded to produce maximum number (12.4 ± 0.66) of shoots per culture. In vitro raised shoots rooted on half-strength MS with 0.5 mg/l IBA + 0.5 mg/l NAA. For acclimation and transplantation, the plantlets in the rooting culture tubes were kept in normal room temperature for 7 days before transplanting in pots where plantlets were reared for three weeks. Auxins and their synthetic analogues are commonly used for rooting of cuttings, but their efficiency depends on experimental set-up and, even more importantly, on species or cultivar, and type of explants investigated. In attempt to improve rooting procedure for highbush blueberries (Vaccinium corymbosum L.), we investigated alanine conjugate of indole-3-butyric acid (IBA-Ala) as potential root-promoting compound and compared with commonly used auxins indole-3-butyric acid (IBA) and indole-3-acetic acid (IAA). The effect of different concentrations of auxins on the rooting of highbush blueberry stem cuttings (var. Bluecrop, Bluetta, Burlington and Jersey) and in vitro-derived microcuttings (var. Jersey) was investigated. Auxin treatments significantly promoted rooting of all four varieties in comparison to control. The most efficient rooting promoter in all varieties appeared IBA-Ala (provided up to 83%, and 93% of the rooted cuttings and microcuttings, respectively). Furthermore, IBA-Ala caused a vigorous, well-branched root system that resulted in better acclimatization and survival of plants. Results suggest the application of IBA-Ala may be beneficial for rooting of difficult-to-root varieties of blueberry and related species and their practical application in the nursery industry.

The procedure for Heuchera villosa ‘Caramel’ propagation was investigated, which involves shoot regeneration, rooting of regenerated shoots, and acclimation of regenerated plantlets. Petioles, as explants, were cultured on MS medium supplemented with 1-naphthylacetic acid (NAA), benzylaminopurine (BA), thidiazuron (TDZ) and callus formed on all media. Shoots were observed to proliferate from callus on media with BA and NAA, whereas no shoots regenerated on media with TDZ and NAA. On media containing 0.5 or 1.0 mg·L−1 BA in combination with NAA, the regenerated shoots showed severe hyperhydricity, whereas on media containing 0.1 mg·L−1 BA in combination with NAA, the regenerated shoots grew normally. The highest shoot induction rate, 90.6%, was obtained on media containing 0.1 mg·L−1 BA and 0.01 mg·L−1 NAA. The effects of indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), and NAA on rooting of H. villosa ‘Caramel’ was explored. The highest rooting rate (95%) was obtained on 1/2 MS medium containing 0.2 mg·L−1 NAA. In the subsequent acclimation experiments, about 85% of rooted plantlets survived and grew normally.

