Background: Fusobacterium nucleatum is a common bacterial in root canal with pulp necrosis and periradicular lesion. A way to eliminate these bacteria from root canal is by root canal irrigation. Root canal irrigation materials that are widely used nowadays has many shortcomings. The pericarp of mangosteen (Garcinia mangostana L) has antibacterial potency. Therefore mangosteen pericarp can be an alternative material which could inhibit and bactericidal function to Fusobacterium nucleatum. Purpose: The aim of this study was to determine the antibacterial potency of mangosteen pericarp extract (Garcinia Mangostana L.) against Fusobacterium nucleatum. Methods: This study was laboratory experimental with pos-test only control group design. A microdilution method was used to determine minimum inhibitory concentration and minimum bactericidal concentration by colony counting bacteriae in Tryptone Soya Agar (TSA) media with drop plate technique. Growth of bacterial colonies in TSA is calculated manually in colony forming unit (CFU/ml). Results: Bacterial colonies growth at concentration 0.78% was 90% less than positive control group and there were no bacterial colonies growth at concentration 0.975%. Conclusion: The Minimum Inhibitory Concentration (MIC) of mangosteen pericarp against Fusobacterium nucleatum was at 0,78% concentration and the Minimum Bactericidal Concentration (MBC) was at 0.975% concentration.

Factor that should be considered in the use of irrigation is the loss of the smear layer on root canal walls. Therefore the prepared root canal can be irrigated using materials, such as EDTA 17% and 2,5% NaOCl and of extract Acanthus ilicifolius 7,5% and 10%. To determine the potential of extract Acanthus ilicifolius leaves as an alternative of root canal irrigation in removing smear layer at a concentratiom of 7,5% and 10%. The type of this study is a laboratory experimental with post-test only control group design. The total sample is 24 teeth samples that have been prepared and irrigated consisting of four groups. 6 samples for irrigation 7,5% extract Acanthus ilicifolius, 6 samples for irrigation 10% extract Acanthus ilicifolius, 6 positive control samples for combination 2,5% NaOCl and EDTA 17%, and 6 negative control samples. Shooting tool used to observe the cleanliness level in the walls of the root canal is a Scanning Electron Microscope (SEM). Data were analyzed using Kappa statistics, followed by Kruskal wallis analysis test and continued with Mann-Whitney analysis. Acanthus ilicifolius 7.5% and 10% extracts can remove the smear layer when compared with the negative control group. The combination of 2.5% NaOCl and 17% EDTA is better in smear layer removal compared to 7.5% and 10% Acanthus ilicifolius extracts. Acanthus ilicifolius was able to clean the smear layer on the root canal and can be used as an alternative root canal irrigation.

ABSTRACTBackground: Aggressive periodontitis is a disease that is mostly suffered by young patients with minimal local factors. The prevalence of this disease is approximately 8% in Indonesia, where Aggregatibacter actinomycetemcomitans bacteria are presented as dominant bacteria in this disease. Aggregatibacter actinomycetemcomitans is a gram-negative bacterium that is sensitive to flavonoids. Ramania leaves are natural substances which contain flavonoids. Flavonoids in ramania leaves can be applied as an antibacterial substance. Objectives: The objectives of this research are to determine and analyze the minimum inhibitory concentration and the minimum bactericidal concentration of flavonoids extract from ramania leaves at the concentration of 0.1%, 0.3%, and 0.5%, and analyze the antibacterial effectiveness on Aggregatibacter actinomycetemcomitans. Methods: True experimental design with pre and post-test with control group design was applied in this study. Antibacterial test using broth and agar dilution method was performed in a total of 5 treatment groups with 6 repetitions. Results:  All exract at 0.1%, 0.3% and 0.5% concentration demonstrated the ability to inhibit bacterial growth, and the minimum bactericidal concentration was found at 0.5% concentration. Result test for One way ANNOVA on minimum inhibitory concentration and Kruskal wallis test on minimum bactericidal concentration exhibited significant value of less than 0.05 presenting significant differences in some groups. Conclusion:  Minimum inhibitory concentration of ramania leaves flavonoid extract is effective on Aggregatibacter actinomycetemcomitans at the concentration of 0.1% and minimum bactericidal concentration of ramania leaves flavonoid extract is more effective at the concentration of 0.5%.Keywords: Aggregatibacter Actinomycetemcomitans, Aggressive Periodontitis, Dilution Method, Flavonoids, Ramania Leaves.