Aklimatisasi merupakan masa transisi sebelum kultur in vitro dapat ditanam di lingkungan ex vitro. Di daerah tropis, stevia seyogianya ditanam di dataran tinggi. Pengembangan klon stevia yang sesuai untuk dataran rendah di kawasan tropis sangat penting untuk memungkinkan penggunaan mekanisasi pada pertanaman stevia yang luas. Tujuan penelitian ini adalah menentukan pengaruh kondisi eksplan dan periode hardening terhadap daya hidup dan pertumbuhan pada tahap aklimatisasi dan pertumbuhan awal stevia klon BS 22 pada area terbuka di dataran rendah di wilayah tropis. Penelitian pertama dilangsungkan menggunakan umur tunas yang berbeda: 0, 1, 2, dan 3 minggu yang dikultur pada media padat sebagai sumber bahan eksplan. Penelitian kedua menggunakan satu buku stevia pada periode hardening dalam media cair selama 1, 4, 7 dan 10 hari. Aklimatisasi dilaksanakan dengan menanam eksplan dalam medium tumbuh campuran pada multi-tray dan diletakkan di dalam sungkup plastik tertutup selama 1 bulan. Tanaman yang berhasil hidup kemudian dipindah ke polibeg pada area terbuka dengan sinar matahari penuh. Pengamatan daya hidup dan pertumbuhan dilakukan pada akhir tahap aklimatisasi dan setelah 2 bulan di area terbuka. Hasil penelitian menunjukkan bahwa eksplan umur 1 minggu yang dikultur pada media padat mempunyai daya hidup tertinggi yakni 83%. Buku tunggal stevia yang dikultur pada medium cair pada tahap hardening selama 4 hari meningkatkan daya hidup menjadi 97% selama aklimatisasi 1 bulan. Setelah aklimatisasi, tinggi tanaman secara rata-rata adalah 2,6 cm dengan 10,6 helai daun. Tanaman yang dipindah ke area terbuka tumbuh pesat dengan tinggi tanaman mencapai 12 cm dengan 30 helai daun dan daya hidup 63% setelah 2 bulan. Hal ini menunjukkan bahwa stevia klon BS 22 mungkin sesuai untuk dataran rendah di daerah tropis.  [Kata kunci: tanaman pemanis, eksplan tunas, tahap hardening, daya hidup, tropika] AbstractAcclimatization is a transition period before in vitro culture can be planted in ex vitro environment. In the tropical region, stevia is should be planted at high altitude areas. The development of stevia clones suitable for low land area in the tropics is important to make it possible to apply mechanization in a large scale stevia plantation. The purpose of this research was to determine the effect of explant conditions and hardening period on survival rate and growth during acclimatization stage and early growth of stevia clone B 22 in an open area at low altitude area in the tropics. The first experiment was conducted using different shoot ages: 0, 1, 2 and 3 weeks cultured on solid media as an explant material source. The second experiment was using single node of stevia in different hardening periods in liquid media for 1, 4, 7 and 10 days.  Acclimatization was carried out by planting the explants on a mixture growing medium in multi-trays and placed inside a closed plastic tunnel for 1 month. The survival rate and growth parameters were observed at the end of acclimatization stage and after 2 months in the open area. The results show that 1-week explant age on solid media had the highest survival rate at 83%. Hardening single node of shoot in a liquid medium for 4 days increased the survival rate to 97% in 1 month acclimatization stage.  After acclimatization, the plant height on average was 2.6 cm with 10.6 leaves. The survived plants planted in an open area grew rapidly to 12 cm in height with 30 leaves and survival rate 63% within 2 months. It indicates that stevia clone BS 22 may suitable for a low altitude area in the tropics. [Key words: sweetener plant, shoot explant, hardening period, survival rate, tropics]

AbstractIn vitro culture through microcutting technology can be used for clonal propagation of rubber (Hevea brasiliensis Muell. Arg.) rootstocks. Acclimatization of in vitro plantlets to ex vitro conditions is a major bottleneck in the micropropagation of many plants.This research was conducted to study the effect of plastic cover closed period and media composition on the survival rate of rubber plantlets. Plantlets derived from microcutting were planted on plastic pots containing a mixture of soil, cocopeat, dung manure, and sand or zeolite. The plantlets were then placed inside a closed transparent plastic cover that opened after 2, 3, 4 and 6 weeks. The cover was placed under tree canopy. The second experiment used the same media composition with or without cocopeat and with sand or zeolite. At 1.5 month after culture, observation was done on the number of survived plantlets, plantlet height and the percentage of rooted plantlets. The results show that the best coverclosed period was six weeks and the best growing medium was a mixture of soil, cocopeat, dung manure, and zeolite (6:2:1:1v/v). On the two combined treatments, the survival rate was 73.3% after 1.5 month of acclimatization. The use of zeolite and a higher soil percentage gave positive influences on rubber plantlet survival rate. The second experiment results confirmed that the use of zeolite was better than sand and the use of cocopeat was definitely needed. It can be concluded that the best of acclimatization of rubber plantlets from microcutting was on a medium mixture of soil, cocopeat, dung manure, and zeolite (6:2:1:1) and placed inside a closed plastic cover for six weeks before the cover was opened gradually. AbstrakKultur in vitro melalui teknologi microcutting dapat digunakan untuk perbanyakan klonal batang bawah tanaman karet (Hevea brasiliensis Muell. Arg.). Aklimatisasi planlet in vitro ke kondisi ex vitro merupakan hambatan utama pada mikropropagasi berbagai jenis tanaman. Penelitian ini dilakukan untuk mempelajari pengaruh lama penutupan sungkup plastik dan komposisi media tumbuh terhadap daya hidup planlet karet. Planlet karet asal microcutting ditanam pada pot plastik berisi media dengan berbagai campuran tanah, cocopeat, pupuk kandang, dan pasir atau zeolit. Planlet selanjutnya diletakkan di dalam sungkup plastik transparan tertutup rapat yang dibuka setelah 2, 3, 4 dan 6 minggu. Sungkup plastik diletakkan di bawah tajuk pepohonan. Percobaan kedua menggunakan komposisi media serupa dengan atau tanpa cocopeat dan dengan pasir atau zeolit. Pada umur 1,5 bulan, pengamatan dilakukan terhadap jumlah planlet yang hidup, tinggi planlet, dan persentase planlet yang berakar. Hasil penelitian menunjukkan bahwa lama penyungkupan terbaik adalah enam minggu dan media tumbuh terbaik adalah campuran tanah, cocopeat, pupuk kandang, dan zeolit (6:2:1:1 v/v). Pada kombinasi kedua perlakuan tersebut, daya hidup planlet karet mencapai 73,3% setelah 1,5 bulan aklimatisasi. Penggunaan zeolit dan persentase tanah yang lebih tinggi berpengaruh positif terhadap daya hidup planlet karet. Hasil percobaan kedua menegaskan bahwa penggunaan zeolit lebih baik daripada pasir dan penggunaan cocopeat mutlak diperlukan. Dapat disimpulkan bahwa aklimatisasi planlet karet asal microcutting terbaik dilakukan pada media campuran tanah, cocopeat, pupuk kandang, zeolit (6:2:1:1) dan diletakkan di dalam sungkup plastik tertutup selama enam minggu sebelum sungkup dibuka secara bertahap.

Multiple adventitious shoot formed from internodal segments of Cephaelis ipecacuanha cultured 25 days on Gamborg basal medium (GAMBORG et al., 1968) supplemented with 6.66mM 6-benzylaminopurine there was a maximum of nine shoots per segment and an average of five shoots per segment formed. The presence of gibberellic acid in the subculture media promoted shoot elongation in all treatments. The shoots attained 3cm in height and rooting of 100% after 35 days of culturing upon Murashige and Skoog's basal medium (MS), added with 4.92mM indole-3-butyric acid, 0.87m gibberellic acid and 0.1% activated charcoal. Further growth was accelerated after the transfer to 1/2 MS without growth regulators. Rooted plantlets transferred to potting soil could be successfully established.

Rooting responses of rootstocks cvs. JTE-F1, M. 26 and MM. 106 were studied to different concentration of IBA in root induction media and to presence of activated charcoal in root elongation media. High rooting rate (>90%) could be achieved in cvs. JTE-H and M. 26, while cv. MM. 106 showed weak rooting ability at each IBA level tested. Increasing IBA content depressed the rooting only in cv. M. 26. Presence of activated charcoal decreased considerable the rooting rate in cv. M. 26 and decreased the number of roots in cvs. JTE-H and M. 26. These cultivars developed longer roots on media containing activated charcoal, while cv. MM. 106 did not showed any reaction for it.

Two methods of rhizogenesis – in vitro and ex vitro of Spiraea betulifolia subsp. aemiliana (C.K. Schneid.) H. Hara microshoots have been compared. Pulse treatment of microshoots with aqueous solutions of 4% “Heteroauxin” or 2% succinic acid (ex vitro rooting) did not effective -rooting frequency ranged from 3 to 19%. It was established that the in vitro rooting on nutrient media supplemented with auxins was a more effective technique, providing a high percentage of rooted microshoots. The use of half- strength MS medium supplemented with 0.1 μM indolyl-3-butyric acid (rooting frequency 8 8%, root number 3.5 ± 0.3 per plantlet) was found to be the most efective for in vitro rooting. The in vitro rooted regenerated plantlets were successfully acclimatized with 55% of survival rate.