Background. The prevalence of Enterococcus faecalis bacterial infection caused the failure of root canal treatment between 24%-77%. This is due to various factors resistance and virulence of Enterococcus faecalis. This research to find alternative materials that have antibacterial properties and by utilizing natural ingredients that can later be used as a root canal irrigation. Antibacterial activity of the Averrhoa bilimbi linn leaf extract against Enterococcus faecalis bacteria can be determined by Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Purpose. The aim of this study was to determine the concentration of Averrhoa bilimbi linn leaf extract that has antibacterial activity against bacteria Enterococcus faecalis. Method. This research is a labolatory experimental with post test only control group design which use diluted Enterococcus faecalis ATCC 29212 according Mc. Farland standard 1,5 x 108 CFU/ml. With treatment Averrhoa bilimbi linn leaf extract on concentration 50%, 45%, 40%, 35%, 30%, and 25% given to each of 0,05 ml  Enterococcus faecalis and using Brain Heart Infusion Broth (BHIB) as planting media. Result. At the concentration 30% of Averrhoa bilimbi linn leaf extract, showed that colony’s growth less than 10%. At the concentration 35% was not revealed any bacterial growth. Conclusion. The Averrhoa bilimbi linn leaf extract has antibacterial effect on bacteria Enterococcus faecalis. The MIC was at 30% and MBC was at 35%.

ABSTRACTBackground: Mauli banana stem and basil leaves are natural ingredients that have the potential to be used as a treatment for Candida albicans infections. The methanol extract of mauli banana stem with 25% concentration is merely fungistatic which is still not capable to induce fungicidal effect, while the 100% concentration is known to be less effective than 0.2% chlorhexidine gluconate. Adding basil leaves extract to mauli banana stem extract is expected to increase antifungal activities against Candida albicans. Purpose: to analyze the antifungal activity of mauli banana stem and basil leaves extract mixture with concentrations of 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% and 100% against Candida albicans by dilution method. Method: This research used a true experimental method with a pretest–posttest control group design consisting of 11 treatments to obtain Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Result: The 10% concentration was identified as the Minimum Inhibitory Concentration (MIC), and no concentration could be classified as the Minimum Bactericidal Concentration (MBC) because all treatment group still exhibited Candida albicans growth on the SDA medium. Conclusion: Mauli banana stem and basil leaves extract mixture has an MIC value of 10% and does not demonstrate any value for the MBC. Keywords: Basil leaf, Candida albicans, Mauli banana stem, MBC, MIC

Cinnamon (Cinnamomum burmannii) is widely used by the public as a food ingredient and contains chemical compounds such as alkaloids, flavonoids, polyphenols, saponins, and terpenoids which function as an antibacterial against Staphylococcus aureus. This study aims to determine the Minimum Inhibitory Concentration and Minimum Bactericidal Concentration of cinnamon ethanol extract on Staphyloccocus aureus. This research is experimental with posttest only control group design through the tube dilution method. The results of the Minimum Inhibitory Concentration (MIC) study showed no clarity at concentrations of 30% and 40%. The results of the Minimum Bactericidal Concentration (MBC) obtained the number of colonies at a level of 10% by 51 CFU / plate, 20% by 27 CFU / plate, 30% by 6 CFU / plate and 40% by 0 CFU / plate. Based on the results of this study concluded that MIC cinnamon ethanol extract was 30%, and MBC cinnamon ethanol extract was 40%.

Background: Dental caries and periodontal disease can be caused by bacteria that attack hard and soft tissues in the oral cavity like Staphylococcus aureus and Streptococcus mutans bacteria One way to prevent infections caused by the bacteria are by using mouthwash. Currently, mouthwash that is widely used is mouthwash containing chlorhexidine, which has side effects in the form of tooth staining when used prolonged. The need for alternative medicines from nature that effectively cope with polymicrobial infections. One of the marine products that has pharmaceutical properties is squid, especially the ink it produces.Methods: This research is a pure experimental study (true experimental design) with a randomized pretest-posttest control group design. Squid ink Loligo sp. extract, obtained by extracting squid ink Loligo sp., with maceration method using 96% ethanol solvent. This research uses serial dilution method with spectrophotometric testing method.Results: After measuring the turbidity value in each treatment tube, it was found that at a concentration of 1.56% squid ink Loligo sp. extract began to inhibit the growth of Staphylococcus aureus and Streptococcus mutans bacteria.  This is proven because the absorbance value after and before incubation is fixed.Conclusions: The minimum inhibitory concentration of squid ink Loligo sp. extract on the growth of Staphylococcus aureus and Streptococcus mutants at a concentration of 1.56%.

Background. Failure of endodontic treatment is commonly caused by the persistent microorganisms remaining in the root canal such as Enterococcus faecalis. Enterococcus faecalis can form a biofilm in tough environmental conditions within the root canals and caused biofilm-mediated infections which needs more complicated treatment due to the increasing of antimicrobial resistance. The biofilm formation initial and most important step is bacteria adherence to the solid surface that is mediated by glucan. NaOCl 2.5% is a commonly used root canal medicaments but can cause injury of periapical tissue. Mangosteen pericarp extract contains flavonoid, tannin, and xanthone have mechanism for inhibiting adherence of bacterial biofilm. Difference of antibacterial activity between NaOCl 2.5% and mangosteen pericarp extract 0.09% can be determined by experimental laboratory to determine the adherence of bacteria in each treatment. Purpose. The aim of this study was to assess the difference of antiglucan activity between NaOCl 2.5% and mangosteen pericarp extract 0.09% on Enterococcus faecalis. Method. This study was designed as an experimental laboratory study with post  test only control group design using Enterococcus faecalis ATCC 29212. Mangosteen pericarp was extracted using maceration method. Adherence analysis was observed after 24 hours by examining the viable cells in suspension. These viable cells are measured by UV-Vis spectrophotometer to compare the suspensions’ turbidity. Using the Independent T-Test, significantly less bacteria were found adhering to the mangosteen pericarp extract. Results. Absorbancy difference level by mangosteen pericarp extract 0.09% is significantly greater than the NaOCl 2.5% (p<0.05). Conclusion. Antiglucan activity that generated by mangosteen pericarp extract 0.09% is greater than NaOCl 2.5%

Background: Root canal irrigation seeks the elimination of bacteria and its products from the root canal. If debris is left in the root canal, it can prohibit adaptation between obturationmaterial and root canal wall. Therefore, it can decrease the sealing ability of obturation material. Purpose: To assess the effectiveness of 2.5% NaOCl and 17% EDTA on the apical sealing ability of resin obturation paste. Method: The roots of 18 human mandibular premolar teeth were selected and the root canal was prepared using a conventional technique. Samples were irrigated with 2.5% NaOCl, and17% EDTA, and sterile aquadest (Aqua Deastilata) as a control group. Samples were filled with gutta-percha and top seal. Samples were soaked in 2% methylene blue for 48 hours to measure the depth of microleakage. Result: The mean apical leakage after irrigation with 2.5% NaOCl; 17% EDTA; and sterile aquadest in the sequence were 0.92 mm; 3.6 mm; and 8.08 mm. Conclusion: 2.5% NaOClhas less depth of microleakage than 17% EDTA. However, the depth of microleakage cannot demonstrate the exact sealing ability of the resin paste due to some factors.

Abstract: Noni (Morinda cittrifolia L.) leaves contain antraquinon, atsiri oil, saponin, tannin, alkaloid, flavonoid, polifenol, and sterol that have been proved can inhibit the growth of Candida albicans. This study was aimed to establish the minimum inhibitory concentration (MIC) of noni leaf extract against Candida abicans. This was a true experimental study with a randomized pretest-posttest control group design. We used serial dilution method with turbidimetry and spectrophotometry tests. Noni leaves were extracted by using maceration with 96% ethanol. Candida albicans fungi were obtained from Microbiology Laboratory of Faculty of Mathematics and Natural Sciences, Sam Ratulangi University. The turbidimetry test using three repetitions showed that the MIC of noni leaf extract against Candida albicans was 6.25% meanwhile the spectrophotometry test established 12.5% as the MIC of noni leaf extract. Conclusion: Minimum inhibitory concentration of noni (Morinda cittrifolia L.) leaf extract against the growth of Candida albicans was 12.5%.Keywords: noni (Morinda citrifolia L.), Candida albicans, minimum inhibitory concentration (MIC) Abstrak: Daun mengkudu (Morinda citrifolia L.) mengandung antraquinon, minyak atsiri, saponin, tannin, alkaloid, flavonoid, polifenol dan sterol yang terbukti dapat menghambat pertumbuhan Candida albicans. Penelitian ini bertujuan untuk mendapatkan konsentrasi hambat minimum (KHM) dari ekstrak daun mengkudu terhadap Candida albicans. Jenis penelitian ialah eksperimental murni dengan randomized pretest-posttest control group design. Metode yang digunakan dalam penelitian yaitu serial dilusi dengan pengujian turbidimetri dan spektrofotometri. Daun mengkudu diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Jamur Candida albicans diambil dari stok jamur Laboratorium Mikrobiologi Program Studi Farmasi Fakultas MIPA Universitas Sam Ratulangi. Hasil penelitian menunjukkan bahwa pengujian turbidimetri dengan tiga kali perlakuan mendapatkan KHM pada konsentrasi 6,25% sedangkan pengujian spektrofotometri mendapatkan KHM pada konsentrasi 12,5%. Simpulan: Konsentrasi hambat minimum (KHM) ekstrak daun mengkudu (Morinda citrifolia L.) terhadap pertumbuhan Candida albicans terdapat pada konsentrasi 12,5%.Kata kunci: mengkudu (Morinda citrifolia L.), Candia albicans, konsentrasi hambat minimum (KHM)

Tifoid fever caused by Salmonella typhi (S. typhi) is endemic disease in Indonesia. S. typhi begin to resist to chloramphenicol, so it is necessary to study about natural material as traditional medicine such as tempuyung. The aim of this research was to study antibacterial activity and Minimum Inhibitory Concentration (MIC) of ethanolic extract from tempuyung leaf (Sonchus arvensis L.) againts growth of S. typhi. This research used quasy experimental methods with post test only control group design. Treatment consisted of 8 concentrations with 5 replications was 2,5 µg/disc, 5 µg/disc, 10 µg/disc, 20 µg/disc, 30 µg/disc, 40 µg/disc, 60 µg/disc, and 80 µg/disc. As much as 10 µl from each concentration was dropped into disc and placed in Mueller Hinton Agar. Then incubated for 18 hours to 37oC. Observation of bacterial inhibition zone by using a caliper. Result of this research that the leaf extract at a concentration 10 µg/disc, 20 µg/disc, 30 µg/disc, 40 µg/disc, 60 µg/disc, and 80 µg/disc can inhibit the growth of Salmonella typhi. Minimum Inhibitory Concentration of ethanolic extract from tempuyung leaf was 10 µg/disc qualitatively and 4,43 µg/disc quantitatively.
